以实验室分离的产壳聚糖酶的LS菌株出发,制备壳聚糖酶粗酶液,经(NH_4)_2SO_4盐析、透析、DEAE-Sephadex A25阴离子交换、Sephadex G-100凝胶过滤分离纯化后,SDS-PAGE显示为一条带,壳聚糖酶被纯化了22.4倍,回收率为34.2%。同时对其酶学性质进行了初步研究,发现该酶在低于35℃和pH 6.O~7.5范围内较稳定,最适反应温度为55℃,反应pH 5.0~5.5;Zn~(2 )、Ag~ 、Ca~(2 )、Co~(2 )、Mn~(2 )对该酶有明显的促进作用,而Fe~(3 )、Hg~(2 )对该酶有强烈的抑制作用;该酶的米氏常数(K_m)为2.50 mg/mL,最大反应速度(V_(max))为4.19μmol/(mL·min);SDS-PAGE测定的相对分子质量为30.9×10~3。  相似文献

【目的】利用筛选培养基,从福建沿海潮间带泥样中分离筛选产壳聚糖酶的菌株,并研究菌株的产酶特性。【方法】通过形态学观察,结合26S rDNA序列进行分类鉴定,采用DNS法测定酶活力。【结果】筛选得到产壳聚糖酶的菌株KQ-1002与草酸青霉(Penicillium oxalicum)的同源性为99%,并初步鉴定为青霉属的一种。发酵培养的最适温度为30°C,最适碳源为1.0%水溶性壳聚糖,最适氮源为1.87%(NH4)2SO4,最适pH为6.0。该菌株液体发酵培养72 h产壳聚糖酶活性最高,经优化后最高产酶量为18 U/mL。纯化后的壳聚糖酶经SDS-PAGE分析其分子量约40 kD。酶促反应最适pH为5.0,最适反应温度为55°C,Km值为1.293 g/L。在离子浓度为1.0×10 3mol/L时,金属离子Cu2+、Hg2+、Ag+对酶的活性均有强烈的抑制作用。壳聚糖酶对不同底物及脱乙酰度的壳聚糖具有不同的降解作用。【结论】筛选获得产壳聚糖酶的真菌菌株KQ-1002的壳聚糖酶活力经优化后提高了约7倍,是一株具有研究和应用潜力的产壳聚糖酶菌株。  相似文献

目的:克隆壳聚糖酶基因于大肠杆菌中实现高表达,制备壳寡糖。方法:以枯草芽孢杆菌总DNA为模板扩增壳聚糖酶基因(CSN),克隆至载体pET23a(+)上,转化菌株BL21(DE3)。重组子经0.5 mmol/L IPTG诱导后,SDS-PAGE和质谱检测与鉴定重组酶。酶纯化后水解壳聚糖,薄层色谱分析其水解产物。结果:质谱证明壳聚糖酶(31.5kDa)成功表达,表达量占菌体总蛋白的45%左右。纯化后重组酶浓度为900 mg/L,纯度95%、回收率85%,酶活力为10 000 U/mg。壳聚糖降解产物为壳二糖至壳四糖。结论:原核表达载体pET23a(+)-CSN构建正确,壳聚糖酶表达量与活性高,适用于水解壳聚糖制备壳寡糖。  相似文献

分离纯化从烟台近海土壤筛选的链霉菌来源壳聚糖酶,并对其酶学性质进行研究。通过(NH4)2SO4分级沉淀分离得粗酶,透析后经Sephadex G-100柱纯化,得到2种壳聚糖酶(ChA和ChB)。SDS-聚丙烯酰胺凝胶电泳及Sephadex G-75凝胶过滤确定ChA的相对分子质量,研究ChA的最适底物水解条件、热稳定性、水解动力学及金属离子对酶活性影响。结果表明:ChA为单亚基蛋白,相对分子质量为4.16×104,在220和280 nm处呈现两个紫外吸收峰,催化水解壳聚糖的最适pH为5.0~5.5,最适温度为55℃。热稳定性实验表明:30℃温育1 h后酶活为初始酶活的33.3%,40℃温育1 h后酶活为初始酶活的22.2%。ChA的酶促反应初速率为6.2×10-3μmol/(mL.min),Vmax为0.318μmol/(mL.min),Km为1×10-2mg/mL,且对底物表现相对专一性。K+、Na+、Li+、Mg2+、Ca2+、Ba2+Zn2+、Cu2+和Co2+对ChA活力均表现为抑制作用,过渡金属离子Mn2+对酶有激活作用,重金属离子Hg2+、Ag+、Cd2+和Pb2+对酶均有较强的抑制作用。Mn2+和Zn2+的动力学研究表明,Mn2+对酶为混合型激活作用,Zn2+对酶为竞争性抑制作用。  相似文献

从PBCV\|1感染小球藻NC64A的细胞裂解液中提取了Lysin的粗制剂,酶活底物范围分析表明,几丁质酶、壳聚糖酶和β\|1,3\|葡萄糖苷酶是Lysin活性的主要组成部分,并与小球藻细胞壁的组成成分相吻合。其中几丁酯酶和壳聚糖酶,特别是几丁酯酶在裂解小球藻细胞壁的过程中发挥了重要的作用。Lysin粗制剂经FPLC分离纯化得到分子量分别为52kD、56kD的两个几丁质酶(Chil和Chi2)和一个分子量为36kD的壳聚糖酶。  相似文献

枯草芽孢杆菌SA-22β-甘露聚糖酶的纯化及其特性   总被引:10,自引:0,他引:10  
利用硫酸铵沉淀、羟基磷灰石柱层析、SephadexG 75凝胶过滤和DEAE 5 2离子交换柱层析的方法 ,将枯草芽孢杆菌SA 2 2 β 甘露聚糖酶纯化了 30 75倍 ,同时 ,该酶比活达到 34780 5 6u mg ,收率达到 2 3 4 3%。利用SDS PAGE凝胶电泳和SephadexG 75凝胶过滤的方法测得枯草芽孢杆菌SA 2 2 β 甘露聚糖酶的分子量分别为 38kD和34kD。实验发现该酶的最适pH为 6 5 ,在pH 5~ 10的范围内稳定 ;该酶最适温度为 70℃ ,在 5 0℃保温 4h后其活力不变 ,在 6 0℃保温 4h后剩余酶活为 74 2 % ,70℃的酶活半衰期为 3h。实验还发现Hg2 对酶活力有明显抑制作用。该酶对槐豆胶和魔芋胶的Km 和Vmax值分别为 11 30mg mL ,4 76mg mL和 188 6 8(μmol·mL- 1 ·min- 1 ) ,114 94(μmol·mL- 1 ·min- 1 )  相似文献

从来自拟青霉属真菌Paecilomyces sp.CS-Z的发酵液中获得一种壳聚糖酶,该酶被纯化了9.4倍,产率为48.2%。经SDS-PAGE分析确定为单一条带,分子量为29kDa,其最适pH为6.0–6.5,最适温度为55℃,在80℃处理60min后,能保持较好的热稳定性,Hg2+完全抑制了酶活,对脱乙酰度85%–95%的壳聚糖具有较高的水解活性,而对几丁质和羧甲基纤维素无活性。薄层层析和质谱分析表明该酶是一种内切酶,其水解产物为聚合度大于6的壳寡糖,其理化性质与至今报道的壳聚糖酶有所不同,为壳聚糖酶的开发提供了重要的实验依据。  相似文献

以魔芋精粉为唯一碳源, 对假蜜环菌(Armillariella tabescens) EJLY2098进行培养, 诱导其产生β-甘露聚糖酶。经DEAE-阴离子交换层析后, 分离纯化出β-甘露聚糖酶atMAN47。酶学性质分析: 该酶分子量约为47 kD, 酶的最适反应温度为50°C, 在pH 5.0~6.5之间该酶的稳定性较好; Na+ 和Ba2+ 对该酶有激活作用, 用TLC对酶产物分析, 表明该酶为内切β-甘露聚糖酶。该酶为偏酸性的内切甘露聚糖酶, 适合发展为饲料的酶制剂, 具有良好的应用前景。  相似文献

利用以壳聚糖为唯一碳源的选择性培养基,从自然界中筛选得到一株壳聚糖酶活较高的菌株 ,其壳聚糖酶活为0.59U/mL.经初步鉴定,该菌株为芽孢杆菌属,以A表示.以该芽孢杆菌为出发菌株,经硫酸二乙酯(DES)诱变处理50 min后,筛选得到壳聚糖酶活明显提高的突变株DES-4,其壳聚糖酶活为1.60U/mL,是出发菌株的2.7倍.该突变株经连续传代5次后仍稳定产酶.研究表明,突变株DES-4的壳聚糖酶产生与芽孢形成之间关系密切,当芽孢充分形成后发酵液的壳聚糖酶活力不再增大.  相似文献

黄杆菌(Flavobacteium sp.)在几丁质的诱导下产生几丁质酶.通过(NH_4)_2SO_4沉淀、DEAE纤维素柱层析、Sephacryl 300柱层析及Sephadex G-75柱层析,从Flavobacterium sp.培养上清液中分离纯化了几丁质酶.SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)纯度分析表明,纯化后的几丁质酶达到了均一的程度.用SDS-PAGE测得该酶的分子量约45D00道尔顿.该酶水解几丁质的最适pH为 7.0,最适温度为50℃,-20C贮存两年以上仍有活性.水解几丁质的Km值为5.0mg/ml.金属离子对几丁质酶活性影响较大,Ca~(2+) 、Co~(2+)’和Cu~(2+)对酶有激活作用.而NH_4~-、Ba~(2+)、Mg~(2+)、Mn~(2+)对酶有抑制作用.几丁质酶水解几丁质的产物是几丁质二糖.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献
