The spermatozoa and the sperm aggregates of 13 species belonging to four genera ( Smithsonidrilus, Limnodriloides, Thalassodrilides, Doliodrilus ) in the tubificid subfamily Limnodriloidinae (Annelida, Oligochaeta) were studied and compared with the spermatozoal patterns already described in the subfamily Tubificinae. Two characters considered exclusive for the Tubificinae were found in the more spermatologically variable Limnodriloidinae: the production of two kinds of spermatozoa, eusperm and parasperm, and the presence, in the spermathecae, of sperm aggregates formed by a combination of the two sperm types. A parsimony analysis was performed on the spermatozoal data of the species examined and compared with that based on the somatic characters of the same species: a critical revision of the already codified eusperm characters was carried out and the ultrastructure of parasperm was used as a new subset of spermatozoal characters. In a total evidence approach, a further parsimony analysis was run using a matrix combining both sets of characters. This analysis suggested that the double sperm line and the sperm aggregates composed of both eusperm and parasperm may well be homologous in tubificines and limnodriloidines. It thus supported the previous notion that Tubificinae and Limnodriloidinae are closely related and indicated that these subfamilies may be sister taxa.  相似文献

In many species, males have evolved to produce a sterile sperm (parasperm) in conjunction with fertilizing sperm (eusperm). Here, we document evidence of males depositing two morphologically distinct types of parasperm (1 and 2) into the female reproductive tract in Drosophila pseudoobscura. These parasperm differ in length, shape, amount produced, amount in long‐term storage and may have separate roles in ensuring male fertilization success. Although both parasperm types protect eusperm from female spermicides, only parasperm 2, which has a corkscrew shape, is associated with sperm competition. Increased production of parasperm 2 is also negatively correlated with the eusperm and parasperm 1 production. Thus, selection may be acting on parasperm production in the presence of sperm competition. Our findings show how both sperm competition and cryptic female choice may be acting in conjunction to influence the evolution of ejaculate composition. Our identification and characterization of two distinct parasperm morphs will enhance the ability for further evaluation of parasperm's role in fertilization.  相似文献

Summary

Littorinid parasperm develop from a distinctive lineage of germ cells which exhibit a process of nuclear destruction that has apototic characteristics. Fragments of DNA and other nuclear breakdown products are incorporated into secretion granules in parasperm that ultimately find their way to the female bursa copulatrix. Spermatozeugmata are stored in the seminal vesicles and, if unused during the breeding season, they are recycled by phagocytosis. Attachment between eusperm and parasperm is facilitated by an electrostatic interaction of proteins. Detachment, caused by alkaline prostate fluid, occurs by the time the ejaculate reaches the tip of the penis. Thus transport of eusperm by parasperm to the female is unlikely. parasperm are sterile cells that may function in defense against rival eusperm as suggested by the presence of lysosomes, or they may act as nuptial gifts as they are packed with glycoprotein nutrients. Differences in the reactivity of different parasperm to specific lectins may enable separation of dimorphic sperm by lectin affinity chromatography, thereby facilitating future studies on individual parasperm. In female Littorinidae, sperm are stored incapacitated in storage organs, or rarely in the ovary itself. In L. littorea serotonin caused spawning of unencapsulated eggs, which, in the presence of activated sperm, became polyspermic.  相似文献

Sperm size and number are important determinants of male reproductive success. The genus Drosophila exhibits a remarkable diversity of sperm production strategies, including the production of multiple sperm morphs by individual males, a phenomenon called sperm heteromorphism. Sperm-heteromorphic Drosophila species in the obscura group produce large numbers of infertile "parasperm" in addition to fertile eusperm. Parasperm have been hypothesized to perform a number of roles in place of fertilization, predominantly focused on their potential function in postcopulatory sexual selection. However, the evolutionary significance of parasperm remains unknown. Here we measured several male and female morphological, behavioral, and life-history traits in 13 obscura group species to test competing hypotheses of parasperm function using comparative methods. We found that parasperm size was unrelated to female reproductive tract morphology but was negatively related to our two indices of sperm competition, suggesting that postcopulatory sexual selection may indeed have shaped the evolution of parasperm. We also found abundant coevolution between male and female reproductive traits. Some of these relationships have been found in both sperm-monomorphic and sperm-heteromorphic taxa, but others are dissimilar. We discuss the significance of our results to the evolution of reproductive traits and the elusive function of Drosophila parasperm.  相似文献

Sperm dimorphism is a rare phenomenon in Chelicerata. Until now, it was known only from three species of the opilionid genus Siro (Sironidae, Cyphophthalmi). Fertilizing (eusperm) and nonfertilizing spermatozoa (parasperm) develop in the same cyst and are thus sister cells. The fine structure of the spermatozoa of two species has been examined and is compared here. In contrast to Siro rubens, S. duricorius spermatozoa lack an acrosomal complex. Both sperm types produce a transitional process, a more or less modified flagellum, which is later retracted. Hence, the spermatozoa are aflagellate. Eusperm and parasperm of all three species form highly ordered sperm balls that are stored in the deferent duct. Reviewing and adding new results about the sperm dimorphism in this arachnid taxon provides the basis for some considerations of another enigmatic morphological character found in Uropygi and Amblypygi, i.e., the tubular accessory genital glands that show holocrine extrusion. These glands are suggested to represent modified, infertile derivatives of the testis anlage. Their secretion is produced in a way reminiscent of a strongly degenerated spermatogenesis. Consequently, these products may be regarded as strongly degenerated germ cells representing a line of germ cell development, which has been separated very early in spermatogenesis from the usual line leading to fertilizing sperm cells. This further, although less evident, case of probable dichotomous germ cell development is discussed with respect to the controversial phylogenetic-systematic relationships between Uropygi (Thelyphonida and Schizomida), Amblypygi, and Araneae.  相似文献

Annual changes in testicular development and occurrence of parasperm were investigated using 2-year-old male fourspine sculpins Cottus kazika, based on the histological observation of testes. The male reproductive organ of fourspine sculpins comprised a pair of testes and a sperm duct that functioned as a sperm-storage organ. Male maturity was divided into the following periods: spermatogonial proliferation period (September), early spermatogenic period (October), mid-spermatogenic period (November), late spermatogenic period (December and January), functional maturation period (February and March), and recovery period (April to August). Spermatogenesis rapidly progressed from October to January and continued until the functional maturation period. Parasperm formation, which is known in some cottidae species, was observed in fourspine sculpins. Testicular regression of cultured fourspine sculpins progressed slowly during the recovery period when residual parasperm and empty spaces occupied the testis. The parasperm were immotile and oval and slightly concave on one side; additionally, they stained strongly with hematoxylin and PAS. Seminal lobules of the testis were filled with parasperm during the spawning period; in contrast, the sperm duct was filled with eusperm. These findings were observed in both cultured and wild fish. In this study, the functions of parasperm with regard to reproduction in fourspine sculpins are discussed.  相似文献

In many species of animals, males normally produce parasperm (dimorphic sperm) along with eusperm (normal sperm) during spermatogenesis. In the present study, to clarify the role of parasperm of the non-copulatory sculpin Hemilepidotus gilberti, whose reproduction is characterized by polyandrous oviposition involving sneaking by neighboring territorial males, we observed the movements of parasperm. Parasperm could not move by themselves, but they were transported in solutions by passive movement due to collisions with actively swimming eusperm. In the viscous ovarian fluid (OF), which isolates eggs from seawater by covering them during spawning, parasperm did not exhibit any movement. However, they could be transported by eusperm movement in solutions with dissolved OF, partly because the viscosity of the fluid become lower. And then, in some solutions parasperm formed lumps. Lump formation of parasperm was also observed at the boundary surface of an egg mass where OF contacted seawater. Eusperm added experimentally to a solution in which parasperm were forming lumps were engulfed in the lumps and never escaped. Thus, lump formation of parasperm would be obstacles for the later arriving eusperm. Although lumps formed against both kin and non-kin eusperm, parasperm are thought to be available to overcome sperm competition which would occur during spawning that involves sneaking being almost concurrent with lump formation. The territorial male eusperm reach the eggs while his parasperm hinder other males' eusperm from reaching the eggs. Thus, we concluded that parasperm of H. gilberti play a role on protection of paternity by blocking rival eusperm physically from approaching eggs.  相似文献

Sperm polymorphism, a phenomenon in which more than one type of sperm is produced within a species, occurs widely in animals from invertebrates to vertebrates. Sperm in this phenomenon can be categorized into fertile sperm and nonfertile sperm on the basis of fertilization ability. Nonfertile sperm can be further classified into parasperm and aberrant deformed sperm. Parasperm are sperm produced through a constant developmental process along with normal fertile eusperm, and they are readily distinguished from deformed sperm, which are irregularly crippled by unpredictable errors at certain stages during spermatogenesis. Sperm identified as parasperm occur widely in invertebrates but are presently quite limited in vertebrates. This may be the result of the deficiency both of studies on parasperm and of clear criteria to identify parasperm in vertebrates. Some vertebrates show unique spermatogenesis, such as symplastic spermatid and semicystic spermatogenesis. Thus, parasperm must be identified by comparing cells in each cyst and in semen, because irregularly shaped cells in the seminal duct could be either parasperm or normal spermatids. Although parasperm are identified by clear criteria in vertebrates, only in cottoid fishes to date, it is possible for parasperm to be discovered in other vertebrates. Recently, roles related to sperm competition have been reported in several species (e.g., the marine cottoid fish Hemilepidotus gilberti), and, with some of them, parasperm production is influenced by an intraspecific factor such as a sex ratio or the density of a population. Sperm competition is one of the important candidates for influencing evolution of parasperm functions, but not all parasperm seem to have a relation to sperm competition. Parasperm function may relate to the ecological conditions of each species that produces parasperm. Studies on parasperm function will be advanced by an ecological approach concerning male fertilization success as well as cytological investigation for parasperm. Sperm competition and cryptic female choice profoundly affect sperm morphology, producing diversity within both species and individuals. One type of within-individual sperm variation is sperm heteromorphism, in which each male produces two or more distinct types of sperm simultaneously, only one of which is typically fertile (the "eusperm"). The adaptive significance of nonfertile "parasperm" types is poorly understood, although numerous sperm-heteromorphic species are known from many disparate taxa. This paper examines in detail two female-centred hypotheses for the evolution and maintenance of this unconventional sperm production strategy. First, we use game theoretical models to establish that parasperm may function to protect eusperm from female-generated spermicide, and to elucidate the predictions of this idea. Second, we expand on the relatively undeveloped idea that parasperm are used by females as a criterion for cryptic female choice, and discuss the predictions generated by this idea compared to other hypotheses proposed to explain sperm heteromorphism. We critically evaluate both hypotheses, suggest ways in which they could be tested, and propose taxa in which they could be important.  相似文献

Sperm morphology and spermatogenesis were examined in the oligochaete annelid Isochaetides arenarius, a species belonging to the subfamily Tubificinae inhabiting the sediments of Lake Baikal. As all tubificines, Isochaetides produces two types of spermatozoa, named eusperm and parasperm. The eusperm are the fertilizing male gametes and consist, in sequence, of an acrosome, a nucleus, a mitochondrial mid-piece, and a tail. The parasperm have the same general architecture, but differ in cytological details: the acrosome is shorter, devoid of a perforatorium, and the acrosome vesicle has a different, simpler, shape. The nucleus is much shorter and rectilinear (the eusperm nucleus is twisted). The mid-piece mitochondria are less numerous but their overall volume is larger. The flagellum has a plasma membrane largely separated from the axoneme, and is devoid of glycogen granules. After mating, the two sperm types gather in the spermathecae to form spermatozeugmata; in these structures the parasperm form an external sheath involving the centrally located eusperm and their tails are connected by conspicuous septate junctions. Parasperm nuclei are produced through a process of fragmentation of the 'spermatocytes', whereas the flagellar basal bodies are produced by a process similar to that giving rise to basal bodies in ciliated epithelia.  相似文献

The testis produces male gametes in the germinal epithelium through the development of spermatogonia and spermatocytes into spermatids and immature spermatozoa with the support of Sertoli cells. The flow of spermatozoa into the epididymis is aided by testicular secretions. In the epididymal lumen, spermatozoa and testicular secretions combine with epididymal secretions that promote sperm maturation and storage. We refer to the combined secretions in the epididymis as the sperm-milieu. With two-dimensional-PAGE matrix-assisted laser desorption ionization time-of-flight MS analysis of healthy testes from fertile accident victims, 725 unique proteins were identified from 1920 two-dimensional-gel spots, and a corresponding antibody library was established. This revealed the presence of 240 proteins in the sperm-milieu by Western blotting and the localization of 167 proteins in mature spermatozoa by ICC. These proteins, and those from the epididymal proteome (Li et al. 2010), form the proteomes of the sperm-milieu and the spermatozoa, comprising 525 and 319 proteins, respectively. Individual mapping of the 319 sperm-located proteins to various testicular cell types by immunohistochemistry suggested that 47% were intrinsic sperm proteins (from their presence in spermatids) and 23% were extrinsic sperm proteins, originating from the epididymis and acquired during maturation (from their absence from the germinal epithelium and presence in the epididymal tissue and sperm-milieu). Whereas 408 of 525 proteins in the sperm-milieu proteome were previously identified as abundant epididymal proteins, the remaining 22%, detected by the use of new testicular antibodies, were more likely to be minor proteins common to the testicular proteome, rather than proteins of testicular origin added to spermatozoa during maturation in the epididymis. The characterization of the sperm-milieu proteome and testicular mapping of the sperm-located proteins presented here provide the molecular basis for further studies on the production and maturation of spermatozoa. This could be the basis of development of diagnostic markers and therapeutic targets for infertility or targets for male contraception.  相似文献

Sperm form and size is tremendously variable within and across species. However, a general explanation for this variation is lacking. It has been suggested that sperm size may influence sperm competition, and there is evidence for this in some taxa but not others. In addition to normal fertilizing sperm, a number of molluscs and insects produce nonfertile sperm that are also extremely morphologically variable, and distinct from fertilizing forms. There is evidence that nonfertile sperm play an indirect role in sperm competition by decreasing female remating propensity in Lepidopterans, but in most taxa the function of parasperm is unknown. We investigated the role of nonfertile (oligopyrene) sperm during sperm competition in the fresh water snail Viviparus ater. Previous studies found that the proportion of oligopyrene sperm increased with the risk of sperm competition, and hence it seems likely that these sperm influence fertilization success during competitive matings. In mating experiments in which females were sequentially housed with males, we examined a range of male characteristics which potentially influence fertilization success. We found that the size of oligopyrene sperm was the best predictor of fertilization success, with males having the longer sperm siring the highest proportion of offspring. Furthermore, we found a positive shell size and sperm concentration effect on paternity, and females with multiply sired families produced more offspring than females mating with only one male. The dimorphic sperm of Hemilepidotus gilberti, i.e., haploid eusperm and diploid parasperm, have different morphologies corresponding to their own roles in fertilization. To estimate how these specific sperm morphologies were established, we focused on the nuclear morphologies and examined their changing processes in dimorphic spermiogenesis. Electron microscopic observation revealed that, in euspermatids, chromatin condensation first appeared as a mosaic pattern of moderate electrodense material in the peripheral region of the round nucleus. Those materials spread across the whole area to form a uniformly condensed nucleus. Chromatin condensation began similarly in paraspermatids to that in euspermatids. These became localized to one side of a nucleus and further condensed to form strong electrodense chromatin clusters, which are a specific feature of parasperm. From the remodeled nuclei of eusperm and parasperm, we found five and three kinds of sperm-specific basic proteins (SBPs), respectively, substituted to histones. The N-terminus amino acid sequences of the SBPs suggest that, in parasperm, one major SBP and two minor ones were distinct from each other. In eusperm nuclei, two kinds of specific SBPs were detected in addition to the homologs of parasperm SBPs. The specific SBPs had homologous amino acid sequences with huge arginine clusters, and one of them was most dominant among the five kinds of SBPs. The different combinations of SBPs in the eusperm and parasperm may cause a specific pattern of chromatin condensation in the dimorphic sperm nuclei of H. gilberti.  相似文献

It is known that tubificine oligochaetes produce two types of spermatozoa: eusperm, fertilizing sperm with regular haploid DNA content; and parasperm, with a much lower DNA content, protecting and carrying the eusperm. Whereas mature spermatozoa and spermatids of the two lines are easily recognized by their morphology and DNA content, little is known about the first steps of differentiation of the two lines. This subject is addressed here in two ways: we have measured DNA content by a new method based on confocal laser microscopy and found that the total DNA content of parasperm cysts is extremely variable and equal or lower than total DNA content of eusperm cysts. Then we focused on the spermatocytes, and we found that the cells which will form paraspermatids undergo a peculiar kind of nuclear fragmentation which differ greatly from a regular cell division. During fragmentation DNA is distributed unevenly among the spermatids and this gives rise to a great and variable number of parasperm with variable DNA content. Immunocytochemical assays revealed that a proper meiotic spindle is never formed during fragmentation and that actin may play an important role in the chromatin division. Electron micrographs showed that the centrioles undergo a phenomenon of mass reproduction similar to that found in ciliated epithelia which supplies each of the numerous paraspermatids of its basal body. We used an artificial semen emission test to examine the semen transporting role of parasperm (unflagellated sperm), which are produced along with eusperm (normal sperm) by an incomplete meiosis in the marine sculpin Hemilepidotus gilberti. After separation of contents of semen (eusperm, parasperm, and seminal plasma) by centrifugation, the distance traveled by semen discharged vertically from a syringe into seawater was compared among various test semen; eusperm semen (eusperm re-mixed with seminal plasma), parasperm semen (parasperm re-mixed with seminal plasma), normal semen (eusperm and parasperm re-mixed with seminal plasma), and natural semen (unmodified semen). Parasperm semen traveled more than 1.5 times as far as the eusperm semen. The lateral dispersion width of normal semen after emission was significantly narrower than that of eusperm semen. These findings indicate that parasperm can reduce the lateral dispersion and prolong the distance semen travels. The eusperm ratio in the lower portion of the emitted semen did not differ from that in the upper portion, indicating that eusperm evenly distribute within the ejaculate and reach the lower portion of semen. Since males cannot closely approach eggs that are deposited in the narrow space between the female's belly and the spawning substrate, restraint of lateral dispersion and prolongation of the distance semen traveled would increase the number of eusperm arriving at eggs, despite reduction of eusperm in the ejaculate.  相似文献

On the sperm surface, glutathione S-transferases (GSTs) exist as oocyte binding proteins but their detoxification function in this unique cell type is not known. Using H(2)O(2)- and 4-hydroxynonenal-induced sperm dysfunction models, this study demonstrates that the sperm surface GSTs are able to use extracellular reduced glutathione to inhibit the loss of functional competence of goat spermatozoa; however, in the presence of GST inhibitors, they are unable to do so. In the context of susceptibility of spermatozoa to oxidative stress, this finding that strategically located sperm surface GSTs are important for maintaining the functional competence of sperm is relevant to studies on male infertility.  相似文献

To assess the relationship between mouse sperm head morphology and karyotype, sperm heads with either a normal or an abnormal morphology were injected individually into enucleated mouse oocytes that were karyotyped at the metaphase of the first cleavage. BALB/c male mice that produce an unusually high proportion of morphologically abnormal spermatozoa were used as sperm donors. Abnormal karyotypes were found in a significantly higher proportion of eggs injected with severely misshapen sperm heads (36-38%) as compared to those injected with normal and quasi-normal heads (15-21%) (p < 0.01). Most karyotype abnormalities were structural rather than numerical, the most common being breaks and exchanges of chromosome type in both normal and abnormal spermatozoa.  相似文献

Spermicide (i.e., female-mediated sperm death) is an understudied but potentially widespread phenomenon that has important ramifications for the study of sexual conflict, postcopulatory sexual selection, and fertility [1, 2]. Males are predicted to evolve adaptations against spermicide, but few antispermicidal mechanisms have been definitively identified. One such adaptation may be the enigmatic infertile sperm morphs or "parasperm" produced by many species, which have been hypothesized to protect their fertile brother "eusperm" from spermicide [2, 3]. Here, we show that female Drosophila pseudoobscura reproductive tracts are spermicidal and that the survival of eusperm after exposure to the female tract is highest when males produce many parasperm. This study clarifies the adaptive significance of infertile sperm castes, which has remained elusive in Drosophila and other taxa despite much recent interest [2-8]. We suggest that spermicide and male countermeasures against it are more common than is appreciated currently and discuss how spermicide could drive the evolution of several key male traits, including sperm size and number.  相似文献

Sperm surface changes during in vitro capacitation were examined with the help of an assay system using lectincoated agarose beads. The nature and intensity of binding of epididymal spermatozoa to beads depended entirely on the particular stage of capacitation and the type of lectin attached to the bead surface. Fresh epididymal spermatozoa bound readily to beads coated with Con A, LCA, WGA, and PNA, but not with seven other lectins. During capacitation there was a constant decline in sperm binding to beads, and spermatozoa cultured for 4–5 hr bound only to those coated with Con A. A dramatic increase in sperm binding to Con A-coated agarose beads occurred between 4.5 and 5 hr, when large numbers of hyperactivated spermatozoa adhered, predominantly through their flagellae, to form large clumps on the beads. The clumping of spermatozoa on Con A-coated beads was enhanced in the presence of stimulators of capacitation (i.e., taurine, hypotaurine, and phosphodiesterase inhibitors) and was suppressed in the presence of various metabolic inhibitors (i.e., sodium azide and local anesthetics). The implications of these results are that the carbohydrate components of the entire surface of spermatozoa undergo striking changes during capacitation, and a close relationship may exist between the sperm surface and the metabolic changes occurring within capacitating spermatozoa. Sperm-bead binding assays are clearly able to recognize surface changes in asynchronous populations of motile spermatozoa and, due to their simplicity and speed, should prove to be valuable in gaining a greater understanding of the biochemistry of sperm capacitation.  相似文献

Heat shock protein A4L (HSPA4L), which is highly expressed in the testis, is correlated with male fertility. However, the relationship between HSPA4L expression and sperm quality remains unknown. In the present study, a systematic characterization of HSPA4L expression on spermatozoa was performed. HSPA4L is highly expressed in the human testis, characterized by abundant localization in testicular spermatocytes and round spermatids. Compared with the testis from young adults (aged 27–36 years old), downregulated expression of HSPA4L in the testis from elderly adults (aged 78–82 years old) was observed. Immunofluorescence quantification demonstrated the localization of HSPA4L in the middle piece of sperm. Compared with mature spermatozoa, a similar lower intensity and localization percentage of HSPA4L in immature and asthenozoospermic spermatozoa were observed, and the consistently decreased expression of HSPA4L in immature and asthenozoospermic spermatozoa was validated by western blot analysis. Functional analysis revealed a correlation between HSPA4L and sperm motility by Spearman correlation analysis and its involvement in sperm–oocyte penetration by the human sperm–hamster egg penetration test. The current study demonstrates that HSPA4L is a promising marker for the assessment of sperm quality and provides clues for exploring biomarkers for the molecular diagnosis and treatment of male infertility.  相似文献
