The role wild bird species play in the transmission and ecology of avian influenza virus (AIV) is well established; however, there are significant gaps in our understanding of the worldwide distribution of these viruses, specifically about the prevalence and/or significance of AIV in Central and South America. As part of an assessment of the ecology of AIV in Guatemala, we conducted active surveillance in wild birds on the Pacific and Atlantic coasts. Cloacal and tracheal swab samples taken from resident and migratory wild birds were collected from February 2007 to January 2010.1913 samples were collected and virus was detected by real time RT-PCR (rRT-PCR) in 28 swab samples from ducks (Anas discors). Virus isolation was attempted for these positive samples, and 15 isolates were obtained from the migratory duck species Blue-winged teal. The subtypes identified included H7N9, H11N2, H3N8, H5N3, H8N4, and H5N4. Phylogenetic analysis of the viral sequences revealed that AIV isolates are highly similar to viruses from the North American lineage suggesting that bird migration dictates the ecology of these viruses in the Guatemalan bird population.  相似文献

Cloacal and tracheal swabs were collected from 1,409 hunter-killed ducks in Cameron Parish, Louisiana, during the 1986 and 1987 waterfowl seasons. Thirty avian paramyxoviruses (PMV's) were isolated from 605 blue-winged teal (Anas discors), 75 mottled ducks (A. fulvigula), 375 gadwalls (A. strepera), 334 green-winged teal (A. crecca), and 20 mallards (A. platyrhynchos). Prevalence of PMV decreased (P = 0.042) from September (4%) through November (2%) to December and January (1%). Juveniles had a higher prevalence of PMV (P less than 0.0001) than adults. An isolate from resident mottled ducks documents transmission of PMV's on the coastal wintering areas of Louisiana. The four serotypes isolated, PMV-1, PMV-4, PMV-6, and PMV-8, were typical of PMV's commonly associated with free-living waterfowl.  相似文献

Waterfowl from northwestern Minnesota were sampled by cloacal swabbing for Avian Influenza Virus (AIV) from July-October in 2007 and 2008. AIV was detected in 222 (9.1%) of 2,441 ducks in 2007 and in 438 (17.9%) of 2,452 ducks in 2008. Prevalence of AIV peaked in late summer. We detected 27 AIV subtypes during 2007 and 31 during 2008. Ten hemagglutinin (HA) subtypes were detected each year (i.e., H1, 3-8, and 10-12 during 2007; H1-8, 10 and 11 during 2008). All neuraminidase (NA) subtypes were detected during each year of the study. Subtype diversity varied between years and increased with prevalence into September. Predominant subtypes during 2007 (comprising ≥5% of subtype diversity) included H1N1, H3N6, H3N8, H4N6, H7N3, H10N7, and H11N9. Predominant subtypes during 2008 included H3N6, H3N8, H4N6, H4N8, H6N1, and H10N7. Additionally, within each HA subtype, the same predominant HA/NA subtype combinations were detected each year and included H1N1, H3N8, H4N6, H5N2, H6N1, H7N3, H8N4, H10N7, and H11N9. The H2N3 and H12N5 viruses also predominated within the H2 and H12 subtypes, respectively, but only were detected during a single year (H2 and H12 viruses were not detected during 2007 and 2008, respectively). Mallards were the predominant species sampled (63.7% of the total), and 531 AIV were isolated from this species (80.5% of the total isolates). Mallard data collected during both years adequately described the observed temporal and spatial prevalence from the total sample and also adequately represented subtype diversity. Juvenile mallards also were adequate in describing the temporal and spatial prevalence of AIV as well as subtype diversity.  相似文献

Although wild ducks are recognized reservoirs for avian influenza viruses (AIVs) and avian paramyxoviruses (APMVs), information related to the prevalence of these viruses in breeding and migratory duck populations on North American wintering grounds is limited. Wintering (n=2,889) and resident breeding (n=524) ducks were sampled in North Carolina during winter 2004-2006 and summer 2005-2006, respectively. Overall prevalence of AIV was 0.8% and restricted to the winter sample; however, prevalence in species within the genus Anas was 1.3% and was highest in Black Ducks (7%; Anas rubripes) and Northern Shovelers (8%; Anas clypeata). Of the 24 AIVs, 16 subtypes were detected, representing nine hemagglutinin and seven neuraminidase subtypes. Avian paramyxoviruses detected in wintering birds included 18 APMV-1s, 15 APMV-4s, and one APMV-6. During summers 2005 and 2006, a high prevalence of APMV-1 infection was observed in resident breeding Wood Ducks (Aix sponsa) and Mallards (Anas platyrhynchos).  相似文献

Waterbirds represent the major natural reservoir for low pathogenic (LP) avian influenza viruses (AIV). Among the wide diversity of subtypes that have been described, two of them (H5 and H7) may become highly pathogenic (HP) after their introduction into domestic bird populations and cause severe outbreaks, as is the case for HP H5N1 in South-Eastern Asia. Recent experimental studies demonstrated that HP H5N1 AIV infection in ducks does not necessarily have significant pathological effects. These results suggest that wild migratory ducks may asymptomatically carry HP AIV and potentially spread viruses over large geographical distances. In this study, we investigated the potential spreading distance of HP AIV by common teal (Anas crecca), mallard (A. platyrhynchos), and Eurasian pochard (Aythya ferina). Based on capture-mark-recapture method, we characterized their wintering movements from a western Mediterranean wetland (Camargue, South of France) and identified the potential distance and direction of virus dispersal. Such data may be crucial in determining higher-risk areas in the case of HP AIV infection detection in this major wintering quarter, and may serve as a valuable reference for virus outbreaks elsewhere.  相似文献

为了解华东地区家鸭内禽流感病毒的遗传进化情况,对2002~2006年分离自华东地区家鸭的3种主要N1亚型的禽流感病毒:2株H1N1、10株H3N1和14株H5N1,共26株病毒的NA基因进行了遗传进化分析。结果表明,华东地区家鸭中的N1亚型的禽流感病毒正处于不断进化状态中。14株H5N1禽流感病毒均在NA的茎部缺失20个氨基酸(49~68位),而其他N1亚型的禽流感病毒的NA都未见发生此缺失。H3N1病毒可能与H1N1病毒发生了NA基因的重排,但是目前还没有直接证据表明华东地区家鸭中H5N1禽流感参与了基因重排。  相似文献

Since 1997, outbreaks of highly pathogenic (HP) H5N1 and circulation of H9N2 viruses among domestic poultry in Asia have posed a threat to public health. To better understand the extent of transmission of avian influenza viruses (AIV) to humans in Asia, we conducted a cross-sectional virologic study in live bird markets (LBM) in Hanoi, Vietnam, in October 2001. Specimens from 189 birds and 18 environmental samples were collected at 10 LBM. Four influenza A viruses of the H4N6 (n = 1), H5N2 (n = 1), and H9N3 (n = 2) subtypes were isolated from healthy ducks for an isolation frequency of over 30% from this species. Two H5N1 viruses were isolated from healthy geese. The hemagglutinin (HA) genes of these H5N1 viruses possessed multiple basic amino acid motifs at the cleavage site, were HP for experimentally infected chickens, and were thus characterized as HP AIV. These HA genes shared high amino acid identities with genes of other H5N1 viruses isolated in Asia during this period, but they were genetically distinct from those of H5N1 viruses isolated from poultry and humans in Vietnam during the early 2004 outbreaks. These viruses were not highly virulent for experimentally infected ducks, mice, or ferrets. These results establish that HP H5N1 viruses with properties similar to viruses isolated in Hong Kong and mainland China circulated in Vietnam as early as 2001, suggest a common source for H5N1 viruses circulating in these Asian countries, and provide a framework to better understand the recent widespread emergence of HP H5N1 viruses in Asia.  相似文献

Recent repeated isolation of H14 hemagglutinin subtype influenza A viruses (IAVs) in the New World waterfowl provides evidence to suggest that host and/or geographic ranges for viruses of this subtype may be expanding. In this study, we used genomic analyses to gain inference on the origin and evolution of H14 viruses in New World waterfowl and conducted an experimental challenge study in mallards (Anas platyrhynchos) to evaluate pathogenicity, viral replication, and transmissibility of a representative viral strain in a natural host species. Genomic characterization of H14 subtype IAVs isolated from New World waterfowl, including three isolates sequenced specifically for this study, revealed high nucleotide identity among individual gene segments (e.g. ≥95% shared identity among H14 HA gene segments). In contrast, lower shared identity was observed among internal gene segments. Furthermore, multiple neuraminidase subtypes were observed for H14 IAVs isolated in the New World. Gene segments of H14 viruses isolated after 2010 shared ancestral genetic lineages with IAVs isolated from wild birds throughout North America. Thus, genomic characterization provided evidence for viral evolution in New World waterfowl through genetic drift and genetic shift since purported introduction from Eurasia. In the challenge study, no clinical disease or lesions were observed among mallards experimentally inoculated with A/blue-winged teal/Texas/AI13-1028/2013(H14N5) or exposed via contact with infected birds. Titers of viral shedding for mallards challenged with the H14N5 IAV were highest at two days post-inoculation (DPI); however shedding was detected up to nine DPI using cloacal swabs. The distribution of viral antigen among mallards infected with H14N5 IAV was largely restricted to enterocytes lining the villi in the lower intestinal tract and in the epithelium of the bursa of Fabricius. Characterization of the infectivity of A/blue-winged teal/Texas/AI13-1028/2013(H14N5) in mallards provides support for similarities in viral replication and shedding as compared to previously described waterfowl-adapted, low pathogenic IAV strains in ducks.  相似文献

Over the past 20 yr, recurrent late-summer mortality of dabbling ducks in southern Québec has been attributed to the digenean parasite Cyathocotyle bushiensis. This study attempted to determine whether this parasite was still implicated in the epizootics, and examined aspects of the ecology of the parasite in the definitive host. Comparison of prevalence and mean intensity of infection of all species of Digenea between salvaged carcasses and hunter-shot ducks revealed that C. bushiensis and a second digenean Sphaeridiotrema globulus were both significantly more prevalent in salvaged ducks. Mean intensity of infection was consistently higher for S. globulus in the salvaged carcasses than in hunter-shot ducks. These data provide strong circumstantial support for the hypothesis that C. bushiensis continues to contribute to the annual mortality, but also strongly suggest that S. globulus may be involved. Comparison of the relative numbers of each species of duck between the salvaged carcass sample and the hunter-shot sample revealed that blue-winged teal (Anas discors) occurred significantly more frequently in the salvaged carcass sample whereas wood ducks (Aix sponsa) were common in the hunter-shot sample but were never found in the salvaged carcass sample. A comparison of susceptibility to experimental infection with C. bushiensis revealed that mallards (Anas platyrhynchos), black ducks (Anas rubripes), blue-winged teal, pintail (Anas acuta) and lesser scaup (Aythya affinis) had similar susceptibilities but that wood ducks were significantly less susceptible to infection. Experimental infections also showed that age of mallards significantly influenced the number of parasites obtained 7 days postinfection; ducklings were most heavily infected. Significant differences were detected in the gross pathology caused by C. bushiensis among duck species. Data obtained from sentinel ducks placed in the field for 24 hr revealed that transmission of both C. bushiensis and S. globulus occurs at least between mid-May and mid-September and that large, potentially lethal, infections can be acquired in as little as 24 hr. Finally evidence is presented to suggest that ducks acquire resistance to challenge infections of C. bushiensis.  相似文献

Ten adult blue-winged teal (Anas discors) and six Canada goose (Branta canadensis) goslings were inoculated with liver tissue from a natural case of duck plague in a wild mallard (Anas platyrhynchos). Four additional teal were placed in contact with the inoculated ducks. Inoculated teal died 63.5-68 hr after inoculation; two of the contact teal died 161-162 hr after exposure. Three of the goslings died 119-133 hr after inoculation, the others were killed when moribund 90-133 hr postinfection. The clinical course of disease was extremely rapid in both species. Signs were limited to sudden onset of profound weakness, ataxia, tremors and terminal convulsions. The only consistent gross lesion in the teal was a small dark spleen; half the teal also had inconspicuous foci of epithelial necrosis in the distal esophagus and in the cloaca. Goslings had more severe lesions, with focal hepatic necrosis visible in all, and mucosal necrosis over the intestinal lymphoid tissue, and intestinal hemorrhage in three birds each. Microscopic lesions in both species were similar to those reported in mallards. Duck plague might be overlooked at necropsy of blue-winged teal, because of the paucity of gross lesions.  相似文献

The nonmigratory and endemic Florida mottled duck (Anas fulvigula fulvigula) is facing conservation threats from the combined effects of urbanization and introgressive hybridization with feral mallards (Anas platyrhynchos) and mallard x mottled duck hybrids. In the past, the status of the Florida mottled duck population was assessed during annual aerial surveys and most brown ducks (mottled ducks, mallards, and hybrids of them) detected during the survey would have been mottled ducks. But the release of domesticated mallards for aesthetic purposes has led to increases in the prevalence of mallards-hybrids (mallards or mallard x mottled duck hybrids) throughout peninsular Florida, USA, and because it is impossible to differentiate among mottled ducks, female mallards, and hybrids during aerial surveys, helicopter surveys were halted in 2009 until state researchers could conduct a range-wide study to determine what proportion of brown ducks are mottled ducks versus mallards-hybrids. We used plumage keys and high-resolution photography to categorize brown ducks from 557 wetland grid points as either mottled ducks or mallards-hybrids. Of the 5,179 brown ducks categorized, 40.1% were mottled ducks and 59.9% were mallards-hybrids. We used logistic regression analysis to model the interactive effect of a site's latitude and level of urbanization (urban gradient value within a 2-km buffer) to generate a predictive raster surface (1-km resolution) of the study area with values corresponding to the probability that a brown duck observed within a cell is a pure mottled duck. Predicted values will be used as correction factors when estimating final mottled duck population abundance from brown-duck survey data. Additionally, the predictive raster surface will be used to identify wetlands where mottled ducks remain predominant so that these sites can be targeted for preservation. Overall, mallards-hybrids outnumbered mottled ducks throughout most of peninsular Florida, especially in more urbanized regions, and their current prevalence rate presents a serious conservation threat, via hybridization, to extant mottled duck populations.  相似文献

Migratory waterfowl of the world are the natural reservoirs of influenza viruses of all known subtypes. However, it is unknown whether these waterfowl perpetuate highly pathogenic (HP) H5 and H7 avian influenza viruses. Here we report influenza virus surveillance from 2001 to 2006 in wild ducks in Alberta, Canada, and in shorebirds and gulls at Delaware Bay (New Jersey), United States, and examine the frequency of exchange of influenza viruses between the Eurasian and American virus clades, or superfamilies. Influenza viruses belonging to each of the subtypes H1 through H13 and N1 through N9 were detected in these waterfowl, but H14 and H15 were not found. Viruses of the HP Asian H5N1 subtypes were not detected, and serologic studies in adult mallard ducks provided no evidence of their circulation. The recently described H16 subtype of influenza viruses was detected in American shorebirds and gulls but not in ducks. We also found an unusual cluster of H7N3 influenza viruses in shorebirds and gulls that was able to replicate well in chickens and kill chicken embryos. Genetic analysis of 6,767 avian influenza gene segments and 248 complete avian influenza viruses supported the notion that the exchange of entire influenza viruses between the Eurasian and American clades does not occur frequently. Overall, the available evidence does not support the perpetuation of HP H5N1 influenza in migratory birds and suggests that the introduction of HP Asian H5N1 to the Americas by migratory birds is likely to be a rare event.  相似文献

Since the recent spread of highly pathogenic (HP) H5N1 subtypes, avian influenza virus (AIV) dispersal has become an increasing focus of research. As for any other bird-borne pathogen, dispersal of these viruses is related to local and migratory movements of their hosts. In this study, we investigated potential AIV spread by Common Teal (Anas crecca) from the Camargue area, in the South of France, across Europe. Based on bird-ring recoveries, local duck population sizes and prevalence of infection with these viruses, we built an individual-based spatially explicit model describing bird movements, both locally (between wintering areas) and at the flyway scale. We investigated the effects of viral excretion duration and inactivation rate in water by simulating AIV spread with varying values for these two parameters. The results indicate that an efficient AIV dispersal in space is possible only for excretion durations longer than 7 days. Virus inactivation rate in the environment appears as a key parameter in the model because it allows local persistence of AIV over several months, the interval between two migratory periods. Virus persistence in water thus represents an important component of contamination risk as ducks migrate along their flyway. Based on the present modelling exercise, we also argue that HP H5N1 AIV is unlikely to be efficiently spread by Common Teal dispersal only.  相似文献

[目的]为了对途经三江保护区的野生迁徙水禽携带禽流感病毒(AIV)和新城疫病毒(NDV)的状况进行有效监测.[方法]在2005年10月、2006年4月、2006年10月3个候鸟的迁徙季节从三江保护区采集了158只野鸟的咽拭子和肛拭子样本.应用SPF鸡胚盲传、血凝和血凝抑制试验和RT-PCR等方法进行了病毒的分离和鉴定.[结果]结果共分离到20株AIV和13株NDV.20株AIV均来自2006年10月采集的样品,经常规血清学分型鉴定分为12个亚型,11个亚型来源于绿头鸭,分别为H2N2(2/20),H2N6(2/20),H3N4(1/20),H3N6(2/20),H3N7(2/20),H3N8(2/20),H6N2(2/20),H11N2(1/20),H11N3(1/20),H11N5(2/20),H11N6(1/20),另外一株来源于白眉鸭,为H5N2(1/20).13株NDV则来自3个迁徙季节的5种不同水禽采,其中包括绿头鸭(8/13),豆雁(1/13),白额雁(1/13),绿翅鸭(1/13)和鸳鸯(2/13).[结论]这一结果表明,拥有极大种群数量、在世界范围内广泛分布的绿头鸭,被认为可能是AIV和NDV最重要的自然宿主之一,并在病毒的传播上比其他野生鸟类具有更为重要的生态学意义.  相似文献

[目的]为了研究近年来华东地区家鸭中禽流感病毒的亚型分布情况.[方法]对2002-2006年分离自华东地区家鸭的180株禽流感病毒的HA亚型和其中88株禽流感病毒的NA亚型分别进行了测定.[结果]近年来华东地区家鸭中至少存在9种HA亚型和6种NA亚型组成的H1N1,H3N1,H3N2,H3N8,H4N6,H5N1,H5N2,H6N2,H6N8,H8N4,H9N2,H10N3,H11N2共13种亚型的禽流感病毒.[结论]华东地区家鸭中有多种亚型的禽流感病毒分布,应加强家鸭禽流感的监测和防制工作.  相似文献

Macrocysts of the protozoan parasite Sarcocystis spp. occurred in 34 of 241 (14%) adult and 6 of 70 (8.6%) immature mottled ducks (Anas fulvigula) that were collected in southwestern Louisiana from 1987 to 1989. The prevalence of Sarcocystis spp. among immature ducks was higher than previously reported, possibly because of a longer exposure period. No differences in carcass composition variables were found between heavily infected and uninfected adult mottled ducks during the winter period of the annual cycle. We concluded that macrocysts of Sarcocystis spp. did not influence carcass composition and probably exert negligible effects on survival of parasitized mottled ducks during winter.  相似文献

Twenty-five species of helminths, recovered from the gastrointestinal tracts of 129 Mexican ducks from Mexico and the United States, were all new host records. The species included: Echinoparyphium recurvatum, Echinostoma revolutum, Hypoderaeum conoideum, Notocotylus attenuatus, Prosthogonimus cuneatus, Zygocotyle lunata, Anomotaenia ciliata, Cloacotaenia megalops, Diorchis bulbodes, Diorchis sp., Drepanidotaenia lanceolata, Echinocotyle rosseteri, Fimbriaria fasciolaris, Fimbriarioides sp., Hymenolepis sp. 1, Hymenolepis sp. 2, Sobolevicanthus gracilis. Corynosoma constrictum, Polymorphus minutus, Amidostomum acutum, Echinuria sp., Epomidiostomum crami, Hystrichis varispinosus, Rusguniella arctica, and Tetrameres sp. Fimbriarioides sp. occurred predominantly in ducks from south-central Chihuahua, Mexico. The distributions for the other 24 species of the helminths were not significantly different among the four collecting areas. The helminth fauna for the 32 complete specimens and 97 intestinal tracts was distributed relatively evenly among the hosts with a calculated mean evenness of 0.77 +/- 0.15 and 0.89 +/- 0.03 respectively. The parasite fauna was more similar to those of the black duck Anas rubripes Brewster of eastern North America (53%), the mallard, Anas platyrhynchos (L.) (49%), and the mottled duck, Anas fulvigula Ridgway, from Florida (45%), than to the green-winged teal, Anas crecca (L.) (36%), the gadwall, Anas strepera (L.) (30%), and the American wigeon, Anas americana Gmelin (26%), collected in the Southwest.  相似文献

Interspecific hybridization has been implicated in population declines for some waterfowl species within the mallard complex, and hybridization with mallards (Anas platyrhynchos) is currently considered the largest threat to mottled ducks (A. fulvigula), one North American member of that complex. We assessed genetic variation among 225 mottled ducks and mallards using five microsatellite loci, and detected significant overall differences between these species within two geographic areas. We characterized hybridization in Florida, where mottled ducks are endemic and mallards are beginning to appear on the breeding grounds, and in South Carolina, where mottled ducks were introduced outside their native range. We used Bayesian genetic mixture analysis in an attempt to distinguish between these closely related species. In Florida, we detected two distinct genetic groups, and 10.9% of our samples from Florida mottled ducks were inferred to have been hybrids. In contrast only 3.4% of Florida mallards were inferred to have been hybrids, suggesting asymmetric hybridization. Populations from different geographic areas within Florida exhibited hybridization rates ranging from 0% to 24%. These data indicate a genetic component would be appropriate in actively managing interspecific hybridization in Florida mottled ducks. In contrast, South Carolina mottled ducks and mallards cannot be differentiated.  相似文献

In wild aquatic birds and poultry around the world, influenza A viruses carrying 15 antigenic subtypes of hemagglutinin (HA) and 9 antigenic subtypes of neuraminidase (NA) have been described. Here we describe a previously unidentified antigenic subtype of HA (H16), detected in viruses circulating in black-headed gulls in Sweden. In agreement with established criteria for the definition of antigenic subtypes, hemagglutination inhibition assays and immunodiffusion assays failed to detect specific reactivity between H16 and the previously described subtypes H1 to H15. Genetically, H16 HA was found to be distantly related to H13 HA, a subtype also detected exclusively in shorebirds, and the amino acid composition of the putative receptor-binding site of H13 and H16 HAs was found to be distinct from that in HA subtypes circulating in ducks and geese. The H16 viruses contained NA genes that were similar to those of other Eurasian shorebirds but genetically distinct from N3 genes detected in other birds and geographical locations. The European gull viruses were further distinguishable from other influenza A viruses based on their PB2, NP, and NS genes. Gaining information on the full spectrum of avian influenza A viruses and creating reagents for their detection and identification will remain an important task for influenza surveillance, outbreak control, and animal and public health. We propose that sequence analyses of HA and NA genes of influenza A viruses be used for the rapid identification of existing and novel HA and NA subtypes.  相似文献
