以从疏花水柏枝(Myricaria laxiflora)中分离的1株内生真菌QY-1为研究对象,为分析其体内及体外的抗氧化活性,对其发酵液及其提取物的活性进行了综合评价。对nr DNA内转录间隔区进行测序鉴定。分别采用总抗氧化力试剂盒、自由基清除试剂盒和还原力测定试剂盒评价发酵液及粗提物的体外抗氧化能力活性;对发酵液提取物及其主成分进行了大肠埃希菌、人神经母瘤细胞SH-SY5Y的体内抗氧化损伤保护作用分析。结果表明QY-1是1株球毛壳菌(Chaetomium globosum),其粗提物具有很高的抗氧化活性,总抗氧化活力、自由基清除率及总还原力均接近维生素C的50%,远高于国际报道水平。且其发酵液提取物有促进细胞增殖和抗氧化保护作用,显著提高大肠埃希菌细胞和神经细胞在H2O2胁迫下的存活率。其粗提物可保护神经细胞膜的完整性,有效减低乳酸脱氢酶(LDH)的渗漏率。从粗提物中分离到一种主成分SF2,可显著抑制凋亡蛋白Caspase 3和Caspase 9的活性。结果表明内生真菌球毛壳菌QY-1不管在体内还是体外都具备较强的抗氧化能力,是一种具有潜在开发价值的天然抗氧化剂资源。  相似文献

采用体外α-葡萄糖苷酶抑制模型对一株球毛壳菌H6的发酵液和菌丝体两种乙酸乙酯提取物进行活性评价,结果表明,发酵液乙酸乙酯提取物对α-葡萄糖苷酶具有较强的抑制活性,其IC50值为(510.76±23.46)μg/mL。采用硅胶、Sephadex LH-20、半制备高效液相等色谱技术从其发酵液乙酸乙酯提取物中分离得到12个化合物。通过各种光谱分析,依次鉴定为chaetoviridins A-B(1-2),chaetoglobosins A-D(3-6),chaetoglobosin J(7),chaetoglobosin Q(8),prochaetogobosinsⅠ-Ⅲ(9-11),vibratilicin(12),其中化合物12为首次从毛壳属中分离得到。对化合物进行α-葡萄糖苷酶抑制活性测定发现,化合物12显示弱的抑制活性,其IC50为(1 182.75±19.14)μg/mL。  相似文献

结合形态特征和分子生物学方法,将一株分离自川贝母鳞茎的内生真菌CBY11鉴定为Fusarium tricinctum。分别研究其发酵液石油醚提取物(PEE)、乙酸乙酯提取物(EAE)、正丁醇提取物(BAE)和乙醇提取物(ETE)的DPPH自由基清除活性、总抗氧化活性(以ABTS法测定)和总还原力(FRAP法测定),以及其总多酚含量(TPC)、总黄酮含量(TFC)和总皂苷(TSC)含量。结果发现EAE、BAE和ETE均具有较强的DPPH自由基清除活性(IC50分别为1.95mg/m L、1.19mg/m L、1.64mg/m L)和总抗氧化活性(IC50分别为0.43mg/m L、0.58mg/m L、0.43mg/m L),但总还原能力相对较弱;PEE的抗氧化活性整体较低。此外,EAE、BAE和ETE的TPC、TFC及TSC显著高于PEE(P0.05)。可见TPC、TFC及TSC与抗氧化活性呈现出一定的正相关性。HPLC分析表明EAE含有多种具有较强抗氧化活性的酚酸等物质,如芹菜素。GC/MS分析表明PEE中含有少量抗氧化酚类物质,最主要成分是烷烯烃类。本研究表明川贝母内生真菌CBY11具有产生丰富抗氧化活性成分的能力,具有多种利用价值。  相似文献

为探究林下参内生真菌球毛壳菌(Chaetomium globosum)FS-01菌株对人参病原菌的抑菌作用,该研究在实验室条件下,测定了FS-01菌株菌丝、发酵液和孢子悬浮液对人参黑斑病菌(Alternaria panax)、人参菌核病菌(Sclerotinia schinseng)、人参灰霉病菌(Botrytis cinerea)、人参立枯病菌(Rhizoctonia solani)、人参根腐病菌(Fusarium solani)5种人参病原菌的抑制作用。结果表明:内生真菌球毛壳菌FS-01对5种病原菌均有抑制作用,其中,对人参黑斑病菌的抑制作用最高,为30.80%,其次是人参立枯病菌、人参菌核病菌、人参根腐病菌和人参灰霉病菌; 发酵液抑菌实验结果表明,在加入内生真菌球毛壳菌FS-01菌株发酵液的PDA培养基上,对人参灰霉病菌的抑制作用最高,为82.09%,其次是人参菌核病菌、人参黑斑病菌、人参立枯病菌和人参根腐病菌; 孢子抑菌实验结果表明,在加入内生真菌球毛壳菌FS-01菌株孢子悬浮液的PDA培养基上,对人参黑斑病菌的抑制作用最高,为83.72%,其次是人参灰霉病菌、人参立枯病菌、人参菌核病菌和人参根腐病菌。综上结果认为,内生真菌球毛壳菌FS-01菌株对人参病原菌均有很高的抑菌作用,可作为人参病原菌的生防菌株资源。  相似文献

测定处于不同生长期的杜仲内生真菌DZJ03胞外多糖含量、发酵液中3种核苷含量,并对其菌株发酵液抑菌效果进行了研究,按50μg/mL的浓度测定了发酵液的石油醚、乙酸乙酯和甲醇等3种提取物对水稻恶苗病菌、棉花枯萎病菌2种植物病原菌以及烟草青枯病菌、金黄色葡萄球菌的抑制活性。结果表明:内生真菌DZJ03胞外多糖含量最高可达到2.0410g/L,其发酵液中所含腺苷、尿苷以及鸟苷的含量分别为1.2647 mg/g、0.8586 mg/g、1.0493 mg/g;发酵液乙酸乙酯相具有较强的抑菌活性,其对水稻恶苗病菌的抑制率为71.92%,抗烟草青枯病菌的抑菌圈直径达到19.21 mm。  相似文献

忍冬木层孔菌是木层孔菌属的一种药用真菌,目前对它的研究少有报道。以液体培养为基础,研究忍冬木层孔菌培养过程中生物量、胞内外多酚含量的变化,以及胞内外提取物抗氧化活性的变化。结果显示,菌丝体生物量在培养168–216h时达到最大值(8.33g/L);胞内多酚和胞外多酚含量分别在216h和168h时达到最大值(15.60mg GAE/g和106.76mg GAE/L);液体培养胞内外提取物的抗氧化活性与多酚含量正相关,多酚含量高时提取物对DPPH[1,1-Diphenyl-2-picrylhydrazyl radical 2,2-Diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl]自由基的抑制率也高。  相似文献

用55%乙醇回流提取散沫花叶中的活性物质,采用比色法测定提取物中的总黄酮及多酚含量,测定了其对DPPH、羟自由基、超氧阴离子自由基的清除能力和还原力等抗氧化活性指标。结果表明,散沫花叶提取物中的总黄酮(以芦丁计)、多酚(以没食子酸计)含量分别为39.42 mg/g、116.20 mg/g,提取物具有一定的抗氧化活性,且抗氧化活性与提取物浓度存在量效关系,本研究为散沫花叶作为低毒高效的天然抗氧化剂应用提供了依据。  相似文献

采用55%的乙醇分别回流提取凤仙花的茎、叶、花中的活性物质,采用比色法测定各部位的多酚及总黄酮含量,测定各提取物对DPPH、羟自由基、超氧阴离子自由基的清除能力和还原力等抗氧化活性指标。结果表明,凤仙花各部位的多酚(以没食子酸计)、总黄酮(以芦丁计)含量由高到低依次为凤仙花的叶、花、茎,多酚含量分别为72.81、66.07、33.07 mg/m L,总黄酮含量分别为112.13、108.06、30.62 mg/m L。各种提取物均有不同程度的抗氧化活性,且抗氧化活性与提取物浓度存在量效关系,其中叶提取物清除DPPH自由基、羟自由基、超氧阴离子自由基活性以及还原力均最高,本研究为凤仙花作为低毒高效的天然抗氧化剂应用提供了依据。  相似文献

本文探讨了益智(Alpinia oxyphylla Miquel)超临界CO2提取物及其渣的水提物、正丙醇提取物和乙酸乙酯提取物的抗氧化作用,测定了总酚含量、黄酮含量、抗氧化力、还原能力、DPPH清除率.结果表明,益智超临界CO2提取物和正丙醇提取物的总酚含量最高,均为5.53%,乙酸乙酯提取物的总酚含量为4.04%,水提物总酚含量最低,为O.89%.抗氧化力与酚含量相关(R2=0.703).四种提取物中黄酮含量顺序为:乙酸乙酯提取物(6.29%)>丙醇提取物(5.81%)>水提取物(4.85%)>超临界CO2提取物(4.70%).在还原能力、清除DPPH自由基和羟自由基方面,乙酸乙酯提取物表现出了很强的抗氧化能力,呈现剂量依赖关系.  相似文献

Cancer prevention and treatment using traditional Chinese medicines have attracted increasing interest. This study characterizes antioxidant activity and phenolic compounds of traditional Chinese medicinal plants associated with anticancer, comprising 112 species from 50 plant families. The improved ABTS(*+) method was used to systematically assess the total antioxidant capacity (Trolox equivalent antioxidant capacity, TEAC) of the medicinal extracts. The TEAC values and total phenolic content for methanolic extracts of herbs ranged from 46.7 to 17,323 micromol Trolox equivalent/100 g dry weight (DW), and from 0.22 to 50.3 g of gallic acid equivalent/100 g DW, respectively. A positive, significant linear relationship between antioxidant activity and total phenolic content (all R(2) values>/=0.95) showed that phenolic compounds were the dominant antioxidant components in the tested medicinal herbs. Major types of phenolic compounds from most of the tested herbs were preliminarily identified and analyzed, and mainly included phenolic acids, flavonoids, tannins, coumarins, lignans, quinones, stilbenes, and curcuminoids. These medicinal herbs exhibited far stronger antioxidant activity and contained significantly higher levels of phenolics than common vegetables and fruits. Traditional Chinese medicinal plants associated with anticancer might be potential sources of potent natural antioxidants and beneficial chemopreventive agents.  相似文献

Introduction – Polyphenolic phytochemicals in traditionally used medicinal plants act as powerful antioxidants, which aroused an increasing interest in their application in functional food development. Objective – The effect of extraction time (5 and 15 min) and hydrolysis on the qualitative and quantitative content of phenolic compounds and antioxidant capacity of six traditionally used medicinal plants (Melissa officinalis L., Thymus serpyllum L., Lavandula officinalis Miller, Rubus fruticosus L., Urtica dioica L., and Olea europea L.) were investigated. Methodology – The content of total phenols, flavonoids, flavan‐3‐ols and tannins was determined using UV/Vis spectrophotometric methods, while individual phenolic acids, flavones and flavonols were separated and detected using HPLC analysis. Also, to obtain relevant data on the antioxidant capacity, two different assays, (2,2‐azino‐bis(3‐ethylbenzthiazoline‐6‐sulphonic acid) (ABTS) radical scavenging and ferric reducing/antioxidant power (FRAP) assays were used. Results – The extraction efficiency of phenolics, as well as the antioxidant capacity of plant extracts, was affected by both prolonged extraction and hydrolysis. The overall highest content of phenolic compounds was determined in hydrolyzed extract of blackberry leaves (2160 mg GAE/L), followed by the non‐hydrolyzed extract of lemon balm obtained after 15 min of extraction (929.33 mg GAE/L). The above extracts also exhibited the highest antioxidant capacity, while extracts of olive leaves were characterized with the lowest content of phenolic compounds, as well as the lowest antioxidant capacity. The highest content of rosmarinic acid, as the most abundant phenolic compound, was determined in non‐hydrolyzed extract of lemon balm, obtained after 15 min of extraction. Although the hydrolysis provided the highest content of polyphenolic compounds, longer extraction time (15 min) was more efficient to extract these bioactives than shorter extraction duration (5 min). Conclusion – The distribution of detected phenolic compounds showed a wide variability with regard to their botanical origin. Low meadow-rue (Thalictrum minus L.) antioxidant complex was studied in cell extracts and culture medium. Its activity was expressed as total polyphenol content in ferulic acid equivalents. In these model systems (cell extracts and culture medium) the inhibition of lipid oxidation and diphenylpicrylhydrazine reduction (EC50 = 12–15 μg/ml) were observed. At the phenolic compound concentration of 8–15 μg/ml, the reducing capacity of cell extracts was equivalent to 1.5 mM ascorbic acid. At the same time, berberine, a major alkaloid synthesized by the culture, manifested a low antioxidant activity. The analysis of phenolic acid composition in low meadow-rue showed that one of the main components of its antioxidant system were caffeic, gallic, chlorogenic, and ferulic acids.  相似文献

Fruits of Broussonetia papyrifera from South China were analyzed for their total chemical composition, and antioxidant activities in ethanol and aqueous extracts. In the fruit of this plant, the crude protein, crude fat and carbohydrates was 7.08%, 3.72% and 64.73% of dry weight, respectively. The crude protein, crude fat and carbohydrates were 15.71%, 20.51% and 36.09% of dry weight, respectively. Fatty acid and amino acid composition of the fruit were analyzed. Unsaturated fatty acid concentration was 70.6% of the total fatty acids. The percentage of the essential amino acids (EAAs) was 40.60% of the total amino acids. Furthermore, B. papyrifera fruit are rich in many mineral elements and vitamins. Total phenolic content was assessed using the Folin-Ciocalteau assay, whereas antioxidant activities were assessed by measuring the ability of the two extracts to scavenge DPPH radicals, inhibit peroxidation, and chelate ferric ions. Their reducing power was also assessed. Results indicated that the aqueous extract of B. papyrifera was a more potent reducing agent and radical-scavenger than the ethanol extract. GC-MS analysis of the ethanol extract showed the presence of some acid-containing compounds. The changes in total phenolic content and antioxidant capacity in B. papyrifera from four different regions grown under normal conditions were assessed. The antioxidant activity of different extracts was positively associated with their total phenolic content. These results suggest that the fruit of B. papyrifera could be used in dietary supplement preparations, or as a food additive, for nutritional gain, or to prevent oxidation in food products.  相似文献

ABSTRACT

This study investigated the contents of saponins and phenolic compounds in relation to their antioxidant activity and α-glucosidase inhibition activity of 7 colored quinoa varieties. The total saponin content was significantly different among 7 varieties and ranged from 7.51 to 12.12 mg OAE/g DW. Darker quinoa had a higher content of phenolic compounds, as well as higher flavonoids and antioxidant activity than that of light varieties. Nine individual phenolic compounds were detected in free and bound form, with gallic acid and ferulic acid representing the major compounds. The free and bound phenolic compounds (gallic acid and ferulic acid in particular) exhibited high linear correlation with their corresponding antioxidant values. In addition, the free phenolic extracts from colored quinoa exhibited higher inhibitory activity against α-glucosidase than the bound phenolic extracts. These findings imply that colored quinoa with abundant bioactive phytochemicals could be an important natural source for preparing functional food.  相似文献

The methanolic extracts of Lactarius deterrimus, Suillus collitinus, Boletus edulis, Xerocomus chrysenteron were analyzed for their antioxidant activities in different test systems namely beta-carotene/linoleic acid, DPPH free radical scavenging, reducing power and metal chelating activities in addition to their total phenolic and flavonoid contents. In beta-carotene/linoleic acid and DPPH systems, L. deterrimus and B. edulis showed the strongest activity patterns. Their activities were as strong as the positive controls. The reducing power of the species was excellent. Chelating capacity of the extracts was increased with the increasing concentration. On the other hand, B. edulis found to have the highest phenolic content. Total flavonoid content of S. collitinus found the superior to the other mushrooms.  相似文献

Three underutilized leafy vegetables Sarcochlamys pulcherrima (Roxb.) Gaudich (SP), Ipomoea aquatica Forssk. (IA) and Zanthoxylum rhetsa (Roxb.) DC (ZR) were extracted with different solvents viz. 95 % ethyl alcohol, methanol and hot water. The extracts were evaluated for their antioxidant potential via DPPH, ABTS and FRAP assay along with electroanalytical studies using cyclic voltammetry. The antidiabetic potential was determined by recording their α-amylase and α-glucosidase inhibitory assay. The total phenolic content (TPC), total flavonoid content (TFC) and the liquid chromatography-mass spectrometry (LC/MS) based phytochemical profiles of the extracts were also determined. All three extracts of SP exhibited significant antioxidant capacity. The antidiabetic potential of the IA and ZR extracts was found to be higher than or at par with that of standard acarbose. LC/MS studies reveal the presence of hitherto reported antioxidant and antidiabetic compounds like gamma-aminobutyric acid, cinnamic acid, caffeic acid, α-viniferin, piperlonguminine, niacin, kaempferol, etc., in the extracts.  相似文献

Epidemiology studies have shown that consumption of fruits and vegetables is associated with the prevention of chronic diseases such as cancer and cardiovascular disease. Induction of cellular phase II detoxifying enzymes is associated with cancer preventive potential. Phenolsulfotransferases (PSTs) are traditionally known as phase II drug-metabolizing or detoxifying enzymes that facilitate the removal of drugs and other xenobiotic compounds. Phenolic acids are known to increase the activities of PSTs. In the present study, human HepG2 cells were used as model to investigate the influence of twenty vegetables on human PST activity and to evaluate the relationships to their antioxidant activity and total phenolics content. The result showed that PST-P activity was significantly (p < 0.01) induced by asparagus, broccoli, cauliflower, celery and eggplant, whereas PST-M activity was induced by asparagus, broccoli, carrot, eggplant and potato at a concentration of 100 microg/ml. The vegetable extracts that induced both forms of PSTs activities were found to have higher antioxidant capacities and total phenolic content in the oxygen radical absorbance capacity (ORAC) and Folin-Ciocalteu assay. The major polyphenols in broccoli, the most potential inducer in both forms of PSTs activities, was antioxidant phenolic acids. HPLC retention times and standard spiked indicated the presence of gallic acid, p-hydroxybenzoic acid, p-coumaric acid, gentisic acid and ferulic acid in broccoli. The overall effect of vegetables tested on the activity of PST-P was well correlated to their ORAC value and total phenolics content (r= 0.82, p < 0.05 and r = 0.78, p < 0.05). These results imply that vegetables have a capability of inducing PST activity, and the PST induction may be possibly ascribed to antioxidant phenolic acids in vegetable extracts.  相似文献

Two phenolic compound parameters (total phenolic and flavonoid contents) and 5 antioxidant parameters (DPPH [2, 2-diphenyl-1-picrylhydrazyl] radical scavenging activity, HRSC (hydroxyl radical scavenging capacity), FRAP (ferric ion reducing antioxidant power), CUPRAC (cupric ion reducing antioxidant capacity), and MCA (metal chelating activity) were measured in bulbs and bolts of 43 garlic cultivars. The bulbs of cultivar ‘74-x’ had the highest phenolic content (total phenolic, flavonoids) and the strongest antioxidant capacity (DPPH, FRAP, and CUPRAC), followed by bulbs of cultivar ‘Hanzhong purple’; the bulbs of cultivar ‘Gailiang’ had the lowest phenolic content and antioxidant capacity (FRAP, CUPRAC, MCA). The bolts of ‘Hanzhong purple’ also had higher phenolic content. Principal components analysis (PCA) separated the cultivars into 3 groups according to phenolic and flavonoid contents and strength of antioxidant activity. The first group had higher HRSC, FRAP, and flavonoid content; the second group had higher total phenolic content and MCA; some cultivars in the third group had higher HRSC and FRAP. All 8 test garlic bulb extracts successfully prevented Human Vascular Endothelial Cell death and significantly prevented reactive-oxygen species (ROS) formation in oxidative stress model, in which cultivar ‘74-x’ had highest protection capability, following by cultivar ‘Hanzhong purple’, and the bulbs of cultivar ‘No. 105 from Korea’ had the lower protection capability against cell death and ROS formation. The protection capability in vivo of these garlic cultivars was consistent with their phenolic content and antioxidant capacity.  相似文献
