Electrochemical and nanogravimetric DNA-hybridization biosensors have been developed for sensing single mismatches in the probe-target ssDNA sequences. The voltammetric transduction was achieved by coupling ferrocene moiety to streptavidin linked to biotinylated tDNA. The mass-related frequency transduction was implemented by immobilizing the sensory pDNA on a gold-coated quartz crystal piezoresonators oscillating in the 10 MHz band. The high sensitivity of these sensors enabled us to study DNA damage caused by representative toxicants and environmental pollutants, including Cr(VI) species, common pesticides and herbicides. We have found that the sensor responds rapidly to any damage caused by Cr(VI) species, with more severe DNA damage observed for Cr2O72− and for CrO42− in the presence of H2O2 as compared to CrO42− alone. All herbicides and pesticides examined caused DNA damage or structural alterations leading to the double-helix unwinding. Among these compounds, paraoxon-ethyl and atrazine caused the fastest and most severe damage to DNA. The physico-chemical mechanism of damaging interactions between toxicants and DNA has been proposed. The methodology of testing voltammetric and nanogravimetric DNA-hybridization biosensors developed in this work can be employed as a simple protocol to obtain rapid comparative data concerning DNA damage caused by herbicide, pesticides and other toxic pollutants. The DNA-hybridization biosensor can, therefore, be utilized as a rapid screening device for classifying environmental pollutants and to evaluate DNA damage induced by these compounds.  相似文献

In the 45 years after legislation of the Clean Air Act, there has been tremendous progress in reducing acidic air pollutants in the eastern United States, yet limited evidence exists that cleaner air has improved forest health. Here, we investigate the influence of recent environmental changes on the growth and physiology of red spruce (Picea rubens Sarg.) trees, a key indicator species of forest health, spanning three locations along a 100 km transect in the Central Appalachian Mountains. We incorporated a multiproxy approach using 75‐year tree ring chronologies of basal tree growth, carbon isotope discrimination (?13C, a proxy for leaf gas exchange), and δ15N (a proxy for ecosystem N status) to examine tree and ecosystem level responses to environmental change. Results reveal the two most important factors driving increased tree growth since ca. 1989 are reductions in acidic sulfur pollution and increases in atmospheric CO2, while reductions in pollutant emissions of NOx and warmer springs played smaller, but significant roles. Tree ring ?13C signatures increased significantly since 1989, concurrently with significant declines in tree ring δ15N signatures. These isotope chronologies provide strong evidence that simultaneous changes in C and N cycling, including greater photosynthesis and stomatal conductance of trees and increases in ecosystem N retention, were related to recent increases in red spruce tree growth and are consequential to ecosystem recovery from acidic pollution. Intrinsic water use efficiency (iWUE) of the red spruce trees increased by ~51% across the 75‐year chronology, and was driven by changes in atmospheric CO2 and acid pollution, but iWUE was not linked to recent increases in tree growth. This study documents the complex environmental interactions that have contributed to the recovery of red spruce forest ecosystems from pervasive acidic air pollution beginning in 1989, about 15 years after acidic pollutants started to decline in the United States.  相似文献

目前国内外许多城市面临严重的空气污染问题,严重制约城市发展、影响人体健康。同时,部分城市(如深圳)空气质量已经达到并保持在良好的水平。探究这些城市空气质量演变过程及其调控,可为我国大量仍面临严重空气污染问题的城市提供参考和借鉴。以深圳市为案例,利用环境质量公报数据和统计年鉴数据,通过分析多个社会经济因子与典型污染物的相关关系,探究其空气质量演变特征与调控经验。结果表明,深圳市各类型空气污染物与城市社会经济发展均符合EKC模型假说,但不同污染物所处阶段不同。颗粒物、SO2和NO2均处于下降阶段,而O3目前处于高水平平稳阶段。总结深圳市空气质量改善历程及调控措施,发现主要有两大方面:宏观上严格把控;微观上精准治理。宏观上,重点放在产业结构和能源结构的快速调整上,对于空气质量的改善起到了非常明显的作用。而在微观精细化的管理上,对于空气污染的治理政策要具有持续性、精准性和及时性。当下,针对首要的O3污染问题,深圳市应重点关注城市人为排放VOCs和植物释放VOCs对O3生成的影响,以及城市热环境对O3浓度的影响。  相似文献

Grains of wheat ( Triticum aestivum L. cv. Starke II) were treated with SAN-9789 (Norflurazon), and grown in darkness for 6 days. The SAN treatment resulted in an inhibition of the carotenoid synthesis at the level of phytoene. The plastids of SAN-treated plants contained enlarged and non-osmiophilic plastoglobuli, compared to the plastoglobuli of the control. The plastoglobuli were isolated and purified by means of a flotation technique, and their lipid composition was determined. Efforts were made to avoid contamination of epicuticular soluble lipids in the plastoglobuli suspension during isolation. The most suitable method was found to be a mechanical removal of the lipids from the surface of the intact leaves prior to homogenization. Membrane lipids, i.e. galacto-, phospho- and sulpholipids could not be detected in plastoglobuli from either SAN-treated or control plants, indicating that contamination with membranes was negligible. In plastoglobuli of SAN-treated plants, large amounts of phytoene and, to a lesser extent, phytofluene accumulated. The proportion of triacylglycerols to quinones was lower than in the control. The main lipids in control plastoglobuli were triacylglycerols, plastoquinones and α-tocopherol. The possible function of plastoglobuli in etioplasts is discussed.  相似文献

BACKGROUND: Gastroschisis is a rare congenital defect of the abdominal wall. Its occurrence is noted primarily in the offspring of young mothers who often smoke during pregnancy. The incidence of gastroschisis has been increasing in many countries in recent years. The etiology of gastroschisis is not known. METHODS: Pregnant mice of CD‐1 strain were maintained on 17 and 9% protein diets mixed with deficient, normal, or supplemental zinc levels throughout gestation. The dams in each protein‐zinc diet group were randomly divided in two groups. One group was exposed to air (control) and the other to 500 ppm carbon monoxide (CO) in air, in environmental chambers, from gestation days (GD) 8–18. The dams were sacrificed by carbon dioxide asphyxiation on GD 18, and data on malformations was collected. RESULTS: The rates of fetal mortality and malformations were increased by protein and zinc deficiencies. Carbon monoxide exposure also increased fetal mortality. In the low protein group, the rate of fetal mortality was inversely related to the dietary zinc level, and the rate of fetal malformations was highest in the zinc deficient group. The incidence of gastroschisis in the low protein/zinc deficient/CO exposed group was 47%, and 60% of the litters were affected. The incidence of gastroschisis in the rest of the low protein/zinc diets/air or CO groups was 0. CONCLUSION: The data indicates that gastroschisis is caused by the combination of protein‐zinc deficiencies and carbon monoxide exposure during gestation. The finding may be relevant to human populations that experience protein and zinc deficiencies during gestation, and are exposed to CO pollution, or cigarette, or marijuana smoke during pregnancy. Exposure to air pollution has been clearly associated with a range of adverse health effects, including reproductive toxicity. However, a limited amount of research has been conducted to examine the association between air pollution and male reproductive outcomes, specially semen quality. We performed a systematic review (up to March 2017) to assess the impact of environmental and occupational exposure to air pollution on semen quality. Epidemiological studies focusing on air pollution exposures and male reproduction were identified by a search of the PUBMED, MEDLINE, EBSCO and TOXNET literature bases. Twenty-two studies were included which assess the impact of air pollutants (PM2.5, PM10, SO2, NOx, O3, PAHs) on main semen parameters (sperm concentration, motility, morphology), CASA parameters, DNA fragmentation, sperm aneuploidy and the level of reproductive hormones. The number of studies found significant results supporting the evidence that air pollution may affect: DNA fragmentation, morphology and motility.In summary, most studies concluded that outdoor air pollution affects at least one of the assessed semen parameters. However the diversity of air pollutants and semen parameters presented in the studies included in the review and different study design caused lack of consistency in results and difficulties in comparison.  相似文献

Malformations in fish embryos have been monitored for several years in the southern part of the North Sea. Their occurrence was thought to be related to pollution because malformation rates were highest in coastal waters, known to receive high pollution loads. For the embryos of all species synchronous trends for the fluctuation of the occurrence of malformations over time were registered in the areas covered, with intermediate prevalences at the beginning of the studies in 1984 and maxima in 1987. Thereafter,malformations in all species decreased significantly,followed by an abrupt increase in 1996. It was found that in addition to a close correlation between the concentrations of environmental pollutants in the water and in the fish a significant negative correlation existed between surface water temperature and the occurrence of malformed embryos of dab (Limanda limanda). These same correlations were also observed for other fish species over time and space. This temperature-related correlation became increasingly visible with decreasing concentrations of organochlorines in fish. From these findings it is concluded that aside from environmental pollutants,natural factors such as temperature may predispose developing fish embryos to the impact of pollutants. Needles of four spruce trees showing different degrees of novel kinds of forest decline were investigated by electron microscopy. Green needles appearing at least superficially still intact were selected for the present investigation. Most of the mesophyll appeared to be undamaged. However, groups of atypical mesophyll cells were found close to the endodermis or the hypodermis. The chloroplasts of the apparently damaged cells were particularly affected. Changes in the matrix of the chloroplasts, i.e,. increased affinity to osmium, occurrence of extensive nests of plastoglobuli, as well as damage to the membranes, i.e. lesions in the envelope and abnormal thylakoid membranes, were observed. Signs of decomposition of other cellular structures including mitochondria were also detectable. There appeared to be a close correlation between the degree of damage at the whole tree level and the degree of damage occurring at the cellular level. It is concluded that particularly the lipids and the proteinsof, the membranes are affected by anthropogenic air pollutants and natural stressors. The altered membrane structure may for instance cause abnormal osmotic conditions for the cellular compartments and may impair transport processes and thus lead to lossof function not only of the cells but also of the whole needle.  相似文献

With the economic development of China, air pollutants are also growing rapidly in recent decades, especially in big cities of the country. To understand the relationship between economic condition and air pollutants in big cities, we analysed the socioeconomic indictorssuch as Gross Regional Product per capita (GRP per capita), the concentration of air pollutants (PM10, SO2, NO2) and the air pollution index (API) from 2003 to 2012 in 31 provincial capitals of mainland China. The three main industries had a quadratic correlation with NO2, but a negative relationship with PM10 and SO2. The concentration of air pollutants per ten thousand yuan decreased with the multiplying of GRP in the provinical cities. The concentration of air pollutants and API in the provincial capital cities showed a declining trend or inverted-U trend with the rise of GRP per capita, which provided a strong evidence for the Environmental Kuznets Curve (EKC), that the environmental quality first declines, then improves, with the income growth. The results of this research improved our understanding of the alteration of atmospheric quality with the increase of social economy and demonstrated the feasibility of sustainable development for China.  相似文献

空气中的细颗粒物(PM2.5)是我国城市空气污染的主要污染物之一,严重威胁着城市居民的健康,限制城市发展的可持续性。PM2.5去除的自然途径有两种,分别是干沉降和湿沉降,其中干沉降占据主导作用,且干沉降的过程和效率与城市森林紧密关联。目前针对城市森林对干沉降作用的研究主要是在小尺度中从不同树种、不同群落结构、不同景观类型等角度来估算并比较其滞尘量,较少关注其占空气污染总量的比率,从而可能影响对城市森林滞尘服务能力的判断。因此,利用城市森林效益(Urban Forest Effect, UFORE)模型中的大气污染干沉降模块的核心算法,以2015年为例,估算了我国主要城市辖区的城市森林一年内对大气中的PM2.5削减量以及其占空气中PM2.5污染总量的比重。结果显示:(1)2015年全国主要城市单位绿地面积日均滞尘量较高地区主要集中在华北地区、华东地区、以及东北地区。其中北京30.47mg/m2,苏州24.63mg/m2,沈阳28.55mg/m2  相似文献

The use of passive air samplers (PAS) in recent years to monitor the occurrence of persistent organic pollutants (POPs) in air has been increasing, not only globally, but also at regional and/or local scales. The present study was aimed at analyzing the airborne concentrations of the POPs: polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), and polychlorinated naphthalenes (PCNs) in the vicinity of a municipal solid waste incinerator (MSWI) in Tarragona (Catalonia, Spain). Mean air levels of PCBs, PBDEs, and PCNs were 44.1, 65.0, and 6.65 pg/m3, respectively. No differences were noted according to the distance or direction from the plant for any chemical. Furthermore, this information was used to estimate the air inhalation of POPs for the local population. Environmental exposure to POPs through air accounted for only <2% of the total when compared with the dietary intake. The overall results indicate that the MSWI here assessed does not mean a relevant increase on human health risks derived from inhalation of POPs. PAS can also be utilized for human health risk assessment studies, being also suitable tools to control the environmental burden of non-regulated chemicals, for which no emission thresholds have yet been derived.  相似文献

One of the most dangerous inhaled pollutants is particulate matter (PM). PM in inhaled air have a negative impact on human wellbeing and health, and may even cause deaths. Where pollutants have been emitted into the outdoor atmosphere, the only possible method for cleaning the air is through phytoremediation, a form of environmental biotechnology, where plants act as biological filters for pollutants. This study compared PM levels on the leaves of Tilia cordata Mill. trees growing in locations at increasing distances from the source of the PM emission. Significant differences between individual trees growing at a distance of between 3 m and 500 m from the road edge were found in: (i) the mass of PM that accumulated on leaves (total, surface, in-wax and the three determined size fractions) and (ii) the amount of waxes deposited on leaves.  相似文献

目的探讨大气污染物对呼吸道的病理变化,为预防大气污染对人体健康的损害提供理论依据。方法采用模拟大气污染方法,用大鼠制备大气污染物模型,对大鼠气管、肺进行透射电镜及气管扫描电镜检查。结果大气污染大鼠动物模型的气管、肺与对照组相比,实验组气管及肺泡上皮有渐进性损伤,且随量的蓄积损伤逐渐加重,最终发生肺实变。结论吸入的污染粉尘可造成气管及肺泡上皮的渐进性损伤,形态学变化与吸入粉尘的量呈一致性,并随吸人量的累积而逐渐变得严重,最终变为肺实变,残留肺组织为代偿性气肿。  相似文献

After a 5 d cultivation in solutions with different concentrations of cadmium, changes in chloroplasts of the leaf and stem parenchyma from Myriophyllum spicatum L. were observed: swelling and merging of the grana thylakoids in various degrees, coagulation of the stroma as well as the formation of crystal-like structures in it, accompanied by changes in the plastoglobuli and starch. Concentrations of 3.5 and 6.0 g(Cd2+) m-3 caused a vast destruction of the whole plastid apparatus. Chloroplasts in the parenchyma of stems showed a reduction in the inner membrane system, and an increased number of plastoglobuli. A comparative analysis that we carried out indicated that the changes were not uniform and metal-specific.  相似文献

The Iraí Reservoir, a water supply in Brazil, is constantly impacted by anthropogenic activities such as waste inputs from agriculture, hospitals and urbanization, resulting toxic cyanobacterial blooms causing economic, social and environmental problems. This study assessed the concentration of some common contaminants of the Iraí Reservoir, namely paracetamol, diclofenac and microcystin-LR and tested whether a laboratory scale Green Liver System® would serve as a suitable technology to remove these contaminants. Further, the study investigated whether the pollutants caused adverse effects to the macrophytes using catalase as a biomarker for oxidative stress and investigated whether biotransformation (glutathione S-transferase) was a main route for detoxification. Egeria densa, Ceratophyllum demersum and Myriophyllum aquaticum were exposed to a mixture of the three contaminants for 14?days in a concentration range similar to those detected in the reservoir. The plants removed 93% of diclofenac and 100% of MC-LR after 14?days. Paracetamol could not be detected. Catalase and glutathione S-transferase enzyme activities remained unaltered after the 14-day exposure, indicating that the mixture did not cause oxidative stress. The study showed that the aquatic macrophytes used are suitable tools to apply in a Green Liver System® for the remediation of mixed pollutants.  相似文献

HSPA1A (HSP70-1) is a highly inducible heat shock gene up-regulated in response to environmental stresses and pollutants. The aim of our study was to evaluate the sensitivity of the stable metabolically competent HepG2 cells containing a human HSPA1A promoter-driven luciferase reporter (HepG2-luciferase cells) for assessing the toxicity of organic pollutants present in air. The HepG2-luciferase cells were validated by heat shock treatment and testing three organic compounds (pyrene, benzo[a]pyrene, and formaldehyde) that are ubiquitous in the air. The maximal level of HSPA1A (HSP70-1) and relative luciferase activity induced by heat shock were over three and nine times the control level, respectively. Pyrene, benzo[a]pyrene, and formaldehyde all induced significantly elevated levels of relative luciferase activity in a dose-dependent manner. Extractable organic matter (EOM) from urban traffic and coke oven emissions in ambient air were tested on the HepG2-luciferase cells. The traffic EOM induced significant increase in relative luciferase activity at concentrations of picogram per liter. The coke oven EOM produced a strong dose-dependent induction of relative luciferase activity up to six times the control value. Significant increases in relative luciferase activity were observed at concentrations that were as low, or lower than the concentrations that the tested organic pollutants decreased cell viability, and increased malondialdehyde concentration, Olive tail moment, and micronuclei frequency. Therefore, we conclude that the HepG2-luciferase cells are a valuable tool for rapid screening of the overall toxicity of organic pollutants present in air.  相似文献

Among the seven plant bioassays reviewed by the US Environmental Protection Agency (EPA) Gene-Tox program in 1980, the Allium/Vicia root tip chromosome aberration assay and the Tradescantia stamen hair mutation and micronucleus assays were adopted by the International Program on Plant Bioassays (IPPB) for monitoring or testing environmental pollutants. These assays are highly sensitive and capable of detecting mutagens, clastogens and carcinogens from the environment. They are effective in situ monitors. These bioassays were validated and their protocols were standardized through a program under the International Program on Chemical Safety (IPPB), the precursor of the IPPB program which currently is in operation under the auspices of the United Nations Environment Program (UNEP). Six different categories of environmental studies, ranging from wastewater, surface or ground water, soil samples, ambient air, pesticides, and radiation, were carried out and are reported in this special issue. The mission of the IPPB/UNEP is to use these bioassays to monitor or test environmental mutagens and clastogens in the air, water, and soil to safeguard the quality of these essential elements in life and to use these simple and clear indicators of pollution damage as the tool for environmental education for the general public. The published reports of the monitoring and testing results will establish the database for environmental conditions in a number of locations around the world. An international monitoring network for the detection of genotoxicity of environmental pollutants is being established under the auspices of UNEP to protect the global environment.  相似文献

Phytoremediative Capacity of Plants Enriched with Melatonin   总被引:1,自引:0,他引:1  
Melatonin is an environmentally friendly-molecule with broad spectrum antioxidant capacity. Melatonin is widely present in the plant kingdom. High levels of melatonin exist in an aquatic plant, the water hyacinth, which is highly tolerant of environmental pollutants. Elevated levels of melatonin probably help plants to protect against environmental stress caused by water and soil pollutants. To investigate the potential relationships between melatonin supplementation and environmental tolerance in plants, pea plants were treated with high levels of copper in the soil. The results show that copper contamination kills pea plants; however, melatonin added to the soil significantly enhanced their tolerance to the copper contamination and, therefore, increased their survival. Based on the theory and these preliminary data, we speculate that melatonin could be used to improve the phytoremediative efficiency of plants against different pollutants. Since melatonin is safe to animals and humans as well as being inexpensive, it may be a feasible and cost-effective approach to clean environmental contaminations.Key Words: phytoremediation, plants, melatonin, antioxidant  相似文献

Scope  In this study, a dynamic model was built in which LCA and PBM were integrated to quantitatively assess the total environmental impacts induced by the product population in a society over time. Specifically, a determination was carried out concerning how Japan’s air conditioner population is used (lifetime distribution, number of units, etc.) and an assessment was made concerning the Global Warming Potential (GWP) associated with the air conditioner population. Methods  The proposed dynamic model was applied to air conditioners for analyzing the total GWP caused by the air conditioner population in Japan from 1990 to 2010. To create a trend forecast model for future environmental load, scenarios for air conditioner production up to 2010 were formulated and the total GWP from the air conditioner population was predicted. Conducted also were sensitivity analyses whose parameters were air conditioner performance, lifetime and the rate of refrigerant recovery when retired units are processed. Results and Discussion  Applying the PBM to the air conditioner population in 2000, it was found that 81.5 million units consumed 5.94 x 10p10 kWh in that year, which was a 6.1 % increase in the total annual power consumption in 1990. In both a stationary scenario and a steady growth (1.5% annual increase), it was found that the total GWP would be 27.7% higher than in 1990 under the stationary scenario and 37.8% higher under the steady growth scenario. The improvements in air conditioner performance will have a small effect on reducing the total GWP from that population. Furthermore, in connection with the average lifetime, it was found that the GWP, due to refrigerant releases when units are disposed of, would be relatively large in 2000 and the following years. Conclusions  Thus, shorter product lifetimes will spur a replacement of air conditioners with new units, a situation that will only lead to the reduction of GWP if the recovery rate of refrigerant is to be achieved to more than 50% under the stationary scenario. Recommendations and Outlook  To meet COP3 targets for Japan in 2010 (i.e. to reach the same level as in 1990 for household appliances), our study shows that it will be vital to raise the refrigerant recovery rate. If the number of air conditioners in use remains unchanged, recovery would have to be 45.7%, but under the steady growth scenario it would have to be at least 60.4%. Therefore, it will be difficult to meet COP3 targets unless the refrigerant recovery rate is strongly increased. This method is applicable to assess not only the GWP of air conditioners, but also other environmental impacts caused by a variety of product populations, which will be quite effective for setting targets of products’ performance, policymaking, etc.  相似文献
