Willows (e.g. Salix caprea L.) are deciduous and richly branched shrubs or small trees. Salix caprea shows a high adaptability to different habitat conditions. One way of evaluating this adaptability is to measure willow biomass and production. Young plants of S. caprea were sampled from the bottom of an artificial lagoon in which sediments removed from the local Vajgar fishpond were deposited. The bottom of the lagoon was overgrown by vegetation dominated by seedlings of the willows S. caprea and S. aurita. Willows grew in the lagoon at average density of 58 plants per m2. The biomass production and growth of S. caprea were determined for 15 samples (collected from 315 individuals) during the growing season. Annual net dry matter production in the whole community was estimated for 2.7 kg m−2. Willows are generally considered to be fast-growing plants. The highest RGR of willows recorded by us was 0.03 to 0.04 g g−1 d−1 both in the stems and roots. This value was often recorded from July to August.  相似文献

Abstract. Broadleaved forest communities degenerated through strong pressure from large herbivores. Relief of this pressure led to regeneration, in particular of Salix caprea and other light-seeded pioneer trees: Populus tremula, Betula pendula and B. pubescens. This regeneration proceeded following conservation protection of degenerate stands in a nature reserve and later in Biaowiéza National Park. The emergence and development of the Salix caprea population proceeded following the expansion of Picea abies, which coincided with the period of enhanced animal pressure on the broadleaved forest. Salix caprea filled all the gaps in the tree stand after the destruction of trees and undergrowth by herbivores (in the years 1892–1915). The species also appeared abundantly in old, at the time unforested, clearings and felled areas. Here, S. caprea developed large populations with certain trees in good condition, with a growth form typical of forest trees and attaining considerable heights. The majority of trees were 50 - 60 yr old at the time of death, although some individuals reached 74 yr of age. The process of extinction of the Salix population — observed over 19 yr on permanent plots with marked trees — proceeded very quickly, especially in the first decade of observation. It led to the almost complete disappearance of S. caprea from the forest communities of Biaowiéza National Park. The death of individual trees is preceded by impairment of their health and reduced annual increments in the last 4–9 years of their life. The development of populations of permanent constituents of the forest, notably Carpinus betulus, Tilia cordata, Acer platanoides and Ulmus glabra, under the canopy of light-seeded trees, and the absence of a new generation of pioneer trees points to the end of the process of regeneration in the forest communities of BiaHwiéza National Park.  相似文献

Summary Samples of 17 plant species (one herb, dwarf shrubs and trees) were collected monthly throughout the winter 1976/77. Samples were frozen down to selected temperatures and hardiness was estimated by visual observations of oxidative browning and bud break. In most species hardiness rapidly increased from September to November and dehardening started in February–March. In six species (Betula pubescens Ehrh., Picea abies (L.) Karst., Salix caprea L., Sorbus aucuparia L., Vaccinium myrtillus L., V. vitis-idaea L.) hardening developed faster at the continental than at the coastal location. Differences in maximum hardiness between sampling locations were generally small and there were no real differences in dehardening of the plants between the locations. Both twigs and buds of Salix pentandra L. and Populus tremula L. survived liquid nitrogen in January–February.  相似文献

Floral scents of male and female inflorescences of three dioeciousSalix species were collected by head-space adsorption, and analysed by GC-MS. InSalix caprea andS. cinerea 1,4-dimethoxy benzene was the main compound, and male and female scents showed a high degree of resemblance. No dominant compound was found inS. repens and malefemale scent similarity was low. Floral scent inSalix is likely a strong orientation cue, guiding pollinators between male and female plants ensuring pollen transfer and pollination. We suggest that a high degree of male-female floral scent resemblance is coupled to a high degree of insect pollination. Floral scent does not promote reproductive isolation betweenS. caprea andS. cinerea.  相似文献

Phytoextraction can be a cost-efficient method for the remediation of contaminated soils. Using species mixtures instead of monocultures might improve this procedure. In a species mixture, an effect of heavy metals on the species' performance can be modified by the presence of a co-occuring species. We hypothesised that (a) a co-occuring species can change the effect of heavy metals on a target species, and (b) heavy metal application may modifiy the competitive behaviour between the plants. We investigated these mechanisms in a greenhouse experiment using three species to serve as a model system (Carex flava, Centaurea angustifolia and Salix caprea). The species were established in pots of monocultures and mixtures, which were exposed to increasing concentrations of Ni and Zn, ranging from 0 to 2,500 mg/kg. Increased heavy metal application reduced the species' relative growth rate (RGR); the RGR reduction being generally correlated with Ni and Zn concentrations in plant tissue. S. caprea was an exception in that it showed considerable Zn uptake but only moderate growth reduction. In two out of six cases, competitors significantly modified the influence of heavy metals on a target species. The interaction can be explained by an increased uptake of Zn by one species (in this case S. caprea) that reduced the negative heavy metal effect on a target species (C. flava). In two further cases, increasing heavy metal application also altered competitive effects between the species. The mechanisms demonstrated in this experiment could be of relevance for the phytoextraction of heavy metals. The total uptake of metals might be maximised in specific mixtures, making phytoextraction more efficient.  相似文献

In our experiments the effect of different plants on microbial activities resulting in degradation and PCB removal from long-term contaminated soil was evaluated. Total bacteria and bacteria representing the dominating microflora within rhizosphere of individual plant species – tobacco (Nicotiana tabacum), black nightshade (Solanum nigrum), horseradish (Armoracia rusticana) and goat willow (Salix caprea) planted in PCB contaminated soil as well as from the same, but non-vegetated PCBs soil, were isolated and biochemically characterized. PCB bacterial degraders, stimulated by root exudates of individual plants, were detected after isolation from rhizosphere soil and precultivation on minimal medium with biphenyl as the sole carbon source. Detection of BphA1 gene (first gene of bacterial aerobic PCB degradative pathway) in genomes of rhizosphere microorganisms was performed by nested PCR technique using previously designed specific primers. Rhizosphere of individual plants contained different bacterial species, mostly gram-negative non-fermenting bacteria of Pseudomonas, Agrobacterium, Ochrobactrum and other species. Gene BphA1 was identified in genome of only several of them. From tested species, S. caprea and A. rusticana have shown to be promising candidates for rhizoremediation purposes.  相似文献

The influence of a new micrografting method in vitro was tested by using long shoot meristems of adult 140 year old European larch trees grafted onto larch seedling rootstocks in sterile peat pellets. Five months after grafting and transfer of grafts to ex vitro conditions, new shoots from sprouting scions were re-established into tissue culture. The propagation behavior of shoots derived from these explants was compared with shoots established via bud culture from the same donor trees. Shoot explants from micrografts multiplied more rapidly than shoots explants from the original adult donor trees. Rooting experiments with cuttings from six micrografted clones resulted in 49.9 ± 11.9% rooting. Cuttings from donor trees invariably showed no root formation. The results confirm a rejuvenating influence of micrografting in larch.  相似文献

A simple micropropagation method is reported for Primula scotica, a rare plant, endemic to the North of Scotland. The technique involves the clonal proliferation of seed-derived plantlets on either hormone free tissue culture medium or on medium containing benzyl amino purine and indole acetic acid. Average multiplication rates of 4–6 were obtained for plants grown on plant growth regulator supplemented medium. The micropropagation method was applied to four different clones of P. scotica and clonal differences were observed in relation to media type. Some plants from certain clones did display hyperhydricity, however, this was circumvented by using frequent sub-culturing intervals and transferring the plants to hormone-free medium. Plantlets rooted on both media types and displayed normal, true-to-type rosette morphology. The cultures did not callus and development proceeded via shoot and root production only. The in vitro-grown plants could be transferred to ex vitro conditions and a range of growth substrates were assessed for their efficacy in supporting ex vitro growth, with a view to developing longer-term strategies for the transfer and reintroduction of micropropagated P. scotica plants into natural habitats. The simple method described in this paper may offer the potential of being applied to other endangered Primula spp.  相似文献

Summary A transformation system that allows regeneration of transgenic pea plants from calli selected for antibiotic resistance was developed. Explants from axenic shoot cultures and seedling epicotyls were cocultivated with nononcogenic Agrobacterium tumefaciens strains, and transformed callus could be selected on callus-inducing media containing either 15 mg/l hygromycin or 75 mg/l kanamycin. After several passages on regeneration medium, shoot organogenesis could be reproducibly induced on hygromycin-resistant calli, but not on the calli selected for kanamycin resistance. Regenerated shoots could subsequently be rooted and transferred into the greenhouse. In addition, the effects of different callus-inducing and growth media on organogenesis were investigated. The transformation of the calli and regenerated plants was confirmed by DNA analysis.  相似文献

Melampsora willow rusts are the most important fungal pathogens in short rotation coppices of biomass willows. In the past, breeding programmes for rust resistant biomass willows concentrated on the distinction of races within the forma specialis Melampsora larici-epitea f. sp. larici-epitea typica that colonized Salix viminalis and related clones. In a new breeding program that is based on a wider range of willow species it is necessary to identify further Melampsora species and formae specialis that are pathogens of willow species other than S. viminalis. Therefore, three stock collections with Salix daphnoides, Salix purpurea, and other shrub willow species (including S. viminalis) species were sampled in north-eastern Germany. A fourth stock collection in central Germany contributed rusts of tree willows (Salix fragilis and Salix alba) and the large shrub Salix caprea. Out of 156 rust samples, 149 were successfully sequenced for ITS rDNA. A phylogenetic analysis combining Neighbour-Joining, Maximum-Likelihood and Bayesian analysis revealed six species: Melampsora ribesii-purpureae, Melampsora allii-salicis-albae, Melampsora sp. aff. allii-fragilis, Melampsora larici-pentandrae, Melampsora larici-caprearum, and Melampsora larici-epitea. The first four species were found exclusively on the expected hosts. Melampsora larici-caprearum had a wider host range comprising S. caprea and S. viminalis hybrids. Melampsora larici-epitea can be further differentiated into two formae speciales. The forma specialis larici-epitea typica (59 samples) colonized Salix viminalis clones, Salix purpurea, Salix×dasyclados, and Salix×aquatica. In contrast to this relatively broad host range, f. sp. larici-daphnoides (65 samples) was found exclusively on Salix daphnoides. With the distinction and identification of the rust species/formae speciales it is now possible to test for race-specific resistances in a more targeted manner within the determined pairings of rust and willow species.  相似文献

Coffee leaf rust due to Hemileia vastatrix is one of the most serious diseases in Arabica coffee (Coffea arabica). A resistance gene (SH3) has been transferred from C. liberica into C. arabica. The present work aimed at developing sequence-characterized genetic markers for leaf rust resistance. Linkage between markers and leaf rust resistance was tested by analysing two segregating populations, one F2 population of 101 individuals and one backcross (BC2) population of 43 individuals, derived from a cross between a susceptible and a SH3-introgressed resistant genotype. A total of ten sequence-characterized genetic markers closely associated with the SH3 leaf rust resistance gene were generated. These included simple sequence repeats (SSR) markers, sequence-characterised amplified regions (SCAR) markers resulting from the conversion of amplified fragment length polymorphism (AFLP) markers previously identified and SCAR markers derived from end-sequences of bacterial artificial chromosome (BAC) clones. Those BAC clones were identified by screening of C. arabica genomic BAC library using a cloned AFLP-marker as probe. The markers we developed are easy and inexpensive to run, requiring one PCR step followed by gel separation. While three markers were linked in repulsion with the SH3 gene, seven markers were clustered in coupling around the SH3 gene. Notably, two markers appeared to co-segregate perfectly with the SH3 gene in the two plant populations analyzed. These markers are suitable for marker-assisted selection for leaf rust resistance and to facilitate pyramiding of the SH3 gene with other leaf rust resistance genes.  相似文献

In order to genotype hybrid genomes of distant asymmetric somatic hybrids, we synthesized hybrid calli and plants via PEG-mediated protoplast fusion between recipient tall fescue (Festuca. arundinacea Schreb.) and donor wheat (Triticum aestivum L.). Seventeen and 25 putative hybrid clones were produced from the fusion combinations I and II, each with the donor wheat protoplast treated by UV light for 30 s and 1 min, respectively. Isozyme and RAPD profiles confirmed that ten hybrid clones were obtained from combination I and 19 from combination II. Out of the 29 hybrids, 12 regenerated hybrid plants with tall fescue phenotype. Composition and methylation-variation of the nuclear and cytoplasmic genomes of some hybrids, either with or without regenerative ability, were compared by genomic in situ hybridization, restriction fragment length polymorphism, and DNA methylation-sensitive amplification polymorphism. Our results indicated that these selected hybrids all contained introgressed nuclear and cytoplasmic DNA as well as obvious methylation variations compared to both parents. However, there were no differences either in nuclear/cytoplasmic DNA or methylation degree between the regenerable and non-regenerable hybrid clones. We conclude that both regeneration complementation and genetic material balance are crucial for hybrid plant regeneration.  相似文献

杨柳科树木是我国华北区域植树造林的主要树种,但春季的杨柳飞絮问题既影响区域大气环境质量,也对人体健康造成一定的危害,因此准确预报杨柳科树木的春季物候期具有重要现实意义。基于中国气象局农业气象观测网和中国科学院地理科学与资源研究所中国物候观测网提供的华北区域1963—2018年59个站点的4种杨柳科树木(垂柳、旱柳、加拿大杨、毛白杨)的4个春季物候期(开花始期、盛期、末期和果实或种子成熟期)观测数据,对3种基于逐日气温序列构建的春季物候过程模型(简单积温模型、温度三基点模型和冷激-三基点模型)进行了参数率定和优选,分别对这16个基于物种-物候期的最优春季物候模型进行了外部检验。内部模拟结果表明,各模型对同一物种-物候期模拟效果相差不大,均方根误差(RMSE)介于5.5—11.6d,各模型对同一物种发生较早的物候期模拟效果相对更好。通过赤池信息准则(AIC)判定,上述16个基于物种-物候期的最优春季物候模型中,有11个选择简单积温模型,有5个选择冷激-三基点模型,其中,垂柳和旱柳的最优春季物候模型都选择了简单积温模型,加拿大杨的各春季物候期对2种物候模型均有选择,毛白杨的各春季物候期则全部...  相似文献

Asymbiotic seedling propagation and introduction of seedlings into a natural habitat were achieved for Cephalanthera falcata. For immature seeds collected 65 days after pollination, high germination rate (av. 50%) was achieved on Hyponex agar medium plates. Root development occurred in about 10% of the protocorms 5 months after seed sowing. Rooted protocorms were transferred to a culture bottle containing 100 ml of the Hyponex agar medium and incubated continually. In about 30% of the transferred individuals, shoot height reached 1.5–2 cm 8 months after the transfer. After acclimatization in wet vermiculite at 4°C for 6 months, 135 individuals were planted in a natural stand of C. falcata in mid February 2001. Shoot appearance rate was 44.4% at the first year and flowering was noted in some plants. At the fifth year, shoots with an average height of 21.6 cm still appeared in four plants, and flowering was noted in three of them. Colonization of mycorrhizal fungi was examined in two of them as well as one wild plant, in which the mycorrhizal fungi were identified to be in Thelephoraceae or Russulaceae. These fungi are known to form ectomycorrhiza with trees, and thus a tripartnership symbiosis consisting of C. falcata, mycorrhizal fungi and trees was suggested. The involvement of ectomycorrhizal fungi might be the reason for the low survival rate in the field experiment, because the distribution of ectomycorrhizal fungi relevant to this orchid is assumed to be uneven. The possibility of introducing artificially propagated orchids into natural habitats was discussed.  相似文献

The relationship between shoot hydraulic conductance (L) and stomatal sensitivity to changes in leaf water status was studied in the saplings of six deciduous tree species. L increased significantly in sequence: Acer platanoides < Tilia Cordata < Padus avium = Quercus robur < Salix caprea = Populus tremula. L was higher in the trees grown in soil with a higher nitrogen content and lower in the trees grown under mild water stress or kept in darkness for several days. L was higher in July than in September in all the species. L correlated positively with maximum photosynthesis, stomatal conductance and stomatal sensitivity to an increase in leaf water potential, but negatively with stomatal sensitivity to a decrease in leaf water potential. The correlations between L and any other parameter were approximated by three different curves: data for water-stressed plants fit to the first, data for plants kept in darkness fit to the second and all the other data fit to the third curve. The reasons of the differences of shoot hydraulic conductance in the different experimental sets and the mechanisms which may cause the correlation between L and the other characteristics are discussed.  相似文献

Summary We describe an in vitro propagation protocol for Zingiber petiolatum (Holttum), I. Theilade, a rare species from the southern part of Thailand. Fruits were surface-sterilized and seeds germinated on Murashige and Skoog medium (MS) medium supplemented with 3% sucrose. Three-month-old seedlings were used as initial plant material for in vitro propagation. Terminal buds of the plants were inoculated on MS medium containing 6-benzylaminopurine (BA; 2.2–35.5 μM) alone or in combination with 1-naphthaleneacetic acid (0.5 μM). Eight weeks after inoculation, the cultures were transferred to MS medium without plant growth regulators for 4wk. The cultures transferred from MS medium with 17.8 μM BA revealed the highest shoot induction rate of 6.1±0.7 shoots per explant. Rooting was spontaneously achieved in MS medium without plant growth regulators. Rooted plants were successfully transplanted to soil.  相似文献

The aim of this study was to assess the responses of herbivores and pathogens to hybrid plants under controlled conditions. F1 hybrids and parental species, produced by hand-pollinating willows in the field, were potted and kept in an experimental field under controlled conditions. In 1997, plant growth and survival were measured along with densities of insects and the degree of pathogen infection on the willows. The survival rate was higher for S. repens than for the hybrids and lowest for S. caprea. Densities of the sawflies Pontania pedunculi and P. brigmanii and the leaf-galling midge Iteomyia capreae were higher on hybrids and on S. caprea than on S. repens. The densities of Crepidodera fulvicornis (Chrysomelidae), chrysomelid larvae and the bud-galling midge Dasineura rosaria did not differ between any of the plant categories. Hybrids were more severely infected by rust (Melampsora sp.) than S. caprea and the totally resistant S. repens. Densities of herbivores on hybrid willows were consistent with the dominance hypothesis (i.e. herbivore densities were similar to densities on one of the parental species) or supported the no-difference hypothesis. Furthermore, herbivore densities on hybrid plants were most similar to densities on the more susceptible parent. The breakdown in rust resistance in hybrid plants suggests that resistance traits are severely disrupted by the genetic re-arrangement in hybrids and that this increased susceptibility could select against hybridisation. Alternate host plants of cereal rust fungi are necessary for studying the rust sexual cycle and pathogenicity. These plants are usually difficult to propagate through cloning, while seed-propagated plants may have variable responses to the pathogen. To overcome these obstacles, tissue culture, under controlled and aseptic conditions, was utilized for clonal propagation and in vitro inoculation of the following species: Rhamnus palaestinus Boiss., the alternate host of oat (Avena spp.) crown rust (Puccinia coronata Corda); Thalictrum speciosissimum L., the alternate host of brown leaf rust of wheat (Puccinia recondita f. sp. tritici Eriks. & Henn.); and Lycopsis arvensis L., the alternate host of rye (Secala spp.) leaf rust (Puccinia recondita f. sp. recondita Rob. & Desm.). Shoot culture procedures for initial establishment and proliferation were developed for all three alternate host species. Shoot cultures were multiplied at rates ranging from 0.3 to 1.7 shoots/week. Successful infection following inoculation with teliospores of the corresponding rust fungi was obtained for R. palaestinus and T. speciosissimum but not for L. arvensis. The hardening and acclimatization efficiency of rooted T. speciosissimum and L. arvensis was of 80–90%. The propagation efficiency for R. palaestinus was not successful because of the low rate and poor quality of its rooting. It is concluded that the in vitro system might be used as an alternative method for inoculation and multiplication of alternate hosts of cereal rusts, although more experimentation is needed to define accurately the appropriate conditions for the proper infection response. Three clones of enset (Ensete ventricosum Welw. Cheesman) from southwestern Ethiopia (Keffa-Shaka zone) were investigated for their potential for micropropagation and regeneration in tissue culture. Corm and leaf tissue of greenhouse-grown plants as well as in vitro germinated zygotic embryos were used as starting material for micro-propagation and regeneration studies. Embryos were excised from disinfected seeds and cultured in vitro. Multiple shoots from both corm- and embryo-explants were obtained using regeneration medium supplemented with 10 μM or 20 μM BAP. Rooting of shoots was achieved using medium with 5 μM IBA, 1 μM BAP and 1 g l−1 activated charcoal. Plantlets obtained by this process were transferred to soil under greenhouse conditions. Optimal conditions, which were determined for clonal propagation of three different genotypes of enset, allowing both in vitro micropropagation and regeneration, are described. This protocol makes for conservation of enset clones, rapid propagation of selected disease-free germplasm and more efficient breeding procedures.
