促卵泡刺激素对小鼠卵巢玻璃化冻存的抗凋亡作用   总被引:1,自引:0,他引:1       下载免费PDF全文
目的:观察小鼠卵巢组织促卵泡刺激素(FSH)在小鼠卵巢组织玻璃化冻存过程中的抗凋亡作用。方法:将4周龄C57BL/6J雌性小鼠卵巢组织,分为新鲜对照组(CG)、玻璃化冻存对照组(VCG)、0.3 IU/ml FSH全程干预玻璃化冻存组(OG-FSH),玻璃化冻存条件为小鼠卵巢入培养液在37℃二氧化碳培养箱培养60 min,接着入1.5 mol/L乙二醇中预渗透平衡8 min,再移入EG5.5-30玻璃化液渗透平衡3.5 min,之后投入-196℃的液氮罐中保存1 d,全过程在22~24℃下进行。每组20枚卵巢,10枚进行HE染色后的正常卵泡百分比计数,10枚进行免疫组织化学法观察。结果:与对照组(CG)比较,玻璃化冻存对照组(VCG)正常卵泡百分比显著降低,active caspase-3的表达量显著升高(P<0.05);与玻璃化冻存对照组(VCG)比较,0.3 IU/ml FSH全程干预玻璃化冻存组(OG-FSH)组的正常卵泡百分比明显升高,active caspase-3的表达量显著降低(P<0.05)。结论:小鼠卵巢玻璃化冻存全程添加FSH干预可有效抵抗凋亡执行蛋白active caspase-3的表达,有利于卵巢组织形态结构的保护。  相似文献

年龄增长、疾病以及抗肿瘤治疗等均可能会造成女性生育力的下降甚至丧失。随着诊疗技术的发展,癌症死亡率逐渐下降,因此患者的生育力保存需求较前增加。胚胎冻存、卵母细胞冻存和卵巢组织冻存是目前常用的女性生育力保存方式。卵巢组织冻存与移植至今仅有20余年历史,现已成为临床上具备医学指征的生育力保存方式,有助于重建患者的内分泌和生育功能,已有超200多例患者通过此技术成功完成生育。为提高冻存卵巢组织再移植的安全性,现有研究将卵母细胞体外成熟技术、人工卵巢技术与之相结合,避免携癌风险高的冻存卵巢组织移植后的癌症复发风险。较多的研究均已证实了卵巢组织冻存与移植技术的有效性和安全性。尽管卵巢组织冻存与移植技术得到了快速发展,但是该技术仍面临如何通过减少卵泡储备损失进而延长卵巢组织移植后有效性等挑战,尚需要更多的基础和临床研究促进卵巢组织冻存和移植技术的进步与发展。  相似文献

目的观察去势大鼠经新鲜、冷冻卵巢移植后体内激素代谢变化情况。方法24只去势大鼠经右下腹皮下移植新鲜卵巢(3 d之内乳鼠卵巢)和经过冻存培养后的卵巢(3 d之内乳鼠卵巢)后,检测移植前、移植后1个月2、个月的三碘甲腺原氨酸(T3)、甲状腺素(T4)、促甲状腺激素(TSH)、睾酮(T)、雌二醇(E2)、孕酮(P)、黄体生成素(LH)及卵泡生成素(FSH)。结果冻存卵巢移植前与新鲜卵巢移植前比较:T值升高,有显著性差异(P〈0.01);冻存卵巢移植后1个月与新鲜卵巢移植后1个月比较差异无显著性(P〉0.05);冻存卵巢移植后2个月与新鲜卵巢移植后2个月比较:E2升高明显,差异具有显著性(P〈0.01;新鲜卵巢移植后2个月与移植前比较:T4,T,E2,P,LH均有明显升高(P〈0.01;P〈0.05);冻存卵巢移植后2个月与移植前比较:E2,P增高明显(P〈0.01;P〈0.05)。结论冷冻卵巢移植较新鲜卵巢移植功能恢复明显。  相似文献

目的探讨人胎儿卵巢组织异体移植的最佳条件。方法观察胎儿卵巢进行异种移植后的组织学变化,并计算各组移植卵巢内各期卵泡的发育情况及卵泡构成比。结果新鲜的卵巢组织、体外培养的卵巢组织以及冷冻复苏后经体外培养的卵巢组织移植后,其内有原始卵泡发育,而且可见丰富的血管网;未见明显的淋巴细胞浸润现象。结论人胎儿卵巢组织(包括新鲜的卵巢组织、体外培养的卵巢组织以及冷冻复苏后经体外培养的卵巢组织)异种移植后原始卵泡在受体体内能进一步发育,生长卵泡分泌的雌激素有调节子宫内膜周期性变化的作用。  相似文献

为确定水牛腔前卵泡的有效分离方法 ,作者取用来自屠宰场 31头水牛的 6 1个卵巢 ,先用剪刀去掉髓质部 ,然后分别用梳刮法、剪碎法和显微分离法分离回收腔前卵泡。结果发现 ,使用梳刮法时 ,平均每个水牛卵巢回收所得的腔前卵泡数 (15 2 35± 4 4 81)明显高于剪碎法 (32 6 2± 14 81)和显微分离法 (8 95± 3 4 4 ) ,且其平均每个卵巢的处理时间 (39 0 5± 4 2 7min)明显少于剪碎法 (4 6 4 3± 4 15min)和显微分离法 (4 4 5 5± 7 82min)。梳刮法分离所得的腔前卵泡中 ,以原始卵泡最多 (占 4 1 2 5 % ) ,其次为初级卵泡 (占 38 79% ) ,而次级卵泡最少 (仅占 19 95 % )。用梳刮法获得的腔前卵泡在体外培养 72h后 ,存活率为 5 6 38% ,与剪碎法(4 8 91% )及显微分离法 (5 9 34% )没有显著差异。用梳刮法处理 10头黄牛的 2 0个卵巢 ,平均每个卵巢可分离获得 1195 2 0± 6 85 0 0个腔前卵泡 ,明显多于水牛的 15 2 35± 4 4 81个腔前卵泡。由此可见 ,梳刮法是水牛腔前卵泡的有效分离方法  相似文献

探讨山羊卵母细胞体外成熟培养的影响因素,为优化山羊体外受精程序奠定基础。比较从屠宰场采摘的山羊卵巢运输温度和体外培养促成熟因子对卵母细胞体外成熟培养和体外受精效果的影响。结果表明,在运输温度接近室温(25±2)℃时的卵母细胞成熟率显著高于接近体温(36±2)℃时的运输温度(成熟率分别为53.8%,35.0%,P0.01),两种运输温度的卵巢卵母细胞成熟培养后体外受精率无显著差异(分别为37.9%,36.6%,P0.05)。与在体外成熟基础培养液(M1:M199 media+1μg/mL E2+10μg/mL FSH+10μg/mL LH+20%EGS)中进行成熟培养相比,在基础培养液中仅添加20 ng/mL EGF(M2)和另外添加10%GFF(M3)进行体外成熟培养的卵母细胞成熟率均有明显提高,其中成熟率在M1为53.8%,极显著低于M2的69.5%和M3的72.6%,P0.01;M2和M3间差异不显著;卵母细胞成熟培养后体外受精率在三者间无明显差异分别为37.9%,39.5%和40.6%,P0.05。山羊体外受精时从屠宰场采摘的卵巢宜在室温条件下进行运输,在体外成熟培养体系中加入EGF和GFF均可有效促进山羊卵母细胞的体外成熟,但EGF起关键作用。  相似文献

将1日龄小鼠卵巢移植入成年雄鼠肾囊下,分别于移植后18d、36d回收移植卵巢进行形态学、组织学观察,以评价卵巢移植体在成年雄性受体小鼠体内生长及卵泡发育潜能。结果表明:移植体生长增大,有各级生长卵泡发育;18日龄移植体平均直径为1881.1μm±204.7μm,与1日龄卵巢相比差异极显著(P<0.01),卵泡发育到有腔卵泡阶段;36日龄移植体平均直径达2575.3μm±466.4μm,显著大于18日龄移植体(P<0.01),有成熟卵泡出现,未观察到黄体;从移植体分离到GV期卵母细胞和卵丘卵母细胞复合体。研究表明1日龄小鼠卵巢移植体在雄性受体生理环境中具有正常生长发育和形成成熟卵泡的潜能。  相似文献

目的 本实验通过对小鼠卵巢组织进行冻存研究 ,掌握卵巢的低温生物学特性 ,摸索出一种简便有效的组织器官冻存法 ,为卵巢移植及器官冷冻提供有用的技术方法。方法 通过对小鼠卵巢组织进行慢速程序法与快速液氮蒸汽法冻存 ,比较分析了不同方法所需保护剂种类、浓度、渗透平衡时间。采用对解冻后卵巢组织超微结构观察、组织化学染色、激素测定及自体、异体移植后动情期的恢复作为评价指标。结果与结论 通过上述实验表明用同种冷冻保护剂 ,液氮蒸汽法冻存的卵巢组织超微结构保存良好 ;组织化学染色示其活性与程序法冻存组织相同 ;自体、异体移植后 ,小鼠动情周期的恢复率及血清雌二醇水平各项指标均与慢速程序法冷冻无显著性差异  相似文献

目的:探讨玻璃化冻存延期回植技术用于大面积撕脱皮肤伤的临床效果。方法:选择2015年6月至2018年6月收治的大面积皮肤撕脱伤的患者16例,根据处理方式分为撕脱皮肤玻璃化冻存二期回植组和撕脱皮肤一期回植组,每组8例患者。皮肤冻存后回植前,采用MTT法检测皮肤活力,比较两组皮片回植后的成活率、感染率、平均住院时间。结果:玻璃化冻存组皮肤冻存后平均活力为92.56%±7.20%,皮片成活率为81.74%±5.78%,感染率12.5%,平均住院时间33.13±3.91 d;一期皮肤回植组皮片成活率21.91%±5.28%,感染率100%,平均住院时间47.50±1.93 d。玻璃化皮肤保存组皮片成活率、感染率、平均住院时间均优于一期皮肤回植组(P0.05)。结论:皮肤玻璃化冻存延期回植技术用于无法一期回植皮肤撕脱伤的临床效果较好。  相似文献

目的:探讨冻融小鼠卵巢同种异体移植后细胞凋亡及血管内皮生长因子表达的变化及意义。方法:收集C57BL/6j雌鼠和BALB/c雄鼠杂交后F1代4周龄小鼠卵巢,慢冻速融后移植至杂交后F1代8~12周雄鼠的肾被膜下,分别于移植后1d(24h)、2d(48h)和7d回收移植物,将冻融以及移植后不同时间段的卵巢组织进行HE染色、全卵巢卵泡计数、电镜观察、免疫组织化学分析细胞凋亡及RT-PCR检测VEGF基因表达。结果:冻融小鼠卵巢移植后随着时间的推移、各级卵泡数和卵泡存活率逐渐下降;移植后48h内细胞凋亡指数最高;电镜观察发现小鼠卵巢组织移植后损伤主要发生在移植后48h内;移植后VEGF的表达有上升的趋势,至第7d仍维持较高水平;移植后48hVEGF120mRNA和VEGF188mRNA水平明显升高(P0.05),至7d下降恢复至移植前水平,而VEGF164mRNA水平移植后无明显变化(P0.05)。结论:小鼠卵巢组织移植后48h内细胞凋亡最为严重,移植后引起大量卵泡的丢失;在移植后血管化的过程中VEGFmRNA表达量增加,VEGF120mRNA和VEGF188mRNA可能参与卵巢移植后早期血管化过程。  相似文献

采用X线局部照射大鼠的双侧卵巢,建立了雌激素缺乏的大鼠动物模型。实验发现X线照射卵巢三个月后大鼠血清雌二醇(E_2)含量明显降低,大鼠淋巴细胞的增殖及其产生白细胞介素2(IL—2)的能力升高。统计学分析表明,大鼠血清E_2含量与大鼠淋巴细胞产生IL—2的能力呈负相关。  相似文献

Summary A high-molecular-weight glycoprotein with a sedimentation coefficient of 22.6 has been isolated and characterized from the accessory cells in the previtellogenic ovary of the echinoid Dendraster excentricus. This glycoprotein is similar to the major yolk glycoprotein of the mature egg in its electrophoretic mobility under non-denaturing conditions, high mannose-type glycan, amino acid composition, constitutive glycopeptides, and immunological determinants. Previous histological and electron microscopical analyses led to the hypothesis that vitellogenesis involves a translocation of material from the accessory cell in the ovary to the oocyte. Because of the close similarities of the accessory cell glycoprotein to the yolk glycoprotein of the mature egg, we conclude that the glycoprotein in the accessory cell is a precursor to the major glycoprotein of the egg yolk. This conclusion is further supported by our additional finding that the accessory cell of another echinoid, Strongylocentrotus purpuratus, also contains a high-molecular-weight (24 S) glycoprotein which shows similarities to the yolk glycoprotein of the mature egg in the carbohydrate moiety and the constitutive glycopeptides.  相似文献

Summary The lateral oviduct and calyx of nulliparous Aedes aegypti on a sucrose diet are both flattened sacs, lacking a well defined lumen. Both are formed of an inner epithelial and an outer muscular layer, each one cell thick. The lateral oviduct is surrounded by a circular muscle sheath which is continuous with the ovarian sheath. Each ovariolar sheath is continuous with the outer layer of the calyx. The structure of both the lateral oviduct and the calyx is greatly modified after the initial blood meal. A distinct lumen develops; there is an extensive development of the outer muscular layers, and the inner epithelial layers become invaginated forming deep crypts lined with extensive microvilli. The follicular stem, which joins the primary follicle to the calyx in each ovariole, is not hollow and does not mark the opening into the calyx through which the mature egg can pass. The eggs gain access to the oviductal system after the calyx extends around the follicular epithelium of the primary follicle, when breaks appear in the calyx wall opposed to the follicular epithelium, until the mature eggs, eventually lie in a highly distended thin-walled sac of calyx from which they have direct and easy access to the lateral oviduct. After oviposition, this sac contracts to occupy once more a compact axial position in the ovary. Remnants of the follicular epithelium, containing many lysosomes are attached to the calyx at this time.  相似文献

Short day (SD) lengths delay puberty, suppress ovulation, inhibit sexual behavior, and decelerate reproductive aging in female Siberian hamsters (Phodopus sungorus). To date, the modulation of the age-associated decline in reproductive outcomes has only been demonstrated in female hamsters experiencing different day lengths during development. To determine if developmental delay is necessary for photo-inhibition to decelerate reproductive aging, hamsters raised in LD were transferred to SD as young adults and remained there for 6 months. Females that demonstrated the most immediate and sustained photo-inhibition were found to have greater numbers of ovarian primordial follicles at advanced ages (9 and 12 months) than did females held in LD, nonresponders to SD, and females with a marginal SD-response. Similarly, for females raised in SD from conception to 6 months of age, prolonged developmental delay was associated with greater numbers of primordial follicles at later ages as compared to hamsters that became refractory to SD. A robust response to SD in juvenile and adult hamsters is associated with decelerated reproductive aging, which may result in greater reproductive success in older females as compared to age-matched individuals demonstrating a more modest response to SD.  相似文献

Summary L-fucose-3H was injected intravenously into mice which were killed at several time intervals after injection and semi-thin sections of their ovaries were processed for radioautography and analysed quantitatively. At the same time the specific activity of serum glycoproteins was determined. Glycoprotein biosynthesis was demonstrated in the oocytes, granulosa and stromal cells. The silver grain density of the follicular fluid in large follicles reached a peak at 4 h, remained high at 8 h after injection and decreased steadily at the subsequent intervals. It was demonstrated that the labeling pattern of the follicular fluid depends on the secretory activity of the granulosa cells and also on the specific activity of serum glycoproteins. The collapsed zonae pellucidae which represent the highest degree of follicular atresia are able to take up glycoprotein macromolecules. Based on this finding and also on the labeling pattern of the large follicles it was shown that there is very little synthesis of specific glycoproteins for the zona pellucida in large follicles. A more specific labeling of the zona pellucida occurred in the medium follicles. Following the growth of these follicles having a previously labeled zona pellucida, it was demonstrated that this extracellular structure is secreted by the oocyte.  相似文献

目的研究血管内皮生长因子(VEGF)在人胚胎卵巢组织发生过程中的表达特征,探讨其在卵巢发生中的作用。方法采用HE染色和SP免疫组织化法学法检测VEGF在不同胎龄卵巢组织中的表达变化。结果VEGF在胎儿卵巢初级卵母细胞、卵泡细胞、部分基质细胞呈阳性表达,在卵母细胞的染色程度均强于卵泡细胞和基质细胞,基质小血管内皮也有阳性表达。其在卵母细胞中以胎24w阳性细胞多且表达量强,此后呈逐渐下降趋势。结论胎儿卵巢存在局部调节因子,VEGF表达于人胎卵巢中,以自分泌或旁分泌方式参与卵母细胞生长,在卵巢发生、发育过程中起着一定的作用。  相似文献

对3例雌性毛脚的卵巢结构进行组织形态学研究。采用常规方法制片,H.E染色。结果显示卵巢分叶呈条索状,卵泡发育中没有出现卵泡腔,卵泡细胞始终为单层,卵泡膜随卵泡发育成熟由单层转变为多层,闭锁卵泡在卵泡发育的各个不同时期都有出现。  相似文献
