In CO2-free air, the CO2 postirradiation burst (PIB) in wheat leaves was measured with an IRGA in an open gas exchange system to ascertain its potential role in alleviating photoinhibition of photorespiratory carbon oxidation (PCO) under a CO2 deficiency. A pre-photosynthesized leaf having been transferred into CO2-free air exhibited a typical CO2 PIB following darkening which could last, with a rate substantially higher than that of dark respiration, over a long time period (at least more than 2 h) of continuously alternate irradiation (2 min)-dark (2 min)-light transitions. The rate and the time of PIB maintenance, although unaffected by the exogenous dark respiration inhibitor iodoacetic acid, were stimulated largely by increasing irradiance and O2 level, and suppressed by DCMU and N-ethyl-maleimide (NEM). They also showed a large photosynthates-loading dependence. In a darkened leaf, the irradiation-induced PIB in the CO2-free air was clearly revealed and it was characterized by an initial net uptake of respiratory CO2. The light-induced PIB was accelerated by increasing irradiance, and delayed by prolonging the period of darkening the leaves. Hence, the origin of carbon needed for a long-term CO2 evolution in the CO2-free air might not only be derived directly from the pool of intermediates in the Calvin cycle, but it might also arise indirectly from a remotely fixed reserve of photosynthates in the leaf via a PCO-mediated, yet to be further clarified, mobilization process. Such mobilization of photosynthates probably exerted an important role in coordination of photochemical reactions and carbon assimilation during photosynthesis in C3 plants under the photoinhibitory conditions.  相似文献

In past studies, it was hypothesized that reductions in chloroplast isoprene emissions at high atmospheric CO(2) concentrations were caused by competition between cytosolic and mitochondrial processes for the same substrate, possibly phosphoenolpyruvate (PEP). We conducted field and laboratory experiments using leaves of white poplar (Populus alba L.) to identify whether an inverse relationship occurs between the dark respiration rate (a mitochondrial process) and the isoprene emission rate. Field experiments that were carried out in a free-air CO(2)-enriched (FACE) facility showed no clear effect of elevated CO(2) on either isoprene emission rate or respiration rate by leaves. In young, not yet fully expanded leaves, low isoprene emission and high dark respiration rates were measured in both ambient and elevated CO(2). In these leaves, isoprene emission was inversely correlated with dark respiration. It is possible to interpret from these results that, in young leaves, high rates of growth respiration compete with isoprene biosynthesis for the same substrate. However, it is also possible that the negative correlation reflects the contrasting reductions in growth respiration and increases in expression of the enzyme isoprene synthase at this final stage of leaf maturation. In contrast to our observations on young leaves, respiration rate and isoprene emission rate were positively correlated in older, fully expanded leaves (8 and 11 from apex). A positive correlation was also found between respiration rate and isoprene emission rate when these parameters were modulated using different ozone exposure, growth light intensity, growth temperature and exposure to different leaf temperatures in laboratory experiments. These data show that competition for substrate between isoprene biosynthesis and leaf respiration does not determine the rate of isoprene emission in most circumstances that affect both processes. A negative correlation was observed across all experiments between isoprene emission rate and the activity of phosphoenolpyruvate carboxylase (PEPc), a cytosolic enzyme that competes with isoprene biosynthesis for substrate. The cytosolic metabolite, PEP, occurs at a metabolic branch point from which substrate flows into three processes: (1) the production of pyruvate for mitochondrial respiration, (2) the production of oxaloacetate (OAA) by PEPc for anabolic support of mitochondrial respiration and (3) transport into the chloroplast to support chloroplastic demands for pyruvate, including isoprenoid biosynthesis. The results of our observations suggest that only the second process competes for substrate with isoprenoid synthesis, while the partitioning of PEP between mitochondrial respiration and chloroplast isoprenoid biosynthesis is controlled in a way that retains balance in substrate demand.  相似文献

C3 and C4 plants were grown in open-top chambers in the field at two CO2 concentrations, normal ambient (ambient) and normal ambient + 340 [mu]LL-1 (elevated). Dark oxygen uptake was measured in leaves and stems using a liquid-phase Clark-type oxygen electrode. High CO2 treatment decreased dark oxygen uptake in stems of Scirpus olneyi (C3) and leaves of Lindera benzoin (C3) expressed on either a dry weight or area basis. Respiration of Spartina patens (C4) leaves was unaffected by CO2 treatment. Leaf dry weight per unit area was unchanged by CO2, but respiration per unit of carbon or per unit of nitrogen was decreased in the C3 species grown at high CO2. The component of respiration in stems of S. olneyi and leaves of L. benzoin primarily affected by long-term exposure to the elevated CO2 treatment was the activity of the cytochrome pathway. Elevated CO2 had no effect on activity and capacity of the alternative pathway in S. olneyi. The cytochrome c oxidase activity, assayed in a cell-free extract, was strongly decreased by growth at high CO2 in stems of S. olneyi but it was unaffected in S. patens leaves. The activity of cytochrome c oxidase and complex III extracted from mature leaves of L. benzoin was also decreased after one growing season of plant exposure to elevated CO2 concentration. These results show that in some C3 species respiration will be reduced when plants are grown in elevated atmospheric CO2. The possible physiological causes and implications of these effects are discussed.  相似文献

The temperature dependence of quantum yields of electron transport from photosystem II (PSII) ([phi]II, determined from chlorophyll a fluorescence) and CO2 assimilation ([phi]CO2, apparent quantum yield for CO2 assimilation) were determined simultaneously in vivo. With C4 species representing NADP-malic enzyme, NAD-malic enzyme, and phosphoenolpyruvate carboxykinase subgroups, the ratio of [phi]II/[phi]CO2 was constant over the temperature range from 15 to 40[deg]C at high light intensity (1100 [mu]mol quanta m-2 s-1). A similar response was obtained at low light intensity (300 [mu]mol quanta m-2 s-1), except the ratio of [phi]II/[phi]CO2 increased at high temperature. When the true quantum yield for CO2 fixation ([phi]CO2*) was calculated by correcting for respiration in the light (estimated from temperature dependence of dark respiration), the ratio of [phi]II/[phi]C02* remained constant with varying temperature and under both light intensities in all C4 species examined. Because the [phi]II/[phi]CO2* ratio was the same in C4 monocots representing the three subgroups, the ratio was not affected by differences in the bio-chemical mechanism of concentrating CO2 in the bundle sheath cells. The results suggest that PSII activity is closely linked to the true rate of CO2 fixation in C4 plants. The close relationship between [phi]II and [phi]CO2* in C4 species under varying temperature and light intensity conditions is apparently due to a common low level of photorespiration and a primary requirement for reductive power in the C3 pathway. In contrast, in a C3 plant the [phi] II/[phi]CO2* ratio is higher under normal atmospheric conditions than under nonphotorespiratory conditions and it increases with rising temperature. This decrease in efficiency in utilizing energy derived from PSII for CO2 fixation is due to an increase in photorespiration. In both the C3 and C4 species, photochemistry is limited under low temperature, and thus excess energy must be dissipated by nonphotochemical means.  相似文献

We developed a new method using 13CO2 and mass spectrometry to elucidate the role of photorespiration as an alternative electron dissipating pathway under drought stress. This was achieved by experimentally distinguishing between the CO2 fluxes into and out of the leaf. The method allows us to determine the rates of gross CO2 assimilation and gross CO2 evolution in addition to net CO2 uptake by attached leaves during steady-state photosynthesis. Furthermore, a comparison between measurements under photorespiratory and non-photorespiratory conditions may give information about the contribution of photorespiration and mitochondrial respiration to the rate of gross CO2 evolution at photosynthetic steady state. In tomato (Lycopersicon esculentum Mill. cv Moneymaker) leaves, drought stress decreases the rates of net and gross CO2 uptake as well as CO2 release from photorespiration and mitochondrial respiration in the light. However, the ratio of photorespiratory CO2 evolution to gross CO2 assimilation rises with water deficit. Also the contribution of re-assimilation of (photo) respiratory CO2 to gross CO2 assimilation increases under drought.  相似文献

We analyzed the impact of growth at either 350 (ambient) or 700 (elevated) microL L(-1) CO(2) on key elements of the C(4) pathway (photosynthesis, carbon isotope discrimination, and leaf anatomy) using the C(4) cereal sorghum (Sorghum bicolor L. Moench.). Gas-exchange analysis of the CO(2) response of photosynthesis indicated that both carboxylation efficiency and the CO(2) saturated rate of photosynthesis were lower in plants grown at elevated relative to ambient CO(2). This was accompanied by a 49% reduction in the phosphoenolpyruvate carboxylase content of leaves (area basis) in the elevated CO(2)-grown plants, but no change in Rubisco content. Despite the lower phosphoenolpyruvate carboxylase content, there was a 3-fold increase in C isotope discrimination in leaves of plants grown at elevated CO(2) and bundle sheath leakiness was estimated to be 24% and 33%, respectively, for the ambient and elevated CO(2)-grown plants. However, we could detect no difference in quantum yield. The ratio of quantum yield of CO(2) fixation to PSII efficiency was lower in plants grown at elevated CO(2), but only when leaf internal was below 50 microL L(-1). This suggests a reduction in the efficiency of the C(4) cycle when [CO(2)] is low, and also implies increased electron transport to acceptors other than CO(2). Analysis of leaf sections using a transmission electron microscope indicated a 2-fold decrease in the thickness of the bundle sheath cell walls in plants grown at elevated relative to ambient CO(2). These results suggest that significant acclimation to increased CO(2) concentrations occurs in sorghum.  相似文献

We investigated the effects of elevated CO(2) (EC) [ambient CO(2) (AC) + 190 ppm] and elevated temperature (ET) [ambient temperature (AT) + 3.6 degrees C] on net ecosystem exchange (NEE) of seedling Douglas fir (Pseudotsuga menziesii) mesocosms. As the study utilized seedlings in reconstructed soil-litter-plant systems, we anticipated greater C losses through ecosystem respiration (R(e)) than gains through gross photosynthesis (GPP), i.e. negative NEE. We hypothesized that: (1) EC would increase GPP more than R(e), resulting in NEE being less negative; and (2) ET would increase R(e) more than GPP, resulting in NEE being more negative. We also evaluated effects of CO(2) and temperature on light inhibition of dark respiration. Consistent with our hypothesis, NEE was a smaller C source in EC, not because EC increased photosynthesis but rather because of decreased respiration resulting in less C loss. Consistent with our hypothesis, NEE was more negative in ET because R(e) increased more than GPP. The light level that inhibited respiration varied seasonally with little difference among CO(2) and temperature treatments. In contrast, the degree of light inhibition of respiration was greater in AC than EC. In our system, respiration was the primary control on NEE, as EC and ET caused greater changes in respiration than photosynthesis.  相似文献

Ten species of plants were grown at ambient (350μmol CO2·mol-1 air) and doubled (700 μmol CO2·mol-1 air) CO2 concentrations at ambient temperature and illumination in order to examine changes of dark respiration of whole seedlings or detached leaves. Effects of CO2 on dark respiration were determined by brief exposure ( ≤ 5 min) to corresponding CO2 concentration and temperatures ( 15,20,25,30 and 35 ℃ ) with infrared CO2 analyzer. The reductions in dark respiration on a weight base for leaves of East-Liaoning oak (Quercus liaotungensis Koidz. ) at 15,20 and 25 ℃ and of soybean ( Glycine max L. ) at 20,25,30 and 35 ℃ and for whole seedlings of three- tcoloured amaranth (Amaranthus tricolor L. ) at 15 and 20 ℃ and cucumber ( Cucumis sativus L. ) at 15 cE measured at elevated concentration relative to the ambient CO2 concentration were observed. No significant difference in respiration responded was observed to elevated or ambient CO2 concentrations at 15 ℃ in maize (Zea mays L. ) seedlings and alfalfa (Medicago sativa L. ) leaves, at 35 ℃ in East-Liaoning oak leaves and at 20,25 and 30 ℃ in three-coloured amaranth seedlings. However CO2 efflux in leaves of weeping willow (Salix babylonica L. ), simon poplar (Populus simonii Carr. ) and eucommia (Eucommia ulmoides Oliv. ) at 15,20,25,30 and 35 ℃, alfalfa at 20,25,30 and 35 ℃, East-Liaoning oak at 30 ℃, maize at 15 ℃, seedlings of common buckwheat (Fagotrytum esculentum Moench) at 15,20,25,30 and 35 ℃, cucumber and maize at 20,25,30 and 35 ℃ and three-coloured amaranth at 35 ℃ showed an increase at elevated in contrast to ambient CO2 concentration. In general, at lower temperatures (i. e. 15, 20 ℃ ) there was no significant difference between elevated and ambient CO2 concentration for dark respiration, while at higher temperatures (i. e. 30,35 ℃ ) elevated CO2 concentration positively stimulate clark respiretion. It has not yet been described that double CO2 concentration could enhance plant dark respiration at 30 and 35 ℃. Impacts of the characteristics in dark respiration on the future changes of vegetation and its mechanism were discussed.  相似文献

We investigated whether leaf dark respiration (nonphotorespiratory mitochondrial CO2 release) is inhibited by light in several Poa species, and whether differences in light inhibition between the species are related to differences in the rate of leaf net photosynthesis. Four lowland (Poa annua L., Poa compressa L., Poa pratensis L., and Poa trivialis L.), one subalpine (Poa alpina L.), and two alpine (Poa costiniana Vick. and Poa fawcettiae Vick.) Poa species differing in whole plant relative growth rates were grown under identical controlled conditions. Nonphotorespiratory mitochondrial CO2 release in the light (Rd) was estimated according to the Laisk method. Photosynthesis was measured at ambient CO2 partial pressure (35 Pa) and 500 [mu]mol photons m-2 s-1. The rate of photosynthesis per unit leaf mass was positively correlated with the relative growth rate, with the slow-growing alpine Poa species exhibiting the lowest photosynthetic rates. Rates of both Rd and respiration in darkness were also substantially lower in the alpine species. Nonphotorespiratory CO2 release in darkness was higher than Rd in all species. However, despite some variation between the species in the level of light inhibition of respiration, no relationship was observed between the level of inhibition and the rate of photosynthesis. Similarly, the level of inhibition was not correlated with the relative growth rate. Our results support the suggestion that rates of leaf respiration in the light are closely associated with rates in darkness.  相似文献

The spatial arrangement of tissues within the ears of cereals,and gas exchange measurements on intact ears of barley and durumwheat suggest that respiratory CO2 associated with grain-fillingprocesses, may be refixed close to its site of evolution. Apparentrefixation of respiratory CO2 in intact ears was compared withthat in flag leaves, by feeding both organs with 14C-labelledsucrose and trapping 14CO2 released by respiration. Apparentrefixation in the ears was twice that measured in flag leafblades of durum wheat genotype Durelle. In ears, the capacityof refixation of respiratory CO2 at 210 mmol mol–1 O2ranged from 55% in barley genotype Roxana to 75% in barley genotypeHatif, and 60% in duwm wheat genotype Bidi 17. A low O2 concentrationincreased refixation of respiratory CO2 by up to 90%, 92% and82%, respectively. 以长期生长于350和700μmolCO_2·mol~(-1)空气的开顶式培养室的杜仲(Eucommia ulmoides Oliv.)、紫花苜蓿(Medicago sativa L.)、玉米(Zea mays L.)等10种植物的离体成熟叶片或整株为材料,研究不同测定温度(15~35℃)下,CO_2浓度倍增对植物暗呼吸的影响。结果表明:在较低温度(15℃、20℃)下,CO_2浓度倍增对植物暗呼吸没有显著效应,在较高温度(30℃、35℃)下多数被测植物的暗呼吸显著增强。讨论了实验所得结果在未来全球气候变化中的可能的意义。  相似文献

Acclimation of 25 degrees C-grown Arabidopsis thaliana at 5 degrees C resulted in a marked increase of leaf respiration in darkness (Rd) measured at 5 degrees C. Rd was particularly high in leaves developed at 5 degrees C. Leaf respiration (non-photorespiratory intracellular decarboxylation) in the light (Rl) also increased during cold acclimation, but less so than did Rd. The ratio Rd/Pt (Pt - true photosynthesis) was higher in more acclimated or cold-developed leaves, while the ratio Rl/Pt remained unchanged. In cold-acclimated leaves, Rl did not correlate with 3-phosphoglycerate and pyruvate nor with hexose phosphate pools in the cytosol. Rl in A. thaliana leaves was probably not limited by the substrate during cold acclimation. Under the conditions tested, Rd was more sensitive to low temperature stress than Rl.  相似文献

The CO? respired by darkened, light-adapted, leaves is enriched in 13C during the first minutes, and this effect may be related to rapid changes in leaf respiratory biochemistry upon darkening. We hypothesized that this effect would be evident at the ecosystem scale. High temporal resolution measurements of the carbon isotope composition of ecosystem respiration were made over 28 diel periods in an abandoned temperate pasture, and were compared with leaf-level measurements at differing levels of pre-illumination. At the leaf level, CO? respired by darkened leaves that had been preadapted to high light was strongly enriched in 13C, but such a 13C-enrichment rapidly declined over 60-100 min. The 13C-enrichment was less pronounced when leaves were preadapted to low light. These leaf-level responses were mirrored at the ecosystem scale; after sunset following clear, sunny days respired CO? was first 13C enriched, but the 13C-enrichment rapidly declined over 60-100 min. Further, this response was less pronounced following cloudy days. We conclude that the dynamics of leaf respiratory isotopic signal caused variations in ecosystem-scale 12CO?/13) CO? exchange. Such rapid isotope kinetics should be considered when applying 13C-based techniques to elucidate ecosystem carbon cycling.  相似文献

To model the effect of increasing atmospheric CO2 on semi-arid grasslands, the gas exchange responses of leaves to seasonal changes in soil water, and how they are modified by CO2, must be understood for C3 and C4 species that grow in the same area. In this study, open-top chambers were used to investigate the photosynthetic and stomatal responses of Pascopyrum smithii (C3) and Bouteloua gracilis (C4) grown at 360 (ambient CO2) and 720 micro mol mol-1 CO2 (elevated CO2) in a semi-arid shortgrass steppe. Assimilation rate (A) and stomatal conductance (gs) at the treatment CO2 concentrations and at a range of intercellular CO2 concentrations and leaf water potentials (psileaf) were measured over 4 years with variable soil water content caused by season and CO2 treatment. Carboxylation efficiency of ribulose bisphosphate carboxylase/oxygenase (Vc,max), and ribulose bisphosphate regeneration capacity (Jmax) were reduced in P. smithii grown in elevated CO2, to the degree that A was similar in elevated and ambient CO2 (when soil moisture was adequate). Photosynthetic capacity was not reduced in B. gracilis under elevated CO2, but A was nearly saturated at ambient CO2. There were no stomatal adaptations independent of photosynthetic acclimation. Although photosynthetic capacity was reduced in P. smithii growing in elevated CO2, reduced gs and transpiration improved soil water content and psileaf in the elevated CO2 chambers, thereby improving A of both species during dry periods. These results suggest that photosynthetic responses of C3 and C4 grasses in this semi-arid ecosystem will be driven primarily by the effect of elevated CO2 on plant and soil water relations.  相似文献

Measurement of the internal CO(2) concentration (Ci) in tobacco leaves using a fast-response CO(2) exchange system showed that in the light, switching from 350 microLL(-1) to a low CO(2) concentration of 36.5 microLL(-1) (promoting high photorespiration) resulted in the Ci oscillating near the value of CO(2) compensation point (Gamma*). The oscillations are highly irregular, the range of Ci varying by 2-4 microLL(-1) in substomatal cavities with a period of a few seconds. The statistical properties of the time series became stationary after a transient of approximately 100s following transfer to low CO(2). Attractor reconstruction shows that the observed oscillations are not chaotic but exhibit stochastic behavior. The period of oscillations is consistent with the duration of photorespiratory post-illumination burst (PIB). We suggest that the observed oscillations may be due to a similar mechanism to that which leads to PIB, and may play a role in switching mitochondrial operation between oxidation of the photorespiratory glycine and of the tricarboxylic acid cycle substrates.  相似文献

依托FACE技术平台,采用稳定13C同位素技术,通过将小麦(C3作物)种植于长期单作玉米(C4作物)的土壤上,研究了大气CO2浓度升高和不同氮肥水平对土壤排放CO2的δ13C值及根际呼吸的影响.结果表明:种植小麦后土壤排放CO2的δ13C值随作物生长逐渐降低,CO2浓度升高200 μmol·mol-1显著降低了孕穗、抽穗期(施氮量为250 kg·hm-2,HN)与拔节、孕穗期(施氮量为150 kg·hm-2,LN)土壤排放CO2的δ13C值,显著提高了孕穗、抽穗期的根际呼吸比例.拔节至成熟期,根际呼吸占土壤呼吸的比例在高CO2浓度下为24%~48% (HN)和21% ~48% (LN),在正常CO2浓度下为20% ~36% (HN)和19%~32%(LN).不同CO2浓度下土壤排放CO2的δ13C值和根际呼吸对氮肥增加的响应不同,CO2浓度与氮肥用量在拔节期对根际呼吸的交互效应显著.  相似文献

In C(4) photosynthesis, a part of CO(2) fixed by phosphoenolpyruvate carboxylase (PEPC) leaks from the bundle-sheath cells. Because the CO(2) leak wastes ATP consumed in the C(4) cycle, the leak may decrease the efficiency of CO(2) assimilation. To examine this possibility, we studied the light dependence of CO(2) leakiness (phi), estimated by the concurrent measurements of gas exchange and carbon isotope discrimination, initial activities of ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) and pyruvate, orthophosphate dikinase (PPDK), the phosphorylation state of PEPC and the CO(2) assimilation rate using leaves of Amaranthus cruentus (NAD-malic enzyme subtype, dicot) plants grown in high light (HL) and low light (LL). phi was constant at photon flux densities (PFDs) >200 micromol m(-2) s(-1) and was around 0.3. At PFDs <150 micromol m(-2) s(-1), phi increased markedly as PFD decreased. At 40 micromol m(-2) s(-1), phi was 0.76 in HL and 0.55 in LL leaves, indicating that the efficiency of CO(2) assimilation at low PFD was greater in LL leaves. The activities of Rubisco and PPDK, and the phosphorylated state of PEPC all decreased as PFD decreased. Theoretical calculations with a mathematical model clearly showed that the increase in phi with decreasing PFD contributed to the decrease in the CO(2) assimilation rate. It was also shown that the 'conventional' quantum yield of photosynthesis obtained by fitting the straight line to the light response curve of the CO(2) assimilation rate at the low PFD region is seriously overestimated. Ecological implications of the increase in phi in LL are discussed.  相似文献

采用Li-6400P光合测定仪对比测定了大气CO2浓度和短期CO2浓度倍增下不同季节厚壁毛竹的光合特性,结果表明:CO2浓度加倍促使最大净光合速率、净光合速率、水分利用率、光合量子效率和光饱和点升高,年平均增幅分别为62.79%、48.74%、94.41%、8.70%和16.67%;CO2浓度加倍促使蒸腾速率、暗呼吸速率和光补偿点下降,年平均降幅分别为17.60%、37.25%和40.50%。不同季节厚壁毛竹光合生理特性参数在CO2浓度加倍后的增加幅度或降低幅度与叶片生理活性和气候变化密切相关。CO2浓度的倍增并未明显改变厚壁毛竹光合特性的季节变化规律,除光补偿点外,其它光合参数的季节大小顺序仍与大气CO2浓度下的相同。厚壁毛竹光合作用对短期CO2浓度升高的响应特征与C3植物光合作用对短期CO2浓度升高响应的普遍规律相符。  相似文献

The acclimation responses of walnut leaf photosynthesis to the irradiance microclimate were investigated by characterizing the photosynthetic properties of the leaves sampled on young trees (Juglans nigraxregia) grown in simulated sun and shade environments, and within a mature walnut tree crown (Juglans regia) in the field. In the young trees, the CO(2) compensation point in the absence of mitochondrial respiration (Gamma*), which probes the CO(2) versus O(2) specificity of Rubisco, was not significantly different in sun and shade leaves. The maximal net assimilation rates and stomatal and mesophyll conductances to CO(2) transfer were markedly lower in shade than in sun leaves. Dark respiration rates were also lower in shade leaves. However, the percentage inhibition of respiration by light during photosynthesis was similar in both sun and shade leaves. The extent of the changes in photosynthetic capacity and mesophyll conductance between sun and shade leaves under simulated conditions was similar to that observed between sun and shade leaves collected within the mature tree crown. Moreover, mesophyll conductance was strongly correlated with maximal net assimilation and the relationships were not significantly different between the two experiments, despite marked differences in leaf anatomy. These results suggest that photosynthetic capacity is a valuable parameter for modelling within-canopies variations of mesophyll conductance due to leaf acclimation to light.  相似文献
