Abstract

Isolates of Pseudomonas spp. collected from the rhizosphere of sugarcane and cane stalks were screened for their antagonistic activity against Colletotrichum falcatum causing red rot disease in sugarcane. Talc formulations of the selected Pseudomonas spp. isolates improved the sugarcane vegetative sett germination and sugarcane growth under field conditions. Optimal talc formulations were assessed for their effect on induction of systemic resistance against the pathogen in the canes under artificial inoculation. All the four isolates CHAO, EP1, KKM1 and VPT4 were effective in inducing systemic resistance against C. falcatum in two seasons. In other studies, the bacterial formulations were assessed to induce resistance in sugarcane in a sick plot situation. In pathogen-infested soil the isolates KKM1 and CHAO suppressed the red rot disease development in susceptible sugarcane cultivar. Pseudomonas strains also protected sugarcane in a disease-endemic location. Pseudomonas spp treatment substantially improved the cane juice quality parameters affected by the pathogen infection. Standardization of talc formulations and application methods in the field offers potential for large-scale application of biocontrol formulations for the management of red rot disease in sugarcane growing regions.  相似文献

The viability of axillary buds and the growth and potential yield of lateral shoots at nodes of red raspberry (Rubus idaeus) infected naturally by Didymella applanata or Botrytis cinerea were measured on excised nodes, decapitated nursery canes or on canes from fruiting plantations. In comparison with lesion-free nodes, buds at infected nodes were smaller and fewer of them were capable of growth when excised and ‘forced’, although the difference in growth between infected and uninfected nodes decreased during late winter. After February, those buds at infected nodes which were capable of forced growth did so as early and with a similar growth rate as those at lesion-free nodes. In April, 70% of buds at infected nodes were capable of growth compared with 94% of those at lesion-free nodes. When nursery canes of cv. Mailing Delight were decapitated above infected nodes the emergence of lateral shoots from the terminal infected node did not differ significantly from that at lesion-free nodes. On a range of farm sites in Scotland the emergence of shoots at infected nodes in the cropping region of canes was significantly poorer than from uninfected nodes but substantially better at infected nodes of cv. Glen Clova than at those of cvs Mailing Jewel and Mailing Orion. It is suggested that cv. Glen Clova is relatively tolerant of spur blight and cane botrytis. The length and potential yield of laterals which developed at infected nodes in the cropping region of canes in these three cultivars did not differ significantly from those at lesion-free nodes. In all tests there was no significant difference in growth at nodes infected by D. applanata and B. cinerea which may indicate a common mechanism for suppression of buds.  相似文献

Populations of the planthopper vector Perkinsiella saccharicida on sugarcane cultivars resistant (cvs QUO and Q87), moderately resistant (cvs Q90 and Q124) and susceptible (cvs NCo310 and Q102) to Fiji disease with known field resistance scores were monitored on the plant (2000–2001) and ratoon (2001–2002) crops. In both crops, the vector population remained very low, reaching its peak in the autumn. The vector population was significantly higher on cultivars susceptible to Fiji disease than on cultivars moderately resistant and resistant to Fiji disease. The number of P. saccharicida adults, nymphs and oviposition sites per plant increased with the increase in the Fiji disease susceptibility. The results suggest that under low vector density, cultivar preference by the planthopper vector mediates Fiji disease resistance in sugarcane. To obtain resistance ratings in the glasshouse that reflect field resistance, glasshouse‐screening trials should be conducted under both low and high vector densities, and the cultivar preference of the planthopper vector recorded along with Fiji disease incidence.  相似文献

Abstract

A search for plant activators capable of inducing systemic resistance in sugarcane showed that plants pre-treated with synthetic signal inducers confer a high degree of resistance to Colletotrichum falcatum – the red rot pathogen. Among the various treatments, Acibenzolar S- methyl (ASM) was found to be very effective in restricting the pathogen colonization inside the inoculated cane stalk tissues. The induction of resistance was accompanied by a significant increase in peroxidases and polyphenoloxidases activities. A considerable decrease of pathogen titre in the pre-treated tissues as determined by ELISA, clearly demonstrated the restriction of pathogen colonization and proliferation in the sensitized cane stalks. Specific induction of new isoforms of peroxidases and polyphenoloxidases in C. falcatum elicitor treatment indicates the pathogen elicitor induced specific cellular response of sugarcane suspension-cultured cells.  相似文献

The induction of pathogenesis-related (PR) proteins in sugarcane (Saccharum officinarum L.) leaves and suspension-cultured cells in response to treatment with a glycoprotein elicitor isolated from Colletotrichum falcatum (the red rot pathogen) was investigated. Treatment of leaves and cells with the elicitor resulted in a much marked increase in the activities of chitinase and β-1,3-glucanase in red rot resistant (BO 91) than susceptible (CoC 671) sugarcane cultivar. SDS-PAGE analysis revealed that C. falcatum elicitor induced the accumulation of several proteins in suspension-cultured cells of resistant cultivar (BO 91); among them the 35 kDa protein was predominant. Whereas, a 27 kDa protein was induced predominantly in the cells of susceptible cultivar upon treatment with the elicitor. When sugarcane leaves were treated with C. falcatum elicitor, two proteins with apparent molecular masses of 25 and 27 kDa were induced both in the resistant and susceptible cultivars. However, the induction was stronger in the resistant than the susceptible cultivar. Immunoblot analysis for chitinase indicated that a protein with an apparent molecular mass of 37 kDa cross-reacting with barley chitinase antiserum was strongly induced in the suspension cultured cells of both the cultivars. The induction of 37 kDa chitinase was more in the cells of resistant cultivar than in the susceptible cultivar. Western blot analysis revealed that a 25 kDa thaumatin-like protein (TLP) cross-reacting with bean TLP antiserum was strongly induced in leaves and cultured cells of both resistant and susceptible cultivars due to elicitor treatment.  相似文献

Serial thermotherapy with hot water involving different treatment series was followed in two sugarcane cultivars namely Co 740 and CoC 671 for assessing its effect on sugarcane mosaic virus (SCMV) titre in the regenerated plants. Observations on virus titre and sett regeneration were recorded at different intervals after planting the treated setts. The direct antigen coating (DAC) ELISA method was followed with the polyclonal antisera raised against the widespread local SCMV strain. All the thermo treatments affected the sett regeneration in both the cultivars. Although none of the treatment series produced virus free plants, the SCMV titre was comparatively reduced in the regenerated plants at an early phase (4 weeks after planting) in severe hot water treatment series but drastic reduction in sett germination was observed. However, the SCMV titre picked up later (8 weeks after planting) in all the treatment series. The early reduction in SCMV titre could be utilized for obtaining more virus free plants in meristem culture.  相似文献

The interactions between barley yellow dwarf virus (BYDV) and Fusarium head blight (FHB), caused by Fusarium graminearum, were studied in the two winter wheat cultivars (cvs.), Agent (susceptible to FHB) and Petrus (moderately resistant to FHB), using ultrastructural and immunocytochemical methods. Infections of wheat plants of both cvs. by BYDV increased susceptibility to FHB. BYDV infection caused numerous cytological changes in lemma tissue of both cvs. such as formation of vesicles in the cytoplasm, degradation of fine structures of chloroplasts of both cvs. and accumulation of large starch grains in the chloroplasts. Electron microscopical studies showed that the development of F. graminearum on spike surfaces was not affected in BYDV‐infected plants. After penetration and intercellular growth in lemma tissue, defence responses to Fusarium infections were markedly reduced in BYDV‐diseased plants compared to the tissue of virus‐free plants. At sites of contact of fungal cells with host tissue, depositions of cell wall material were distinctly less pronounced than in tissues of virus‐free plants of cv. Petrus. Detection of β‐1,3‐glucanases and chitinases in lemma tissue of cv. Agent revealed no appreciably increased accumulation of both defence enzymes in F. graminearum‐infected virus‐free and BYDV‐infected tissues compared to the non‐infected control tissue. On the other hand, in cv. Petrus, infection with F. graminearum induced a markedly enhanced activity of both enzymes 3 days after inoculation. The increase of both enzyme activities was less pronounced in BYDV‐infected plants than in tissue exclusively infected with F. graminearum. Cytological studies suggest that in contrast to the susceptible cv. Agent postinfectional defence responses may play still an important role in the resistance of the moderately resistant cv. Six commercial carnation cultivars were inoculated with Fusarium oxysporum f. sp. dianthi race 2, and grown under three different temperature regimes. Colonization by the pathogen and development of wilt symptoms were assessed at intervals up to 40 days. No symptoms and very little colonization were seen in any of the cultivars at 14–15°C. At a temperature of 22°C, the cultivars were clearly differentiated into three groups: those with resistance, partial resistance or susceptibility to the pathogen depending on the severity of symptoms and the extent of fungal colonization. Symptom severity was associated with the extent of colonization. This differentiation was not seen at 26°C, when all cultivars except the most resistant, cv.‘Carrier 929′, rapidly became diseased and died by 23 days after inoculation. ‘Carrier 929’ also showed some wilt symptoms at this temperature and was colonized throughout the height of the stem after 40 days. The pathogen caused disease at 26°C by a combination of vascular wilting and stem base and root rotting. Fungal colonization was assayed at 22°C by the dilution plate/homogenization method and by estimation of fungal chitin in a highly resistant (‘Carrier 929′) and in a highly susceptible (‘Red Baron’) cultivar. Both methods of assay gave similar results. In ‘Red Baron’, colonization increased slowly up to 20 days after inoculation then progressed rapidly, closely following the development of severe wilt symptoms. In ‘Carrier 929’, colonization remained very low. The low level of fungal biomass in ‘Carrier 929’ compared with ‘Red Baron’ indicated that the former cultivar showed true resistance as opposed to tolerance to the disease.  相似文献

In addition to cultural practices, the application of the fungicide mefenoxam is an important disease management tactic used to control both pink rot and leak on potato tubers grown in the USA. Mefenoxam resistance has been identified in many of the potato growing regions, and therefore resistance management strategies are very important for retaining this fungicide as a tool to manage these storage rot diseases. The relationship between mefenoxam efficacy and cultivar susceptibility to pink rot and leak was assessed in post‐harvest inoculation studies. Mefenoxam was applied to potato (Solanum tuberosum) cultivars known to express varying levels of susceptibility to pink rot and leak caused by Phytophthora erythroseptica and Pythium ultimum, respectfully. Tubers harvested from plants treated with in‐furrow and foliar applications of mefenoxam were inoculated with isolates sensitive to the fungicide. Incidence and severity of both diseases ranged widely among cultivars. Russet Norkotah was the most susceptible to infection by P. erythroseptica, while cvs Pike and Atlantic were the most resistant. Cultivars Dark Red Norland, Russet Norkotah, Goldrush and Russet Burbank were most susceptible to infection by P. ultimum whereas Snowden was most resistant. Control of pink rot differed significantly among cultivars following mefenoxam treatment, ranging from 28% (cv. Goldrush) to 67% (cv. Snowden) and generally provided the greatest level of disease control on susceptible and moderately susceptible cultivars such as Russet Norkotah and Snowden, respectively. In contrast, the impact of mefenoxam on leak development was minimal and disease control did not differ significantly among the cultivars. The fungicide failed to control leak in the susceptible cvs Atlantic and Pike and control ranged from 1.7% to 5.2% in cvs Goldrush, Russet Norkotah, Dark Red Norland, Russet Burbank and Kennebec. The greatest level of leak control was achieved with the moderately resistant cv., Snowden, at 12.7%. Cultivars most likely to benefit from mefenoxam treatments should be targeted as part of a pink rot management programme. Judicious use of the fungicide, when matched with the level of cultivar susceptibility, may prove to be an efficient and effective approach to reduce infection rates and possibly manage mefenoxam resistance thereby maintaining longevity of the compound.  相似文献

Abstract

A study was conducted in the greenhouse to examine the resistance of three soybean cultivars against root-infecting fungi, and to determine the role of five strains of Pseudomonas aeruginosa in protecting the roots from these fungal pathogens. In this study soybean cv RAWAL was found to be less susceptible against charcoal rot fungus Macrophomina phaseolina than cvs PARC and BRAGG. Most of the strains of P. aeruginosa used as seed dressing significantly reduced M. phaseolina and Rhizoctonia solani infection on all three cvs PARC, BRAGG and RAWAL (p < 0.05). Most of the strains of P. aeruginosa were effective on cv PARC against Fusarium solani infection, while on cv BRAGG P. aeruginosa strain Pa3, and on cv RAWAL strain Pa5 were effective. Both strains Pa3 and Pa22 gave maximum plant height and fresh weight of shoots, respectively on cvs PARC and BRAGG than other strains. These characteristics make these P. aeruginosa strains good candidates for use as biocontrol agents against soil-borne plant pathogens.  相似文献

Yellow leaf (YL) of sugarcane caused by Sugarcane yellow leaf virus (ScYLV, a Polerovirus of the Luteoviridae family) is a serious disease affecting the crop production and productivity in India. Although impact of the disease on cane growth is observed, no systematic study has been done so far from the tropical Asian region to establish its impact on various physiological parameters, cane yield and juice quality. We have assessed physiological parameters in symptomatic and asymptomatic plants of ten different cultivars and a genotype. In addition, similar comparisons were made between virus-infected and virus-free plants derived through meristem culture. Our studies established that among several physiological parameters, photosynthetic rate (A), stomatal conductance (g s) and SPAD metre values were significantly reduced in cultivars severely infected with ScYLV. Virus-infected cultivars exhibited significant reduction in growth/yield parameters, viz. stalk height, stalk thickness and number of internodes. Plant growth reductions were found to be 42.9, 42.3 and 38.9 % in susceptible cultivars CoPant 84211, Co 86032 and CoC 671, respectively. In addition to reduction in stalk weight, height and girth, YL disease also reduced juice yield in the affected canes up to 34.15 %. Similarly, comparison of diseased (virus-infected) and virus-free plants derived through meristem culture also revealed a drastic reduction in cane growth/physiological parameters and juice yield due to virus infection. The present study is the first comprehensive report demonstrating that YL disease caused by ScYLV seriously affects cane and juice yield in major sugarcane varieties under tropical climatic conditions (India). Consequently, this situation warrants a massive programme to provide healthy seed material and initiate breeding for YL resistance in sugarcane.  相似文献

Comparisons were made between Botrytis cinerea and Didymella applanata, which occupy the same ecological niche on canes of red raspberry. Isolates of B. cinerea from diverse localities within the British Isles were pathogenic to the SCRI selection M30 when internodal wounds on young canes were inoculated. A single inoculation frequently caused the buds at several nodes to fail the following spring. Differences in the lengths of the stem lesions that formed indicated differences in isolate pathogenicity, but these were not related to isolate origin. Bud suppression and lateral shoot failure also occurred when petioles of cvs Mailing Orion, Mailing Jewel and Glen Clova were wound inoculated with B. cinerea up to late August. The relative resistance of seven cultivars to three isolates of each pathogen was determined. Principal components analysis of data from five estimates of resistance to each pathogen showed that 40% of the variation described a common resistance to the two diseases. Further analysis showed that cvs Chilcotin and Meeker had the strongest common resistance and that cvs Glen Prosen and Mailing Jewel had the weakest. The remaining variation described cultivar differences in relative bud length after petiole inoculation with either pathogen, and differences in the relative importance of spring and autumn symptoms. Only 7% of the variation indicated independent resistance to the two pathogens and this was not influenced by cultivar differences.  相似文献

Abstract

Inoculation of sugarcane plants, cv. Jaronu 60-5, with teliospores of Ustilago scitaminea increased the production of sugarcane glycoproteins of high molecular mass (HMMG) and decreased the amount of those of mid-molecular mass (MMMG) recovered from stalks cell-free extracts. Whereas sugarcane glycoprotein of healthy plants totally inhibited the production of fungal mycelium from teliospores, mycelium growth was slightly affected by MMMG, and drastically diminished by HMMG obtained from inoculated plants. The adhesion of sugarcane glycoproteins to fungal teliospores produced cell aggregation. This effect was clearly reduced by incubating teliospores in buffer containing HMMG or MMMG previously digested with invertase. Sugarcane arginase associated to MMMG was retained by teliospores, whereas the enzymatic activity associated to HMMG from inoculated plants increased. Chitinase was not significantly retained by teliospores. Only HMMG and MMMG obtained from healthy plants were able to inhibit cell polarity in some extent. Smut induced changes in the composition of HMMG and MMMG. Some of these new glycoproteins are able to inhibit cell polarity. Since receptors, isolated from the cell wall of smut teliospores, were eluted from activated agarose beads by N-acetyl-D-glucosamine, we concluded that the peptide fraction of HMMG and MMMG bind to this amino sugar in the polysaccharide moiety of smut ligands.  相似文献

Abstract

Two families of sugarcane glycoproteins differing in their molecular mass have been isolated from sugarcane stalks. These glycoproteins specifically bind to cell wall receptors of Xanthomonas albilineans, a sugarcane pathogen, producing bacterial agglutination. Bound glycoproteins can be desorbed from bacterial cell walls by galactitol, a component of the glycosidic moiety of the sugarcane protein. This indicates that sugarcane glycoproteins bind through their glycosidic rest to the peptide moiety of the bacterial receptor. Several cell wall receptors have been isolated by affinity chromatography and separated by capillary electrophoresis.  相似文献

Resistance to Sitobion avenue and Metopolophium dirhodum was assessed in the glasshouse by releasing immature aphids on uncovered wheat and barley plants at the tillering stage. Six cultivars of winter wheat were tested with S. avenue and of these cv. Kador was resistant and cv. Maris Huntsman susceptible. Among seven cultivars of barley, cv. CI16145 was more resistant to S. avenue than any other whilst cvs Tyra and Nirakei 10 were the most susceptible, but the cultivars did not differ significantly in the numbers of M. dirhodum on them, although there were ranking differences similar to those observed with S. avenue.  相似文献

【目的】甘蔗鞭黑粉菌(Sporisorium scitamineum)引起的甘蔗黑穗病是我国甘蔗生产重要的病害。示踪甘蔗鞭黑粉菌侵染甘蔗的过程将有助于揭示其致病性和甘蔗抗黑穗病机制,为抗病品种的选育以及黑穗病的防治奠定基础。【方法】利用农杆菌介导的遗传转化技术对甘蔗鞭黑粉菌进行黄色荧光标记,对转化子进行配合及致病力检测,将标记菌株接种甘蔗感病品种ROC22及抗病品种中蔗1号、中蔗6号和中蔗9号并进行早期可视化观察。【结果】组成型表达的eYFP不影响标记菌株的配合及致病能力,而且黄色荧光性状能通过冬孢子稳定遗传。激光共聚焦显微观察表明,注射接种病原菌第5天,在感病品种ROC22的生长点已可见荧光菌丝及少量聚集状菌丝体,在抗病品种中蔗1号、中蔗6号和中蔗9号中可见少量单一丝状菌丝,无聚集状菌丝体。接种后35 d,在ROC22中可见大量聚集状菌丝体,但在中蔗品种中的聚集状菌丝体明显较少,而以中蔗1号最少。【结论】成功构建了甘蔗鞭黑粉菌侵染甘蔗的荧光示踪系统,并发现中蔗系列品种存在抑制甘蔗鞭黑粉菌菌丝体在细胞间扩展的机制。  相似文献

Virulence of 31 Kenyan isolates of Fusarium oxysporum obtained from bananas showing symptoms of Panama disease was tested against the differential banana cvs Bluggoe, Gros Michel, Dwarf Cavendish, and two other local cvs Muraru and Wang'ae. Seventeen isolates were assigned to either race 1 or race 2 of F. oxysporum f.sp. cubense (FOC). Race 4 was not apparent in this sample of 31 isolates from Kenya as none were pathogenic to cv. Cavendish, and no wilted Cavendish have been observed in field surveys in Kenya. Races could not be assigned to 12 isolates as they were virulent on more than one differential cultivar, and two were apparently not pathogenic. All isolates assigned to races 1 and 2 belonged to the VCG bridging complex 0124/5/8/20, but some other isolates belonging to this VCG complex could not be assigned to race. All five isolates assigned to VCG 01212 could not be assigned to known races. Considerable variability thus exists within FOC isolates within this region. Local cultivars of banana showed differential resistance to the pathogen. The interaction of cultivars and isolates on the level of disease was significant. Overall, cv. Wang'ae was the most susceptible to most of the isolates tested, regardless of their race, and could therefore be used as a reference cultivar in pathogenicity tests of isolates of FOC in the East African region. Of the cultivars tested that are widely grown on smallholder farms in Kenya, Muraru was the least susceptible.  相似文献

Colletotrichum acutatum is a cosmopolitan and damaging plant pathogen of temperate, subtropical, and tropical fruits and causes anthracnose on olive (Olea europaea L.). Three olive cultivars showing a variable response to infection by C. acutatum were selected to a preliminary study of pathogen development. Fruit samples, from susceptible and tolerant cultivars, were taken at 0, 24, 48, 72, and 192 h after inoculation for a microscopic and histological study of the infection and colonization process. The aim of this study was to compare the infection process: conidial germination, germ tube and appressorium formation, hyphal growth, and mesocarp colonization in susceptible and tolerant olive cultivars as a condition for further exploration of disease development, which is required to develop cultivars with improved resistance to anthracnose. The rate of mesocarp colonization differed between the susceptible and tolerant cultivars, and both intracellular hemibiotrophy and subcuticular intramural necrotrophy were observed. Hemibiotrophic infection predominated in the moderately tolerant cultivar.  相似文献

【背景】寄生帚梗柱孢霉是花生黑腐病的病原菌,被我国列为重要的进境植物检疫性有害生物。该病菌2009年已入侵我国广东,造成花生植株基部腐烂而死亡,严重威胁花生生产安全。筛选与种植抗病品种是防控该病害的重要措施。【方法】收集广东推广种植的15个主要花生品种,通过人工接种方法,鉴定这些品种对花生黑腐病菌的抗性水平。【结果】15个供试花生品种中,湛红2号、湛油62等2个品种表现为抗病;湛油75、湛油82、粤油390、粤油410、仲恺花44、仲恺花99、汕油诱1号等7个品种表现为中抗;花育33号、汕油523、汕油辐1号、粤油18、湛油53等5个品种表现为感病;仲恺花332表现为高感。【结论与意义】目前广东生产上推广种植的花生品种多数对黑腐病菌表现为抗病或中抗水平,部分品种表现为感病或高感。该结果可为我省花生品种的推广与布局提供依据。  相似文献

Fifteen wheat varieties commercially grown in Kenya were tested for their susceptibility to head blight and mycotoxin accumulation after inoculation with Fusarium graminearum in pot experiments. The strains of the pathogen used had been isolated from wheat collected in different growing areas of Kenya. Head blight susceptibility was assessed as the percentage of spikelets bleached and area under disease progress curve; kernel colonization by fungal mycelium was determined as ergosterol content. All varieties were found to be moderately to highly susceptible. However, the varieties differed in head blight susceptibility (29–68% of spikelets bleached; mean 54%), fungal colonization (67–187  μ g/g ergosterol content; mean 111  μ g/g) and the resulting mycotoxin contamination [deoxynivalenol (DON) 5–31  μ g/g; mean 13.5  μ g/g]. Grain weight reductions due to head blight ranged from 23 to 57% (mean 44%). The varieties could be therefore divided into partially resistant and highly susceptible genotypes. The kernels of highly susceptible varieties had higher mycotoxin and ergosterol contents. However, the kernels of some varieties contained more fungal mycelium (ergosterol) without the corresponding high amounts of DON, suggesting that they possess some resistance to DON accumulation. Less susceptible varieties showed resistance to fungal spread, as indicated by a slow disease development and lower content of fungal biomass.  相似文献
