Accurate and highly sensitive evaluation of the sirtuin 1 (SirT1) level is becoming increasingly important for understanding the contribution of SirT1 in metabolism pathways. Here, a novel electrochemical immunoassay of SirT1 based on crosslinked hyperbranched azo-polymer decorated with gold colloids (Au–HAP) as sensing platform and titanium dioxide (TiO2)–Au nanocomposites to immobilize secondary antibody–horseradish peroxidase (Ab2–HRP) as electrochemical labels has been designed. Greatly enhanced sensitivity was achieved by exploiting the excellent conductivity of Au nanoparticle, the amplification effect of Au–HAP and TiO2–Au, and the favorable catalytic ability of HRP. The nanocomposites of Au–HAP and TiO2–Au could attach numerous capture antibodies on the surface for significant immune recognition efficiency. Meanwhile, the TiO2–Au-labeled Ab2–HRP using an HRP–thionine–H2O2 (hydrogen peroxide) detection system could further induce signal readout. Under optimal conditions, the signal intensity was linearly related to the concentration of SirT1 in the range of 1–500 ng ml−1, and the limit of detection was 0.28 ng ml−1. The developed biosensor exhibits attractive performance for the analysis of SirT1, with rapid response, high sensitivity, and high accuracy, and could become a promising technique for protein detection.  相似文献

In this work, a multiplexed electrochemical immunosensor was developed for sensitive detection of carcinoembryonic antigen (CEA) and α-fetoprotein (AFP) using silver nanoparticles (Ag NPs) or gold nanoparticles (Au NPs) coated-carbon nanospheres (CNSs) as labels. CNSs were employed as the carrier for the immobilization of nanoparticles (Ag NPs or Au NPs), thionine (Thi), and secondary antibodies (Ab2) due to their good monodispersity and uniform structure. Au NPs reduced graphene oxide (rGO) nanocomposites were used as sensing substrate for assembling two primary antibodies (Ab1). In the presence of target proteins, two labels were attached onto the surface of the rGO/Au NPs nanocomposites via a sandwich immunoreaction. Two distinguishable peaks, one at +0.16 V (corresponding to Ag NPs) and another at −0.33 V (corresponding to Thi), were obtained in differential pulse voltammetry (DPV). The peak difference was approximately 490 mV, indicating that CEA and AFP can be simultaneously detected in a single run. Under optimal conditions, the peak currents were linearly related to the concentrations of CEA or AFP in the range of 0.01–80 ng ml−1. The detection limits of CEA and AFP were 2.8 and 3.5 pg ml−1, respectively (at a signal-to-noise ratio of 3). Moreover, when the immunosensor was applied to serum samples, the results obtained were in agreement with those of the reference method, indicating that the immunosensor would be promising in the application of clinical diagnosis and screening of biomarkers.  相似文献

An electrochemical immunosensor for quantitative detection of α-fetoprotein (AFP) in human serum was developed using graphene sheets (GS) and thionine (TH) as electrode materials and mesoporous silica nanoparticles (MSNs) loaded with ferroferric oxide (Fe3O4) nanoparticles and horseradish peroxidase (HRP) as labels for signal amplification. In this study, the compound of GS and TH (GS–TH) was used as a substrate for promoting electron transfer and immobilization of primary antibody of AFP (Ab1). MSNs were used as a carrier for immobilization of secondary antibody of AFP (Ab2), Fe3O4, and HRP. The synergistic effect occurred between Fe3O4 and HRP and greatly improved the sensitivity of the immunosensor. This method could detect AFP over a wide concentration range from 0.01 to 25 ng ml−1 with a detection limit of 4 pg ml−1. This strategy may find wide potential application in clinical analysis or detection of other tumor markers.  相似文献

A label-free electrochemical immunosensor based on Nafion/carbon nanotubes (CNTs)/charged pyridinecarboxaldehyde composite film was developed for the detection of hepatitis B surface antigen. Nafion/CNTs/charged pyridinecarboxaldehyde nanocomposites were prepared by dispersing charged pyridinecarboxaldehyde and CNTs in Nafion solution. The nanocomposites were cast on the electrode surface to form aldehyde-terminated composite film that can covalently bind antibody on the film without using other reagent. The immunosensor response was linearly changed with hepatitis B surface antigen concentration in the range from 0.1 to 25 ng ml−1 with a detection limit (signal/noise ratio = 3) of 0.04 ng ml−1. Some important advantages such as simple preparation, good stability, reproducibility, and selectivity of the immunosensor were achieved.  相似文献

This paper reports a label-free electrochemical immunosensor for the determination of aflatoxin B1 (AFB1), which is based on a gold electrode modified by a biocompatible film of carbon nanotubes/poly(diallyldimethylammoniumchloride)/Pd–Au nanoparticles (CNTs/PDDA/Pd–Au). The nanocomposite was characterized by transmission electron microscopy and the electrochemical behavior of modified electrodes was investigated by cyclic voltammetry. The CNTs/PDDA/Pd–Au nanocomposites film showed good electron transfer ability, which ensured high sensitivity to detect AFB1 in a range from 0.05 to 25 ng mL−1 with a detection limit of 0.03 ng mL−1 obtained at 3σ (where σ is the standard deviation of the blank solution, n = 10). The proposed immunosensor provides a simple tool for AFB1 detection. This strategy can be extended to any other antigen detection by using the corresponding antibodies.  相似文献

A new electrochemical immunoassay of alpha-fetoprotein (AFP) was developed on an organic–inorganic hybrid nanostructure-functionalized carbon electrode by coupling with magnetic bionanolabels. Multi-walled carbon nanotubes (CNTs), single-stranded DNA, thionine and AFP were utilized for the construction of the immunosensor, while the core–shell Fe3O4-silver nanocomposites were employed for the label of horseradish peroxidase-anti-AFP conjugates (HRP-anti-AFP-AgFe). Electrochemical measurement toward AFP was carried out by using magnetic bionanolabels as traces and H2O2 as enzyme substrate with a competitive-type immunoassay mode. Experimental results indicated that the immunosensors with carbon nanotubes and DNA exhibited better electrochemical responses than those of without carbon nanotubes or DNA. Under optimal conditions, the electrochemical immunosensor by using HRP-anti-AFP-AgFe as signal antibodies exhibited a linear range of 0.001–200 ng mL−1 AFP with a low detection limit of 0.5 pg mL−1 at 3sB. Both intra- and inter-assay coefficients of variation were 7.3%, 9.4%, 8.7% and 10.2%, 7.8%, 9.4% toward 0.01, 30, 120 ng mL−1 AFP, respectively. The specificity and stability of the electrochemical immunoassay were acceptable. In addition, the methodology was validated for 12 clinical serum specimens including 9 positive specimens and 3 normal specimens, receiving a good correlation with the results obtained from the referenced electrochemiluminescence assay.  相似文献

In this article, a novel, label-free, and inherent electroactive redox immunosensor for carcinoembryonic antigen (CEA) based on gold nanoparticles (AuNPs) and Nile blue A (NB) hybridized electrochemically reduced graphene oxide (NB–ERGO) is proposed. The composite of NB–graphene oxide (NB–GO) was prepared by π–π stacking interaction. Then, chronoamperometry was adopted to simultaneously reduce HAuCl4 and nanocomposites of NB–GO for synthesizing AuNPs/NB–ERGO. The immunosensor was fabricated by capturing CEA antibody (anti-CEA) at this nanocomposite modified electrode. The immunosensor determination was based on the fact that, due to the formation of antigen–antibody immunocomplex, the decreased response currents of NB were directly proportional to the concentrations of CEA. Under optimal conditions, the linear range of the proposed immunosensor was estimated to be from 0.001 to 40 ng ml−1 and the detection limit was estimated to be 0.00045 ng ml−1. The proposed immunosensor was used to determine CEA in clinical serum samples with satisfactory results. The proposed method may provide promising potential application in clinical immunoassays with the properties of facile procedure, stability, high sensitivity, and selectivity.  相似文献

In this work, a simple electrochemical immunosensor was developed for the detection of carcinoembryonic antigen (CEA) based on rhombic dodecahedral Cu2O nanocrystals–graphene oxide–gold nanoparticles (rCu2O–GO–AuNPs). GO as the template and surfactant resulting in rCu2O exhibit improved rhombic dodecahedral structure uniformity and excellent electrochemical performance. Moreover, GO was found to be able to effectively improve the long stability of rCu2O on the electrode response. Under optimal conditions, the immunosensor showed a low limit of detection (0.004 ng ml−1) and a large linear range (0.01–120 ng ml−1). This work presents a potential alternative for the diagnostic applications of GO-supported special morphology materials in biomedicine and biosensors.  相似文献

In this work, a novel thiol aromatic aldehyde was synthesized. It can be used as a substrate to directly immobilize antibodies on a gold electrode, for which no additional chemical cross-linker is required. It was also applied as a linker to prepare Fe3O4@Au/PAMAM/Ab2–horseradish peroxidase bioconjugates, which introduced multiple enzymes onto a sensing interface owing to the high surface-to-volume ratio of Fe3O4@Au nanoparticles and many functional groups of the poly(amidoamine) dendrimer (PAMAM). The introduced multiple enzymes greatly improved the detection signal. Under optimal conditions, the proposed electrochemical immunosensor exhibited desirable performance for detection of IgG in the range 0.005–50 ng ml−1 with a detection limit of 3 pg ml−1 based on a signal-to-noise ratio of 3. It has great potential application in the area of clinical analysis.  相似文献

In this study, a novel sensitive electrochemiluminescence (ECL) immunosensor was constructed by carboxyl graphene (GR) for enhancing luminol–O2 system emission. Here, carboxyl GR was used to enhance the ECL intensity of luminol that had excellent electron transfer ability and good solubility. The sensing platform was constructed by depositing carboxyl GR on electrodes and immobilizing antibodies on the surface of carboxyl GR through amidation. The specific immunoreaction between α-fetoprotein (AFP) and antibodies resulted in a decrease of ECL intensity, and the intensity decreased linearly with AFP concentrations in the range of 5 pg ml−1 to 14 ng ml−1 with a detection limit of 2.0 pg ml−1. The proposed immunosensor exhibits high specificity, good reproducibility, and longtime stability. It may become a promising technique for protein detection.  相似文献

A direct electrochemical immunoassay system based on the immobilization of α-1-fetoprotein antibody (anti-AFP), as a model system, on the surface of core-shell Fe2O3/Au magnetic nanoparticles (MNP) has been demonstrated. To fabricate such an assay system, anti-AFP was initially covalently immobilized on to the surface of core-shell Fe2O3/Au MNP. Anti-AFP-modified MNP (bio-nanoparticles) were then attached to the surface of carbon paste electrode with the aid of a permanent magnet. The performance and factors influencing the performance of the resulting immunosensor were studied. α-1-Fetoprotein antigen was directly determined by the change in current or potential before and after the antigen–antibody reaction versus saturated calomel electrode. The electrochemical immunoassay system reached 95% of steady-state potential within 2 min and had a sensitivity of 25.8 mV. The linear range for AFP determination was from 1 to 80 ng AFP ml−1 with a detection limit of 0.5 ng AFP ml−1. Moreover, the direct electrochemical immunoassay system, based on a functional MNP, can be developed further for DNA sensor and enzyme biosensor. Prostate-specific antigen (PSA), as the specificity of prostate cancer markers, has been widely used in prostate cancer diagnosis and screening. In this study, we fabricated an electrochemical immunosensor for PSA detection using the amino-functionalized graphene sheet–ferrocenecarboxaldehyde composite materials (NH2-GS@FCA) and silver hybridized mesoporous silica nanoparticles (Ag@NH2-MCM48). Under optimal conditions, the fabricated immunosensor showed a wide linear range with PSA concentration (0.01–10.0 ng·ml−1). Low detection limit (2 pg·ml−1) proved the high sensitivity. In addition, the immunosensor possessed good stability and reproducibility. Moreover, the application to PSA analysis in serum samples yielded satisfactory results.  相似文献

A multiplexing electrochemical immunosensor was developed for ultrasensitive detection of cancer related protein biomarkers. We employed disposable screen-printed carbon electrode (SPCE) array as the detection platform. A universal multi-labeled nanoprobe was developed by loading HRP and goat-anti-rabbit IgG (secondary antibody, Ab2) onto multiwalled carbon nanotube (MWNT). This universal nanoprobe was available for virtually any sandwich-based antigen detection and showed superiority in several areas. By using the SPCE array and the universal nanoprobe, we could detect as low as 5 pg mL−1 of prostate specific antigen (PSA) and 8 pg mL−1 of Interleukin 8 (IL-8) with the electrochemical immunosensor. We also demonstrated simultaneous detection of two protein biomarkers with this platform. With these attracted features, our immunoassay system shows promising applications for in-field and point-of-care test in clinical diagnostics.  相似文献

An electrochemical immunosensor was developed for the detection of hepatitis B surface antigen (HBsAg). The biotinylated hepatitis B surface antibody was immobilized on streptavidin magnetic nanoparticles and used for targeting the HBsAg. By the addition of horseradish peroxidase conjugated with secondary antibody (HRP–HBsAb), a sandwich-type immunoassay format was formed. Aminophenol as substrate for conjugated HRP was enzymatically changed into 3-aminophenoxazone (3-APZ). This electroactive enzymatic production (3-APZ) was transferred into an electrochemical cell and monitored by cyclic voltammetry. Under optimal conditions, the cathodic current response of 3-APZ, which was proportional to the HBsAg concentration, was measured by a glassy carbon electrode. The immunosensor response was linear toward HBsAg in the concentration range from 0.001 to 0.015 ng/ml with a detection limit of 0.9 pg/ml at a signal/noise ratio of 3.  相似文献

A convenient and effective strategy for fabrication of hydrogen peroxide biosensor based on sodium alginate (SA) and polyvinyl butyral (PVB) as matrices was reported in this paper. The horseradish peroxidase (HRP) and SA were electro-co-deposited onto the surface of gold electrode, and the HRP–SA/Au electrode was further coated with PVB. The interaction between HRP and SA was characterized by UV–vis absorption spectroscopy, and the fabricating process of biosensor was characterized by electrochemical impedance spectroscopy (EIS). The electrochemical characteristics of the biosensor were studied by cyclic voltammetry and chronoamperometry. Experimental conditions were investigated which influence the performance of the biosensor, such as pH, and applied potential. The biosensor showed a linear response to H2O2 over a concentration range from 7.0 × 10−6 to 4.1 × 10−3 M with a detection limit of 1.8 × 10−6 M based on a signal-to-noise ratio of 3 under optimum conditions. The value of HRP in the composite was evaluated to be 1.38 mM. The biosensor obtained from this study possesses high sensitivity, good reproducibility, and long-term stability.  相似文献

An electrochemical biosensor for determination of hydrogen peroxide (H2O2) was fabricated, based on the electrostatic immobilization of horseradish peroxidase (HRP) with one-dimensional gold nanowires (Au NWs) and TiO2 nanoparticles (nano-TiO2) on a gold electrode. The nano-TiO2 can give a biocompatible microenvironment and compact film, and the Au NWs can provide fast electron transferring rate and greatly add the amount of HRP molecules immobilized on the electrode surface. Au NWs were characterized by ultraviolet–visible spectra and transmission electron microscope. The electrode modification process was probed by cyclic voltammetry and electrochemical impedance spectroscopy. Chronoamperometry was used to study the electrochemical performance of the resulting biosensor. Under optimal conditions, the linear range for the determination of H2O2 was from 2.3 × 10−6 to 2.4 × 10−3 M with a detection limit of 7.0 × 10−7 M (S/N = 3). Moreover, the proposed biosensor showed superior stability and high sensitivity.  相似文献

A novel strategy for the fabrication of sensitive immunosensor to detect α-fetoprotein (AFP) in human serum has been proposed. The immunosensor was prepared by immobilizing AFP antigen onto the glassy carbon electrode (GC) modified by gold nanoparticles and carbon nanotubes doped chitosan (GNP/CNT/Ch) film. GNP/CNT hybrids were produced by one-step synthesis based on the direct redox reaction. The electrochemical properties of GNP/CNT/Ch films were characterized by impedance spectroscopy and cyclic voltammetry. It was indicated that GNP/CNT nanohybrid acted as an electron promoter and accelerated the electron transfer. Sample AFP, immobilized AFP, and alkaline phosphatase (ALP)-labeled antibody were incubated together for the determination based on a competitive immunoassay format. After the immunoassay reaction, the bound ALP label on the modified GC led to an amperometric response of 1-naphthyl phosphate (1-NP), which was changed with the different antigen concentrations in solution. Under the optimized experimental conditions, the resulting immunosensor could detect AFP in a linear range from 1 to 55 ng ml−1 with a detection limit of 0.6 ng ml−1. The proposed immunosensor, by using GNP/CNT/Ch as the immobilization matrix of AFP, offers an excellent amperometric response of ALP-anti-AFP to 1-NP. The immunosensor provided a new alternative to the application of other antigens or other bioactive molecules.  相似文献

A novel series of elliptical gold (Au0) nanoparticles (18–40 nm) embedded antimony glass (K2O-B2O3-Sb2O3) dichroic nanocomposites have been synthesized by a single-step melt-quench in-situ thermochemical reduction technique. X-ray and selected area electron diffractions manifest growth of Au0 nanoparticles along the (111) and (200) crystallographic planes. The transmission electron microscopic image reveals elliptical Au0 nanoparticles having an aspect ratio varying in the range 1.2–2.1. The dichroic behavior of the nanocomposites arises due to elliptical shape of the Au0 nanoparticles. These nanocomposites show strong surface plasmon resonance (SPR) band of Au nanoparticles in the range 610–681 nm and it exhibit red-shifts with increasing Au concentration. They, when co-doped with Sm2O3 and excited at 949 nm, exhibit about sevenfold enhancement of the upconverted red emission transition of 4G5/26H9/2 at 636 nm due to local electric field enhancement effect of Au0 nanoparticles induced by its SPR. These nanocomposites are the promising materials for laser, display, and various nanophotonic applications.  相似文献

A novel amperometric immunosensor based on chitosan–gold nanoparticles (Chit–GNPs) composite film and thionine (Thi) was prepared for the determination of α-1-fetoprotein (AFP). The immunosensor was prepared by electro-depositing a Chit–GNPs composite matrix on the surface of the glass carbon electrode, then Thi was immobilized onto the Chit–GNPs film using glutaraldehyde as a cross-linker. Furthermore, the GNPs were chemisorbed onto Thi film for immobilization of α-1-fetoprotein antibody. The procedure of the immunosensor was characterized by means of cyclic voltammograms. The performance and influencing factors of the resulting immunosensor were studied in details. Under optimal conditions, the immunosensor was highly sensitive to AFP and the linear range covered from 0.40 to 200.0 ng mL−1 with a detection limit of 0.24 ng mL−1 at three times background noise. Moreover, the simple and controllable electro-deposition method overcame the irreproducibility for preparing Chit-based immunosensor systems and the proposed immunosensor displayed a satisfactory reproducibility and stability.  相似文献

Electrically active magnetic nanocomposites (EAMNCs), Au nanoparticles/self-doped polyaniline@Fe3O4 (AuNPs/SPAN@Fe3O4) with well-defined core/shell structure, were first synthesized by a simple method. The morphology and composition of the as-synthesized AuNPs/SPAN@Fe3O4 nanocomposite have been characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), Fourier transform infrared (FT–IR), ultraviolet–visible (UV–Vis), X-ray powder diffraction (XRD), and thermogravimetric analysis (TGA). Horseradish peroxidase (HRP)–AuNPs/SPAN@Fe3O4 biocomposites were immobilized onto the surface of indium tin oxide (ITO) electrode to construct an amperometric hydrogen peroxide (H2O2) biosensor. The effects of HRP dosage, solution pH, and the working potential on the current response toward H2O2 reduction were optimized to obtain the maximal sensitivity. Under the optimal conditions, the proposed biosensor exhibited a linear calibration response in the range of 0.05 to 0.35 mM and 0.35 to 1.85 mM, with a detection limit of 0.01 mM (signal-to-noise ratio = 3). The modified electrode could virtually eliminate the interference of ascorbic acid (AA) and uric acid (UA) during the detection of H2O2. Furthermore, the biosensor was applied to detect H2O2 concentration in real samples, which showed acceptable accuracy with the traditional potassium permanganate titration.  相似文献
