昆明山海棠根部水抽提物对体外微管蛋白聚合的影响   总被引:2,自引:0,他引:2  
本研究利用猪脑中分离纯化的微管蛋白聚合和解聚反应,分析了非整倍体诱发剂昆明山海棠根部水抽提物(THH)对微管蛋白聚合状态的影响,从该角度探讨了THH诱发哺乳动物非整倍体的机制。秋水仙素(COL)为本研究的阳性对照物。结果发现THH能显著抑制体外微管蛋白的聚合,该抑制效应呈明显的剂量——效应关系。研究结果与我们以往关于THH为非整倍体诱发剂的实验证据相吻合并进一步提示THH可以抑制微管蛋白聚合作为诱发非整倍体的途径之一。  相似文献

本文以小鼠着丝粒次要卫星DNA探针FISH和抗着丝粒CREST染色,研究了可疑的非整倍体毒剂丙烯酰胺(AA)诱导的小鼠NIH3T3细胞微核(MN)的着丝粒组成情况和小鼠骨髓染色体畸变(CA)情况。结果发现AA在100—400μg/ml诱导的MN约52.7%—71.6%为FISH阳性,60.5%—68.2%的MN为CR-EST阳性,两种结果均显示AA具有较强的非整倍体诱发效应。小鼠骨髓CA的FISH表明,AA既能诱导染色体结构畸变,又能诱导非整倍体形成,而以非整倍体诱发效应更为明显。  相似文献

用0.05%和0.1%秋水仙素溶液对洋葱Allium cepa根尖进行控时处理,研究不同秋水仙素浓度和不同处理时间对洋葱根尖细胞有丝分裂畸变的影响。结果显示,秋水仙素有促进细胞有丝分裂染色体停滞在中期的能力;不同固定时间对洋葱根尖细胞有丝分裂有影响;秋水仙素对洋葱根尖细胞染色体有畸变效应,诱导产生畸变的能力受溶液浓度和处理时间的影响。  相似文献

微核直径测试作为非整倍体诱发剂的分析手段   总被引:3,自引:0,他引:3  
以小鼠骨髓红细胞微核直径测试,比较了秋水仙素(COL)与昆明山海棠(THH)、对苯二酚(HQ)在哺乳动物体细胞中的非整倍体诱发效应。丝裂霉素C(MMC)作为多功能染色体断裂剂引入实验,为诱发非整倍体的阴性对照。结果发现,COL组,71%的微核直径(d)大于或等于所在细胞直径(D)的五分之一(d≥D/5);THH诱发微核中,54%的微核d≥D/5;HQ及MMC组,分别有47%及14%的微核相对直径达此阈值。暗示THH及HQ具有类似COL的某种非整倍体诱发效应。微核直径测试可作为非整倍体诱发剂检测的辅助手段。 The relative diameters of micronucleus induced by colchicines(COL),Triptergium hypoglaucum(Level)Hutch(THH)and hydroquinone(HQ)were compared to evaluate their aneugenic activities in mouse bone marrow erythrocytes.MitomycinC(MMC)was taken as the negative control in the experiment because it is a multifunctional clastogen without aneugenic potential.Diameters of the cytoplasm(D)and the micronucleus(d)of each micronucleated erythrocytes were measured with a micrometer in a microscope.The frequency of relatively large micronuclei(d≥D/5)was found(71%) in COL treated group.In the THH and HQ treated groups,the relatively large micronuclei were 54% and 47%,respectively.Such micronuclei were infrequent(14%)in the MMC treated group.The results implied that THH and HQ may possess some aneugenic potential like COL.  相似文献

本研究利用猪脑中分离纯化的微管蛋白聚合和解聚反应,分析了具有争议的非整倍体诱发剂2(4′噻唑)苯丙咪唑(thiabendazole,TBZ)对微管蛋白聚合状态的影响。秋水仙素(colchicine)为本研究的阳性对照物。结果发现2(4′噻唑)苯丙咪唑能显著抑制体外微管蛋白的聚合,并呈明显的剂量效应关系。研究表明,TBZ可能通过抑制微管蛋白聚合来影响染色体正常分离,诱发非整倍体。  相似文献

以黑腹果蝇的非整倍体测试品系评价了云南地方中草药昆明山海棠水抽提物(THH)在动物生殖细胞中的非整倍体诱发效应,秋水仙素(COL)为本试验的阳性对照物。结果发现,在所有雌雄成虫口饲染毒组中,THH(10-80mg /ml)及COL(2.5-20μg/ml)均显著诱发生殖细胞非整倍体,从而导致X0及X XY例外子代频率显著升高(P<0.001-0.5)。试验表明:THH及COL均能够在蝇生殖细胞形成过程中导致染色体的丢失或不分离;在本研究的受试剂量范围内,雄性果蝇对受试物诱发生殖细胞X染色体遗失效应较为敏感,而雌性果蝇则易检出诱发生殖细胞性染色体不分离的化合物。  相似文献

研究了1,2,4-三氯苯(TCB)对蚕豆幼苗生长、根尖细胞分裂及染色体畸变的影响.结果表明,随TCB浓度增加和处理时间延长,蚕豆幼苗根长的生长及根尖细胞有丝分裂指数降低甚至停止.TCB诱发蚕豆根尖细胞有丝分裂过程中染色体数目畸变和结构畸变.50-100μg.g^-1TCB胁迫12-24h,蚕豆根尖染色体的主要损伤形式为c-有丝分裂、染色体桥和不均匀排列,其出现百分率达1.0%--10.3%.300μg.g^-1TCB胁迫12-96h,蚕豆根尖细胞中染色体粘连(S)、S+染色体断裂(S+B)、S+染色体环(S+R)、S+染色体不均匀排列(S+A)及S+染色体桥(S+Be)出现的百分率达47.9%--88.9%,各种类型染色体断裂出现的百分率仅为18.1%--29.6%,说明蚕豆根尖细胞染色体畸变分析可作为TCB土壤污染监测的敏感生物监测指标.  相似文献

平阳霉素三种成份诱发染色体畸变的研究   总被引:8,自引:1,他引:7  
本文报道了抗癌新药平阳霉素(PYM)三种成份(A_2、A_5、A_6)诱发蚕豆根尖细胞染色体畸变的效应。结果表明PYM的主要成份A_5的诱变能力显著高于A_2和A_6,从而找出了平阳霉素诱变能力大于博莱霉素的原因。同时观察到平阳霉素可诱发出大量特异的散碎型染色体畸变。上述三种成份诱发的畸变绝大多数属于染色体畸变,很少有染色单体畸变。另外并出现大量的微核,证明PYM很可能是一种不依赖S期效应的诱变剂。  相似文献

Osteocytes are the most abundant cells in bone and there is increasing evidence that they control bone remodeling via direct cell-to-cell contacts and by soluble factors. In the present study, we have used the MLO-Y4 cell line to study the effect of osteocytes on the proliferation, differentiation and bone-forming capacity of bone marrow mesenchymal stem cells (MSC). Conditioned media (CM) from osteocytic MLO-Y4 and osteoblastic MC3T3-E1 cell lines were collected and added on mouse bone marrow cultures, in which MSC were induced to osteoblasts. There was a significant increase in alkaline phosphatase activity and osteocalcin expression in the presence of MLO-Y4 CM. No such stimulus could be observed with MC3T3-E1 CM. There was almost 4-fold increase in bone formation and up to 2-fold increase in the proliferation of MSC with MLO-Y4 CM. The highly proliferating bone marrow cells were negative for ALP and OCN, suggesting that they could represent early osteoblast precursors. MLO-Y4 CM did not enhance the viability of mature osteoblasts nor protected them of apoptosis. This is the first study to describe soluble signals between osteocytes and osteoblasts and there most likely are several still unidentified or unknown factors in osteocyte CM. We conclude that osteocytes have an active stimulatory role in controlling bone formation.  相似文献

The genotoxic effects of fungicide Conan 5FL (containing 50 g/L hexaconazole) in mouse bone-marrow cells and human lymphocytes have been evaluated. Three different concentrations of Conan 5FL (17.50, 35.00 and 70.00 microg/mL for human lymphocytes and 17.50, 35.00 and 70.00 mg/kg for mouse bone marrow cells) were studied. Conan 5FL induced significant increases (except 17.50 mg/kg for mouse bone marrow) in the frequency of chromosomal aberrations (CAs) in both test systems. This fungicide caused structural and numerical abnormalities in both mammalian cells. These are sister chromatid union, chromatid and chromosome breaks, fragments, dicentric and ring chromosomes, and polyploidy. Significant increase was found in induction and in minimum-maximum numbers of sister chromatid exchanges (SCEs) at all treatments compared with the negative control. Conan 5FL did not affect the replication index (RI) in human lymphocyte cultures, however, it significantly decreased the mitotic index (MI) in all treatment concentrations in both test systems. Using of Conan 5FL should be reconsidered due to its possible cytotoxic, clastogenic and mutagenic effects.  相似文献

本文对几种化学诱变剂诱发小鼠体内脾脏、骨髓和精原细胞的SCE进行了比较研究,同时分析了几类常见化合物在小鼠脾脏细胞中诱发SCE的活力。结果显示诱变剂在脾脏细胞中诱发SCE比骨髓和精原细胞敏感。几类化合物都能显著地诱发小鼠脾脏SCE的增加,与对照相比差异显著(P<0.05)或极显著(P<0.01),说明利用小鼠脾脏细胞检测环境诱变物是相当灵敏的。  相似文献

Possible interactions between hydroquinone (HQ) and phenol (PHE), 2 known benzene metabolites, in inducing micronuclei in mouse bone marrow cells were investigated. HQ and PHE administered alone gave weak and negative results, respectively, at the doses tested. However, simultaneous administration of both compounds caused a considerable increase in the induction of micronuclei as well as an increase in bone marrow toxicity. Using 3 different statistical methods, it was shown that the observed joint effect was significantly higher than additive interaction, and was close to multiplicative interaction. These findings bring further support to the hypothesis that the toxic and genotoxic effects of benzene are produced by several metabolites acting synergistically.  相似文献

Clastogenic effects of acrylamide in mouse bone marrow cells   总被引:5,自引:0,他引:5  
Acrylamide, known to induce dominant-lethal mutations (Shelby et al., 1986; Smith et al., 1986) and heritable translocations (Shelby et al., 1987) in rodent germ cells, was hitherto a questionable clastogen in rodent bone marrow (Shiraishi, 1978). Therefore, it was tested for chromosomal aberrations in mouse bone marrow cells, spermatogonia and by the micronucleus test. The intraperitoneally injected doses ranged from 50 to 150 mg/kg. In the chromosomal bone marrow test and the micronucleus assay positive results were obtained with acrylamide, and in the latter test the effect increased linearly with dose. Chromosomal aberrations were not induced in differentiating spermatogonia by the acute acrylamide treatment. Cisplatin was used as a positive control and gave the expected positive response in all 3 tests. The present results demonstrate that acrylamide is no exception among clastogens. It breaks chromosomes not only in mammalian germ cells but also in somatic cells.  相似文献

Benzene toxicity is considered to be elicited by its metabolites and phenolic metabolites of benzene are known to induce apoptosis in leukemia cells in culture and in human bone marrow progenitor cells. One potential mechanism of apoptosis induced by benzene metabolites that has not been examined is the production of pro-apoptotic cytokines such as endothelial IL-8 from endothelial cells in bone marrow stroma. In this study, we utilized HL-60 cells which are known to produce the endothelial form of IL-8 (elL-8) and human bone marrow endothelial cells (HBMEC) as model systems. Hydroquinone (HQ), Catechol (Cat) and benzenetriol (BT) all induced eIL-8 production and apoptosis in HL-60 cells. HQ induced a marked 50-70-fold stimulation of eIL-8 levels and HL-60 cells were shown to have the eIL-8 receptor, CXCR I thus enabling an autocrine pathway of apoptosis. However, treatment with recombinant elL-8 failed to induce apoptosis in HL-60 cells as previously reported and antibodies to either IL-8 or CXCRI did not significantly abrogate benzene metabolite-induced apoptosis. HQ and Cat but not BT also induced stimulation of elL-8 production in HBMEC. These data demonstrate that although metabolites of benzene induce marked stimulation of eIL-8, this is unlikely to be responsible for apoptosis induced in HL-60 cells. Our data also demonstrates that phenolic metabolites of benzene stimulate the production of eIL-8 from HBMEC suggesting that higher levels of endothelial-derived cytokines may occur in bone marrow after benzene exposure.  相似文献

Benzene is an occupational toxicant and an environmental pollutant that potentially causes hematotoxicity and leukemia in exposed populations. Epidemiological studies suggest an association between an increased incidence of childhood leukemia and benzene exposure during the early stages of pregnancy. However, experimental evidence supporting the association is lacking at the present time. It is believed that benzene and its metabolites target hematopoietic stem cells (HSCs) to cause toxicity and cancer in the hematopoietic system. In the current study, we compared the effects of hydroquinone (HQ), a major metabolite of benzene in humans and animals, on mouse embryonic yolk sac hematopoietic stem cells (YS-HSCs) and adult bone marrow hematopoietic stem cells (BM-HSCs). YS-HSCs and BM-HSCs were isolated and enriched, and were exposed to HQ at increasing concentrations. HQ reduced the proliferation and the differentiation and colony formation, but increased the apoptosis of both YS-HSCs and BM-HSCs. However, the cytotoxic and apoptotic effects of HQ were more apparent and reduction of colony formation by HQ was more severe in YS-HSCs than in BM-HSCs. Differences in gene expression profiles were observed in HQ-treated YS-HSCs and BM-HSCs. Cyp4f18 was induced by HQ both in YS-HSCs and BM-HSCs, whereas DNA-PKcs was induced in BM-HSCs only. The results revealed differential effects of benzene metabolites on embryonic and adult HSCs. The study established an experimental system for comparison of the hematopoietic toxicity and leukemogenicity of benzene and metabolites during mouse embryonic development and adulthood.  相似文献
