降雨对秦皇岛西浴场细菌总数和可培养菌群组成的影响   总被引:3,自引:0,他引:3  
【目的】研究降雨条件对浴场细菌总数和优势菌群组成的影响。【方法】2014年8月强降雨前后采集秦皇岛西浴场3个站位的海水样品,采用荧光显微镜计数法和平板计数法分别对细菌总数和可培养细菌总数进行计数;对群落结构组成进行分析,并对可培养细菌进行鉴定。【结果】雨前3个站位细菌总数和可培养细菌总数平均值分别为5.6×10~9 CFU/L和8.3×10~7 CFU/L,雨后分别为9.2×109 CFU/L和2.1×10~8 CFU/L。在可培养菌群中,变形菌门(Proteobacteria,雨前占80%,雨后占73%)是主要的微生物类群,其次为拟杆菌门(Bacteroides,雨前占12%,雨后占13%)、厚壁菌门(Firmicutes,雨前占7%,雨后占11%)等;肠杆菌属(Enterobacter spp.,21株)、海杆菌属(Marinobacter spp.,13株)、弓形菌属(Arcobacter spp.,13株)、假单胞菌属(Pseudomonas spp.,10株)、芽孢杆菌属(Bacillus spp.,10株)和弧菌属(Vibrio spp.,6株)为雨前可培养细菌优势属,而雨后可培养细菌优势属为肠杆菌属(22株)、海杆菌属(21株)、芽孢杆菌属(14株)、不动杆菌属(Acinetobacter spp.,11株)、假单胞菌属(9株)和弓形菌属(5株)等。【结论】降雨对细菌总数有显著的影响,同时降雨后浴场微生物群落结构发生了改变。  相似文献

为了解醉马草内生细菌的组成和多样性,采用液氮研磨法提取醉马草总DNA,利用内生细菌16S rRNA基因特异性引物对醉马草总DNA 进行16S rRNA 基因扩增,构建醉马草内生细菌16S rRNA基因文库。对筛选到249个克隆进行Hae Ш酶切分析,得到57个操作分类单元(OTUs)。系统发育分析将其归为4个门:变形杆菌门(Proteobacteria)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、拟杆菌门(Bacteroidetes)以及1个未知类群。其中74%的克隆与可培养细菌中的37个属具有高的16S rRNA基因序列相似性 (97%-100%),其中芽孢杆菌属(Bacillus spp.)、红球菌属(Rhodococcus spp.)、黄杆菌属(Flavobacterium spp.)、黄单胞杆菌属(Xanthomonas spp.)最为丰富。另外26%的克隆序列与GenBank中已存细菌16S rRNA基因序列相似性小于96%。研究结果表明,醉马草中可培养内生细菌丰富,多样并且可能存在一些潜在新物种或新类群。  相似文献

生物治理有害藻华因其具有无毒、高效、专一性强等特点,已成为富营养化水体治理的研究热点。本文从溶藻细菌种类、溶藻机制和溶藻化合物等三个方面对近十年已报道的溶藻细菌及其化合物的研究进展进行了归纳和总结,并探讨了溶藻细菌工业化应用研究中遇到的问题及其原因,据此提出了建议以期促进溶藻细菌的工业化应用研究。  相似文献

从安徽镜湖腐烂的蓝藻中筛选出1株细菌,命名为R1,该菌株具有较强的耐镉、溶藻以及可降解微囊藻毒素能力,经16S r DNA序列对比分析鉴定,R1属于气单胞菌属(Aeromonas)。该菌株对Cd~(2+)有很好的耐受性,在含有50 mg/L Cd~(2+)的培养基中仍能正常生长;细菌通过分泌胞外物质溶解铜绿微囊藻,且该胞外物质耐高温,属非核酸或多糖类物质,丙氨酸和苯丙氨酸协同作用而溶藻;在0. 1 mg/L Cd~(2+)存在的情况下,细菌溶藻能力增强;微囊藻毒素的初始浓度为1. 84 mg/L时,7 d时间降解率为40. 2%。  相似文献

为了探索新分离到的3株溶藻细菌胞外溶藻活性物质的分离特性,选择了对水华鱼腥藻生长无抑制作用的淀粉培养基培养溶藻细菌.采用透析、乙醇沉淀、有机溶剂萃取、活性炭吸附与解吸等方法对其分离特性进行了研究.溶藻细菌L7的溶藻活性物质的分子量小于3.5 kD,溶藻细菌L8、L18的溶藻活性物质的分子量在3.5 kD~7 kD之间;3株溶藻细菌的胞外溶藻活性物质不能用乙醇沉淀法完全分离;3株溶藻细菌的溶藻活性物质具较好的亲水性和较强的极性,且都不能被活性炭吸附.  相似文献

为了解特定生态环境中晚稻稻种的非致病细菌多样性以更有效地治理水稻主要病害,于1996~2001年间对采自浙江平原、丘陵及山区的606份稻种样本进行了非致病细菌种类、频率及对水稻纹枯病和恶苗病的抑菌作用研究.从中分离出9605个菌株,经致病性测定、菌落形态及部分细菌学特征(革兰氏染色、KMB培养基上的荧光色素和扩散性非荧光色素、YDC培养基上黄色菌落的产生、好气性、鞭毛及芽孢的染色镜检等)测定后,选出代表菌株622个,连同80个对照菌株用Biolog及脂肪酸分析法(FAME)进行测试.鉴定出Pseudomonas 属11个种或型及其他14属的23种非致病细菌,并发现Pseudomonas 属中55%的种及其他属中49%的种存在对水稻纹枯病或恶苗病的拮抗菌株,但不同种间的抑菌率存在较大差异.3类稻区均以Bacillus spp.、Acinetobacter spp.和P. putida等3个种群为主导并同时存在许多其他相似种,其中7个常见种的分离频率在不同稻区有显著差异.  相似文献

研究以草鱼(Ctenopharyngodon idellus)为实验对象, 运用厌氧培养的方法, 研究了饥饿状态下草鱼肠道黏膜固有微生物的类群及其在不同肠段的分布。实验结果显示草鱼前肠、中肠与后肠中细菌的数量分别是3.17×103、1.63×104和1.79×107 cfu/g。研究共分离到274株单菌落, 经16S rRNA鉴定, 分别属于拟杆菌属(Bacteroides spp.)、鲸杆菌属(Cetobacterium spp.)、梭形杆菌属(Fusobacterium spp.)、气单胞菌属(Aeromonas spp.)、希瓦氏菌属(Shewanella spp.)、芽孢杆菌属(Bacillus spp.)、泛菌属(Pantoea spp.)和柠檬酸杆菌属(Citrobacter spp.)8个种类, 其中专性厌氧细菌的数量占9.1%, 兼性厌氧细菌的数量占90.9%。进一步分析发现, 前肠中只分离到兼性厌氧细菌, 中肠与后肠专性厌氧细菌和兼性厌氧细菌都有分布。专性厌氧细菌Bacteroides paurosaccharolyticus和Bacteroides luti在中肠与后肠都有分布, 而Cetobacterium somerae和Fusobacterium ulcerans只在后肠有发现。兼性厌氧细菌是草鱼肠道黏膜的优势菌群, 其中嗜水气单胞菌Aeromonas hydrophila占73.7%。草鱼肠道不同部位固有厌氧微生物组成存在差异, 细菌数量也明显不同, 后肠中具有更高的细菌丰度和多样性。  相似文献

随着全球水体富营养化的加剧,有害藻类水华和赤潮的爆发日趋频繁,所造成的环境和经济问题日益引起人们的重视.溶藻细菌作为水生生态系统生物种群结构和功能的重要组成部分,对维持藻的生物量平衡具有非常重要的作用.目前在物理、化学和其它生物方法治理水华和赤潮都不够理想的情况下,利用溶藻细菌治理水华,可保持水环境生态平衡,从而达到防止水华的目的.而且随着溶藻细菌的生物学活性不断被发现,以茵治藻将作为水华和赤潮生物防治的一个新对策,具有广泛的研究前景.本文通过查阅近年来国内外有关水生生态系统中溶藻细菌研究的最新研究成果并对溶藻细菌防治有害藻类的种类、分离方法、作用方式及可能的作用机理、作用效应、研究前景和存在问题做一阐述,以期对于细菌溶藻方面的研究及开发应用具有一定的参考价值.  相似文献

滇池中溶藻细菌的分离鉴定及其溶藻效应   总被引:1,自引:0,他引:1  
【背景】藻类水华或赤潮在世界范围内频发,带来各种危害,亟需找到有效途径控制水华或赤潮。溶藻细菌具有杀死藻类控制藻类生物量的能力,可以作为防治水华和赤潮的有效工具。【目的】分离并鉴定滇池中的铜绿微囊藻(Microcystisaeruginosa)及其溶藻细菌,对溶藻菌作用于铜绿微囊藻的溶藻效应进行研究,初步了解其溶藻特性与溶藻机制。【方法】采用LB平板稀释涂布,再经多次划线分离纯化细菌,测定16SrRNA基因序列以鉴定细菌种类;采用毛细管分离的方法分离铜绿微囊藻,并测定其cpcBA基因序列以鉴定蓝藻种类;采用热乙醇法提取叶绿素a,从而计算溶藻效率;基于过氧化氢酶(CAT)、还原型谷胱甘肽(GSH)和丙二醛(MDA)探究藻细胞在溶藻菌处理下的抗氧化系统响应。【结果】共分离获得11株微囊藻和17株针对铜绿微囊藻的高效溶藻菌。选取其中一株生长速度最快的铜绿微囊藻DCM4和一株溶藻效果最好的溶藻菌Sp37 (Bacillus siamensis)进行后续研究。Sp37对DCM4的4 d溶藻率达到92.4%±1.5%,且对微囊藻属的水华微囊藻(M. flos-aquae)和惠氏微囊藻(M.wesenbergii)均有溶藻效果,而对绿藻没有溶藻效果。Sp37的原菌液和无菌滤液对DCM4的4d溶藻率分别为86.8%±4.3%和81.1%±2.2%,两者没有显著差异(P0.05)。Sp37菌体对DCM4的溶藻率为25.4%±7.3%。Sp37无菌滤液经不同温度和pH处理之后的溶藻率与未经处理的无菌滤液的溶藻率无明显差异。Sp37无菌滤液处理藻细胞会使藻细胞的CAT、GSH和MDA含量发生变化。【结论】菌株Sp37对铜绿微囊藻DCM4具有高效的溶藻作用,而且对微囊藻属具有一定的溶藻特异性。Sp37是通过分泌胞外物质间接溶藻,且溶藻物质具有热稳定性和酸碱稳定性。Sp37无菌滤液处理藻细胞会触发藻细胞抗氧化系统,并且会损伤藻细胞膜。Sp37无菌滤液很可能是通过对藻细胞造成氧化胁迫,最终导致藻细胞死亡的。  相似文献

Aims: This work is aiming at investigating algicidal characterization of a bacterium isolate DHQ25 against harmful alga Alexandrium tamarense. Methods and Results: 16S rDNA sequence analysis showed that the most probable affiliation of DHQ25 belongs to the γ‐proteobacteria subclass and the genus Vibrio. Bacterial isolate DHQ25 showed algicidal activity through an indirect attack. Xenic culture of A. tamarense was susceptible to the culture filtrate of DHQ25 by algicidal activity assay. Algicidal process demonstrated that the alga cell lysed and cellular substances released under the visual field of microscope. DHQ25 was a challenge controller of A. tamarense by the above characterizations of algicidal activity assay and algicidal process. Conclusion: Interactions between bacteria and harmful algal bloom (HAB) species proved to be an important factor regulating the population of these algae. Significance and Impact of Study: This is the first report of a Vibrio sp. bacterium algicidal to the toxic dinoflagellate A. tamarense. The findings increase our knowledge of the role of bacteria in algal–bacterial interaction.  相似文献

The aim of this study was to isolate and identify algicidal bacteria against the dinoflagellate Cochlodinium polykrikoides, and to determine the algicidal activity and algicidal range. During the declining period of C. polykrikoides blooms, seven algicidal bacteria were isolated. The algicidal bacteria against C. polykrikoides were enumerated using the most probable number (MPN) method. The number of algicidal bacteria was high (3.7 × 103 mL−1). Algicidal bacteria were identified on the basis of biochemical and chemotaxonomic characteristics, and analysis of 16S rDNA sequences. Seven algicidal bacteria isolated in this study belonged to the genera Bacillus, Dietzia, Janibacter, and Micrococcus. The most algicidal bacterium, designated Micrococcus luteus SY-13, is assumed to produce secondary metabolites. When 5% culture filtrate of this strain was applied to C. polykrikoides cultures, over 90% of C. polykrikoides cells were destroyed within 6 h. M. luteus SY-13 showed significant algicidal activities against C. polykrikoides and a wide algicidal range against various harmful algal bloom (HAB) species. Taken together, our results suggest that M. luteus SY-13 could be a candidate for controlling HABs.  相似文献

Interactions between bacteria and species of harmful and/or toxic algae are potentially important factors affecting both the population dynamics and the toxicity of these algae. Recent reports of bacteria lethal to certain harmful algal bloom (HAB) species, coupled with a rapidly evolving interest in attempting to minimize the adverse effects of HABs through various prevention, control, and mitigation strategies, have focused attention on defining the role of algicidal bacteria in bloom termination. The aim of the present study was to determine whether algicidal bacteria active against Gymnodinium breve Davis, a dinoflagellate responsible for frequent and protracted red tides in the Gulf of Mexico, are present in the waters of the west Florida shelf. To date, we have isolated two bacterial strains from this region lethal to G. breve and have begun to characterize the algicidal activity of one of these strains, 41-DBG2. This bacterium, a yellow-pigmented, gram-negative rod, was isolated from waters containing no detectable G. breve cells, suggesting that such bacteria are part of the ambient microbial community and are not restricted to areas of high G. breve abundance. Strain 41-DBG2 produced a dissolved algicidal compound(s) that was released into the growth medium, and the algicide was effective against the four Gulf of Mexico G. breve isolates tested as well as a closely related HAB species that also occurs in this region, Gymnodinium mikimotoi Miyake et Kominami ex Oda. Nonetheless, data showing that a nontoxic isolate of Gymnodinium sanguineum Hirasaka from Florida Bay was not affected indicate that the algicidal activity of this bacterium does exhibit a degree of taxonomic specificity. Our efforts are currently being directed at resolving several critical issues, including the identity of the algicide(s), the mechanisms regulating its production and ability to discriminate between target algal species, and how the growth rate of 41-DBG2 is affected by the presence of G. breve cells. We have also proposed a conceptual model for interactions between algicidal bacteria and their target species to serve as a testable framework for ensuing field studies.  相似文献

Summary Bacterial isolates from contaminated mint shoot cultures were characterized and identified as a preliminary step in determining an elimination treatment. The 22 bacteria were characterized using biochemical and morphological tests and subjected to sensitivity tests with four antibiotics. The isolates were compared with known organisms and assigned to genera according to similarities in characteristics. Seven isolates were analyzed by fatty acid analysis carried out by a commercial laboratory. Six were classified asAgrobacterium radiobacter; eight asXanthomonas; one each asPseudomonas fluorescens, Micrococcus spp.,Corynebacterium spp., andCurtobacterium spp.; four could not be assigned to genera. Inhibition of growth of the bacteria by most antibiotics was best at pH 7.5. Minimal inhibitory concentration and minimal bactericidal concentrations of gentamicin, rifampicin, streptomycin sulfate, and Timentin varied with genotype.  相似文献

从水体中分离得到一株具有溶藻能力的细菌,命名为NP23。经形态特征、生理生化鉴定和16S rDNA序列分析表明,该菌株属于肠杆菌属(Enterobacter)。研究了该菌株对湖泊中优势藻的溶藻效果,初步探讨了其溶藻方式及溶藻物质。结果表明,该菌株对小球藻、惠氏微囊藻、栅藻和蛋白核小球藻具有一定的去除效果,叶绿素a的去除率分别为64.1%、53.1%、87.2%和84.4%,而且在10-108CFU/mL菌浓度范围内,藻的去除率与菌液的浓度成正相关;该菌株对小球藻、栅藻和蛋白核小球藻是间接溶藻,对惠氏微囊藻是直接溶藻;该菌株对栅藻的溶藻物质是蛋白类物质,对蛋白核小球藻的溶藻因子是菌体胞外分泌的具有热稳定性的非蛋白类物质。  相似文献

The structures of bacterial communities were studied in activated sludge samples obtained from the aerobic and anaerobic zones of a wastewater treatment plant showing enhanced phosphorous removal. Samples were analyzed by in situ hybridization with oligonucleotide probes complementary to selected regions of the 16S and 23S ribosomal RNA (rRNA) characteristic for defined phylogenetic entities (genera and larger groups). The microbial community structures revealed by molecular techniques were compared with the compositions of culturable bacterial communities, obtained from the characterization of 255 isolates from tryptone-soy (TS) agar and R2A agar. These isolates were characterized by 89 physiological tests and their cellular fatty acid patterns, and identified. Culture-dependent techniques indicated the following distribution: different Aeromonas spp. (2.7–8.3% on R2A agar; 45.0–63.7% on TS agar), Acinetobacter spp. (5.4–9.0% on R2A agar; 5.0–9.1% on TS agar), Pseudomonas spp. (up to 10% on R2A agar) and Shewanella putrefaciens (up to 3.0% on R2A agar), all members of the gamma subclass of Proteobacteria, were isolated most frequently. The relatively rare isolates of the beta subclass were identified as Acidovorax spp., Alcaligenes spp., and Comamonas spp., The Gram-positive bacteria (high DNA G+C) were assigned mainly to Arthrobacter spp., Microbacterium spp., and Mycobacterium phlei. In order to assess the in situ abundance of the most frequently isolated genus, Aeromonas, two rRNA-targeted oligonucleotide probes were developed. The two gamma proteobacterial genera Aeromonas and Acinetobacter constituted less than 5% of all bacteria. In situ, Proteobacteria belonging to the beta subclass and high G+C Gram-positive bacteria were dominant. From filamentous bacteria, Sphaerotilus spp. and Leptothrix spp. could be detected occasionally. In addition, one sample contained a high proportion of the morphologically distinct filaments of Microthrix parvicella.As for the genus Acinetobacter, the relative abundance of the most frequently gamma-proteobacterial genus Aeromonas was overestimated by the intrinsic selectivity of cultivation. Microfungi were isolated from soils, mosses, algae and lichens in the Windmill Islands region of Antarctica. From a total of 1,228 isolates, 22 genera were identified. The most frequently isolated fungi from mosses were Mycelia sterilia (47% of total isolates), Phoma spp. (18%), Penicillium spp. (11%), Chrysosporium spp. (7%) and Thelebolus microsporus (6%). Mycelia sterilia, Penicillium spp., Mortierella spp., Chrysosporium cf. pannorum and Thelebolus microsporus were also frequently isolated from algae. Fungal distribution and diversity were poor in samples of lichens, compared to samples from mosses and algae. The frequency of occurrence of microfungi was most often associated with strong biotic influence. There was a marked increase in fungal diversity in human-disturbed sites. Twelve taxa were restricted to soils from near the Australian Casey Station, suggesting significant introduction of fungi into this environment by human activities. Away from the station, fungal distribution appeared to be related to substrata and nutrient status rather than dispersal opportunities. Suggestions for future research and the need for constant monitoring to clarify the role of human disturbance on Antarctic fungi are discussed. Summary The importance of algae in the diet of the oligochaetes Lumbriculus variegatus and Rhyacodrilus sodalis was determined from cellections made in a eutrophic bay from April 1977 to April 1978. During the summer, algae represented 70–85% of the gut contents of both species. The most frequently ingested algae were Cymatopleura elliptica, Cymbella spp., Epithemia turgida, Pinnularia spp., and Synedra ulna. Almost all species were consumed in proportion to their abundance in the environment. However, Nostoc pruniforme and Nostoc verrucosum were not eaten due to size selection. The high level of algal consumption exhibited by the oligochaetes was primarily related to the large standing crop of algae on the sediments. During the winter, when algal densities were low in the environment, detritus and associated bacteria were the major food source.  相似文献

The structure of microbial communities of brown algae, red algae, and of the red alga Gracilaria verrucosa, healthy and affected with thallus rot, were comparatively investigated; 61 strains of heterotrophic bacteria were isolated and characterized. Most of them were identified to the genus level, some Vibrio spp., to the species level according to their phenotypic properties and the fatty acid composition of cellular lipids. The composition of the microflora of two species of brown algae was different. In Chordaria flagelliphormis, Pseudomonas spp. prevailed, and in Desmarestia viridis, Bacillus spp. The composition of the microflora of two red algae, G. verrucosa and Camphylaephora hyphaeoides, differed mainly in the ratio of prevailing groups of bacteria. The most abundant were bacteria of the CFB cluster and pseudoalteromonads. In addition, the following bacteria were found on the surface of the algae: Sulfitobacter spp., Halomonas spp., Acinetobacter sp., Planococcus sp., Arthrobacter sp., and Agromyces sp. From tissues of the affected G. verrucosa, only vibrios were isolated, both agarolytic and nonagarolytic. The existence of specific bacterial communities characteristic of different species of algae is suggested and the relation of Vibrio sp. to the pathological process in the tissues of G. verrucosa is supposed.  相似文献
