本工作利用双室系统观察了金褐霉素对平板脂双层(Planar lipid bilayer)的作用。双室系统包括一个带直径为700μm 小孔的 Teflon 薄膜中隔,和由它隔开的两个充满盐溶液的小室。用脂双层(成膜液为卵磷脂和胆固醇的正癸烷溶液,重量比4∶1)覆盖小孔,在电压箝位下,研究脂双层的电学和通透性质。记录金褐霉素产生的电导变化和单通道电流。实验观察到,在将金褐霉素(终浓度10—20μg/ml)加入小室,20min 左右可记录出通道样活动噪音,脂双层膜电阻下降。它们的发生不依赖于跨膜电位差和离子浓度梯度的存在。将加入的金褐霉素的浓度降低至1.4μg/ml,可获得离散的单位电导涨落的记录。在对称100mmol/L 的 KCl 溶液中,这种单通道活动的优势电导为4—6pS。通过改变两小室的离子浓度,测定平衡电位,可由 Goldmann-Hodgkin-Katz 电场方程推算出通道的离子选择性。结果表明,金褐霉素在脂双层形成的离子通道对 K~+比对 Cl~有较高的通透性(P_K:P_(Ol)≈5.2)。这些结果为金褐霉素增加神经末梢的递质释放,降低肌细胞膜电位,以及为它在临床上的抑菌作用提供了解释。  相似文献

利用由卵磷脂和胆固醇(分别为20和5mg/ml)的正癸烷溶液形成的平板脂双层,观察了由金环蛇毒纯化的膜活性多肽B(BMAP B)形成通道的作用。实验结果表明,在膜两侧有一定电位差或盐浓度梯度的条件下,BMAP B参入脂双层形成离子通道,在电压箝位下可记录出不同电导的单位电导涨落,随着更多通道的形成,脂双层稳态膜电阻明显降低;通过在不对称盐溶液中测定平衡电位,确定BMAP B通道的阴阳离子选择性不强,P_K/P_Cl≈1.4;和在生物膜的作用一样,二价金属离子Ba~(2+),Ca~(2+)抑制BMAP B通道的活动。这些结果为金环蛇毒膜活性多肽在生物膜的去极化作用提供了解释。  相似文献

从菠菜(Spinacia oleracea Mill.)叶中分离获得H~ -ATP酶(CF_0-CF_1)复合体。将CF_0-CF_1重组于平板脂双层上,在电压钳位下,研究CF_0~CF_1的质子传导性能,观察到:(1)当CF_0-CF_1重组于平板脂双层上后,平板膜电阻由10~20GΩ立即下降到1GΩ左右。(2)溶液中蛋白质(CF_0-CF_1)浓度在2mg/L下可记录到单通道电流的涨落,单位电导约在5~10pS。(3)通道电流随膜两侧ΔpH变化而改变,在ΔpH为2~4时,膜电流随ΔpH增加而增大,在ΔpH为4.5时膜电流呈现回落。(4)质子传导抑制剂Dicyclohexyl-carbodiimide(DCCD)显示出迅速地且不可逆地阻断通道电流。(5)无金属离子的溶液中,跨膜(BLM)的ΔpH为3时,在0~ 150mV钳位下,镁离子比钙离子所引起的CF_0-CF_1的通道电流要大得多。以上结果不仅表明CF_0-CF_1已成功地组装于人工膜上,而且也显示出镁离子直接参与了质子传导过程。  相似文献

用水两相法纯化的高粱根质膜嵌入平面脂双层膜,可以测得离子通道电流。记录了典型的单离子通道。TritonX—100使质膜蛋白溶解,除去不溶物后,加入AmberliteXAD—2使膜蛋白重组为蛋白脂质体。将蛋白脂质体嵌入脂双层,测定了脂双层上K+通道电流逐渐出现的过程,该典型的通道电流记录的通道电导为112pS。通过改变前后槽KCl浓度,测定蛋白脂质体嵌入脂双层后的稳态电流一电压关系,近似计算稳态逆转电位,由GHK等式得K+和Cl-的选择通透比Pk:Pcl为8.6主要是通透K+。关键词:  相似文献

目的:将提取的猪心室肌细胞膜上K^ 通道重装在磷脂双层上,用电压嵌方法研究离子通道。方法:将猪心室肌制成匀浆,通过蔗糖梯度离心,分离出通道蛋白成分,利用双室系统将其重装在人工膜上,在电压籍位下记录通道电流。结果:提取的通道蛋白成分优势电导为27~31ps,此外还记录到有15,50和100ps的几种通道活动,其中以27~31ps最为常见。经测定反转电位,确定它们为K^ 选择性通道。结论:本研究在国内首次完成了心肌钾通道在人工脂双层膜上的重装,为在单通道基础上研究K^ 通道提供了重要手段。  相似文献

跨膜离子流动在启动配子间的相互作用,如顶体反应过程中有重要作用。.但由于精子太小,无用论细胞内记录或膜片箝技术都难以对哺乳动物精子膜的离子通病进行研究。本工作将通道蛋白重组于脂双层,在电压箝位下记录了人精子膜的Ca^2+通道电流。由12个健康人的精子分离的膜蛋白通过融合重组于脂双层后,在CaCl2溶液中(cis50//trans 10mmol/L)记录到两类单通道活动,它们的平衡电位都接近Ca^2  相似文献

用微电极技术对新生大鼠脊髓横切薄片运动神经元(MN)进行细胞内记录,测得静息电位为-62±4mV(n=26),膜电阻为67±31MΩ,时间常数3.8±1.6ms,动作电位幅度68±7mV(n=26),阈电位-50±8mV,超射值6±4mV。灌流谷氨酸(1~30mmol/L)诱导伴膜电阻降低的缓慢去极化反应,5-羟色胺(50μmol/L)介导伴膜电导降低的电压依赖性内向电流。结果表明新生大鼠脊髓切片MN的细胞内生物电记录是一种稳定可靠的电生理学和药理学研究方法。  相似文献

半夏凝集素(PTL)可与甘露聚糖结合。本文以PTL与~(125)I标记的甘露聚糖的结合活性为指标,观察了一些金属离子对PTL的糖结合活性的影响,井对PTL的糖结合专一性作了较系统的研究。结果表明常见的金属离子或EDTA对其糖结合活性无显著影响,但K~+可明显增加PTL的糖结合活性。大多数单糖,二糖不抑制PTL与甘露聚糖的结合,但一些疏水配基形成的糖苷可产生显著的抑制效应。PTL专一与高甘露糖型糖链结合。  相似文献

用细胞内电位记录技术,以离体蟾蜍交感链为标本,观察了尖吻蝮蛇毒(AAV)的神经毒性作用。结果表明,该蛇毒(10-200μg/ml)对交感神经元的静息电位、膜电阻和膜电容没有显著的作用,对动作电位形状也无可测出的影响,但能使阈电位轻皮升高,即使神经元兴奋性稍降低。AAV(>25μg/ml)对胆碱能性的快兴奋性突触后电位有剂量依从性的,部分可逆性的抑制作用。其作用机制至少应部分归之于AAV对突触后膜上的N型胆碱能受体的阻断作用。本研究发现的AAV神经毒性作用可能有一定实践意义。  相似文献

从半夏(Pinellia ternata)块根中,经95%硫酸铵沉淀和固定化猪甲状腺球蛋白柱亲和层析分离得到一种凝集素,称为半夏凝集素(PTL)。凝胶过滤、SDS-PAGE和免疫双扩散鉴定PTL是均一的样品,分子量约44kD,由四个约12kD的亚基组成。PTL主要存在于半夏的块茎中,茎中也有少量存在。  相似文献

The ligand specificity of the human monocyte receptor that mediates phagocytosis of particulate activators of the human alternative complement pathway was defined by inhibiting the phagocytic response with glycans known to be present in zymosan. When monocytes in monolayers were preincubated with 100 micrograms/ml of beta-glucan and then incubated with 1.25 to 2.5 X 10(6) zymosan particles, the percentage of cells that exhibited phagocytosis was inhibited in a time-dependent manner; maximal inhibition occurred within 20 min of preincubation. beta-Glucan inhibited monocyte phagocytosis of zymosan and rabbit erythrocytes (Er) in a similar dose-dependent fashion and at 100 micrograms/ml reduced monocyte ingestion of 5 X 10(6)/ml zymosan and 2 X 10(8)/ml Er by 63 +/- 8% and 68 +/- 16% (mean +/- SD, n = 3), respectively. The other glycan constituent of zymosan, mannan, was less than 1% as active, and 10 mg/ml of mannan reduced the number of monocytes ingesting zymosan and Er by 56 +/- 12% and 26 +/- 11%, respectively. At concentrations as high as 500 micrograms/ml, beta-glucan had no effect on monocyte Fc, C3b, or fibronectin receptor-mediated functions. Enzymatic hydrolysis of beta-glucan and alpha-mannan with beta-glucosidase or beta-glucanase before their incubation with monocytes abrogated their inhibitory capacity, whereas hydrolysis with alpha-mannosidase or alpha-glucosidase did not. Neither of the two alpha-glucans tested (dextran T-70 and nigeran) affected monocyte ingestion of zymosan particles or sheep erythrocytes (Es) sensitized with rabbit 7S anti-Es (EsIgG) at concentrations as high as 2 mg/ml. In contrast, a number of beta-glucans were active against zymosan but not EsIgG ingestion with a 75% reduction in the number of monocytes ingesting zymosan occurring with 100 micrograms/ml laminarin, 500 micrograms/ml soluble pachyman, and 900 micrograms/ml of soluble pustulan. The galactan, agarose, either in suspensions at 2 mg/ml or in a soluble portion at 600 micrograms/ml failed to affect monocyte ingestion of zymosan particles or Er. Thus, the monocyte receptor for particulate activators that is specifically inhibited by beta-glucan at a rate compatible with a phagocytic process and that recognizes beta-glucans but not alpha-glucans, mannan, or galactan is a beta-glucan receptor.  相似文献

The dependence of the surface potential difference (delta U), transversal elasticity module (E1) and membrane conductivity (G0) on the concentrations of the antiviral drugs, rimantadine and amantadine was studied in the planar bilayer lipid membrane system. The method used was based on independent measurements of the second and third harmonics of the membrane capacitance current. The binding constants of bilayer lipid membranes obtained from the drug adsorption isotherms were 2.1 X 10(5) M-1 and 1.3 X 10(4) M-1 for rimantadine and amantadine, respectively. The changes in G0 took place only after drug adsorption saturation had been achieved. The influence of rimantadine and amantadine on the interaction of bilayer lipid membranes with matrix protein from influenza virus was also investigated. The presence of 70 micrograms/ml rimantadine in the bathing solution resulted in an increase in the concentration of M-protein at which the adsorption and conductance changes were observed. The effects of amantadine were similar to those of rimantadine but required a higher critical concentration of amantadine. The results obtained suggest that the antiviral properties of rimantadine and amantadine may be related to the interaction of these drugs with the cell membrane, which can affect virus penetration into the cell as well as maturation of the viral particle at the cell membrane.  相似文献

The glycans used in an earlier study to define the ligand specificity of the human monocyte phagocytic receptor for unopsonized particulate activators were assessed for their capacities to activate the proteins of the human alternative complement pathway. Normal human serum was preincubated with glycans under conditions of chelation to prevent activation of the classical complement pathway, and the activation-depletion of the alternative complement pathway was determined by the subsequent capacity of the serum to lyse rabbit erythrocytes (Er). When serum was preincubated at a 1/2 dilution in 8 mM EGTA/2 mM Mg with increasing numbers of yeast glucan or zymosan particles, and was evaluated at final serum dilutions of 1/8, its capacity to lyse Er was found to be reduced by 50% with 1.9 X 10(6)/ml yeast glucan particles and 1.4 X 10(6)/ml zymosan particles. At 2 mg/ml of serum diluted 1/2 in 8 mM EGTA/2 mM Mg, nonturbid preparations of mannan, laminarin, or pyrogen-free inulin and turbid suspensions of cellulose, Sephadex, agarose, or purified inulin failed to activate the alternative complement pathway. In contrast, activation-depletion of the alternative pathway was induced by turbid preparations of crude inulin, nigeran, pachyman, barley beta-glucan, and pustulan, which at 700 micrograms/ml, 500 micrograms/ml, 350 micrograms/ml, 60 micrograms/ml, and 27 micrograms/ml, respectively, effected 50% reductions in the subsequent lysis of Er. After centrifugation of 2 mg/ml suspensions of barley beta-glucan at 1100 X G for 5 min and at 15,000 X G for 15 min, the supernatants contained 90 to 92% and 65% of the barley beta-glucan, respectively, as determined by the anthrone method. On a weight basis, the 1100 X G supernatant exhibited the same capacity to activate the alternative pathway as the corresponding original suspension, whereas the 15,000 X G supernatants had less than 3% of the original anti-complementary activity. Preincubation of adherent human monocytes with increasing concentrations of barley beta-glucan suspensions, 100,000 X G supernatants containing 64% of the original beta-glucan, and laminarin all decreased subsequent ingestion of 1.25 X 10(6) zymosan particles in a dose-related fashion. The numbers of monocytes from three different donors phagocytosing zymosan were reduced by 50% after pretreatment with 30 to 65 micrograms/ml, 25 to 48 micrograms/ml, and 12 to 15 micrograms/ml of barley beta-glucan suspensions, 100,000 X G supernatants of barley beta-glucan, and laminarin, respectively, even though the latter two preparations were fully soluble and had no capacity to activate the alternative pathway.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

The mechanism by which synexin mediates calcium-dependent aggregation of medullary cell chromaffin granules and fusion of granule ghosts involves specific interactions with the lipid component of the membrane. To study the details of these interactions we measured synexin-induced changes in capacitance of phosphatidylserine bilayers formed at the tip of a patch pipet using the double-dip method. Provided calcium was present in the solution filling the pipet (10-50 mM) stable phosphatidylserine bilayers were easily formed. Addition of synexin (0.1 microgram/ml) to an external medium lacking added calcium induced no measurable changes in either bilayer resistance (10-30 G omega) or displacement current across the membrane. However, addition of calcium (0.1-2.5 mM) in the presence of synexin in the external solution caused a marked increase in the size and time constant of decay of the displacement current. From the steady-state value of the current we calculated a 5-fold decrease in resistance and from the charge displaced during the voltage-clamp pulses we calculated a 10-fold increase in membrane capacitance (from 20 to 200 fF). The size of the synexin-specific charge displacement in one direction during a pulse was always equal to the charge returning to the original configuration after the pulse. The synexin-specific transfer of charge reached saturation when the pipet potential was taken to a sufficient positive or negative value. These properties of the extra charge movement support our view that in the presence of calcium the cytosolic protein synexin penetrates into the bilayer. It is possible that these properties may be related to the mechanism by which synexin promotes membrane fusion in natural membranes.  相似文献

Analyses of randomly amplified polymorphic DNA (RAPD) and PCR-restriction fragment length polymorphism (PCR-RFLP) were performed in an effort to distinguish between two different origins of Pinellia ternata. To determine whether the origin of Pinellia ternata is in China or Korea. RAPD analysis was carried out using ten 20-mer random primers. Although the coefficients of similarity between the DNA of Chinese and Korean accessions of Pinellia ternata were high, distinguishable band patterns were observed in the reaction performed using primer numbers 3, 6 and 10. Primer 3 produced one (410 bp) and primer 6 produced four (410 bp, 350 bp, 300 bp, 250 bp) Chinese Pinellia-specific fragments. Primer 10 produced one (900 bp) Korean Pinellia rhizome-specific fragment. In addition, using PCR-RFLP analysis, different fingerprints were obtained from Korean and Chinese Pinellia ternata respectively. These results suggest that the analyses with RAPD and PCR-RFLP can be used to authenticate the relevant Chinese and Korean herbal medicines.  相似文献

RNA沉默是真核生物体内由病毒来源的干扰小RNA(virus derived small interfering RNA, vsiRNA)沉默复合物介导目标RNA特异降解的一种保守机制,通过对vsiRNA分析可进行植物病毒病原鉴定。本文利用小RNA深度测序技术对感病半夏叶片进行鉴定,结果发现,表现典型花叶症状的半夏叶片受到大豆花叶病毒(Soybean mosaic virus, SMV)、黄瓜花叶病毒(Cucumber mosaic virus, CMV)、芋花叶病毒(Dasheen mosaic virus, DsMV)、魔芋花叶病毒(Konjac mosaic virus, KoMV)、烟草花叶病毒(Tobacco mosaic virus, TMV)等多种病毒的复合侵染。为明确SMV山西半夏分离物(SMV-SXBX)的进化关系,进行SMV-SXBX全基因组克隆与分析,获得SMV-SXBX全长为9 735 nt,编码一个由3 105个氨基酸组成的多聚蛋白质。通过核苷酸与氨基酸序列比对发现,SMV-SXBX与半夏分离物P同源性最高,分别为91.1%和94.1%,且系统发育分析表明,SMV-SXBX与半夏SMV分离物P聚为一簇。同时,也对vsiRNA进行了系统分析,研究结果有望为半夏SMV的有效防治提供一定科学依据。  相似文献
