The induction of non-disjunction by X-irradiation of the second chromosome in stage-7 oocytes of Drosophila melanogaster has been studied by employing isochromosome stocks. This makes the quantitative recovery possible of progeny resulting from disomic and nullosomic eggs. Determination of egg hatchability has been used to correct for varying degrees of segregation in males carrying different isochromosomes. Even at exposures as low as 250 R the frequency of non-disjunction is significantly higher than in the controls. No evidence has been obtained for the existence of a threshold. In the stage-7 oocytes, the induction of non-disjunction increased linearly with radiation exposure over a range of 250–3000 R and thus seems to reflect a single-hit event. These findings could be of significance for the evaluation of genetic radiation hazards in man. In slightly younger oocyte stages the induction of disomic eggs followed dose-square kinetics. The frequency of nullosomic eggs rises exponentially with radiation exposure, presumably as a consequence of increasing chromosome loss resulting from unrestituted breaks in each of the two maternal isochromosomes. Furthermore, it was observed that the late stage-7 oocytes were more sensitivie to the induction of non-disjunction than earlier stages.  相似文献

The experimental material comprised 7 ostrich families (7 males and 14 females) of which five families were at the age of 7 and two at the age of 5 years. In the course of the entire reproductive season, the following parameters were analysed: length of the laying period, mean laying rate, number of eggs laid by one female, proportion ofhatching eggs, egg weight and shape, egg weight lost during incubation, egg fertilisation, percentage of dead embryos and unhatched chicks, hatchability from fertilised and set eggs. Seven year-old ostriches were characterised by shorter laying period (134 days) but, at the same time, by higher proportions of hatching eggs. This group was also characterised by high egg fertilisation (79.7%) as well as high hatchability indices at simultaneous highest embryo mortality during incubation (11.6 %). Five year-old ostriches exhibited a longer laying period (175 days) during which females laid more eggs (49 pcs.). In addition, this group was characterised by a smaller proportion of hatching eggs, better egg fertilisation indices (83.5%) and hatchability results. Moreover, the determined higher egg shape index indicates that the 5 year-old females laid eggs which were more spherical. Recapitulating, the obtained results indicate that, under Polish conditions, better indices of laying performance, egg fertilisation and hatchability were observed in the group of 5 year-old ostriches.  相似文献

Summary Entire reproductive tracts dissected from live femalePseudoterranova decipiens, some collected from grey seals (Halichoerus grypus) and some raised in vitro, were examined using light and electron microscopy. The reproductive tracts from both samples are similar in that the oogonia accumulate cytoplasmic inclusion granules and remain attached to the rachis until just before entering the oviduct. Sperm stored in the oviduct fertilize the oocytes, which then pass into the uterus where elaboration of the shell occurs. Two polar bodies are evident in recently fertilized eggs, suggesting that reduction division proceeds as in most nematode eggs. The epithelial cells of the oviduct appear to secrete material that surrounds the oocytes, and the epithelial cells of the uterus secrete a fibrous material that adheres to the outside of the egg shell. The two samples differ in that the oocytes of in vivo-raised nematodes contain curious conglomerates of organelles: areas of membranous whorls in association with electron-dense inclusion granules and glycogen granules. The samples differ also in that the ovarian epithelial cells in the in vitro-raised specimens phagocytose necrotic oogonia at the tip of the ovary.  相似文献

早期胚胎显微注射是目前获得转基因家蚕Bombyx mori的主要途径。显微注射操作对蚕卵的损伤导致注射后的蚕卵孵化率降低, 是家蚕转基因工作的主要障碍之一。本研究对不同卵龄的蚕卵进行了开孔或注射实验, 并对产后5 h的蚕卵上背侧、 腹侧、 前极、 后极和中央等5个不同的位置进行了开孔实验, 调查了卵孵化率和体形异常蚕的产生情况。结果表明: 较早卵龄期的注射或从蚕卵背侧的注射可以获得高的孵化率。腹侧注射产生大量的体形异常蚕而背侧注射的蚕完全正常。通过调整注射时期和注射位置避开上述影响可以减少死卵和畸形蚕, 提高孵化率。本研究为改进家蚕转基因操作技术提供了有效的参考。  相似文献

温湿度对稻纵卷叶螟卵的联合作用   总被引:1,自引:0,他引:1  
为了探讨温湿度在稻纵卷叶螟Cnaphalocrocis medinalis (Guenée)种群发展中的作用, 通过室内实验调查了不同温度和湿度组合下该蛾卵的发育历期、 胚胎发育情况、 孵化率和卵粒重量的变化。结果表明: 在相同温度下卵历期随相对湿度的增大而缩短, 孵化率随相对湿度的加大而提高。在22℃下低于46%的相对湿度显著降低了卵的孵化率, 而在25~34℃下低于66%的相对湿度会引起孵化率的显著降低, 37℃下卵无论在何种湿度中均不能孵化。在50%左右的低湿条件下, 温度高于28℃后卵也不能孵化。温度在22~31℃和相对湿度在77%~100%范围内, 卵的孵化率无显著差异, 这属于稻纵卷叶螟卵的适宜温湿度范围。稻纵卷叶螟卵的发育起点温度和有效积温分别为10.1±0.6℃和63.7±3.5日度。卵的孵化率(Y)与温湿系数(RH/T)间呈显著的逻辑斯蒂曲线关系Y=0.8662/[1+exp(17.4084-7.5714×RH/T)]。温湿系数在2.34以下时卵孵化率将低于50%, 而达到3.0左右时孵化率接近最高值。结论认为, 低湿造成的稻纵卷叶螟卵重量显著降低、 卵粒干瘪、 胚胎发育受阻是致死卵的主要原因。  相似文献

Immunological approaches were used to characterize an antigen that is present within the cortical granules of mouse oocytes and eggs. Immunoelectron microscopy shows a specific localization of the antigen to the cortical granules in the cortex of mouse oocytes and eggs. Following in vitro fertilization, the antigen is present in the perivitelline space and is associated with the zona pellucida. No cortical granules and very little antigen are detected in the two-cell embryo. This antiserum detects a protein of Mr = 75,000 (p75) following immunostaining of egg proteins on Western blots, or immunoprecipitation of metabolically labeled oocyte proteins or radio-iodinated egg proteins. p75 is also present in exudates obtained from A23187-treated eggs, as detected by either radio-iodination of the released egg proteins, or maturation and ionophore activation of metabolically labeled oocytes. Two-dimensional gel electrophoresis of radio-iodinated egg proteins reveals four species of p75 with pIs between 4.9 and 5.3, whereas only the most basic form of p75 is detected in metabolically labeled oocytes. Multiple forms of the radio-iodinated p75 are present in the exudate of ionophore-treated eggs. p75 displays a greater electrophoretic mobility under nonreducing conditions, indicating the presence of intramolecular disulfide bonds, a common characteristic of secreted proteins. We conclude that p75 is synthesized in oocytes, modified and packaged into cortical granules, and released from eggs following fertilization or activation.  相似文献

Effects of mutations rad201, mei-9, and mei-41 on cell sensitivity to gamma-radiation in Drosophila oogenesis were studied. Females of the control (Oregon R) and mutant strains were irradiated at a dose of 15 Gy. For 9 days after the irradiation, the number of eggs in consecutive day batches, the frequency of dominant lethals (DLs) among the eggs, and the cytologically recorded distribution of oocytes for stages of their development, and the frequency of egg chamber degeneration in female ovaries were estimated. As a result of joint analysis of the data, different oogenesis stages were characterized with regard to the frequency of two radiation-induced events: appearance of DLs in oocytes and degeneration of egg chambers due to apoptosis of nurse cells. It was shown that the mutations affect these parameters only at particular stages of early oogenesis, at which previtellogenetic growth of egg follicles and meiotic recombination in oocytes occur. Mutation rad201G1 increased the frequency of DLs and egg chamber degeneration, mei-41D5 affected only the DL frequency, and mei-9a, in addition to enhancing the chamber degeneration frequency, promoted radiation "rescue" of some oocytes from the DL induction.  相似文献

Adult female fruitflies exposed to colchicine admixed to the culture medium show a series of dosage-related abnormalities that affect oogenesis and may induce sterility. Among the effects observed were decreased fecundity and hatchability of laid eggs, formation of oocytes lacking chorionic appendages, abnormal distribution and diminution in number of yolk spheres, inhibition of oocyte growth and abnormally located oocyte nuclei. Potentially the most significant effect was the development of egg chambers which contained the normal complement of 16 cells but in which all the cells had the nuclear morphology of nurse cells. The approach provides for the first time an experimental means to divert a potential oocyte into the developmental pathway of the nurse cell in a wild-type fly, and hence should be helpful in the elucidation of factors which control oocyte and nurse cell differentiation. In addition, the results serve to expand the usefulness of oogenesis in Drosophila as a model system for the evaluation of drug-induced metabolic-morphologic abnormalities.  相似文献

Drosophila melanogaster male accessory gland proteins (Acps) that are transferred in the ejaculate with sperm mediate post-mating competition for fertilizations between males. The actions of Acps include effects on oviposition and ovulation, receptivity and sperm storage. Two Acps that modulate egg production are Acp26Aa (ovulin) and Acp70A (the sex peptide). Acp26Aa acts specifically on the process of ovulation (the release of mature eggs from the ovaries), which is initiated 1.5 h after mating. In contrast, sperm storage can take as long as 6-9 h to complete. Initial ovulations after matings by virgin females will therefore occur before all sperm are fully stored and the extra eggs initially laid as a result of Acp26Aa transfer are expected to be inefficiently fertilized. Acp26Aa-mediated release of existing eggs should not cause a significant energetic cost or lead to a decrease in female lifespan assuming, as seems likely, that the energetic cost of egg laying comes from de novo egg synthesis (oogenesis) rather than from ovulation. We tested these predictions using Acp26Aa(1) mutant males that lack Acp26Aa but are normal for other Acps and Acp26Aa(2) males that transfer a truncated but fully functional Acp26Aa protein. Females mating with Acp26Aa(2) (truncation) males that received functional Acp26Aa produced significantly more eggs following their first matings than did mates of Acp26Aa(1) (null) males. However, as predicted above, these extra eggs, which were laid as a result of Acp26Aa transfer to virgin females, showed significantly lower egg hatchability. Control experiments indicated that this lower hatchability was due to lower rates of fertilization at early post-mating times. There was no drop in egg hatchability in subsequent non-virgin matings. In addition, as predicted above, females that did or did not receive Acp26Aa did not differ in survival, lifetime fecundity or lifetime progeny, indicating that Acp26Aa transfer does not represent a significant energetic cost for females and does not contribute to the survival cost of mating. Acp26Aa appears to remove a block to oogenesis by causing the clearing out of existing mature eggs and, thus, indirectly allowing oogenesis to be initiated immediately after mating. The results show that subtle processes coordinate the stimulation of egg production and sperm storage in mating pairs.  相似文献

The initial eggs produced by broiler breeder hens are relatively small compared with later in the production cycle. An evaluation of indices related to hatchability is required when these eggs are to be used for the production of broiler chicks. Two experiments were conducted to evaluate characteristics related to the hatchability of eggs from pullet-to-breeder transition phase, at 25 and 27 weeks of age, and from the peak of production period and five weeks later, at 32 and 37 weeks of age. Eggs from birds 25 weeks had a lesser fertility in Experiment 1. Mortality occurred unevenly in early (1–5 days), middle (6–17 days) and late (18–21 days) incubation, and greater mortality was observed after the internal membrane was ruptured. The younger the hen, the lighter the egg, chick, and shell, and the longer the time required to complete the hatching process. In Experiment 2, greater mortalities were observed at the early period (1–5 days) and after “pipping” of the internal and external membranes. Embryos from heavy eggs of breeder hens 37 weeks of age took less time to complete the hatching process. Results indicated the larger the egg, the heavier the chick and shell, and the lesser the shell percentage. As breeder age advanced, characteristics related to egg fertility and hatchability improved.  相似文献

In the frog, Rana temporaria, the number of eggs in a clutch ranged from about 2500 to 9000/100g body mass and egg diameter ranged from 1.1 to 1.9 mm. There was an inverse relation between number and size of eggs. It is indicated that egg size depends upon the number of oocytes recruited to vitellogenic growth and that the number of oocytes recruited depends upon the number of small oocytes that constitutes the recruitment pool. The total amount of yolk deposited in the ovaries was independent of the number of eggs, i. e. clutch size. During an ovarian cycle, therefore, physiological mechanisms seem to regulate the amount of yolk deposited within the ovaries at large, rather than in the individual vitellogenic oocyte.  相似文献

Abstract Female yellow dung flies have large paired accessory reproductive glands, the function of which remains unclear. However, gland contents are secreted during copula and egg laying. Other female flies produce a range of anti-bacterial substances in their accessory reproductive glands that protect them and their eggs from pathogens, and it is possible that gland secretion acts similarly in yellow dung flies. A series of experiments was conducted to test this idea. Because the volume of secretion remaining in the glands is negatively related to copula duration, egg hatchability and longevity of females was compared in groups that copulated experimentally once or three times. A zone inhibition assay was used to see if gland extract inhibited bacterial growth. Egg number was positively associated with female body size, but the proportion of eggs hatching was not. Neither copula number nor duration influenced egg number or hatch number or proportion. In accordance with this, gland extract did not inhibit bacterial growth. However, female longevity was reduced in females that copulated with more males. This suggests that gland secretion does not serve to protect eggs, but as with a number of taxa, copula is costly to female yellow dung flies.  相似文献

In mammals, the final number of oocytes available for reproduction of the next generation is defined at birth. Establishment of this oocyte pool is essential for fertility. Mammalian primordial germ cells form and migrate to the gonad during embryonic development. After arriving at the gonad, the germ cells are called oogonia and develop in clusters of cells called germ line cysts or oocyte nests. Subsequently, the oogonia enter meiosis and become oocytes. The oocyte nests break apart into individual cells and become packaged into primordial follicles. During this time, only a subset of oocytes ultimately survive and the remaining immature eggs die by programmed cell death. This phase of oocyte differentiation is poorly understood but molecules and mechanisms that regulate oocyte development are beginning to be identified. Heat treatment of food-processing facilities involves using elevated temperatures (50-60 degrees C for 24-36 h) for management of stored-product insects. Heat treatment is a viable alternative to the fumigant methyl bromide, which is phased out in the United States as of 2005 because of its adverse effects on the stratospheric ozone. Very little is known about responses of the cigarette beetle, Lasioderma serricorne (F.) (Coleoptera: Anobiidae), a pest associated with food-processing facilities, to elevated temperatures. Responses of L. serricorne life stages to elevated temperatures were evaluated to identify the most heat-tolerant stage. Exposure of eggs, young larvae, old larvae, and adults during heat treatment of a food-processing facility did not clearly show a life stage to be heat tolerant. In the laboratory, exposure of eggs, young larvae, old larvae, pupae, and adults at fixed times to 46, 50, and 54 degrees C and 22% RH indicated eggs to be the most heat-tolerant stage. Time-mortality responses at each of these three temperatures showed that the time for 99% mortality (LT99) based on egg hatchability and egg-to-adult emergence was not significantly different from one another at each temperature. Egg hatchability alone can be used to determine susceptibility to elevated temperatures between 46 and 54 degrees C. The LT99 based on egg hatchability and egg-to-adult emergence at 46 degrees C was 605 and 598 min, respectively, and it decreased to 190 and 166 min at 50 degrees C and 39 and 38 min at 54 degrees C. An exponential decay equation best described LT99 as a function of temperature for pooled data based on egg hatchability and egg-to-adult emergence. Our results suggest that during structural heat treatments eggs should be used in bioassays for gauging heat treatment effectiveness, because treatments aimed at controlling eggs should be able to control all other L. serricorne life stages.  相似文献

The ability to store different insect stadia for prolonged periods provides considerable flexibility and ability to conduct experiments properly. Therefore, studies were undertaken to determine the effect of storage temperature and duration on viability of eggs of Helicoverpa armigera (Hübner). The percentage egg hatch and incubation period were significantly (P=0.01) influenced by egg age, storage temperature, and storage duration. Egg hatch ranged from 0.0 to 96.8% across temperatures and storage durations. None of the eggs hatched when stored at -20 and 0 degrees C. The regression model with the optimum Mallow Cp statistic for any of the identified linear and quadratic terms did not improve the precision of prediction in egg hatch beyond 67.0%. Forecasting of incubation period based on egg age, storage duration, and durationxtemperature was quite effective (R2=84.2%). Day degrees required for egg hatching decreased with an increase in temperature from 10 to 27 degrees C, and egg age from 0 to 3 days. The day degree requirements were highest for 0-day-old eggs at 10 degrees C, and lowest at 27 degrees C. Although the incubation period was higher, the hatchability was lower for 0- and 1-day-old eggs stored at constant 10 degrees C, these eggs can be stored for 10 days at 10 degrees C, with a hatchability of >75.0%. It was safer to store the H. armigera eggs for 10 days at 10 degrees C, which will hatch within 1.6 to 2.0 days after restoration at 27 degrees C with a hatchability of >75.0%. This information will be useful in planning and execution of experiments involving H. armigera on various aspects of research in entomology.  相似文献

Japanese quail (Coturnix coturnix japonica) embryos were irradiated continuously in ovo with 2.45-GHz continuous wave radiation during the first 12 days of embryogenesis at an incident power of 5 mW/cm2 and a specific absorption rate of 4.03 mW/g. The internal temperature of irradiated and nonirradiated (sham) eggs was 37.5 +/- 0.3 degrees C, which is the optimum temperature for incubating quail eggs. At 35 days after hatching irradiated and sham-irradiated males were paired with irradiated or sham-irradiated females and daily records of reproductive performance were collected through 224 days of age. Progeny were hatched from each of the male-female pairs, and progeny reproductive performance was measured from 35 through 168 days of age. Hatchability was not affected by irradiation during embryogeny. Mortality after hatching, egg production, egg weight, fertility, hatchability of eggs produced, and reproductive performance of the progeny were not affected by irradiation during embryogeny. These observations indicate that irradiation of quail embryos with low-level microwave radiation does not affect the reproductive capacity of the hatchlings or of progeny produced from quail irradiated during incubation.  相似文献

Solar ultraviolet-B (UVB) radiation has deleterious effects on plant-dwelling mites. We assessed the biological effects of UVB radiation on the eggs of the twospotted spider mite, Tetranychus urticae Koch, under both near ambient (UV+) and UV-attenuated (UV−) conditions from spring to autumn and compared them to the effects of temperature and humidity. The ambient daily UVB irradiance increased from January to August and then decreased rapidly until December, whereas egg hatchability under UV+ was lowest in April (10.7%) and increased almost linearly until October (74.9–92.3%). In contrast, hatchability under UV− was consistently high (96.2–99.8%) through all seasons. For UV+, the stepwise multiple linear regression analysis supported the negative correlation of hatchability with cumulative UVB irradiance during egg periods (cumulative dose), but did not support that with the mean daily UVB irradiance (dose rate), suggesting that UVB-induced mortality in T. urticae eggs is cumulative dose dependent rather than dose rate dependent. The high mortality in April may have reflected the slower development caused by the relatively lower temperature and higher UVB radiation, increasing the cumulative dose, while the low mortality in October may have reflected the faster development caused by the relatively higher temperature and lower UVB radiation, decreasing the cumulative dose.  相似文献

The complete life history of Chorda tomentosa Lyngbye from northern Norway has been followed in culture. Under relatively high temperatures (10–15°C) or low irradiance, zoospores develop into filamentous monoecious gametophytes with unlimited vegetative growth. Formation of oogonia and antheridia was induced by transfer to strong white fluorescent light and low temperatures (1–5°C). By variation of these environmental factors the degree of fertility can be controlled. In a light-dark regime, egg release occurs exclusively during the dark periods. Freshly released eggs secrete a sexual hormone which effects explosive discharge of spermatozoids from the antheridia and subsequent chemotaxis towards the egg. Plasmogamy occurs immediately. Chromosome staining reveals interesting nuclear activities during karyogamy. Frequently, unfertilized eggs develop parthenogenetically. The resulting sporophytes are haploid and show the same developmental pattern as those originating from fertilized eggs. All sporophytes produce sporangia and release zoospores within 60 to 90 days after egg discharge.  相似文献

在浙江温岭沿海逐月采样,显微观察了可口革囊星虫体腔中卵细胞大小组成的周年变化。3—5月,卵径30~90μm的卵细胞占大部分。6月,卵径70~130μm的卵细胞占多数。7月,大部分卵细胞卵径达100μm以上,其中130~150μm的卵细胞约占1/3。8月,卵细胞卵径均在110μm以上,其中130~150μm的卵细胞约占2/3。9月,仍多数卵细胞卵径在110μm以上,130~150μm的卵细胞约占1/3。10月,以110~140μm的卵细胞占多,40μm以下的卵细胞较少,大卵细胞开始退化。11月至翌年2月,随着大型卵细胞的逐渐退化,小型及中型卵细胞逐渐增多,2月仍可见少量未退化的大型卵细胞。分析认为,可口革囊星虫在浙江温岭的繁殖期主要在7—9月。  相似文献

The effects of Mycoplasma gallisepticum (MG) infections on egg production, fertility, and hatchability were studied in captive-reared wild turkeys (Meleagris gallopavo). Three groups of adult birds, each consisting of four hens and two toms, were exposed to MG by the respiratory route at the beginning of their breeding season. Fourteen control birds received sterile growth medium. Although no mortality of infected or control birds occurred, egg production during the first breeding season after infection was reduced. The mean number of eggs/hen/day produced by infected groups the first breeding season postexposure (PE) was significantly lower than the control value. The mean number of eggs produced daily by the same hens 1 yr later was unaffected by MG infection. The percentage of fertile eggs produced by infected groups was slightly reduced in both the first and second breeding seasons PE. Hatchability of fertile eggs from infected hens was significantly lower than eggs from control hens. Productivity may be impaired if MG infections occur in free-ranging wild turkey populations.  相似文献
