A system is described for the study of abnormal chromosome segregation in Sordaria brevicolis. The system utilizes two complementing alleles of the b1 locus on linkage group II. Abnormal asci containing black disomic ascospores were detected which fall into two main categories. (a) Non disjunctional asci in which the disomic spores were present together with an equal number of abortive (nullosomic) spores and (b) asci in which an extra replication of the chromosomes had occurred resulting in pseudo-wild types being formed without accompanying spore abortion. Calculations indicate that the non-disjunction frequencies at the first and second divisions of meiosis are 4.25×10−4 and 4.35×10−4 respectively. It is suggested that the system is potentially a valuable one both for the study of meiotic non-disjunction and other causes of aneuploidy.  相似文献

To test whether the induction of two-hit events plays a role in the induction of disomic gametes by exposure to X-rays of the oocytes of females carrying compound second chromosomes, exposures of 1000 R, 2000 R or 3000 R were given as an acute dose or as two equal fractions separated by a 3-h interval. The dose-effect relationship for acute exposure is linear over this exposure range (1000–3000 R) and the yields obtained are remarkebly similar to those recorded in an earlier study by Clark and Sobels. At 3000 R a significant reduction in yield was observed after exposure fractionation. At exposure levels of 2000 and 1000 R, however exposure fractionation tends to enhance the yield od disomics.The dose-effect relationship study was then extended to 125, 250 and 500 R. After an exposure of 125 R the induced frequency of disomics was significantly higher than control, but not different from the frequencies induced by 250, 500 and 1000 R. There is no obvious explanation for this plateau which, together with the linear increase from 1000–3000 R, provide an indication that there may be several mechanisms involved in the induction of disomic gemetes.Treatment with ethyl methanesulfonate (EMS) and ageing of the females for a week did not affect the yield of disomic progeny.  相似文献

A number of genetic systems are described which in yeast may be used to monitor the induction of chromosome aneuploidy during both mitotic and meiotic cell division. Using these systems we have been able to demonstrate the induction of both monosomic and trisomic cells in mitotically dividing cells and disomic spores in meiotically dividing cells after both UV light and X-ray exposure. The frequency of UV-light-induced monosomic colonies were reduced by post-treatment with photoreactivity light and both UV-light- and X-ray-induced monosomic colonies were reduced by liquid holding post-treatment under non-nutrient conditions. Both responses indicate an involvement of DNA-repair mechanisms in the removal of lesions which may lead to monosomy in yeast. This was further confirmed by the response of an excision-defective yeast strain which showed considerably increased sensitivity to the induction of monosomic colonies by UV-light treatment at low doses. Yeast cultures irradiated at different stages of growth showed variation in their responses to both UV-light and X-rays, cells at the exponential phase of growth show maximum sensitivity to the induction of monosomic colonies at low doses whereas stationary phase cultures showed maximum induction of monosomic colonies at high does. The frequencies of X-ray-induced chromosome aneuploidy during meiosis leading to the production of disomic spores was shown to be dependent upon the stage of meiosis at which the yeast cells were exposed to radiation. Cells which had proceeded beyond the DNA synthetic stage of meiosis were shown to produce disomic spores at considerably lower radiation doses than those cells which had only recently been inoculated into sporulation medium. The results obtained suggest that the yeast sustem may be suitable for the study of sensitivities of the various stages of meiotic cell division to the induction of chromosome aneuploidy after radiation exposure.  相似文献

Dominant lethal mutations induced by γ-radiation were measured in stage-7 and stage-14 oocytes of Musca domestica. At both stages the data are consistent with the multi-hit theory on radiation induction of dominant lethals. This conclusion is supported by fractionation experiments which indicate that both] S7 and S14 oocytes are capable of repairing, in defferent periods of time, a similar amount of dominant lethal damage.  相似文献

Experiments were conducted to inquire whether the radioressitance observed in an irradiated laboratory population (RÖI) of Drosophila melanogaster might be associated in some way with recombinational processes. Simultaneously, data were collected on the stage distribution of radioresistance in RÖI by studying the induction of dominant lethals and X-chromosome losses in mature females at various exposure levels of X-irradiation (in eggs sampled from subsequent 12-h broods).The data show that (1) the radiation response of both populations (RÖI and its control + K) is equal in the highly sensitive mature stages, (2) RÖI is resistant relative to +K in the medium-sensitive stage-7 and younger oocytes collected on days 1.0 to 5.5 after exposure, and (3) the difference between the populations disappears again when the sensitivity steeply decreases on days 5.5 to 6.5. Similar brood-pattern experiments indicate that exchanges between homologous chromosomes are induced (by temperature shock or X-irradiation) in eggs sampled after day 5.5. Thus it is evident that the relative radioresistance in RÖI is due to mechanisms which operate in the developing oocyte in the stages of a medium radiosensitivity between that phase in which recombination is inducible and stage-14.The observed temporal sequence of recombination and relative radioresistance in RÖI supports the speculation that the latter might be associated with recombination repair. However, the natural recombination frequencies were equal in +K and RÖI. Likewise, no clear evidence was obtained on differences between the two populations with respect to X-ray-induced modifications of homologous exchanges in various para- and pericentric parts of the genome.  相似文献

Young (0—4-h-old) Drosophila melanogaster females were X-irradiated with single or fractioned exposured over a range up to 6000 R and the induction of dominant lethals in immatuer (stage-7) oocytes was studied. The results show that (i) the frequencies of dominant lethals are higher after single than after fractionated exposures; (ii) at any given exposure level, the higher the number of fractions, the lower is the frequency of dominant lethals; (iii) conserquently, the reduction in dominant lethality relative to single exposures increases with increasing number of fractions; and (iv) this relative reduction in dominant lethality approaches a maximum value when the magnitude of the single X-ray exposure approaches zero (i.e., when tha egg survival after single X-ray exposure approaches 100%); the maximum, however, are different for the different fractionation regimes, being higher with increasing number of fractions.These findings are consistent with the assumed kinetics of X-ray induction of dominant lethality in stage-7 oocytes. It is shown that it is possible to predict the expected relative reduction in dominant lethality after fractionation, from appropriate dominant lethal data from single unfractioned exposures.  相似文献

A unique property of Drosophila melanogaster l(1)ts403 strain with the defect in heat shock protein system (HSP) is high frequency of losses and non-disjunction of sex chromosomes induced by heat shock (HS) (37 degrees C, 1 h). This effect was shown in only 6-14-th stages of oocytes. Anoxia was not effective in induction of these mutations. Successive action of anoxia and HS decreased loss frequency and non-disjunction in comparison with the only action of HS. These findings agree with the data in literature indicating that HSP synthesis was increased in the l(1)ts403 mutant when first anoxia and then HS were administered, in contrast to the action of HS only. The role of HSP in the recovery of HS-induced disruptions (chromosomal proteins and meiotic division apparatus) which can lead to chromosome non-disjunction and losses is discussed.  相似文献

The genetic system that controls the relative radioresistance in an irradiated laboratory population of Drosophila melanogaster (RÖ I) was studied. Comparisons were made between an unirradiated control population (+60, +K), the population RÖ I (after 227–333 generations of irradiation at 2100 R per generation), the sub-population RÖ I0 (derived from RÖ I after 260 generations of irradiation and kept without irradiation for up to 74 generations), the F1 hybrids +60/RÖ I, various homo- and heterozygous carriers of the 3 major chromosomes of RÖ I and +60, respectively, in combination with suitable balancers, and several chromosome substitution stocks of +K and RÖ I. The criteria used to assess the magnitude of radiosensitivity were dominant lethals, X-chromosome loss, and sex-linked recessive lethals induced in stage-7 oocytes at various exposure levels of X-irradiation.The data show that the radioresistance in RÖ I is controlled by a stable and homozygous genetic system. The system is semidominant. With respect to the induction of dominant lethals and sex-linked recessive lethals, the relative resistance is mainly contributed by chromosomes I and II. The effects of the two chromosomes are additive, each contributing about half the relative resistance. Resistance to the X-ray induction of X-chromosome loss is solely contributed by chromosome II.The findings suggest that at least 2 different and independent mechanisms are involved in determining the resistance of the RÖ I population.  相似文献

Effects of mutations rad201, mei-9, and mei-41 on cell sensitivity to gamma-radiation in Drosophila oogenesis were studied. Females of the control (Oregon R) and mutant strains were irradiated at a dose of 15 Gy. For 9 days after the irradiation, the number of eggs in consecutive day batches, the frequency of dominant lethals (DLs) among the eggs, and the cytologically recorded distribution of oocytes for stages of their development, and the frequency of egg chamber degeneration in female ovaries were estimated. As a result of joint analysis of the data, different oogenesis stages were characterized with regard to the frequency of two radiation-induced events: appearance of DLs in oocytes and degeneration of egg chambers due to apoptosis of nurse cells. It was shown that the mutations affect these parameters only at particular stages of early oogenesis, at which previtellogenetic growth of egg follicles and meiotic recombination in oocytes occur. Mutation rad201G1 increased the frequency of DLs and egg chamber degeneration, mei-41D5 affected only the DL frequency, and mei-9a, in addition to enhancing the chamber degeneration frequency, promoted radiation "rescue" of some oocytes from the DL induction.  相似文献

Mature sperm and prophase-1 oocytes of Tetranychus urticae Koch were irradiated with 250-kVp X-rays or 1.5 MeV fast neutrons. The X-ray doses ranged from 0.5 to 24.0 krad, and those of the fast neutrons from 0.1 to 16.0 krad. The genetic endpoint measured was lethality, expressed in the stages from egg to adulthood in the F1 progeny. The frequency of recessive lethals in female germ cells was estimated by comparing survival of fertilized versus unfertilized F1 eggs, after irradiation with the same dosage. X-Rays induce dominant lethals in prophase-1 oocytes by the action of both single hits on single targets and multiple hits on multiple targets. 1.5-MeV neutrons induce these effects predominantly by the action of multiple tracks on multiple targets. Dominant lethals were induced in mature sperm by X-rays and by fast neutrons by the action of both single hits on single targets and multiple hits on multiple targets. Both for prophase-1 oocytes and for mature sperm the low R.B.E. value corresponded with the relatively large multiple-target component of induction of dominant lethals by fast neutrons. The nature of dominant lethality in relation to the kinetochore organization of the chromosome is discussed. A non-linear trend in the dose--effect relationship was observed for both X-rays and fast neutrons for the estimated frequency of recessive lethals induced in prophase-1 oocytes. X-Rays were more effective than neutrons in inducing recessive lethals in prophase-1 oocytes at doses lower than 3 krad.  相似文献

The effects of relatively small doses of X-rays (up to 100 cGy) to immediately preovulatory mouse oocytes have been examined by screening chromosome aberrations at metaphase I. Dose-related responses for the induction of aberrations were found. These were mainly of the quadratic or power-law types, and therefore similar in nature to the dose-responses described elsewhere for dictyate oocytes. The frequencies of the various categories of structural aberration have been compared to the previously determined rates of radiation-induced non-disjunction in immediately preovulatory oocytes in order to examine the potential involvement of structural chromosome aberrations in radiation-induced non-disjunction.  相似文献

The sensitivity of male and female pre-meiotic germ cells of the mouse to the induction of non-disjunction by low doses of X-rays, has been tested. No enhancement with 5 rad was observed over control of values in dictyate oocytes irradiated from young or aged females. In males, a 3-fold increase in overall chromosome abnormalities (aneuploids, polyploids and mosaics) was found following the treatment of germ cells sampled in the 7th week after irradiation (spermatogonia and early primary spermatocytes) with 100 rad. The increase in aneuploidy alone was not however significant at the 5% level of probability. Primary spermatocytes sampled in week 5 after irradiation were generally insensitive to the induction of chromosome abnormalities.  相似文献

In females of Drosophila melanogaster, compound autosomes enter the repulsion phase of meiosis uncommitted to a particular segregation pattern because their centromeres are not restricted to a bivalent pairing complex as a consequence of crossing over. Their distribution at anaphase, therefore, is determined by some meiotic property other than exchange pairing, a property that for many years has been associated with the concept of nonhomologous pairing. In the absence of heterologous rearrangements or a free Y chromosome, C(3L) and C(3R) are usually recovered in separate gametes, that is as products of meiotic segregation. Nevertheless, there is a regular, albeit infrequent, recovery of reciprocal meiotic products (the nonsegregational products) that are disomic and nullosomic for compound thirds. The frequency of these exceptions, which is normally between 0.5 and 5.0%, differs for the various strains examined, but remains constant for any given strain. Since previous studies have not uncovered a cause for this base level of nonsegregation, it has been referred to as the spontaneous frequency. In this study, crosses between males and females whose X chromosomes, as well as compound autosomes, are differentially marked reveal a highly significant positive correlation between the frequency of compound-autosome nonsegregation and the frequency of X-chromosome nondisjunction. However, an inverse correlation is found when the frequency of nondisjunction is related to the frequency of crossing over in the proximal region of the X chromosome. These findings have been examined with reference to the distributive pairing and the chromocentral models and interpreted as demonstrating (1) that nonsegregational meiotic events arise primarily as a result of nonhomologous interactions, (2) that forces responsible for the segregation of nonhomologous chromosomes are properties of the chromocentral region, and (3) that these forces come into expression after the exchange processes are complete.  相似文献

The genetic stability of wheat/rye (‘Chinese Spring’/‘Imperial’) disomic addition lines was checked using the Feulgen method and fluorescent in situ hybridization (FISH). Feulgen staining detected varying proportions of disomic, monosomic, and telosomic plants among the progenies of the disomic addition lines. The greatest stability was observed for the 7R addition line, while the most unstable lines were those with 2R and 4R additions. Chromosome rearrangements were also detected using FISH. Based on the specific hybridization patterns of repetitive DNA probes pSc119.2 and (AAC)5, as well as ribosomal DNA probes (5S and 45S), isochromosomes were identified in the progenies of 1R and 4R addition lines. The results draw attention to the importance of continuous cytological checks on basic genetic materials by using FISH, because this method reveals chromosome rearrangements that could not be detected either with the conventional Feulgen staining technique or with molecular markers.  相似文献

Over the past few years, several groups have used fluorescence in situ hybridization (FISH) to study aneuploidy in human sperm. Several important observations have derived from these studies, including the demonstration of chromosome-specific variation in non-disjunction frequencies, and the possible association of aneuploidy with environmental agents and with increasing paternal age. However, an important technical limitation of these studies has been the inability to distinguish between autosomal non-disjunction occurring at meiosis I and meiosis II. In the present report, we describe a simple FISH-based approach designed to overcome this limitation. Using oligonucleotide probes capable of distinguishing subtle differences in the alpha satellite sequences of chromosome 17, we demonstrate that (in appropriate heterozygotes) it is possible to simultaneously identify disomic sperm and to determine the meiotic stage of origin of the additional chromosome. This novel approach has important implications for future FISH sperm studies, since the ability to distinguish between meiosis I and meiosis II non-disjunction will make it possible to determine whether putative etiological agents affect chromosome segregation at both, or only one, of the two meiotic stages. We report on the investigation of the parental origin and mode of formation of the two isochromosomes, i(2p) and i(2q), detected in a healthy adult male. Conventional cytogenetic analysis revealed the proband's lack of structurally normal chromosomes 2, these being replaced by an i(2p) and an i(2q). Investigation of the parental origin of the isochromosomes revealed a paternal origin of the i(2p) chromosome and a maternal origin of the i(2q) chromosome. Thus, the formation of both isochromosomes, or at least of the paternal i(2p), appears to have occurred postzygotically. Interestingly, whilst a paternal isodisomy was observed for the entire 2p, maternal heterodisomy was detected for two segments of 2q, separated by a segment showing isodisomy. The results are indicative of an initial error (non-disjunction or i(2q) formation) concerning the maternal chromosomes 2 during meiosis I, which likely favored the subsequent mitotic recombination event resulting in the presence of two isochromosomes. To the best of our knowledge this is the first case of an initial meiotic error, followed by postzygotic trisomy rescue through the formation of isochromosomes, resulting in a normal phenotype. A prenatal detection, by cytogenetic and molecular analysis, of such chromosome abnormality would have led to the incorrect conclusion of a most likely poor prognosis for the fetus.  相似文献

Yegorova and colleagues (1978) showed that a mutant strain of Drosophila melanogaster (ebony) was more sensitive to UV-induced killing of embryos and also less proficient in photoreactivating (PR) ability than a wild-type (Canton-S) strain and that the genes governing UV sensitivity and PR ability were different and presumably located on the autosomes. The experiments reported in the present paper were designed to compare the patterns of sensitivity of these 2 strains and their hybrids to X-irradiation. The sensitivity of the larvae to the killing effects of X-irradiation, and of male and female germ-cell stages to the X-ray induction of genetic damage was studied.It was found that the larvae of the ebony strain are more sensitive to X-ray-induced killing than those of the Canton-S strain. The frequencies of radiation-induced dominant lethals and sex-linked recessive lethals are higher in spermatozoa sampled from ebony males than in those of Canton-S males. In spermatozoa sampled from hybrid males, the yields of dominant lethals are no higher than in those sampled from Canton-S males and do not seem to depend on the origin of the X-chromosome. There are no statistically significant differences between the ebony and Canton-S strains in the sensitivity of their spermatozoa to the induction of autosomal translocations.Stage-7 oocytes sampled from ebony females are more sensitive to the X-ray induction of dominant lethality than are those from Canton-S females; oocytes sampled from hybrid females manifest a level of sensitivity that is significantly lower than that in either parental strain. The frequencies of X-chromosome losses induced in in this germ-cell stage are significantly lower in ebony than in Canton-S females at least at the exposure level of 3000 R at which 3 experiments were carried out. There are no measurable differences in the amount of dominant lethality induced in stage-14 oocytes of ebony, Canton-S and hybrid females.When X-irradiated Berlin-K males are mated to ebony or Canton-S females, the yields of dominant lethals are higher when ebony females are used, showing that there is a “maternal effect” for this kind of damage. Such a maternal effect is also found for sex-linked recessive lethals (irradiated Muller-5 males mated to ebony or Canton-S females). However, when irradiated ring-X-chromosome-carrying males are mated to ebony or Canton-S females, the frequencies of paternal sex-chromosome losses (scored as XO males) are lower when ebony females are used.These results have been interpreted on the assumption that the ebony strain is homozygous for recessive, autosomal genes that confer increased radiosensitivity and that the Canton-S strain carries the normal, wild-type alleles for these genes. The higher yields of dominant and recessive lethals in mature spermatozoa and of dominant lethals in stage-7 oocytes are a consequence of an enhanced sensitivity to the mutagenic (in particular, to the chromosome-breaking) effects of X-irradiation and/or of defective repair of radiation-induced genetic damage. The lower yield of XO males from irradiated stage-7 oocytes of ebony females is probably a consequence of a defect in the repair of chromosome-breakage effects, resulting in the conversion of potential X losses in females into dominant lethals. The “maternal effects” for dominant lethals, sex-linked recessive lethals and for the loss of ring-X chromosomes are assumed to have a common causal basis, namely, a defective repair of chromosome-breakage events in the females of the ebony strain.  相似文献

Summary During the past 6 years, 14 cytogenetic studies on human oocytes recovered during in vitro fertilization procedures have been published; they report contradictory results. The present survey has pooled the more than 1500 oocyte chromosome complements examined to date, in order to determine generalized trends in chromosomal abnormalities of female gametes. The overall frequency of abnormalities in mature oocytes is 24.0% with a large majority of aneuploidies (22.8%) over structural aberrations (1.2%), which could be explained by the difficulty in the detection of structural abnormalities in oocyte chromosome sets. An analysis of the distribution of non-disjunction among all chromosomes was also performed. In the A, C, D, and especially in the G groups, there is a significant difference between the observed non-disjunction and the frequencies expected from an equal partitioning of non-disjunction among all chromosomes. These data are discussed with reference to the differences obtained from cytogenetic studies on human sperm and from investigations on spontaneous abortion.  相似文献

A series of X-irradiation experiments was carried out using Drosophila melanogaster females homozygous for a third chromosome mutator gene and females which had a similar genetic background except that the mutator-bearing third chromosomes were substituted by normal wild-type chromosomes. The mutator females had been previously shown by Gold and Green to manifest a higher level of radiation-induced mutability (as measured by the X-ray-induction of sex-linked recessive lethals) in their pre-meiotic germ cells compared to normal females at an exposure of 100 R. In the presence work, the sensitivity of the pre-meiotic germ cells of mutator and normal females to the X-ray induction (2000 R) of sex-linked recessive lethals was studied. In addition, experiments were conducted to examine the sensitivity of the immature (stage 7; prophase I of meiosis) oocytes of both kinds of females to the induction of dominant lethals, X-linked recessive lethals and X-chromosome losses. The result show that in pre-meiotic germ cells, the frequencies of radiation-induced recessive lethals are similar in both kinds of females. However, the proportion of these mutations that occur in clusters of size 3 and higher, is higher in mutator than in normal females. In stage-7 oocytes, the frequencies of radiation-induced dominant lethals and sex-linked recessive lethals were similar in both kinds of females. The X-loss frequencies however, were consistently higher in mutator females although statistical significance was obtained only at higher exposures (3000 and 3750 R) and not at lower ones (750-2250 R). Possible reasons for the discrepancy between the present results and those of Gold and Green with respect to pre-meiotic germ cells are discussed.  相似文献

Different germ-cell stages of Drosophila males with a double marked Y-chromosome and either a normal X- or a ring-X chromosome were irradiated with X-rays, inducing the following aberrations: chromosome loss, chromosome gain (XYX-females), partial Y loss and isochromosomes of the Y-chromosome.Doses of 520 rad in spermatocytes and spermatids and 2600 rad in sperm, produced the same effect in these stages with regard to the chromosome loss in the males with a normal X, and the following results were obtained: (a) The partial Y loss in postmeiotic stages is small in comparison with spermatocytes in both stocks. This could mean that in spermatocytes this aberration is determined by exchange processes which can only be induced and/or detected in premeiotic stages. (b) In spermatocytes and mature sperm of males with a ring-X chromosome, the chromosome loss was 2.9 times greater than in those with a normal X. In spermatids of the males with a ring-X the rate of loss was only 1.5 times greater. In spermatocytes of either males with a ring-X or a normal X a similar high rate of isochromosomes could be induced. However, in spermatids and mature sperm the rate of induction of isochromosomes was found to be very small. These results seem to indicate that in mature sperm the rejoining of breaks in the Y-chromosome takes place before, and in the X-chromosome usually after the replication. If in post-meiotic stages of Drosophila the X- and Y-chromosomes existed as chromatid-like subunits then in spermatids these should behave as a structural unit.In sperm we were able to induce similar frequencies of individuals with a single isochromosome type in all body cells as of individuals with two types of isochromosomes (isochromosome mosaics). This result seems to indicate that after irradiation of sperm one of the first two division nuclei is lethal in an proportion of the zygotes.  相似文献
