双酶电极法测定L-苯丙氨酸的研究   总被引:1,自引:0,他引:1  
本文以Clark氧电极为基础,把水杨酸羟化酶和苯丙氨酸脱氨酶同时固定在氧电极的表面,制成了双酶生物传感器。在磷酸缓冲液中,水杨酸浓度为0.5mmol/L,烟酰胺腺嘌呤二核甘酸(NAD^+)的浓度为1.0mmol/L,其响应电流的变化对应反池中L-苯丙氨酸的浓度在0-0.15mmol/L之内有良好的线性范围。  相似文献

研究了深红酵母As2.279产生L-苯丙氨酸解氨酸(PAL)的条件、转化反式 桂酸(tCa)生成L-苯丙氨酸(L-Phe)的条件以及几种因素对PAL稳定性的影响,结果表明,最佳转化条件为:1.0%t-Ca,8mol/L氨,pH10.0,30℃。在转化液中加入还原剂和充入N2有利于提高酶的稳定性,在此条件下可一次转化64%的t-Ca,保留60%的酶活。生成L-Phe浓度为5.8g/L。  相似文献

野花生豆凝集素(CML)经SephadexG-200测得分子量为103.OkD.用对二甲基氨基苯甲醛(DAB)为显色剂,测得每个CML分子含有5.9个色氨酸残基.在pH5.1,含8mol/L脲的醋酸缓冲液中,N-溴代丁二酰亚胺(NBS)可修饰CML分子中的5.6个色氨酸(Trp)残基,同时使CML的凝血活性完全丧失.用焦碳酸二乙酯(DEPC)和N-乙酰顺丁烯酰胺(NEM)分别修饰CML的组氨酸残基和半胱氨酸巯基后,CML的活性均无变化.CML在天然状态下荧光发射峰位于336nm处,用CML的专一性抑制糖N-乙酰半乳糖胺研究色氨酸的微环境,发现N-乙酰半乳糖胺可以淬灭CML中88%的色氨酸残基萤光,Stern-Volmer常数K=1.73L/mol.同时发现N-乙酰半乳糖胺能够保护CML,避免NBS对CML的修饰作用,表明色氨酸可能是CML维待活性所必需,并直接参与和专一性抑制糖的结合,其微环境较为疏水.  相似文献

用从系统感染TMV(tobacco m osaic virus)的番茄叶胞外蛋白提取液中纯化的β-N-乙酰氨基己糖苷酶为材料,研究了该酶的部分性质。以p-硝基苯基-N-乙酰-β-D-氨基葡萄糖苷(pNP-β-D-GlcNAc)或p-硝基苯基-N-乙酰-β-D-氨基半乳糖苷(pNP-β-D-GalNAc)为底物,该酶的最适pH在4.8—5.0 之间,最适温度在44—47℃之间。对酶的热稳定性研究表明,温度对酶的热变性作用表现为两相变性曲线,Km 值分别为0.36 m m ol/L (pNP-β-D-GlcNAc为底物)和0.67 m m ol/L (pNP-β-D-GalNAc 为底物),N-乙酰氨基葡萄糖(GlcNAc)和N-乙酰氨基半乳糖(GalNAc)是酶活性的竞争性抑制剂,Ag+ 和Hg2+ 是酶活性的强抑制剂,金属离子Fe2+ 、Fe3+ 和Cu2+ 也抑制酶活性。  相似文献

The uncoupled portion of the partially uncoupled oxidation of tetrahydropterins by phenylalanine hydroxylase can be described by the same model as we have recently derived for the fully uncoupled reaction (Davis, M.D. and Kaufman, S. (1989) J. Biol. Chem.264, 8585–8596). Although essentially no hydrogen peroxide is formed during the fully coupled oxidation of tetrahydrobiopterin or 6-methyltetrahydropterin by phenylalanine hydroxylase when phenylalanine is the amino acid substrate, significant amounts of hydrogen peroxide are formed during the partially uncoupled oxidation of 6-methyltetrahydropterin whenpara-fluorophenylalanine orpara-chlorophenylalanine are used in place of phenylalanine. Similarly, during the partially uncoupled oxidation of the unsubstituted pterin, tetrahydropterin, even in the presence of phenylalanine, hydrogen peroxide formation is detected. Rapid filtration assays were used to determine the effects of barium, calcium and an insecticidal δ-endotoxin from Bacillus thuringiensis on sodium and potassium ion gradient dependent phenylalanine accumulation by brush border membrane vescles from the larval midgut of the tobacco hornworm (Manduca sexta). Neither barium nor calcium had a significant effect on sodium ion gradient dependent phenylalanine accumulation by the membrane vesicles. Both barium and calcium inhibited potassium ion gradient dependent phenylalanine accumulation by the membrane vesicles. B. thuringiensis δ-endotoxin inhibited both sodium and potassium ion gradient dependent phenylalanine accumulation by the vesicles. Inhibition of both sodium and potassium ion gradient dependent phenylalanine accumulation increased similarly with increasing δ-endotoxin inhibition of either sodium or potassium dependent phenylalanine accumulation by the vesicles.  相似文献

The in vitro effects of phenylalanine and some of its metabolites on ATP diphosphohydrolase (apyrase, EC 3.6.1.5) activity in synaptosomes from rat cerebral cortex were investigated. The enzyme activity in synaptosomes from rats subjected to experimental hyperphenylalaninemia (-methylphenylalanine plus phenylalanine) was also studied. In the in vitro studies, a biphasic effect of phenylalanine on both enzyme substrates (ATP and ADP) was observed, with maximal inhibition at 2.0 mM and maximal activation at 5.0 mM. Inhibition of the enzyme activity was not due to calcium chelation. Moreover, phenylpyruvate, when compared with phenylalanine showed opposite effects on the enzyme activity, suggesting that phenylalanine and phenylpyruvate bind to two different sites on the enzyme. The other tested phenylalanine metabolites (phenyllactate, phenylacetate and phenylethylamine) had no effect on ATP diphosphohydrolase activity. In addition, we found that ATP diphosphohydrolase activity in synaptosomes from cerebral cortex of rats with chemically induced hyperphenylalaninemia was significantly enhanced by acute or chronic treatment. Since it is conceivable that ATPase-ADPase activities play an important role in neurotransmitter (ATP) metabolism, it is tempting to speculate that our results on the deleterious effects of phenylalanine and phenylpyruvate on ATP diphosphohydrolase activity may be related to the neurological dysfunction characteristics of naturally and chemically induced hyperphenylalaninemia.  相似文献

Polymerizations of DL -phenylalanine NCA by block copolymers of sarcosine and DL -phenylalanine, designated by (Phe)m(Sar)n and capable of reaction at the phenylalanyl terminal, were investigated in nitrobenzene solution at 25°C. With increasing n for constant m (m = 0, 1, 2, and 5), the polymerization rate greatly increased. Previously the acceleration of the initiation reaction in the polymerization of DL -phenylalanine NCA by polysarcosine (m = 0) was reported. The present results showing the acceleration by the copolymers of sarcosine and DL -phenylalanine indicate the presence of the polymer effect in the propagation reaction as well. However, the polymer effect was most marked with polysarcosine (m = 0), and decreased with increasing m. The same polymerizations by sequential copolymers composed of ten sarcosine units and two DL -phenylalanine units were also investigated. Again with these copolymer catalysts the polymerization rate was larger than that by monomeric amines. But the polymer effect decreased sharply when the phenylalanine units take positions near the terminal amine group of the copolymer catalyst. These two deteriorating effects of the phenylalanine unit have been interpreted in terms of the decrease of the flexibility of polymer chain, caused possibly by an intramolecular hydrogen bond of the phenylalanine unit.  相似文献

The effect of nine phenylalanine and tyrosine analogues at various concentrations upon the reproduction of adult olive fruit fly Bactrocera oleae Gmelin (Diptera, Tephritidae), was tested. Fecundity (eggs/female/day) and percentage egg hatchability was significantly reduced by the following anti-amino acids (in parentheses are indicated the antagonized amino acid): p-fluoro- DL -phenylalanine (phe), p-amino- DL - and - L -phenylalanine (tyr), 3-amino- L -tyrosine (tyr) and L -mimosine (tyr), at concentrations of 0.1, 0.25, 0.05 and 0.5% in the diet, respectively. Hatchability was also affected by two other analogues of phenylalanine and tyrosine, p-bromo- DL -phenylalanine at a concentration of 10% and m-fluoro- DL -tyrosine at a concentration of 1.5%. Insect survival was affected only by p-fluorophenylalanine and 3-amino- L -tyrosine at concentrations 0.25 and 6%, respectively. No significant differences were observed between the survival of the two sexes. Finally, β-2-thienyl- DL -alanine (phe) and α-methyl- DL -p-tyrosine (tyr) did not affect any of the parameters tested. Electron microscopy examination of the follicles and the egg-shell structure of eggs laid by females fed with diets containing p-amino- L -phenylalanine and 3-amino- L -tyrosine, revealed abnormalities related to the egg-shell fine structure.  相似文献

SYNOPSIS. Phenylalanine hydroxylase could not be assayed in extracts of Tetrahymena pyriformis strain W in a system by which the enzyme could be assayed in rat liver extracts. Isotopically labelled phenylalanine, however, was converted to tyrosine by growing or washed cells. Growth conditions which allowed limited synthesis of unconjugated tetrahydropteridine severely reduced the ability of the cells to synthesize tyrosine from phenylalanine. The presence of glucose and acetate in the growth medium resulted in elevated free tyrosine pools and an increased capacity of washed cell suspensions to convert phenylalanine to tyrosine. It would appear that the putative phenylalanine hydroxylation system is not subject to the repressive effects of glucose and acetate which apply to the enzymes of tyrosine catabolism. The significance of this distinction is discussed.  相似文献

Production feedback inhibition both on cell growth and on product formation of phenylalanine fermentation might be alleviated by elevated oxygen supply. Batch fermentations by a high phenylalanine producing strain Corynebacterium glutamicum CCRC 18335 at various initial phenylalanine concentrations (P(0)) ranging from 0 to 20 g/L and different oxygen transfer rate coefficients (K(L)a) ranging from 23 to 76 h(-1) were studied. The fermentation parameters with respect to P(0) were strongly dependent on K(L)a. Cell yield favored higher K(L)a and lower P(0). Product yield with respect to varying phenylalanine concentration was evaluated by the relative oxygen availability (ROA). The optimal ROA for phenylalanine formation was strongly dependent on the product concentration. While P(0) was low, the product inhibition was less significant and the maximum product yield occurred while ROA was at 0.5-0.6. While P(0) was high, the product inhibition was significant and the maximum product yield occurred while ROA was at 0.8-0.9. These results suggest that the product feedback inhibition of phenylalanine fermentation processes can be alleviated by a gradual increase in oxygen supply rate while the increasing product concentration is taken into account. The strategy is demonstrated in a fed-batch culture with elevated oxygen supply. The final phenylalanine concentration was 23.2 g/L, which was 45% better than that of the fed-batch fermentation without elevated oxygen supply. Likewise, the maximum productivity was improved by 42% at 0.37 g/(L x h).  相似文献
