【目的】本研究旨在探究菜豆Phaseolus vulgaris植株对不同害虫危害的系统防御机制。【方法】分别以斜纹夜蛾Spodoptera litura、二斑叶螨Tetranychus urticae、西花蓟马Frankliniella occidentalis取食或昆虫针穿刺处理菜豆中部叶片6,24,48,72和96 h后,测定菜豆植株中部处理叶片以及上部和下部未处理叶片的过氧化物酶(POD)、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性的变化。【结果】不同害虫取食后,POD,CAT和SOD活性在受害的菜豆中部叶片发生了明显变化,但不同害虫造成酶活性的变化趋势不同。与未处理的对照植株中部叶片相比,POD活性均是先升高后恢复至对照水平。POD活性在二斑叶螨取食后,于24 h达到最大值,是对照的4.6倍;POD活性在机械损伤和西花蓟马取食后在48 h达到峰值,而斜纹夜蛾取食诱导下POD活性升高最缓慢,直到72 h才达到最大值。中部叶片的CAT活性在机械损伤和3种害虫取食后变化不同,机械损伤24 h后CAT活性明显升高,而3种害虫取食后CAT活性变化较早,在6 h就明显升高,但随时间的延长,CAT活性的变化趋势不同。中部叶片SOD活性在不同处理下变化较大,在机械损伤诱导下仅在96 h明显升高,而二斑叶螨取食24 h以后均明显升高;但斜纹夜蛾及西花蓟马取食后,SOD活性在72 h时受到明显的抑制。在菜豆植株的上部和下部未受害叶片中,3种害虫取食和机械损伤处理后,以上3种酶的活性也发生了明显变化,POD活性在上部未受害叶片不同处理96 h时被抑制,而在下部叶片不同处理下均被激发,但诱导程度不同。机械损伤、斜纹夜蛾、二斑叶螨和西花蓟马处理后,菜豆植株上部和下部未受害叶片CAT活性都分别在24,6,6和24 h被明显诱导。上部和下部未受害叶片SOD活性在机械损伤96 h,斜纹夜蛾和二斑叶螨取食72 h后,均被明显激发;但西花蓟马取食后,SOD活性在48 h和72 h明显被抑制。【结论】斜纹夜蛾、二斑叶螨和西花蓟马取食均能显著诱导菜豆植株受害叶和未受害叶防御酶活性的变化,即引起菜豆植株的系统防御反应,但3种害虫诱导的防御酶活性变化程度不同,表明植物防御反应的时空效应与害虫种类有关。  相似文献

水培大豆和田间生长的大豆,接种根瘤菌 Rhizobium B16-11C 后植株全氮含量、叶片叶绿素含量和净光合速率及种子产量都明显增加。比较 Clark 大豆的结瘤品系和不结瘤品系获类似结果。摘除根瘤后3天内叶片净光合速率无明显变化。大豆植株遮阴、去叶或切掉地上部导致根瘤活性明显下降。但去豆荚不能提高根瘤固氮的比活性。根瘤活性的日变化不能用根瘤蔗糖、淀粉含量或周围温度的变化来解释,其控制因子尚待深入研究。  相似文献

大豆品种吉林3号植株不同节位叶片的上部未完全展开叶的酰脲含量和尿囊素酶活性较高。幼茎、叶柄和叶片酰脲含量及尿囊素酶活性高于成年的茎、叶柄和叶片。荚皮酰脲含量显著高于籽粒,但籽粒中的尿囊素酶活性是荚皮中的2~3倍。  相似文献

大豆植株不同冠层种子活力及其萌发中抗氧化酶活性   总被引:1,自引:0,他引:1  
以吉农15号和吉农24号大豆(Glycine max)品种为材料,根据植株高度平均分为下部、中下部、中部、中上部和上部5个冠层,分析不同冠层种子百粒重、种子活力及萌发7天子叶中抗氧化酶活性和丙二醛含量的变化。结果表明,随着植株冠层的升高,大豆百粒重呈先略上升后逐渐下降的趋势,上部冠层种子的百粒重最小。大豆植株下部与中下部冠层种子的发芽率和发芽势较高;并且该两层的种子活力指数显著高于中上部与上部冠层,种子萌发后幼苗的茎粗也显著大于中上部及上部冠层幼苗。下部与中下部冠层种子萌发时子叶中的抗氧化酶活性高于上部,而丙二醛含量低于上部,说明该两层的大豆种子活力高,萌发后幼苗健壮且子叶新陈代谢旺盛,是优选良种的最佳冠层。  相似文献

研究结果表明 ,不同产地的芦荟植株不同叶位、花器间生理生化量值变化与生境背景水平息息相关。总的变化趋向 :POD活性以野生植株 >棚栽植株 ,下部叶片高于上、中部叶片 2 5 .91%、2 7.42 %;糖类组分含量以总糖>多糖 >葡萄糖 >果糖 ;花器间 4种活性酶含量差异显著 (S .P <0 .0 5 ) ,AST >GPT >β HBDH >LDH H ;蛋白质电泳谱带是上部叶片比下部叶片的带多且色深 ,活性强。  相似文献

水分胁迫下AM真菌对沙打旺生长和抗旱性的影响   总被引:7,自引:0,他引:7

利用盆栽试验研究了水分胁迫条件下接种AM真菌对优良牧草和固沙植物沙打旺(Astragalus adsurgens Pall.)生长和抗旱性的影响。在土壤相对含水量为70%、50%和30%条件下,分别接种摩西球囊霉(Glomus mosseae)和沙打旺根际土著菌,不接种处理作为对照。结果表明,水分胁迫显著降低了沙打旺植株(无论接种AM真菌与否)的株高、分枝数、地上部干重和地下部干重,并显著提高了土著AM真菌的侵染率,对摩西球囊霉的侵染率无显著影响。接种AM真菌可以促进沙打旺生长和提高植株抗旱性,但促进效应因土壤含水量和菌种不同而存在差异。不同水分条件下,接种AM真菌显著提高了植株菌根侵染率、根系活力、地下部全N含量和叶片CAT活性。土壤相对含水量为30%和50%时,接种株地上部全N、叶片叶绿素、可溶性蛋白、脯氨酸含量和POD活性显著高于未接种株;接种AM真菌显著降低了叶片MDA含量;接种土著AM真菌的植株株高、分枝数、地上部和地下部干重显著高于未接种株。土壤相对含水量为30%时,接种AM真菌显著增加了地上部全P含量和叶片相对含水量;接种摩西球囊霉的植株株高、分枝数、地上部和地下部干重显著高于未接种株。水分胁迫40d,接种AM真菌显著提高了叶片可溶性糖含量。水分胁迫80d,接种株叶片SOD活性显著增加。菌根依赖性随水分胁迫程度增加而提高。沙打旺根际土著菌接种效果优于摩西球囊霉。水分胁迫和AM真菌的交互作用对分枝数、菌根侵染率、叶片SOD、CAT和POD活性、叶绿素、脯氨酸、可溶性蛋白、地上部全N和全P、地下部全N和根系活力有极显著影响,对叶片丙二醛和地下部全P有显著影响。AM真菌促进根系对土壤水分和矿质营养的吸收,改善植物生理代谢活动,从而提高沙打旺抗旱性,促进其生长。试验结果为筛选优良抗旱菌种,充分利用AM真菌资源促进荒漠植物生长和植被恢复提供了依据。  相似文献

【目的】本研究旨在探究锉吸式口器害虫西花蓟马Frankliniella occidentalis取食诱导的菜豆Phaseolus vulgaris木植株系统防御反应的分子机制。【方法】利用荧光定量PCR技术,检测西花蓟马分别在稳定取食菜豆植株2 h后的24,48,72和96 h菜豆植株不同部位(上部、中部和下部)叶片中茉莉酸信号转导途径和水杨酸信号转导途径防御相关基因脂氧合酶基因(LOX)、苯丙氨酸解氨酶基因(PAL)和β-1,3-葡聚糖酶基因(PR-2)相对表达量的变化。【结果】西花蓟马取食菜豆植株后,受害叶片及上部和下部健康叶片中LOX,PAL和PR-2均被显著诱导表达,均以受害叶片防御酶基因表达量的变化较大,并且表达量在相同叶片不同时间下的变化不同。中部受害叶片中LOX的相对表达量均显著高于未接虫的对照植株,并随西花蓟马为害时间的延长逐渐升高,在96 h时为对照的209.54倍;随着时间的延长LOX表达量在上部和下部健康叶片呈现升高-降低-升高的趋势。PAL的表达量在西花蓟马为害植株的中部受害叶片和上部健康叶片均于48 h时出现最大值,分别为对照的52.70倍和41.20倍;但在下部健康叶片于72 h时达到最大值,为对照的47.06倍。PR-2表达量在受害株的上部健康叶片于48 h时出现最大值,在中部受害叶片和下部健康叶片均于96 h时达到最大值,但在24 h时在下部健康叶片中受到抑制。【结论】西花蓟马取食能显著诱导菜豆植株茉莉酸和水杨酸信号转导途径相关防御酶基因LOX,PAL和PR-2的表达,并在菜豆植株不同部位叶片产生系统抗性。  相似文献

选择3个相邻的葡萄承包户,测定生长季使用除草剂对巨峰葡萄叶片光合机构及枝条贮藏营养的影响.结果表明:与人工除草(CK)相比,2009年使用除草剂的葡萄园(T1和T2)葡萄叶片净光合速率(Pn)明显降低,其中T1园内第4节和T2园内第6节叶片Pn降幅最大,分别为40.5%和32.1%;而除草剂对叶片气孔导度(Gs)的影响较小.2010年继续喷施除草剂的T1葡萄园叶片Pn仍显著低于对照,而停止喷施除草剂、改为人工除草的T2葡萄园中上部叶片的Pn和Gs稍高于T1,但Pn仍显著低于对照,表现为除草剂残留的后效作用.2009年,使用除草剂降低了葡萄叶片光系统Ⅱ最大光化学效率(Fv/Fm)和性能指数(PI),快速叶绿素荧光诱导动力学曲线(OJIP)中J点和K点荧光上升,叶片反应中心和放氧复合体破坏程度较高;喷施除草剂显著降低了葡萄中部叶片的色素含量,且浓度越大降幅越大.除草剂处理显著提高了所有部位叶片过氧化氢酶(CAT)和过氧化物酶(POD)活性;提高了中部叶片超氧化物歧化酶(SOD)活性,而显著降低了上部和下部叶片的SOD活性;上部叶片的抗坏血酸过氧化物酶(APX)活性显著降低,而中、下部叶片显著提高.使用除草剂加剧了葡萄叶片膜脂过氧化程度,导致枝条可溶性糖、淀粉、游离氨基酸和可溶性蛋白含量明显下降.  相似文献

以不同烤烟品种‘红花大金元’和‘中烟100’为试验对象,研究同一生育期不同部位叶片的无机氮积累及其与氮素代谢和氨挥发的关系。结果表明,烟草植株下部衰老叶片(第5片叶)NO3^-和NH4^+的含量要高于中部叶(第10和第15片叶)和上部叶(第20片叶),并且‘红花大金元’下部叶NO3^-和NH4^+的含量比‘中烟100’显著偏高;‘中烟100’植株各部位叶片的氨气补偿点比‘红花大金元’高。两个品种在第5到第20叶位间的谷氨酰胺合成酶(GS)、硝酸还原酶(NR)和谷氨酸脱氢酶(GDH)活性大小及其变化不一致,是叶片无机氮积累存在品种间差异的生理基础。  相似文献

研究了大田生长条件下,大豆品种──早12花及幼荚脱落过程中植物激素变化。结果表明,自盛花期始,位于大豆第8~12节位的花序,其上、中、下三个部分花荚的脱落率逐渐升高,至最后一次取样时,三部分花荚的脱落率分别为77.4%、54.7%和18.3%,说明同一花序的基部花荚不易脱落。由内源细胞分裂素(iPA,ZRs,DHZRs)的分析发现,基部幼荚在生长前期,其内源iPA、ZRs、DHZRs均出现一个含量高峰,总CTK含量明显高于中、下部,而在后期脱落酸含量则明显较低;后期基部幼荚的生长速率及干物质积累量均高于中、上部。可见,植物激素参与调控了大豆生殖器官的脱落过程。  相似文献

Psophocarpin B1 is a 20,000 Mr protein of winged bean (Psophocarpus tetragonolobus) seeds having chymotrypsin inhibitory activity. Single crystals of this protein suitable for X-ray crystallographic studies have been obtained by the vapour diffusion method using ammonium sulphate. The crystals are hexagonal, space group P6(4)22 or P6(2)22, cell dimensions a = b = 61 A, c = 210 A. They are stable to irradiation with X-rays and diffract to at least 2.6 A resolution.  相似文献

Karyotype morphology of Psophocarpus tetragonolobus and P. scandens is described and the behaviour of chromosomes at meiosis discussed. Both species have a chromosome number of 2n = 18 and similar karyotypes of a sort unusual within the Phaseolinae. Differences between the species in detailed morphology of the chromosomes agree with data from external morphology, palynology and crossability in indicating that P. scandens cannot be the immediate wild ancestor of domesticated P. tetragonolobus . Study of meiotic chromosomes and analysis of qualitative and quantitative variation in Papua New Guinean P. tetragonolobus suggest that there are few restrictions to recombination other than those imposed by predominant self-pollination, The bearing of these data on the taxonomic relationships of Psophocarpus and on chromosomal evolution within the Phaseolinae is discussed.  相似文献

Analysis of Leucaena leucocephala from various localities in Western Nigeria shows that young leaves, pods and seeds contain more crude protein but lower crude fibre and ether extract than mature ones. The crude fat component was higher in the seeds (4.6% and 5.5% for green and ripe seeds, respectively) than in any other edible part of the plant, while the green and brown seed coats contain the least amount of ether extract and total ash. The mineral composition of the dry matter of mature leaves is 2.8% calcium, 0.26% phosphorus, 0.37% magnesium, 1.78% potassium, 0.21% sodium and 0.12% iron.  相似文献

Lysine rich protein (LRP) gene derived from the seed of Psophocarpus tetragonolobus was transformed into Brassica napus, employing cotyledon petiole as explants and by using the Agrobacterium tumefaciens strain LBA4404. Transformation efficiency was found to be closely related with phytohormone concentration, infection incubation, and co-cultured time. A medium containing 4 mg/l 6-benzyladenine (6-BA) and 0.3 mg/l naphthalene acetic acid (NAA) was used for plant regeneration. With infection incubation of A. tumefaciens (OD600 = 0.4) for 20 min and co-culture of infected cotyledon petiole for 3 days, the highest transformation efficiency of 8.5% was obtained. To confirm LRP gene expression, PCR and Southern blot analysis were performed on leaf-isolated DNA from regenerated plants resistant to kanamycin. All transgenic plants of the generation T0 formed fertile seeds, which were sowed for the inheritance study of generational T1 and amino acid analysis. It was found that the lysine content of seeds from T1 generation increased by 16.7% compared with non-transgenic lines.  相似文献

A comparative investigation was undertaken with pigeon pea leaves and attached flower buds/flowers/pods during their developmental stages including senescence in a natural system in experimental plots. Alterations in chloroplast pigments, total soluble proteins, lipid peroxidation, malondialdehyde (MDA) content and activities of guaiacol peroxidase (POD, EC 1.11.1.7) and superoxide dismutase (SOD, EC 1.15.1.1) were studied at 5-day interval from initial to 40-day stage. Chloroplast pigments and proteins of leaves increased upto 15 and 20-day stages respectively followed by a steady decline. Reproductive parts, however, exhibited rise in chloroplast pigments upto 25-day and protein till last stage as developing pods gain the amount from the senescing leaves which are nearest to them. Senescing leaves show very high POD activity than the developing and senescing pods and POD appears to be associated with chlorophyll degradation. Considerably higher activity and amount of LOX and MDA respectively have been noticed in senescing leaves than in flowers and pods. Increase in SOD activity during early stage of leaf growth and maturation indicates protective role that declined at senescent stages. Pods are unique in having very high SOD activity, only last stage of senescence does show a decline.  相似文献

In most experimental hybridizations between oilseed rape (Brassica napus) and weedy B. campestris, either intra- or interspecific pollen has been applied to individual flowers. Under field conditions, however, stigmas will often receive a mixture of the two types of pollen, thereby allowing for competition between male gametophytes and/or seeds within pods. To test whether competition influences the success of hybridization, pollen from the two species was mixed in different proportions and applied to stigmas of both species. The resulting seeds were scored for paternity by isozyme and randomly amplified polymorphic DNA analysis. Using data on the proportion of fully developed seeds and the proportion of these seeds that were hybrids, a statistical model was constructed to estimate the fitness of conspecific and heterospecific pollen and the survival of conspecific and heterospecific zygotes to seeds. B. campestris pollen in B. napus styles had a significantly lower fitness than the conspecific pollen, whereas no difference between pollen types was found in B. campestris styles. Hybrid zygotes survived to significantly lower proportions than conspecific zygotes in both species, with the lowest survival of hybrid zygotes in B. napus pods. This is in contrast to the higher survival of hybrid seeds in B. napus than in B. campestris pods when pollinations are made with pure pollen. Altogether, the likelihood of a foreign pollen grain producing a seed was much lower on B. napus than on B. campestris. In addition, pods on B. napus developed to a lower extent the more heterospecific pollen was in the mix, whereas this had no effect on B. campestris.  相似文献

Dalbergia miscolobium (Papilionaceae) is a tree of the Brazilian cerrado that produces ovaries with two ovules. Only one ovule usually matures, but when two mature, they are smaller than seeds of single-seeded pods. Germination percentage was lower in small seeds than in medium or large seeds. In general, small seeds gave rise to smaller seedlings (dry mass) than did medium and large seeds. The root/shoot ratio, however, was very similar for the first six months, indicating that this ratio is consistent across seedling sizes. There was no correlation between seed size and relative growth rate. Single-seeded fruit production in D. miscolobium resulted in greater seed reserves and increased fruit dispersal distance; single-seeded fruits were lighter than double-seeded ones. Together, these two features should increase the probability of successful seedling establishment of seeds from single-seeded versus double-seeded fruits.  相似文献

大豆开花结荚期,不同发育阶段的幼蕾与花荚的脱落率不同,其中以花后5d内的幼荚脱落最严重。与败育花荚相比,正常花荚中的干物质积累量均较高。细胞分裂素(DHZRs,ZRs,iPA)含量也较高,花后3~5d的幼荚中表现更明显。脱落酸(ABA)则是以败育幼蕾及花后3~5d的幼荚中含量较高。不同发育阶段的大豆生殖器官中,正常开放花中的玉米赤霉烯酮(ZEN)含量最高。  相似文献

Many protease inhibitors have been characterized from leguminous seeds but very little is known about seed proteases which are supposedly regulated by these inhibitors. We have developed an indirect immunoaffinity chromatography system for the purification of cognate proteases from the same source, based on preferential high salt elution of the enzyme from a ternary complex of the protease, the inhibitor, and the anti-inhibitor IgG. Using anti-winged bean chymotrypsin inhibitor (WbCI) IgG as an affinity ligand, a multicatalytic protease complex has been purified from developing winged bean (Psophocarpus tetragonolobus) seeds. The purified preparation resolves into two large proteolytically active components when subjected to gel permeation chromatography under nondenaturing conditions, while SDS/PAGE analysis shows the presence of approximately 15 polypeptide chains in the 20- to 115-kDa range. The preparation cleaves known synthetic peptide substrates of trypsin, chymotrypsin, and V8 protease and it is only partially inhibited by a number of class-specific protease inhibitors. Western blot analysis shows the presence of WbCI in the purified preparation even after its extensive removal by the IgG-Sepharose column. The versatility of the indirect immunoaffinity chromatography system is attested by its extension to the soybean seeds.  相似文献
