为了筛选具有产油能力的微藻,从自然界水体中分离出14株微藻,根据形态特点对它们进行了初步鉴定。对其中12株微藻在自养和异养条件下的生长特性和产油性能进行了比较。通过微藻的生长曲线,生物量和油脂含量等指标,从中筛选出高产藻株并对该藻株进行了分子生物学鉴定。结果表明:藻株Y06在12种微藻中的油脂产量和产率最高,经18S rDNA鉴定确定为栅藻(Scenedesmus abundans)。藻株Y06在自养条件下的油脂产率为9.40 mg/(L.d),在异养条件下的油脂产率为201.29 mg/(L.d)。  相似文献

为了建立一个高效的高产油微藻诱变育种流程,微藻中油脂含量快速和准确的测定在其中具有重要作用。在本研究中,首先利用低场核磁共振技术,建立了直接检测干藻粉和培养液中小球藻油脂含量的方法,其信号强度与细胞中油脂含量存在特异的线性关系,干藻粉和藻液中油脂含量与信号值拟合的R2均高于0.99,说明该方法用于小球藻油脂含量的检测是准确和可行的。同时该方法与传统油脂测量方法相比,具有快速、简便和准确等优点。但其通量不及尼罗红染色法,因此,我们开发了将尼罗红染色法用于初筛,低场核磁共振技术用于复筛的新型高通量藻种复合筛选方法,并将此筛选方法应用于一种异养高产油原壳小球藻的诱变育种过程中。首先从3 098株诱变藻种中初筛得到108株具有较高油脂含量的藻株,然后利用低场核磁共振技术复筛得到9株高产油性能的藻株,其中一株甘油三酯含量超过20%,比原始藻株提高1倍,培养168 h后培养液油脂浓度达到5 g/L,证明此诱变育种流程不仅提高了筛选的效率还可靠且可行。  相似文献

本实验利用尼罗红荧光与叶绿素荧光比值表征小球藻的油脂含量,快速筛选出一株紫外诱变高产油藻株。经过叶绿素荧光分析表明,诱变株M2的生长及光合作用没有显著变化。检测诱变株M2总油脂含量32.1%,比野生藻株的23.1%提高了9个百分点。  相似文献

利用通气培养系统, 对10株经过初步生长筛选的微藻进行培养, 以总脂单位体积产率为主要指标, 筛选具有产油潜力的优良藻种。结果表明, 10株微藻的生物质干重、总脂含量分别为0.81.6 g/L、14.8%39.7%, 总脂单位体积产率大于30 mg/(Ld)的有6株, 其中藻株HY-6总脂单位体积产率达到最高的50.8 mg/(Ld), 是一株具有潜力的产油微藻。运用形态学特征和18S rDNA及ITS系统学分析相结合的方法对藻种HY-6进行分类鉴定。依据形态学特征, HY-6为球状单细胞, 具有1个明显的蛋白核, 杯状色素体周生, 从而初步判断该藻株可能属于小球藻属(Chlorella)或拟小球藻属(Parachlorella); 18S rDNA及ITS系统学分析表明HY-6与小球藻属分为两个不同的进化支, 但与凯氏拟小球藻(Parachlorella kessleri)的亲缘关系较近, 且具有较高的自展支持率, 因此将其鉴定为凯氏拟小球藻(P. kessleri)。研究结果将为产油微藻资源的收集、筛选及后续研究提供基础。    相似文献

产油微藻的分离、筛选及自养培养氮源、碳源的优化   总被引:6,自引:0,他引:6  
从云南滇池的水样中分离筛选得到一株自养产油小球藻(Chlorella vulgaris,C.vulgaris),其油脂产率可达28.6mg/(L·d),进一步考察了不同氮源、氮源浓度和添加无机碳源对其自养生长和油脂积累的影响。结果表明,硝酸钠为优化氮源,氮元素的优化浓度为123mg/L,油脂含量随氮元素浓度升高而降低;添加NaHCO3显著提高了C.vulgaris生物量产率和油脂产率,其优化浓度为800mg/L。在氮源和碳源的优化浓度下,C.vulgaris的最大生物量产率和油脂产率可达332.8mg/(L·d)和100.2mg/(L·d),分别是对照组的3.6和3.4倍。  相似文献

研究了5种光质对小球藻(Chlorella vulgaris)M209256生长和产油的影响。结果表明:蓝光为小球藻的最适生长和产油光质;与其他光质相比,蓝光培养的小球藻生物量和油脂含量均较高,为2.40×107个/mL和28%;红光培养的小球藻生长最慢且油脂含量最低,为1.32×107个/mL和15.13%,表现出明显的"红降"现象。在GCMS分析的基础上,对油脂甲酯化后的十六烷值进行评估,结果发现:蓝光的十六烷值最高;5种光质培养的小球藻所产油脂,甲酯十六烷值均在47以上。因此,小球藻油脂所制备的生物柴油具有较好的燃烧性能。  相似文献

【目的】筛选具有较快生长速率及较强产油能力的微藻,探究所获得微藻的生理生化性能及不同培养方式对其生物量、产油能力、碳消耗等生长特性的影响与藻种对pH的适应能力。【方法】通过磷酸香草醛测定法及尼罗红染色对微藻进行初筛复筛,通过设置光合自养、异养和混养等3种培养方式,并采用气质联用等方法,研究不同培养方式对所获微藻生长特性、所产油脂脂肪酸组成以及碳代谢等方面的影响。【结果】筛选出两株产油能力较强的藻株H、Z_8,其油脂产量分别可达1.14±0.05 g/L和1.33±0.10 g/L,经形态观察和分子生物学鉴定初步表明藻株H属布朗单针藻(Chlorolobion braunii)、藻株Z_8属链带藻(Desmodesmus intermedius)。构建了不同培养方式下微藻动力学模型,H、Z_8属于生长偶联型。当培养环境的pH处于6.0–9.0,对藻株H、Z_8的总脂量与生物量无明显差异(P0.05)。【结论】筛选获得的藻株H、Z_8中C16与C18脂肪酸占总脂肪酸的比率能达到90%以上。藻种在混养条件下生物量积累优于异养,但异养条件下更加有利于油脂的积累,且H、Z_8均具有较为宽泛的pH适应能力,是具有一定产业化应用潜力的优良产油藻株。  相似文献

重要理化因子对小球藻生长和油脂产量的影响   总被引:5,自引:0,他引:5  
本文采用通气培养的方法研究了N、P、Fe3 、盐度、光照强度、温度对小球藻(Chlorella sp. XQ-200419)生长速率、生物量和油脂产量的影响。主要结果如下:N浓度对小球藻的生长和油脂产量均有显著的影响,在KNO3浓度0.05—0.3g/L范围内,小球藻生长速率随N浓度的增加而提高,并积累更多的生物量,而油脂含量随之递减,KNO3浓度为0.3g/L时,油脂产量最高。小球藻对P浓度变化的适应范围很大,K2HPO4浓度在10—160mg/L范围内,对小球藻的生长和油脂产量都没有显著影响。在小球藻培养后期补加不同浓度Fe3 对其生长速率没有显著影响,总脂含量随着Fe3 浓度升高呈现上升的趋势,均比对照有极显著提高,Fe3 浓度为0.75mmol/L时油脂产量最高。盐度对小球藻的生长有一定的抑制作用;油脂含量先随着盐度的增大而提高,当NaCl浓度达到0.6mol/L, 油脂含量又显著降低;油脂含量和油脂产量均在盐度为0.2mol/L时最高。光照强度对处于生长后期的小球藻的生长影响不大,但影响其油脂积累,小球藻的油脂含量和产量随光照强度的增大而显著提高,当光照强度增至280μmolm-2s-1时,油脂含量和油脂产量最高。温度对小球藻的生长速率、生物量、油脂含量和油脂产量都有显著的影响,在15-40℃范围内,随着培养温度的升高,生长速率、生物量、油脂含量和油脂产量都经历了一个先上升然后下降的过程,适合小球藻生长、积累油脂的温度范围是20-35℃,30-35℃时油脂产量最高,40℃时生物量、油脂含量和产量都最低。理化因子对生长和油脂含量的影响分为两种情况:1. 温度、光强、铁浓度和盐度的影响表现为在适宜生长的条件下提高油脂含量,这种模式可以称为“适宜模式”;2. 氮浓度的影响表现为在不利于生长的条件下提高油脂含量,这种模式可以称为“胁迫模式”。两种模式都可以提高油脂含量,但是,只有适宜模式才可以提高油脂产量。在筛选小球藻优良产油藻种时要注意,只有在适宜的培养条件下油脂含量高的藻种才具有高产油潜力。  相似文献

【背景】小球藻由于蛋白含量高、营养丰富,在水产养殖上可直接作为鱼、虾、贝类的优质饵料。【目的】对从养殖环境中分离的小球藻进行诱变,选育生长快、蛋白含量高的突变株,为水产养殖天然饵料生产提供优良藻种资源。【方法】以从养殖环境中筛选的生长相对较快且蛋白含量较高的TX作为出发藻株,对该藻株进行分子鉴定,并对该藻株进行紫外诱变、甲基磺酸乙脂(ethyl methyl sulfonate,EMS)诱变和复合诱变,采用96孔板高通量筛选技术和递进式重复筛选方法选育高生物量、高蛋白突变株。【结果】经18SrRNA基因序列分析,TX鉴定为Chlorella sorokiniana,从540个可能的突变株中筛选到8个遗传稳定且生长较快的突变株,其中H10的总蛋白含量达64.2%,可溶性蛋白含量达0.44g/L,干重达0.72g/L,分别较出发藻株提高3.4%、15.8%和26.2%。【结论】突变株H10蛋白含量高且生长较快,可用于天然饵料生产。  相似文献

Nannochloropsis salina was cultured batch-wise to evaluate the potential of the alga to produce biodiesel. The cells were harvested at the end of the exponential growth phase when the concentration was 18 x 10(6) cells/mL culture. The growth estimated as dry weight from this cell number was (3.8 +/- 0.7) mg/L. The lipid and triglyceride contents were 40% and 12% on a dry weight basis, respectively. The amount of the ratio triglycerides/total lipids was approximately 0.3. The composition of triglyceride fatty acid methyl esters (biodiesel) was analysed by gas-liquid chromatography and identified as: C14:0, C16:0, C16:1, C18:0, C18:1, C18:2, C18:3, C20:1, and C20:5. The ratio of unsaturated to saturated fatty acid contents was approximately 4.4. Additionally, the characterization of each individual fatty acid ester was discussed with regard to the fuel properties of biodiesel produced by the alga.  相似文献

An alga known as “Nannochloropsis”, isolated from a prawn farm in Hainan, China, has been critically investigated and identified as Chlorella, a member of the Chlorophyceae based on fatty acid composition, ultrastructure, and 18S rDNA. Cells of this alga were spherical, measured by 1–6 μm in diameter and were enclosed in thin walls of approximately 0.04 μm thickness. They contained several small mitochondria, two to three thylakoids and had no vacuoles. There were many pyrenoids in the algal cells and their thylakoid lamellae were sparse and not translucent. Many lipid droplets were present in the cytoplasm. The total lipid content of this alga was 3% per gram dry weight and its major fatty acids were C16:0, C18:0, C18:1, C18:2, C18:3 and C20:0. Eicosapentaenoic acid (C20:5, EPA) was not detected. The length of its 18S rDNA sequence was 1,712 bp. 18S rDNA sequence analyses indicated that this alga was a species of Chlorella.  相似文献

Two thioredoxins (named Ch1 and Ch2 in reference to their elution pattern on an anion-exchange column) have been purified to homogeneity from the green alga, Chlamydomonas reinhardtii. In this paper, we described the properties and the sequence of the most abundant form, Ch2. Its activity in various enzymatic assays has been compared with those of Escherichia coli and spinach thioredoxins. C. reinhardtii thioredoxin Ch2 can serve as a substrate for E. coli thioredoxin reductase with a lower efficiency when compared to the homologous system. In the presence of dithiothreitol (DTT), the protein is able to catalyze the reduction of porcine insulin. Thioredoxin Ch2 is as efficient as its spinach counterpart in the DTT or light activation of corn NADP-malate dehydrogenase, but it only activates spinach fructose-1, 6-bisphosphatase at very high concentrations. The complete primary structure of the C. reinhardtii thioredoxin Ch2 was determined by automated Edman degradation of the intact protein and of peptides derived from trypsin, chymotrypsin, clostripain, and SV8 protease digestions. It consists of a polypeptide of 106 amino acids (MW 11,808) and contains the well-conserved active site sequence Trp-Cys-Gly-Pro-Cys. The sequence of the algal thioredoxin Ch2 has been compared to that of thioredoxins from other sources and has the greatest similarity (67%) with the thioredoxin from Anabaena 7119.  相似文献

不同演替阶段臭柏种群的遗传多样性   总被引:2,自引:1,他引:1  
为了阐明分子变异与演替阶段或生态过渡带的联系,应用随机扩增多态性DNA (random amplified polymorphic DNA, RAPD) 标记方法对毛乌素沙地图克境内4个不同演替阶段的臭柏种群进行了分子生态学研究.用17个随机引物扩增出190条清晰谱带,其中173条为多态性谱带,并利用POPGENE 3.2 Version 1.31软件对数据进行处理.结果表明,臭柏种群的遗传多样性较丰富,各种群多态位点百分比在64.21%~74.63%,以演替早期的半固定沙地油蒿+臭柏种群最高(74.63%),演替亚顶极时期的固定沙地臭柏种群最低(64.21%);臭柏种群间分化较小,种群间遗传分化系数Gst= 0.1761, 82.39%的遗传变异存在于种群内;聚类分析显示,演替阶段相近的种群聚集到一起,反映了臭柏种群的遗传分化和演替阶段密切相关.利用Nei指数统计RAPD数据,各种群基因多样度在0.2163~0.2564之间,也证实了大部分的遗传变异存在于种群内.  相似文献

The release of total acetylcholine (ACh) and [3H]ACh was investigated in electrically stimulated cortical slices prepared from 4- and 18-month-old male Wistar rats. The slices were prelabeled with [3H]choline ([3H]Ch) and perfused with Krebs solution containing physostigmine. Total ACh was measured and the nature of the tritium efflux identified by HPLC. The total tritium content in the slices at the end of the incubation period was half as great in the old as in young rats. A linear relationship was found between stimulation frequencies (2, 5, and 10 Hz) and fractional [3H]ACh release in both young and old rats. In the latter the release was significantly smaller. At 10 Hz stimulation frequency the ratio between the two 2-min stimulation periods, S2/S1, was higher in the 18-month-old rats than in the young rats. Specific activity of the evoked ACh release was significantly smaller in S2 than in S1 in 4-month-old rats only. These findings indicate that the young synthetize ACh from endogenous unlabeled Ch more than older rats. In 18-month-old rats both the evoked total ACh and [3H]ACh release, expressed as picograms per minute, showed an approximately 50% decrease in both S1 and S2 stimulation periods, with no significant difference in specific activity. Phosphatidylserine (PtdSer) administration (15 mg/kg, i.p. daily) for 1 week to 18-month-old rats prevented the reduction in total evoked ACh release but not the reduction in evoked [3H]ACh release. Oleaginous microbial strains were cultivated to identify the best oil-producing strain amongst Yarrowia lipolytica (CGMCC 2.1398), Lipomyces starkeyi (CGMCC 2.1608), Rhodosporidium toruloides (CGMCC 2.1389), Mortierella isabellina (CGMCC 3.3410), Cunninghamella blakeleana (CGMCC 3.970), and Mycobacterium QJ311. A method for rapid determination of oil content and fatty acid composition was established to identify the optimum oil-producing strains. This method had a relative standard deviation of 4.09%, an average recovery ratio of 97.09% and a detection limit of 0.1-1.0 g. Mortierella isabellina CGMCC 3.3410 was identified as the best oil-producing strain amongst the six strains tested, with a total biomass of 75 g/10 L and a lipid content of 35%. A rapid screening method of oleaginous microorganisms is discussed for the first time.  相似文献

171,348 mosquitoes and 4,353 other arthropods collected at three centres in Queensland in 1972-1976 yielded 151 strains of 18 viruses. Culex annulirostris was the major source of virus isolation but 42 strains from Aedes normanensis indicate it to be a vector of importance. Ross River and Kokobera viruses were isolated at Kowanyama in the dry season, a finding of interest as being compatible with year-round survival in vector-vertebrate cycles. Culex fatigans has in part replaced Culex annulirostris in peridomestic breeding sites at Kowanyama; the infrequency of virus isolation from it suggests that this replacement may lower arbovirus infection rates. Twelve strains were identified as viruses antigenically distinct from any previously isolated in Australia or New Guinea: Ch16129, showed by the International Reference Centre for Arboviruses to be a previously undescribed member of the Simbu Group (Facey's Paddock virus), Ch16313 (Murweh), Ch19520 (Parker's Farm) and Ch19546 (little Sussex). The remaining strains were identified as viruses previously known in Australia, but included many new host or geographical records.  相似文献

A Proteus vulgaris isolated from external ulcers of the fresh water fish Channa punctatus showed multidrug resistance and heavy metal tolerance. The isolate from the ulcer showed resistance to chloramphenicol (Ch), nalidixic acid (Nx), streptomycin (Str) and tetracycline (Tet) with minimum inhibitory concentration (MIC) values of 750, 150, 75 and 125 microg/ml, respectively. The isolate showed growth in medium containing cadmium (Cd2+), up to a concentration of 2.5 mM indicating its heavy metal tolerance. Resistance to Ch, Str, Tet and Cd2+ of the isolate was lost after plasmid curing. Presence of plasmid DNA in the wild type and its absence in the cured P. vulgaris suggested that the resistance were plasmid mediated.  相似文献

The paper describes the composition of extracellular free fatty acids (FFAs) and intracellular fatty acids (FAs) in the enrichment cultures of the prokaryotic alga Spirulina platensis and the eukaryotic alga Chlorella vulgaris grown at optimal, supraoptimal, and suboptimal growth temperatures. With increasing growth temperature, the degree of unsaturation of the intracellular FAs of both algae decreased, while that of the extracellular FFAs of S. platensis increased. The composition of the extracellular FFAs of C. vulgaris practically did not depend on the growth temperature.  相似文献

Changes in the ordering and motion of lipids in response to changes in the external solute concentration have been studied by using the 5-nitroxide stearate (5NS) and 16-nitroxide stearate (16NS) spin probes in the plasma membrane of the halotolerant unicellular alga Dunaliella salina. Increases in ordering of the 5NS probe and decreases in motion of the 16NS probe were observed in cells equilibrated over 18 h at increasing NaCl concentrations. These changes probably resulted from the influence of the high NaCl concentration on the charged phospholipid head groups of the membrane. A short-term (less than 100 min) decrease in the order parameter, S, of the 5NS probe was observed for cells swollen by exposure to a sudden decrease of NaCl concentration from 5.0 to 2.5 M. After 100 min the value of S for 5NS was close to the value obtained in cells that had been equilibrated in 2.5 M-NaCl for 18 h. Since the cells had regained their original size and shape by 100 min it was assumed that the short-term decrease in S was associated with the swelling. A similar result was obtained when the cells were suddenly changed from 3.0 M- to 1.5 M-sorbitol. Conversely, an increase in S was observed for cells shrunk when the external solute concentration was doubled from 1.5 M- to 3.0 M-NaCl. As the cells regained their original size and shape the value of S decreased to the value observed in cells that had been equilibrated in 3.0 M-NaCl for 18 h. It is suggested that the changes in S are related to the movement of lipid into or out of a reservoir of membrane material as the membrane shrinks or expands. This movement of lipid maintains the tension of the membrane below the value at which it is disrupted. Such changes in lipid ordering could provide a mechanism whereby information about external osmotic-pressure changes is transmitted across the cell wall.  相似文献
