Stylostomes of the trombiculid mite larvae Neotrombicula pomeranzevi (Schluger), Hirsutiella zachvatkini (Schluger), Miyatrombicula esoensis (Sasa and Ogata) and Euschoengastia rotundata (Schluger) (Acariformes: Trombiculidae), formed in the host skin during feeding of the parasites on their natural hosts (voles) were studied histologically and histochemically. A stylostome is a variously shaped tube formed of solidified mite saliva that extends from the mouthparts of the parasite through the epidermis into the dermis of the host, and allows the mite to obtain its liquid food. The first step of stylostome formation is deposition of an eosinophilic cone, to which the larva’s chelicerae are glued. Organization of the stylostome depends on the mite species, and its walls may show weakly expressed longitudinal or transverse stratification. Histochemically, the stylostome is composed of complex glycoprotein with varying tinctorial properties through the width or the length of the stylostome’s walls. Beneath the distal end of the stylostome, irrespectively of its localization either in the epidermis or in the dermis of the host, a feeding cavity is formed as a result of the action of the hydrolytic components of the mite’s saliva forced through the stylostome into the wound. An inflammatory dermal reaction of moderate intensity is evolved during larval feeding and stylostome formation. It is manifested by the infiltration of the foci with neutrophiles, lymphocytes and macrophages and by dilation of capillaries of the terminal vessel bed and filling them by erythrocytes and other blood elements. Around the stylostome, necrosis of the epidermal cells occurs, leucocytes come to the damaged area and fuse with the necrotic epidermal cells, leading to the formation of the large scabs on the surface of the host’s skin. In the case of E. rotundata, single capsules having a terminal opening and containing feeding larva are formed on the abdomen of the hosts. The walls of the capsules are composed of the mite’s saliva flowing upon the surface of the host’s skin. At the bottom of the capsule, a stylostome perforating the epidermis is also present.  相似文献

Water-mite larvae of the subgenus Arrenurus (Acari, Hydrachnellae) act as habitual ectoparasites on zygopteran imagines, mostly attached to the soft membranous cuticle. The powerful larval pedipalp claws grasp the cuticle and the distal sabre-like cheliceral segments tear it, thus obtaining the host's tissue fluids. The chelicerae and palps co-operate to anchor the larva. The attached larva soon produces—in the host's subcuticular epidermis layer, and separated from the haemocoele by the thin sheet of subepidermal connective tissue—an elongated pouch, the stylostome, consisting of an acellular gelatinous substance, apparently secreted by the larva. Presumably, interaction with components in the host's tissue fluids solidifies the stylostome, which becomes firmly fixed to the host's body wall. The stylostome is thought to lengthen by repeated alternation between the suction of tissue fluids from the host and secretion of fresh soft substance forming additional stylostome segments. The resilient stylostome develops in a cleft between the host's cuticle and the subepidermal connective tissue in connection with partial decomposition of the host's epidermis, probably caused by cytotoxins leaking from the stylostome. In attempts to wall off the wound and thus to avoid influx of foreign substances, the host lays down a melanin sleeve around the stylostome at the perforation site. Lodgement of the functional stylostome separated from the haemocoele within the host's tissues might explain why the newly-formed stylostome seems immune from cellular host reactions. However, cellular encapsulation and melanization appear to happen to outworn, non-active stylostomes.  相似文献

Ivy gourd, Coccinia grandis (L.) Voigt, patches throughout Kailua-Kona, Hawaii Island, HI, were identified as persistent sources of melon fly, Bactrocera cucurbitae (Coquillett). These patches had a low incidence of Psyttalia fletcheri (Silvestri), its major braconid parasitoid natural enemy in Hawaii, and were used to evaluate augmentative releases of P. fletcheri against melon fly. In field cage studies of releases, numbers of melon flies emerging from ivy gourd fruit placed inside treatment cages were reduced up to 21-fold, and numbers of parasitoids were increased 11-fold. In open field releases of P. fletcheri into ivy gourd patches, parasitization rates were increased 4.7 times in release plots compared with those in control plots. However, there was no significant reduction in emergence of melon flies from fruit. In subsequent cage tests with sterile melon flies and P. fletcheri, combinations of sterile flies and P. fletcheri produced the greatest reduction (9-fold) in melon fly emergence from zucchini, Cucurbita pepo L. Reductions obtained with sterile flies alone or in combination with parasitoids were significantly greater than those in the control, whereas those for parasitoids alone were not. Although these results suggest that the effects of sterile flies were greater than those for parasitoids, from a multitactic melon fly management strategy, sterile flies would complement the effects of P. fletcheri. Cost and sustainability of these nonchemical approaches will be examined further in an ongoing areawide pest management program for melon fly in Hawaii.  相似文献

Studies were done to determine the effect of 4 fruit ripeness levels on the abundance of Psyttalia fletcheri (Silvestri),a parasitoid of Bactrocera cucurbitae (Coquillett). In addition,an improved trapping method was developed for monitoring massreleased adult parasitoids in the field. A total of 420,000 adult P. fletcheri were released over a period of 15 weeks,in a 0.16 ha experimental field plot containing cucumber,zucchini and tomato in Kauai,HI. We recovered significantly more P. fletcheri from rotting fruits of all fruit types than from immature,commercial grade or oversized fruits. In addition,there was a positive correlation between the number of P. fletcheri and B. cucurbitae recovered from rotting fruits. Average parasitism in rotting tomato,zucchini and cucumber was 5,19 and 36% respectively. Yellow pan traps were as effective as sticky sphere traps for monitoring P. fletcheri,indicating that parasitoids were foraging close to the ground where rotting fruits occurred. The pan traps were much easier to use,less expensive and required less set up time. Recovery of parasitoids from both fruit collections and pan traps indicated that P. fletcheri was least abundant in tomato. Both male and female parasitoids exhibited a weakly aggregated distribution in each crop. We discuss modified sampling and release methods for population estimation and augmentative release studies of P. fletcheri.  相似文献

为分析普通小麦(Triticumaestivum)-天兰冰草(Agropyronintermedium)部分双二倍体──远中2号(2n=54)的染色体构成,用生物素(biotin-16-dUTP)标记天兰冰草染色体组DNA作为探针,以普通小麦品种中国春染色体组DNA为封闭DNA(blockingDNA),与远中2号的有丝分裂中期染色体DNA进行了分子原位杂交。证明远中2号除具有普通小麦的21对染色体外,附加了1对小麦-天兰冰草易位染色体(即天兰冰草染色体片段易位到小麦染色体的两臂端部)、5对天兰冰草染色体。说明小麦-天兰冰草部分双二倍体在形成过程中染色体行为是比较复杂的,不仅可能产生小麦-天兰冰草染色体间易位,而且小麦染色体也可能与天兰冰草染色体的3种染色休组染色体共同参与组建新的染色体组附加到小麦中去。  相似文献

Arrenurus larvae, ectoparasitic on zygopteran imagoes, attach to the host's cuticle and tear it to obtain the host's tissue fluids. Within the host's epidermis, each larval mite produces a feeding device, the stylostome, a narrow gelatinous resilient blind sac. Heavy mite infestation brings about several wounds in close proximity, accompanied by loss of more or less extensive areas of the epidermis. Despite wound repair by congregating hemocytes, local lack of epidermis seems to enfeeble the host, presumably owing to desiccation. Heavily mite-loaded zygopterans have lost the typical agility and are easily caught. A mite-induced mortality seems to exist in zygopteran populations; the infestation contributes to reduced longevity. The study of formation and decline of the arrenurid stylostome in zygopterans renders it possible to trace cellular defence reactions under natural conditions. Most stylostomes seem to thwart the ability of the host to recognize them as foreign bodies. The host's defence appears as a two-step reaction: (1) initial hemolymph clotting and deposition of melanin associated with aggregating hemocytes at the penetration site, (2) occasional subsequent melanization and cellular encapsulation of the stylostome.  相似文献

Larvae of the mite Trombidium newelli Welbourn and Flessel are ectoparasitic on adult alfalfa weevil, Hypera postica (Gyllenhall), an insect pest of alfalfa. The mite larvae are found under the elytra, attached to the dorsal surface of weevils abdomen. T. newelli larvae use their chelicerae to penetrate the pliable and weakly sclerotized areas of the hosts integument, and to hold on firmly to the host. The attachment sites associated with larval mites of different degrees of engorgement were examined using both light and scanning electron microscopy. The ventral inside surface of parasitized host tergites revealed a characteristic injury as spots varying in color from white-yellow to golden-brown depending on the engorged size of the mite. In addition, spots associated with fully engorged mite larvae showed an aggressive dendritically radiating mass expanding outward from the site of cheliceral penetration. This mass, known as feeding tube or stylostome was branched, with each branch ending in a cluster of closed bulbs. Stylostomes appear to exist independently in the hosts tissue. Stylostome mass corresponded with the engorgement state of the mite, increasing as the mite larva increased in size. The possible nature and mechanism of stylostome formation is discussed.  相似文献

The stylostome of larvae of the trombiculids Leptotrombidium scutellare (Nagayo et al.), Leptotrombidium fletcheri (Womersley et Heaslip) and Leptotrombidium deliense (Walch) was studied experimentally at different time intervals after larval attachment using the histological method. The stylostome of these species has the same organization and belongs to the epidermal combined with the mixed type, developing more in width than in length. Neither transverse nor conspicuous longitudinal layers are present within the stylostome walls, which stain predominantly in red with Azan, also showing longitudinal portions with blue staining. Larvae tend to attach closely to each other and scabs, consisting of the hyperkeratotic epidermal layers fusing with migrating inflammatory cells, develop around the attachment sites. The dermis shows inflammatory foci with dilated capillaries and inflammatory cells inserting in the connective tissue layer underneath the stylostome. The feeding cavity, which is moderately expressed, may be found either in the epidermis or in the dermis. It contains inflammatory cells and their debris in the liquefied host tissues. The stylostome length depends on the character of the attachment site (the thicker epidermis or scab the longer the stylostome), and does not directly correspond to the stages of larval feeding. Nevertheless, at the 48-h time interval, nearly all attached larvae are found to be fully fed and their midgut cells are filled with nutritional globules.  相似文献

中间偃麦草麦、小麦和小麦-中间偃麦草2Ai-2附加系Z1、Z2、X6,代换系ZD28等进行RAPD分析,从320个RAPD引物中,鉴定出2Ai-2染色体特异的2个RAPD标记OPO05650和OPMO414000。利用这2个特异OPO05和OPM04,PCR扩增普通小麦CS(ABD)及其近缘植物中间偃麦草(E1E2St)、拟鹅冠草(St),长穗偃麦草(E)、簇毛麦(V)、黑麦(R)、大麦(H)粗山羊草(D)等基因组DNA。结果表明,OPO05650和OPO41400均是2Ai-2染色体上St基因组区域的特异标记。将上棕2个特异片段分离回收、克隆、测序,根据测序结果重新设计、合成特异引物,成功地转换RAPD标记为SCAR(sequence characterizked amplifed region)标记SC-05和SC-M4。利用SCAR标记对不同材料进行分析的结果表明,凡含有2Ai-2染色体的抗黄矮病材料及拟鹅冠草均产生一条扩增带,不含2Ai-2染色体的材料,包括小麦、长穗麦草、簇毛麦、黑麦、在麦、粗山羊草以有含有其他他中间偃麦草染色休的附加系,均没有扩增产物,说明上棕2个SCAR标记是中间偃麦草2Ai-2染色体的特异性PCR标记,且是2Ai-2染色体上St基因组区域的特异性标记。克隆与鉴定中间偃麦草的2个SCAR扩增片段TiSCO5和TiSCM4。结果表明,克隆的中间偃麦草TiSCO5和TiSCM4特异片段,分别是St基因组特异性的寡拷贝序列有多拷贝重复序列,为St基因组遗传研究的新探针。  相似文献

Chigger mites of the genus Leptotrombidium (Acari: Trombiculidae) transmit scrub typhus, caused by Rickettsia tsutsugamushi (= R.orientalis) in South-East Asia. In China, eighty-two species of Leptotrombidium have been recorded; these are listed with the names of Provinces where they were found. Five species, L.deliense, L.insularae, L.kaohuense, L.rubellum and L.scutellare, have been implicated as Chinese vectors of scrub typhus. A brief key is given to the larvae of all but three of the Leptotrombidium mites known in China.  相似文献

Along a line of the four nemertean orders, Archi-, Palaeo-,Hetero-, and Hoplonemertea established by Iwata, formation ofthe foregut is described in detail on the basis of data hithertorecorded. The axiality of the egg, larva and adult worm of aspecies may change as a function of foregut morphogenesis inthe embryo. During foregut formation the mouth moves from thevegetal pole to the ventral side of the body. In Archinemertea,the larva has an angle of 60° formed by the long axis ofthe body and the foregut, while in Palaeonemertea the angleis 45°. This relationship is retained in the adult. Theplacement of the mouth thus obtained in the larva and adultworm seems to be important for the systematics of the Nemertea,especially for the Archinemertea and Palaeonemertea.  相似文献

According to conventional theory, little genomic changes should occur in homozygous and stable amphiploids of the grass family, particularly those involving polyploid wheat as a parent. In the present study, however, extensive genomic changes were detected in two octoploid partial amphiploids of common wheat ( Triticum aestivum L. )-wheatgrass ( Agropyron intermedium (Host) P.B. = Elytrigia intermedia (Host) Nevski = Thinopyrum intermedium (Host) Barkworth and Dewey), namely Zhong 3 and Zhong 5, by RFLP an, analysis using 10 low-copy, wheat chromosome-specific sequences and 33 representative homoeologous group-specific sequences as probes. C, enomic changes involved loss of wheat hybridization fragment (s) and/or acquisition of new fragment(s). Uniformity of the RFLP patterns among 5 individual plants taken respectively from Zhong 3 and Zhong 5 in two successive generations, suggested that genomic changes probably had occurred in the early few generations after octoploid amphiploid formation, and remained essentially static thereafter, The highly similar RFLP patterns between Zhong 3 and Zhong 5, which had identical genomic constitution but differed from each other due to involvement of different wheat varieties as parents imply that genomic changes were probably not at random. Possible causes for the extensive and rapid genomic changes in the newly formed plant amphiploids, as well as their implications for polyploid genome evolution and breeding application are discussed.  相似文献

Diet and foraging behaviour of three species of burrowing huntsman spiders, Leucorchestris arenicola, L. steyni and Carparachne aureoflava , from the Namib dunes were investigated over a three-year period. These nocturnal spiders are polyphagous predators that prey on more than 97 species of insects, arachnids and reptiles. Most prey were nocturnal or crepuscular tenebrionid beetles, moths and weevils. Diet varied regionally owing to faunal differences, but was relatively constant over seasons. Although spiders occasionally captured prey greater than themselves, average prey length was about two-thirds their own length. Prey size was not strongly related to spider size. Larger spiders were both cannibals and intraguild predators. Foraging pattern of L. arenicola was variable with several nights of activity followed by one or several nights of rest. Spiders foraged within 3 m of the burrow, but occasionally pursued prey or neighbouring conspecifics further. Large prey (> 3 mm) were captured approximately every five weeks in summer and every seven weeks in winter, producing an average annual consumption of ≅ 10 prey/spider. Namib huntsmen are sit-and-wait predators within narrow territories waiting for the fortuitous arrival of prey and are thus unlikely to limit prey populations. They compensate for food shortages by cannibalism, thus restricting their own population.  相似文献

研究了菌株DN2降解烟碱的特性和对烟草废弃物中烟碱的降解情况。结果表明,该菌降解烟碱的最适条件为接种量为5 %,温度30 ℃,初始pH值为6.5。在该条件下,对初始烟碱浓度为500 mg/L的降解过程进行考察。结果表明,未经烟碱诱导的降解曲线呈倒S曲线,半衰期为17.43 h;经烟碱诱导的降解曲线符合Eckenfelder动力学模型,半衰期为4.10 h。添加0.1 %(质量分数)葡萄糖,可提高菌株DN2的烟碱耐受浓度,达5000 mg/L。菌株DN2能够降解烟草废弃物水提液中的烟碱(烟碱含量约为2220 mg/L),60 h时烟碱的降解率为95.22 %,表明该菌在治理烟碱污染环境方面具有应用价值。  相似文献

Criteria were established to improve quality control methods for augmentative releases of the opiine parasitoids, Diachasmimorpha longicaudata (Ashmead) and Psyttalia fletcheri (Silvestri). These included correlating puparial weight with adult emergence and sex ratio at the Honolulu rearing facility, and determining the effect of air shipments and field releases on parasitoid emergence and subsequent mortality. There was a positive relationship between the weight of 7–10‐day‐old host puparia and percentage of emergence for both D. longicaudata and P. fletcheri. Standardization of shipping methods included placement of ice blocks in the top levels of the ice chests, prompt shipment and pick‐up of samples, and cooling of puparia before shipment to minimize metabolic heat generated in the samples, and to delay emergence of samples. Before standardization, emergence losses of up to 58% were observed for D. longicaudata and 18% for P. fletcheri. When shipping methods were standardized, emergence was no longer reduced. In addition, low emergence rates were associated with reduced flight propensity of D. longicaudata (Rs = ‐ 0.45 at Kilauea and ‐ 0.54 at Kealia). At the two release sites, 88–95% of adult D. longicaudata that did not escape the release containers were males.  相似文献

Giant cells induced in roots of Impatiens balsamina by Meloidogyne javanica and Meloidogyne incognita have been examined by light and electron microscopy. The first sign of giant-cell formation was division of cells surrounding a larva. Cell plate alignment appeared to proceed normally, but cytokinesis was unsuccessful and binucleate cells formed subsequently. No wall breakdown was evident then or later. The number of nuclei appeared to increase by repeated mitosis without separation by cytokinesis. Although no holes in walls were observed, wall stubs were found, and mechanisms for their formation are suggested.  相似文献

Partial amphiploids between wheat (Triticum aestivum L.) and Thinopyrum species play an important role in the transfer and use of traits from alien species. A wheat-Thinopyrum intermedium partial amphiploid, TAI8335, and its alien parent were characterized by a combination of genomic in situ hybridization (GISH) and cytological observations. Evidence from GISH indicated that the donor parent Th. intermedium possessed seven pairs of S, seven J(s) and 21 J chromosomes. Mitotic observation showed that the majority of TAI8335 plants had 56 chromosomes, but a few had 54 to 55, in some cases with two to three additional telochromosomes. The chromosomes in most pollen mother cells of plants with 2n = 56 formed 28 bivalents, averaging 27.12 in 223 cells, suggesting a basic cytological stability. Sequential GISH patterns using genomic Pseudoroegneria spicata and genomic Th. intermedium DNA as probes revealed that TAI8335 had fourteen chromosomes derived from Th. intermedium and its alien genome consisted of one pair of S-, three pairs of J(s) - and one pair of J-genome chromosomes as well as two translocated chromosome pairs, one being a Robertsonian translocation and another an intercalary translocation, both of which involved J and S genome. Two of the telochromosomes in the aneuploid plants originated from the J genome and one from wheat. Disease screening demonstrated this line was highly resistant to leaf rust, stem rust, stripe rust and powdery mildew. This study showed that the partial amphiploid TAI8335 appears to serve as a novel source for the transfer of resistance genes for multiple fungal pathogens to wheat.  相似文献

Oviposition and the development of larvae in Hydrachna cruenta Müller 1769 was studied using methods of light and scanning electron microscopy. Some features of similarity in the reproductive behavior and development of larvae were noted between H. cruenta and terrestrial trombiculid mites: spermatophore fertilization, the presence of one eggshell, tolerance to short-term egg drying, and formation of the stylostome. The life of H. cruenta larvae in water during the search for a host may be regarded as an adaptation of this species to a new habitat.  相似文献

Mandibular first molars in mice ranging in age from 18 days prenatal to 5 days postnatal were used for light and electron microscopic examinations of the enamel-free area (EFA) during development of the occlusal cusp (mesiobuccal cusp). Notable morphological changes in the inner enamel epithelium and the cells of the stratum intermedium were observed. At prenatal age of 18 days, the inner enamel epithelium of the EFA (EFA epithelium) was composed of a layer of columnar cells and covered by the cells of the stratum intermedium. Two days after birth, the EFA epithelium was made up largely of preameloblasts, with mitochondria located in the proximal side of the cells toward the stratum intermedium. The cells of the stratum intermedium were irregularly shaped, with wide intercellular spaces between them. At a postnatal age of 3 days, most of the EFA epithelial cells resembled maturation-stage ameloblasts, being short and columnar in shape and having nuclei located in their proximal side. Distal cell membranes were folded, and mitochondria were scattered throughout the cytoplasm. In 4-day-old mice, the EFA epithelium was found to be formed of short columnar or cuboidal cells with distinct intercellular spaces. The cells of the stratum intermedium could no longer be detected, and cells of the EFA epithelium could not be distinguished from those of the stellate reticulum. Odontoblasts of the EFA were arranged and polarized parallel to the basal lamina, and odontoblastic processes extended toward the cusp tip. The orientation of thin and thick collagen fibers within predentin and dentin was also parallel to the basal lamina. Even after dentin mineralization, disrupted basal lamina and long, aperiodic, fine fibrils were found between the epithelium and the dentin. Following the disappearance of the basal lamina and fine fibrils, stippled material and crystals appeared on the dentin surface. The mineralized matrix, which x-ray microanalytical energy peaks identified as containing calcium and phosphorus, was continuous with enamel in the distal slope of the cusp at the cusp tip. Thus, the inner enamel epithelium of the EFA differentiated into secretory cells capable of enamel-like matrix formation.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献
