One hundred strains of Rhizobium leguminosarum bv. trifolii were isolated from roots of wheat cultivated in rotation with clover in two different regions of Morocco. The isolates were first screened for their effect on the growth of the cultivar Rihane of wheat cultivated in an agricultural soil under greenhouse conditions. After 5 weeks of growth, 14 strains stimulating the fresh or dry matter yield of shoots were selected and used in a second pot inoculation trial performed with two different agricultural soils. The results show that the strains behaved differently according to the soil used. In the loamy sand Rabat, strain IAT 168 behaved potentially like a plant growth promoting rhizobacteria (PGPR), as indicated by the 24% increases (P < 0.1) observed in wheat shoot dry matter and grain yields. In the silty clay Merchouch, no PGPR activity was observed, and 6 strains showed a significant deleterious effect on yields. These observations suggest that it is very important in a crop rotation system to choose a R. leguminosarum bv. trifolii strain that is effective with clover and shows PGPR activity with wheat to avoid deleterious effects on wheat yields.  相似文献

Standard and locally isolated nodule bacteria and plant growth-promoting rhizobacteria (PGPR) were grown on crude oil and individual pure hydrocarbons as sole sources of carbon and energy. The nodule bacteria included two standard Rhizobium leguminosarum strains, two standard Bradyrhizobium japonicum strains, and one unknown nodule bacterial strain that was locally isolated from Vicia faba nodules. The PGPR included one standard Serratia liquefaciens strain and two locally isolated strains of Pseudomonas aeruginosa and Flavobacterium sp. The pure hydrocarbons tested included n-alkanes with chain lengths from C9 to C40 and the aromatic hydrocarbons benzene, biphenyle, naphthalene, phenanthrene, and toluene. Quantitative gas liquid chromatographic analyses confirmed that pure cultures of representative nodule bacteria and PGPR could attenuate n-octadecane and phenanthrene in the surrounding nutrient medium. Further, intact nodules of V. faba containing bacteria immobilized on and within those nodules reduced hydrocarbon levels in a medium in which those nodules were shaken. It was concluded that legume crops are suitable phytoremediation tools for oily soil, since they enrich such soils not only with fixed nitrogen, but also with hydrocarbon-utilizing microorganisms. Further, legume nodules may have biotechnological value as materials for cleaning oily liquid wastes.  相似文献

Plant growth‐promoting rhizobacteria (PGPR) affect growth of host plants through various direct and indirect mechanisms. Three native PGPR (Pseudomonas putida) strains isolated from rhizospheric soil of a Mentha piperita (peppermint) crop field near Córdoba, Argentina, were characterised and screened in vitro for plant growth‐promoting characteristics, such as indole‐3‐acetic acid (IAA) production, phosphate solubilisation and siderophore production, effects of direct inoculation on plant growth parameters (shoot fresh weight, root dry weight, leaf number, node number) and accumulation and composition of essential oils. Each of the three native strains was capable of phosphate solubilisation and IAA production. Only strain SJ04 produced siderophores. Plants directly inoculated with the native PGPR strains showed increased shoot fresh weight, glandular trichome number, ramification number and root dry weight in comparison with controls. The inoculated plants had increased essential oil yield (without alteration of essential oil composition) and biosynthesis of major essential oil components. Native strains of P. putida and other PGPR have clear potential as bio‐inoculants for improving productivity of aromatic crop plants. There have been no comparative studies on the role of inoculation with native strains on plant growth and secondary metabolite production (specially monoterpenes). Native bacterial isolates are generally preferable for inoculation of crop plants because they are already adapted to the environment and have a competitive advantage over non‐native strains.  相似文献

Two experiments were conducted to investigate the effect of inoculating Vicia faba plants (broad beens) raised in clean and oily sand with nodule-forming rhizobia and plant-growth-promoting rhizobacteria (PGPR) on growth of these plants in sand and to test whether this can improve the phytoremediation potential of this crop for oily desert areas. It was found that crude oil in sand at concentrations < 1.0% (w/w) enhanced the plant heights, their fresh and dry weights, the total nodule weights per plant, and the nitrogen contents of shoots and fruits. Similar enhancing effects were recorded when roots of the young plants were inoculated with nodule bacteria alone, PGPR alone, or a mixture of one strain of nodule bacteria and one of the PGPR. Such plant growth effects were associated with a better phytoremediation potential of V. faba plants for oily sand. The total numbers of oil-utilizing bacteria increased in the rhizosphere and more hydrocarbons were eliminated in sand close to the roots. The nodule bacteria tested were two strains of Rhizobium leguminosarum and the PGPR were Pseudomonas aeruginosa and Serratia liquefaciens. The four strains were found to use crude oil, n-octadecane, and phenanthrene as sole sources of carbon and energy. It was concluded that coinoculation of V. faba plant roots in oily sand with nodule bacteria and PGPR enhances the phytoremediation potential of this plant for oily desert sand through improving plant growth and nitrogen fixation.  相似文献

Bacterial wilt (Ralstonia solanacearum) is one of the production constraints of potato (Solanum tuberosum). The intent of the study was to evaluate potential of bacterial antagonists to suppress bacterial wilt disease development and evaluate the role of the strains as plant growth-promoting rhizobacteria (PGPR) in potato. One hundred-twenty rhizosphere bacterial isolates were screened against virulent strain of Ralstonia solanacearum PPRC-Rs. After in vitro screening, six antagonistic strains (PFMRI, BS-DFS, PF9, PF20, BC, and BS-wly) with inhibition diameter >11 mm were selected and studied further in the greenhouse, in vivo. During in vivo study, the strains were evaluated for their effect in suppressing disease development in terms of area under disease progress curve (AUDPC) and increasing biomass (plant height and dry weight) of potato. Accordingly, PFMRI, BS-DFS, and PF9, significantly reduced AUDPC by 78.6, 66, and 64.3%, and wilt incidence by 82.7, 66.2, and 65.7%, respectively, compared to the control. During the sole application, the strains significantly (P < 0.0001) increased plant height by 35.6, 45.9, and 45%, and dry matter by 111, 130.4, and 129%, respectively compared to non-bacterized control. In the presence of the pathogen strain PFMRI, BS-DFS, and PF9 increased plant height by 66, 50, and 48.2%, and dry matter by 153.8, 96.8, and 92.5%, respectively compared to the pathogen treated control. Hence, the study shows that PFMRI, BS-DFS, and PF9 strains have potential use in potato bioprotection, as PGPR or in an integrated bacterial wilt management; whose effectiveness under a variety of field conditions should be investigated.  相似文献

Sixty-four native bacterial colonies were isolated from mycorrhizal roots of Helianthemum almeriense colonized by Terfezia claveryi, mycorrhizosphere soil, and peridium of T. claveryi to evaluate their effect on mycorrhizal plant production. Based on the phylogenetic analysis of the 16S rDNA partial sequence, 45 different strains from 17 genera were gathered. The largest genera were Pseudomonas (40.8 % of the isolated strains), Bacillus (12.2 % of isolated strains), and Varivorax (8.2 % of isolated strains). All the bacteria were characterized phenotypically and by their plant growth-promoting rhizobacteria (PGPR) traits (auxin and siderophore production, phosphate solubilization, and ACC deaminase activity). Only bacterial combinations with several PGPR traits or Pseudomonas sp. strain 5, which presents three different PGPR traits, had a positive effect on plant survival and growth. Particularly relevant were the bacterial treatments involving auxin release, which significantly increased the root-shoot ratio and mycorrhizal colonization. Moreover, Pseudomonas mandelii strain 29 was able to considerably increase mycorrhizal colonization but not plant growth, and could be considered as mycorrhiza-helper bacteria. Therefore, the mycorrhizal roots, mycorrhizosphere soil, and peridium of desert truffles are environments enriched in bacteria which may be used to increase the survival and mycorrhization in the desert truffle plant production system at a semi-industrial scale.  相似文献

AIMS: Plant growth promoting rhizobacteria (PGPR) are commonly used as inoculants for improving the growth and yield of agricultural crops, however screening for the selection of effective PGPR strains is very critical. This study focuses on the screening of effective PGPR strains on the basis of their potential for in vitro auxin production and plant growth promoting activity under gnotobiotic conditions. METHODS AND RESULTS: A large number of bacteria were isolated from the rhizosphere soil of wheat plants grown at different sites. Thirty isolates showing prolific growth on agar medium were selected and evaluated for their potential to produce auxins in vitro. Colorimetric analysis showed variable amount of auxins (ranging from 1.1 to 12.1 mg l-1) produced by the rhizobacteria in vitro and amendment of the culture media with l-tryptophan (l-TRP), further stimulated auxin biosynthesis (ranging from 1.8 to 24.8 mg l-1). HPLC analysis confirmed the presence of indole acetic acid (IAA) and indole acetamide (IAM) as the major auxins in the culture filtrates of these rhizobacteria. A series of laboratory experiments conducted on two cv. of wheat under gnotobiotic (axenic) conditions demonstrated increases in root elongation (up to 17.3%), root dry weight (up to 13.5%), shoot elongation (up to 37.7%) and shoot dry weight (up to 36.3%) of inoculated wheat seedlings. Linear positive correlation (r = 0.99) between in vitro auxin production and increase in growth parameters of inoculated seeds was found. Based upon auxin biosynthesis and growth-promoting activity, four isolates were selected and designated as plant growth-promoting rhizobacteria (PGPR). Auxin biosynthesis in sterilized vs nonsterilized soil inoculated with selected PGPR was also monitored that revealed superiority of the selected PGPR over indigenous microflora. Peat-based seed inoculation with selected PGPR isolates exhibited stimulatory effects on grain yields of tested wheat cv. in pot (up to 14.7% increase over control) and field experiments (up to 27.5% increase over control); however, the response varied with cv. and PGPR strains. CONCLUSIONS: It was concluded that the strain, which produced the highest amount of auxins in nonsterilized soil, also caused maximum increase in growth and yield of both the wheat cv. SIGNIFICANCE AND IMPACT OF STUDY: This study suggested that potential for auxin biosynthesis by rhizobacteria could be used as a tool for the screening of effective PGPR strains.  相似文献

Most oil from oceanic spills converges on coastal ecosystems, such as mangrove forests, which are threatened with worldwide disappearance. Particular bacteria that inhabit the rhizosphere of local plant species can stimulate plant development through various mechanisms; it would be advantageous if these would also be capable of degrading oil. Such bacteria may be important in the preservation or recuperation of mangrove forests impacted by oil spills. This study aimed to compare the bacterial structure, isolate and evaluate bacteria able to degrade oil and stimulate plant growth, from the rhizospheres of three mangrove plant species. These features are particularly important taking into account recent policies for mangrove bioreme-diation, implying that oil degradation as well as plant maintenance and health are key targets. Fifty-seven morphotypes were isolated from the mangrove rhizospheres on Bushneil-Haas (BH) medium supplemented with oil as the sole carbon source and tested for plant growth promotion. Of this strains, 60% potentially fixed nitrogen, 16% showed antimicrobial activity, 84% produced siderophores, 51% had the capacity to solubilize phosphate, and 33% produced the indole acetic acid hormone. Using gas chromatography, we evaluated the oil-degrading potential of ten selected strains that had different morphologies and showed Plant Growth Promoting Rhizobacteria (PGPR) features. The ten tested strains showed a promising degradation profile for at least one compound present in the oil. Among degrader strains, 46% had promising PGPR potential, having at least three of the above capacities. These strains might be used as a consortium, allowing the concomitant degradation of oil and stimulation of mangrove plant survival and maintenance.  相似文献

A screening for PGPRs was carried out in the rhizosphere of wild populations of Cistus ladanifer. Two hundred and seventy bacteria were isolated, purified and grouped by morphological criteria. Fifty percent of the isolates were selected and tested for aminocyclopropanecarboxylic acid (ACC) degradation, auxin and siderophore production and phosphate solubilisation. Fifty-eight percent of the isolates showed at least one of the evaluated activities, with phosphate solubilisation and siderophore production being the most abundant traits. After PCR-RAPDs (Randomly amplified polymorphic DNA) analysis, 11 groups appeared with 85% similiarity, revealing the low diversity in the system. One strain of each group was tested in a biological assay, and those that enhanced Cistus growth were identified by 16S rDNA sequencing.Although seven of the 11 assayed strains were phosphate solubilisers and able to produce siderophores, only one was really effective in increasing all biometric parameters in Cistus ladanifer seedlings, the lack of effect of the other six probably being due to the rich substrate used. This suggests that other mechanisms apart from nutrient mobilisation might be involved in growth promotion by this strain. However, the low diversity together with the high redundancy detected by PCR-RAPDs and the predominance of strains able to mobilise nutrients in the rhizosphere of Cistus reveals that the plant selects for bacteria that can help to supply scarce nutrients. This type of plant growth promoting rhizobacteria (PGPR) strains should be succesful in reforestation practices.  相似文献

Sediment organic matter is a key stressor for submerged macrophyte growth, which negatively impacts the ecological restoration of lakes. Plant growth-promoting rhizobacteria (PGPR) were screened from the rhizosphere of submerged macrophytes and used due to their promoting effect on Vallisneria natans under a high sediment organic matter load. Root exudates were used as the sole carbon source to obtain the root affinity strains. Eight isolates were selected from the 61 isolated strains, based on the P solubilization, IAA production, cytokinins production and ACC deaminase activity. The analysis of the 16S rDNA indicated that one strain was Staphylococcus sp., while the other seven bacterial strains were Bacillus sp. They were all listed in low-risk groups for safety use in agricultural practices. The plant height significantly increased after inoculation with PGPR strains, with the highest rate of increase reaching 96%. This study provides an innovative technique for recovering submerged macrophytes under sediment organic matter stress.  相似文献

AIMS: Deleterious rhizosphere inhabiting bacteria (DRB) have potential to suppress plant growth. This project focuses on the isolation of DRB with potential for development as commercial products for weed control. METHODS AND RESULTS: Bacteria were isolated from the rhizosphere, rhizoplane, and endorhizosphere of seedlings and mature plants of wild radish (Raphanus raphanistrum), annual ryegrass (Lolium rigidum) and capeweed (Arctotheca calendula) growing in vineyards in the Swan Valley, Western Australia. A majority (81.5%) of the 442 strains was obtained from either rhizospheres or rhizoplanes. Rapid screening techniques were developed to evaluate in the laboratory and glasshouse the effects of bacteria on plants. Strains were screened in the glasshouse for deleterious effects on annual ryegrass, wild radish, grapevine rootlings (Vitis vinifera) and the legume cover crop subterranean clover (Trifolium subterraneum). Three strains were identified using the Biolog system and 16S rRNA gene sequencing as two strains of Pseudomonas fluorescens (WSM3455 and WSM3456) and one strain of Alcaligenes xylosoxidans (WSM3457). One of the P. fluorescens (WSM3455) strain produced hydrogen cyanide, an inhibitor of plant roots and a broad-spectrum antimicrobial compound. CONCLUSIONS: Three strains specifically inhibited wild radish but had no significant deleterious effects on either grapevine rootlings or subterranean clover. SIGNIFICANCE AND IMPACT OF THE STUDY: This study suggested manipulation of the weed seedling rhizosphere using identified DRB as a potential biocontrol agent for wild radish.  相似文献

Iron is one of the essential elements for a proper plant development. Providing plants with an accessible form of iron is crucial when it is scant or unavailable in soils. Chemical chelates are the only current alternative and are highly stable in soils, therefore, posing a threat to drinking water. The aim of this investigation was to quantify siderophores produced by two bacterial strains and to determine if these bacterial siderophores would palliate chlorotic symptoms of iron-starved tomato plants. For this purpose, siderophore production in MM9 medium by two selected bacterial strains was quantified, and the best was used for biological assay. Bacterial culture media free of bacteria (S) and with bacterial cells (BS), both supplemented with Fe were delivered to 12-week-old plants grown under iron starvation in hydroponic conditions; controls with full Hoagland solution, iron-free Hoagland solution and water were also conducted. Treatments were applied twice along the experiment, with a week in between. At harvest, plant yield, chlorophyll content and nutritional status in leaves were measured. Both the bacterial siderophore treatments significantly increased plant yield, chlorophyll and iron content over the positive controls with full Hoagland solution, indicating that siderophores are effective in providing Fe to the plant, either with or without the presence of bacteria. In summary, siderophores from strain Chryseobacterium C138 are effective in supplying Fe to iron-starved tomato plants by the roots, either with or without the presence of bacteria. Based on the amount of siderophores produced, an effective and economically feasible organic Fe chelator could be developed.  相似文献

Fluorescent pseudomonads are among the most influencing plant growth-promoting rhizobacteria in plants rhizosphere. In this research work the plant growth-promoting activities of 40 different strains of Pseudomonas fluorescens and Pseudomonas putida, previously isolated from the rhizosphere of wheat (Triticum aestivum L.) and canola (Brassica napus L.) and maintained in the microbial collection of Soil and Water Research Institute, Tehran, Iran, were evaluated. The ability of bacteria to produce auxin and siderophores and utilizing P sources with little solubility was determined. Four strains of Wp1 (P. putida), Cfp10 (Pseudomonas sp.), Wp150 (P. putida), and Wp159 (P. putida) were able to grow in the DF medium with ACC. Thirty percent of bacterial isolates from canola rhizosphere and 33% of bacterial isolates from wheat rhizosphere were able to produce HCN. The results indicate that most of the bacteria, tested in the experiment, have plant growth-promoting activities. This is the first time that such PGPR species are isolated from the Iranian soils. With respect to their great biological capacities they can be used for wheat and canola inoculation in different parts of the world, which is of very important agricultural implications.  相似文献

Plant growth promoting rhizobacteria Ochrobactrum lupini KUDC1013 and Novosphingobium pentaromativorans KUDC1065 isolated from Dokdo Island, S. Korea are capable of eliciting induced systemic resistance (ISR) in pepper against bacterial spot disease. The present study aimed to determine whether plant growth-promoting rhizobacteria (PGPR) strains including strain KUDC1013, strain KUDC1065, and Paenibacillus polymyxa E681 either singly or in combinations were evaluated to have the capacity for potential biological control and plant growth promotion effect in the field trials. Under greenhouse conditions, the induced systemic resistance (ISR) effect of treatment with strains KUDC1013 and KUDC1065 differed according to pepper growth stages. Drenching of 3-week-old pepper seedlings with the KUDC-1013 strain significantly reduced the disease symptoms. In contrast, treatment with the KUDC1065 strain significantly protected 5-week-old pepper seedlings. Under field conditions, peppers treated with PGPR mixtures containing E681 and KUDC1013, either in a two-way combination, were showed greater effect on plant growth than those treated with an individual treatment. Collectively, the application of mixtures of PGPR strains on pepper might be considered as a potential biological control under greenhouse and field conditions.  相似文献

【背景】挖掘兼具烟碱降解和植物根际促生细菌(Plant Growth-Promoting Rhizobacteria,PGPR)功能的细菌资源,有助于保护土壤质量,实现绿色种植。【目的】分析烤烟根际细菌多样性,筛选可降解高浓度烟碱的PGPR。【方法】采用纯培养法在选择性培养基上分离烟碱降解细菌。通过BOXA1R-PCR分析技术、16SrRNA基因测序及系统发育树构建,对菌株的遗传多样性和分类学地位进行分析。进一步评价了菌株的吲哚乙酸(Indole-3-Acetic Acid,IAA)活性、溶磷能力、病原菌拮抗能力等PGPR指标,以筛选出高效PGPR,最后通过盆栽试验验证其促生效果。【结果】分离得到58株烟碱降解细菌,根据BOXA1R-PCR指纹图谱选取11株菌进行16S rRNA基因序列测定,结果表明,58株菌分别属于芽孢杆菌属(Bacillus)、假单胞菌属(Pseudomonas)、拉乌尔菌属(Raoultella)和短波单胞菌属(Brevundimonas)4个属,以芽孢杆菌属(Bacillus)为优势菌属。58株细菌中48.28%的菌株可产IAA,27.59%具备溶磷能力,37.93%具备纤维素降解能力,G2-13、G2-3及HT2-8因促生与抗病特性突出而被筛选为目标功能菌。盆栽试验结果表明,G2-13菌株对幼苗生长的促进作用明显,可使株高与地上部鲜重分别增加33.05%与53.32%。【结论】烤烟根际存在较为丰富多样的烟碱降解细菌,它们在种植业上具有潜在的应用价值。  相似文献

Thirty-two isolates were obtained from wheat rhizosphere by wheat germ agglutinin (WGA) labeled with fluorescein isothiocyanate (FITC). Most isolates were able to produce indole acetic acid (65.6%) and siderophores (59.3%), as well as exhibited phosphate solubilization (96.8%). Fourteen isolates displayed three plant growth-promoting traits. Among these strains, two phosphate-dissolving ones, WS29 and WS31, were evaluated for their beneficial effects on the early growth of wheat (Triticum aestivum Wan33). Strain WS29 and WS31 significantly promoted the development of lateral roots by 34.9% and 27.6%, as well as increased the root dry weight by 25.0% and 25.6%, respectively, compared to those of the control. Based on 16S rRNA gene sequence comparisons and phylogenetic positions, both isolates were determined to belong to the genus Bacillus. The proportion of isolates showing the properties of plant growth-promoting rhizobacteria (PGPR) was higher than in previous reports. The efficiency of the isolation of PGPR strains was also greatly increased by WGA labeled with FITC. The present study indicated that WGA could be used as an effective tool for isolating PGPR strains with high affinity to host plants from wheat roots. The proposed approach could facilitate research on biofertilizers or biocontrol agents.  相似文献

植物根际促生菌是一类可促进植物生长的有益细菌,有效的根际促生菌剂可以减少化肥施用。以束村氏菌属(Tsukamurella sp.)P9、伯克霍尔德氏菌属(Burkholderia sp.)P10、以及P9和P10混合菌液作为接种菌株,研究促生菌对花生生长、植株及土壤营养、根际土壤微生物类群及功能的影响。30 d盆栽实验结果表明,接种组的花生鲜重、株高及根长均显著提高;根际土壤细菌总数、固氮菌和溶磷菌数均明显高于未接种组;氮循环功能菌群数量有不同程度提高,土壤蔗糖酶、脲酶及过氧化氢酶均高于对照;土壤碱解氮及速效钾显著提高,植株营养指标有所提升,尤以P10接种效果更优。本研究初步结论表明2株促生菌通过活化土壤微生物、提高植株的有效营养元素含量,促进了花生的生长。  相似文献

【背景】植物-微生物联合修复土壤重金属污染日渐兴起,获取与超富集植物高效互作的微生物是实现联合修复技术进步的关键。龙葵在镉农田污染修复中广泛应用。【目的】筛选可促进龙葵生长及富集镉的耐镉根际促生菌。【方法】从龙葵根际土分离耐镉菌株,筛选具有良好促生特性的菌株,水培试验考察镉胁迫下菌株对植物生长及镉富集能力的影响,确定可促进龙葵生长及富集镉的微生物菌株,通过生理生化特性和16S rRNA基因序列分析进行菌株的初步鉴定。【结果】分离得到NT1、AXY1、AW2和AW1四株强化龙葵富集镉促生菌,经鉴定分别为Lysinibacillus sp.、Beijerinckia fluminensis、Achromobacter animicus和Herbaspirillum huttiense。上述菌株均可有效促进龙葵生长,增加其株高和干物质积累,提高地上部镉富集量。其中,NT1可使株高、地上部干重分别增加31.33%和62.65%,AW2可使地上部镉富集量增加37.29%。【结论】筛选所得菌株可为提高植物修复效率提供实践依据,为研制田间施用生态功能菌剂做铺垫,用于农田镉污染的微生物-龙葵联合原位修复。  相似文献

One of the major mechanisms utilized by plant growth-promoting rhizobacteria (PGPR) to facilitate plant growth and development is the lowering of ethylene levels by deamination of 1-aminocyclopropane-1-carboxylic acid (ACC) the immediate precursor of ethylene in plants. The enzyme catalysing this reaction, ACC deaminase, hydrolyses ACC to α -ketobutyrate and ammonia. Several bacterial strains that can utilize ACC as a sole source of nitrogen have been isolated from rhizosphere soil samples. All of these strains are considered to be PGPR based on the ability to promote canola seedling root elongation under gnotobiotic conditions. The treatment of plant seeds or roots with these bacteria reduces the amount of ACC in plants, thereby lowering the concentration of ethylene. Here, a rapid procedure for the isolation of ACC deaminase-containing bacteria, a root elongation assay for evaluating the effects of selected bacteria on root growth, and a method of assessing bacterial ACC deaminase activity are described in detail. This should allow researchers to readily isolate new PGPR strains adapted to specific environments.  相似文献
