AIMS: Plant growth promoting rhizobacteria (PGPR) are commonly used as inoculants for improving the growth and yield of agricultural crops, however screening for the selection of effective PGPR strains is very critical. This study focuses on the screening of effective PGPR strains on the basis of their potential for in vitro auxin production and plant growth promoting activity under gnotobiotic conditions. METHODS AND RESULTS: A large number of bacteria were isolated from the rhizosphere soil of wheat plants grown at different sites. Thirty isolates showing prolific growth on agar medium were selected and evaluated for their potential to produce auxins in vitro. Colorimetric analysis showed variable amount of auxins (ranging from 1.1 to 12.1 mg l-1) produced by the rhizobacteria in vitro and amendment of the culture media with l-tryptophan (l-TRP), further stimulated auxin biosynthesis (ranging from 1.8 to 24.8 mg l-1). HPLC analysis confirmed the presence of indole acetic acid (IAA) and indole acetamide (IAM) as the major auxins in the culture filtrates of these rhizobacteria. A series of laboratory experiments conducted on two cv. of wheat under gnotobiotic (axenic) conditions demonstrated increases in root elongation (up to 17.3%), root dry weight (up to 13.5%), shoot elongation (up to 37.7%) and shoot dry weight (up to 36.3%) of inoculated wheat seedlings. Linear positive correlation (r = 0.99) between in vitro auxin production and increase in growth parameters of inoculated seeds was found. Based upon auxin biosynthesis and growth-promoting activity, four isolates were selected and designated as plant growth-promoting rhizobacteria (PGPR). Auxin biosynthesis in sterilized vs nonsterilized soil inoculated with selected PGPR was also monitored that revealed superiority of the selected PGPR over indigenous microflora. Peat-based seed inoculation with selected PGPR isolates exhibited stimulatory effects on grain yields of tested wheat cv. in pot (up to 14.7% increase over control) and field experiments (up to 27.5% increase over control); however, the response varied with cv. and PGPR strains. CONCLUSIONS: It was concluded that the strain, which produced the highest amount of auxins in nonsterilized soil, also caused maximum increase in growth and yield of both the wheat cv. SIGNIFICANCE AND IMPACT OF STUDY: This study suggested that potential for auxin biosynthesis by rhizobacteria could be used as a tool for the screening of effective PGPR strains.  相似文献

Plant interactions with plant growth-promoting rhizobacteria (PGPR) are highly dependent on plant genotype. Modern plant breeding has largely sought to improve crop performance but with little focus on the optimization of plant × PGPR interactions. The interactions of the model PGPR strain Pseudomonas kilonensis F113 were therefore compared in 199 ancient and modern wheat genotypes. A reporter system, in which F113 colonization and expression of 2,4-diacetylphloroglucinol biosynthetic genes (phl) were measured on roots was used to quantify F113 × wheat interactions under gnotobiotic conditions. Thereafter, eight wheat accessions that differed in their ability to interact with F113 were inoculated with F113 and grown in greenhouse in the absence or presence of stress. F113 colonization was linked to improved stress tolerance. Moreover, F113 colonization and phl expression were higher overall on ancient genotypes than modern genotypes. F113 colonization improved wheat performance in the four genotypes that showed the highest level of phl expression compared with the four genotypes in which phl expression was lowest. Taken together, these data suggest that recent wheat breeding strategies have had a negative impact on the ability of the plants to interact with PGPR.  相似文献

This study assessed the possible role of different traits in selected plant growth-promoting rhizobacteria (PGPR) for improving wheat growth and yield under natural conditions. Rhizobacteria exhibiting 1-aminocyclopropane-1-carboxylate (ACC)-deaminase activity were isolated and screened for their growth-promoting activity in wheat under axenic conditions. Five isolates belonging to Pseudomonas and one Burkholderia caryophylli isolate that showed promising performances under axenic conditions were selected and characterized for in vitro ACC-deaminase activity, chitinase activity, auxin production, P solubilization, and root colonization. These isolates were then used as inocula for wheat cultivated under natural conditions in pot and/or field trials. Significant increases in root elongation, root weight, tillers per pot, 1,000-grain weight, and grain and straw yields were observed in response to inoculation with PGPR in the pot trials. Inoculation with these PGPR was also effective under field conditions and increased the wheat growth and yield significantly. However, the efficacy of the strains was inconsistent under the axenic, pot, and field conditions. Pseudomonasfluorescens (ACC50), which exhibited a relatively high in vitro ACC-deaminase activity, chitinase activity, auxin production, and P solubilization and more intensive root colonization, was the most efficient isolate under the field conditions. Therefore, these results demonstrated that ACC-deaminase activity is an efficient parameter for the selection of promising PGPR under axenic conditions. However, additional traits of PGPR, including auxin production, chitinase activity, P solubilization, and root colonization, are also important for selecting PGPR as biofertilizers.  相似文献

One hundred strains of Rhizobium leguminosarum bv. trifolii were isolated from roots of wheat cultivated in rotation with clover in two different regions of Morocco. The isolates were first screened for their effect on the growth of the cultivar Rihane of wheat cultivated in an agricultural soil under greenhouse conditions. After 5 weeks of growth, 14 strains stimulating the fresh or dry matter yield of shoots were selected and used in a second pot inoculation trial performed with two different agricultural soils. The results show that the strains behaved differently according to the soil used. In the loamy sand Rabat, strain IAT 168 behaved potentially like a plant growth promoting rhizobacteria (PGPR), as indicated by the 24% increases (P < 0.1) observed in wheat shoot dry matter and grain yields. In the silty clay Merchouch, no PGPR activity was observed, and 6 strains showed a significant deleterious effect on yields. These observations suggest that it is very important in a crop rotation system to choose a R. leguminosarum bv. trifolii strain that is effective with clover and shows PGPR activity with wheat to avoid deleterious effects on wheat yields.  相似文献

【背景】挖掘兼具烟碱降解和植物根际促生细菌(Plant Growth-Promoting Rhizobacteria,PGPR)功能的细菌资源,有助于保护土壤质量,实现绿色种植。【目的】分析烤烟根际细菌多样性,筛选可降解高浓度烟碱的PGPR。【方法】采用纯培养法在选择性培养基上分离烟碱降解细菌。通过BOXA1R-PCR分析技术、16SrRNA基因测序及系统发育树构建,对菌株的遗传多样性和分类学地位进行分析。进一步评价了菌株的吲哚乙酸(Indole-3-Acetic Acid,IAA)活性、溶磷能力、病原菌拮抗能力等PGPR指标,以筛选出高效PGPR,最后通过盆栽试验验证其促生效果。【结果】分离得到58株烟碱降解细菌,根据BOXA1R-PCR指纹图谱选取11株菌进行16S rRNA基因序列测定,结果表明,58株菌分别属于芽孢杆菌属(Bacillus)、假单胞菌属(Pseudomonas)、拉乌尔菌属(Raoultella)和短波单胞菌属(Brevundimonas)4个属,以芽孢杆菌属(Bacillus)为优势菌属。58株细菌中48.28%的菌株可产IAA,27.59%具备溶磷能力,37.93%具备纤维素降解能力,G2-13、G2-3及HT2-8因促生与抗病特性突出而被筛选为目标功能菌。盆栽试验结果表明,G2-13菌株对幼苗生长的促进作用明显,可使株高与地上部鲜重分别增加33.05%与53.32%。【结论】烤烟根际存在较为丰富多样的烟碱降解细菌,它们在种植业上具有潜在的应用价值。  相似文献

Fluorescent pseudomonads are among the most influencing plant growth-promoting rhizobacteria in plants rhizosphere. In this research work the plant growth-promoting activities of 40 different strains of Pseudomonas fluorescens and Pseudomonas putida, previously isolated from the rhizosphere of wheat (Triticum aestivum L.) and canola (Brassica napus L.) and maintained in the microbial collection of Soil and Water Research Institute, Tehran, Iran, were evaluated. The ability of bacteria to produce auxin and siderophores and utilizing P sources with little solubility was determined. Four strains of Wp1 (P. putida), Cfp10 (Pseudomonas sp.), Wp150 (P. putida), and Wp159 (P. putida) were able to grow in the DF medium with ACC. Thirty percent of bacterial isolates from canola rhizosphere and 33% of bacterial isolates from wheat rhizosphere were able to produce HCN. The results indicate that most of the bacteria, tested in the experiment, have plant growth-promoting activities. This is the first time that such PGPR species are isolated from the Iranian soils. With respect to their great biological capacities they can be used for wheat and canola inoculation in different parts of the world, which is of very important agricultural implications.  相似文献

Summary Treatment of wheat (Triticum aestivum L.) seedlings with elicitors originating from either plant or fungal cell walls induces about a 2-fold increase of wheat germ agglutinin (WGA) in the roots. While the WGA content in roots of healthy plants normally decreases as a function of germination time, a transient accumulation of WGA could be observed in plants challenged with different fungi, including Rhizoctonia solani, Fusarium culmorum, Pythium ultimum and Neurospora crassa. Peak levels in challenged roots were 2 to 5 times as high as in control plants. Most of this induced WGA could be released from the roots by soaking them in a solution of the hapten N-acetylglucosamine. On the basis of the results obtained it is postulated that WGA may be involved in the defence of wheat against fungal attack.  相似文献

目的研究小麦PGPR(植物根际促生菌)菌株的个体生态学及其在小麦根圈的定植动态。方法采用三亲本杂交法将发光酶基因luxAB转人具有固氮能力的小麦根际促生菌Azotobacter N2106菌株中,获得标记菌株N2106-L,将标记菌株接种到灭菌和非灭菌的黄褐土、红壤和黄潮土中研究其存活状况,采用根盒试验追踪标记菌株在小麦根圈的定植动态。结果标记菌株N2106-L具有发光活性和对km、str、tet三种抗生素的抗性,且具有较好的遗传稳定性。N2106-L在灭菌土壤中的数量稍高于非灭菌土壤;在3种土壤中的数量依次为:黄褐土〉黄潮土〉红壤。N2106-L在小麦根表定植密度大于根际定植密度;在小麦根际,小麦播种10d时标记菌株在0-2cm深度根际土壤定植达到最大值(2.17±0.25)×10^6CFU/g土,20d时在2-4cm深度的根际土壤中达到最高定植水平(3.92±0.47)×10^5CFU/g土;在小麦根表,标记菌株在小麦播种10d时在所有深度的根段均达到最高定植水平,0-2cm根段定植密度为(3.60±0.60)×10^6CFU/g鲜根,12cm以下根段达到(2.78±0.56)×10^4CFU/g鲜根。结论标记菌株随着根的伸长不断向根尖方向扩散,且较为稳定地在小麦根圈定植,研究结果为小麦PGPR菌株的应用提供了可靠实验数据。  相似文献

Heavy metal contamination of agricultural soils has increased along with industrialization. Mercury is a toxic heavy metal and a widespread pollutant in the ecosystem. Mercury-tolerant and plant growth-promoting rhizobacteria (PGPR) HG 1, HG 2, and HG 3 were isolated from the rhizosphere of plants growing in a mercury-contaminated site. These isolates were able to grow in the presence of mercury ranging from 10 to 200 µM in minimal medium and 25 to 500 µM in LB medium. The strains were characterized by morphological, biochemical, and plant growth-promoting traits. In the present study, these PGPR strains were analyzed for their involvement in metal stress tolerance in Triticum aestivum (wheat). Two bacterial strains, namely, Enterobacter ludwigii (HG 2) and Klebsiella pneumoniae (HG 3), showed better growth promotion of T. aestivum seedlings under metal stress. Different growth parameters like, water content and biochemical properties were analyzed in the PGPR-inoculated wheat plants under 75 µM HgCl2. Shoot length, root length, shoot dry weight, root dry weight and relative water content (RWC) were significantly higher in inoculated plants compared to uninoculated plants under stress condition. Proline content, electrolyte leakage, and malondialdehyde content (shoots and roots) were significantly lower in inoculated plants with respect to uninoculated plants under mercury stress. Therefore, it could be assumed that all these parameters collectively improve plant growth under mercury stress conditions in the presence of PGPR. Hence, these PGPRs can serve as promising candidates for increasing plant growth and also have immense potential for bioremediation of mercury-contaminated soils.  相似文献

Salinity is one of the major environmental threats for successful crop production, hampering plant growth due to the osmotic effect and nutritional and hormonal imbalances. The application of naturally occurring plant growth-promoting rhizobacteria (PGPR) is an emerging technology aimed at ameliorating the negative impact of salinity. However, the results obtained in the laboratory can sometimes not be reproduced in the field. The aim of the study reported here was to evaluate the effect of PGPR inoculation on seed germination in a saline environment under axenic conditions and on enhancement of the growth and yield of wheat under natural salt-affected field conditions. Wheat seeds were inoculated with pre-isolated strains of Pseudomonas putida, Enterobacter cloacae, Serratia ficaria, and Pseudomonas fluorescens and sown at different salinity levels (1, 2, 3, 6, 9, 12, 15 dS m-1). Inoculation with these strains was found to enhance the germination percentage, germination rate, and index of wheat seeds up to 43, 51, and 123 %, respectively, over the uninoculated control at the highest salinity level. The potential of these PGPR for improving the growth and yield of wheat was also evaluated at two natural salt-affected sites. Inoculation with PGPR resulted a significant increase in the growth and yield parameters of wheat at both sites. The inoculated plants also improved the nutrient status of the wheat plants. The inoculated plants had low sodium and high nitrogen, phosphorus, and potassium contents. Our results show that such rhizobacterial strains may be used as an effective tool for enhancing plant growth under salinity stress and for maximizing the utilization of salt-affected soils.  相似文献

Two plant growth‐promoting rhizobacterial (PGPR) strains, Bacillus subtilis SU47 and Arthrobacter sp. SU18, were found to tolerate 8% NaCl. Wheat co‐inoculated with these two PGPR strains, and grown under different salinity regimes (2–6 dS m?1), showed an increase in dry biomass, total soluble sugars and proline content. Wheat sodium content was reduced under co‐inoculated conditions but not after single inoculation with either strain or in the control. The activity of antioxidant enzymes in wheat leaves decreased under salinity stress after PGPR co‐inoculation, suggesting these PGPR species could be used for amelioration of stress in wheat plants. Activity of three antioxidant enzymes in wheat grown with both PGPR strains was reduced, most notably that of catalase activity at a salinity of 6 dS m?1, when compared with the control. The results indicate that co‐inoculation with B. subtilis and Arthrobacter sp. could alleviate the adverse effects of soil salinity on wheat growth.  相似文献

In this study, we isolated bacteria from rhizosphere and endorhizophere of wheat crops of the central region of Argentina. The isolates were phenotypically characterized and the restriction patterns of 16S rDNA (ARDRA) using endonuclease AluI were analysed. Representative isolates were used to evaluate the effect of the inoculation on the growth of wheat under greenhouse conditions. The effects of plant growth-promoting bacteria on wheat plants were studied by evaluating shoot fresh and dry weights and root fresh and dry weights. One native strain increased the shoot and root dry biomass by 23% and 45% respectively. Other strains increased the shoot dry biomass. A 1.5 kb fragment of the 16S rRNA gene of one isolate was sequenced. This isolate showed high identity with different species of Pseudomonas.  相似文献

Although plant growth-promoting rhizobacteria (PGPR) have been reported to influence plant growth, yield and nutrient uptake by an array of mechanisms, the specific traits by which PGPR promote plant growth, yield and nutrient uptake were limited to the expression of one or more of the traits expressed at a given environment of plant–microbe interaction. We selected nine different isolates of PGPR from a pool of 233 rhizobacterial isolates obtained from the peanut rhizosphere on the basis of ACC-deaminase activity. The nine isolates were selected, initially, on the basis of germinating seed bioassay in which the root length of the seedling was enhanced significantly over the untreated control. All the nine isolates were identified as Pseudomonas spp. Four of these isolates, viz. PGPR1, PGPR2, PGPR4 and PGPR7 (all fluorescent pseudomonads), were the best in producing siderophore and indole acetic acid (IAA). In addition to IAA and siderophore-producing attributes, Pseudomonas fluorescens PGPR1 also possessed the characters like tri-calcium phosphate solubilization, ammonification and inhibited Aspergillus niger and A. flavus in vitro. P. fluorescens PGPR2 differed from PGPR1 in the sense that it did not show ammonification. In addition to the traits exhibited by PGPR1, PGPR4 showed strong in vitro inhibition to Sclerotium rolfsii. The performances of these selected plant growth-promoting rhizobacterial isolates were repeatedly evaluated for 3 years in pot and field trials. Seed inoculation of these three isolates, viz. PGPR1, PGPR2 and PGPR4, resulted in a significantly higher pod yield than the control, in pots, during rainy and post-rainy seasons. The contents of nitrogen and phosphorus in soil, shoot and kernel were also enhanced significantly in treatments inoculated with these rhizobacterial isolates in pots during both the seasons. In the field trials, however, there was wide variation in the performance of the PGPR isolates in enhancing the growth and yield of peanut in different years. Plant growth-promoting fluorescent pseudomonad isolates, viz. PGPR1, PGPR2 and PGPR4, significantly enhanced pod yield (23–26%, 24–28% and 18–24%, respectively), haulm yield and nodule dry weight over the control in 3 years. Other attributes like root length, pod number, 100-kernel mass, shelling out-turn and nodule number were also enhanced. Seed bacterization with plant growth-promoting P. fluorescens isolates, viz. PGPR1, PGPR2 and PGPR4, suppressed the soil-borne fungal diseases like collar rot of peanut caused by A. niger and PGPR4 also suppressed stem rot caused by S. rolfsii. Studies on the growth patterns of PGPR isolates utilizing the seed leachate as the sole source of C and N indicated that PGPR4 isolate was the best in utilizing the seed leachate of peanut, cultivar JL24. Studies on the rhizosphere competence of the PGPR isolates, evaluated on the basis of spontaneous rifampicin resistance, indicated that PGPR7 was the best rhizoplane colonizer and PGPR1 was the best rhizosphere colonizer. Although the presence of growth-promoting traits in vitro does not guarantee that an isolate will be plant growth promoting in nature, results suggested that besides ACC-deaminase activity of the PGPR isolates, expression of one or more of the traits like suppression of phytopathogens, solubilization of tri-calcium phosphate, production of siderophore and/or nodulation promotion might have contributed to the enhancement of growth, yield and nutrient uptake of peanut.  相似文献

In the roots of bread wheat (Triticum aestivum L.) seedlings, the effects of pretreatment with 28 nM wheat germ agglutinin (WGA) and successive action of 1 mM cadmium acetate on growth, phytohormone balance, lignin deposition, and also cadmium accumulation and distribution were studied. Priority data on cadmium-induced ABA-mediated reversible accumulation of WGA in the roots, which was accompanied by its excretion in the medium of seedling incubation, were obtained. Pretreatment with WGA exerted a clear protective effect on seedling growth in the presence of cadmium, which was based on a decrease in the amplitude of stress-induced shifts in the balance between IAA and ABA and preventing the reduction in the cytokinin level. Acceleration of lignification of the cell walls in the basal parts of roots of seedlings pretreated with WGA and subjected to stress is shown, and this limits cadmium entry into the plant.  相似文献

Sixty-four native bacterial colonies were isolated from mycorrhizal roots of Helianthemum almeriense colonized by Terfezia claveryi, mycorrhizosphere soil, and peridium of T. claveryi to evaluate their effect on mycorrhizal plant production. Based on the phylogenetic analysis of the 16S rDNA partial sequence, 45 different strains from 17 genera were gathered. The largest genera were Pseudomonas (40.8 % of the isolated strains), Bacillus (12.2 % of isolated strains), and Varivorax (8.2 % of isolated strains). All the bacteria were characterized phenotypically and by their plant growth-promoting rhizobacteria (PGPR) traits (auxin and siderophore production, phosphate solubilization, and ACC deaminase activity). Only bacterial combinations with several PGPR traits or Pseudomonas sp. strain 5, which presents three different PGPR traits, had a positive effect on plant survival and growth. Particularly relevant were the bacterial treatments involving auxin release, which significantly increased the root-shoot ratio and mycorrhizal colonization. Moreover, Pseudomonas mandelii strain 29 was able to considerably increase mycorrhizal colonization but not plant growth, and could be considered as mycorrhiza-helper bacteria. Therefore, the mycorrhizal roots, mycorrhizosphere soil, and peridium of desert truffles are environments enriched in bacteria which may be used to increase the survival and mycorrhization in the desert truffle plant production system at a semi-industrial scale.  相似文献

Drought stress substantially impedes crop productivity throughout the world. Microbial based approaches have been considered a potential possibility and are under study. Based on our prior screening examination, two distinct and novel biofilm-forming PGPR strains namely Bacillus subtilis-FAB1 and Pseudomonas azotoformans-FAP3 are encompassed in this research. Bacterial biofilm development on glass surface, microtiter plate and seedling roots were assessed and characterized quantitatively and qualitatively by light and scanning electron microscopy. Above two isolates were further evaluated for their consistent performance by inoculating on wheat plants in a pot-soil system under water stresses. Bacterial moderate tolerance to ten-day drought was recorded on the application of individual strains with wheat plants; however, the FAB1 + FAP3 consortium expressively improved wheat survival during drought. The strains FAB1 and FAP3 displayed distinct and multifunctional plant growth stimulating attributes as well as effective roots and rhizosphere colonization in combination which could provide sustained wheat growth during drought. FAB1 and FAP3-induced alterations cooperatively conferred improved plant drought tolerance by controlling physiological traits (gs, Ci, E, iWUE and PN), stress indicators (SOD, CAT, GR, proline and MDA content) and also maintained physico-chemical attributes and hydrolytic enzymes including DHA, urease, ALP, protease, ACP and β glucosidase in the soil. Our findings could support future efforts to enhance plant drought tolerance by engineering the rhizobacterial biofilms and associated attributes which requires in-depth exploration and exploiting potential native strains for local agricultural application.  相似文献

Plant growth-promoting rhizobacteria (PGPR) are known to influence plant growth by various direct or indirect mechanisms. A total of 216 phosphate-solubilizing bacterial isolates were isolated from different rice rhizospheric soil in Northern Thailand. These isolate were screened in vitro for their plant growth-promoting activities such as solubilization of inorganic phosphate, ammonia (NH3), catalase and cell wall-degrading enzyme activity. It was found that 100% solubilized inorganic phosphate, 77.77% produced NH3 and most of the isolates were positive for catalase. In addition, some strains also produced cell wall-degrading enzymes such as protease (7%), chitinase (1%), cellulase (3%) and β-glucanase (3%), as evidenced by phenotypic biochemical test and quantitative assay using spectrophotometry. The isolates could exhibit more than two or three plant growth-promoting (PGP) traits, which may promote plant growth directly or indirectly or synergistically. Part of this study focused on the effect of NaCl, temperature, and pH on a specific the bacterial isolate Acinetobacter CR 1.8. Strain CR 1.8 was able to grow on up to 25% NaCl, between 25 and 55°C, and at pH 5–9. Maximum solubilization of tricalcium phosphate and aluminium phosphate was obtained at neutral pH, and 37°C. Strain CR 1.8 had protease activity but no cellulase, β-glucanase and cellulase activities.  相似文献
