Acetylation of pumpkin (Cucurbita pepo, lady godiva variety) polysaccharide using acetic anhydride with pyridines as catalyst under different conditions was conducted to obtain different degrees of acetylation on a laboratory scale. Furthermore, antioxidant activities and cytoprotective effects of pumpkin polysaccharide and its acetylated derivatives were investigated employing various established in vitro systems. Results showed that addition of pyridine as catalyst could increase the degree of substitution, whereas volume of acetic anhydride had little effect. The acetylated polysaccharides in DPPH scavenging radical activity assay, superoxide anion radical activity assay and reducing power assay exhibited higher antioxidant activity than that of unmodified polysaccharide. H2O2-induced oxidative damages on rat thymic lymphocyte were also prevented by pumpkin polysaccharide and its acetylated derivatives and the derivatives presented higher protective effects. On the whole, acetylated polysaccharide showed relevant antioxidant activity both in vitro and in a cell system.  相似文献

Porphyran extracted from Porphyra haitanensis is a sulfated polysaccharide, which possesses excellent antioxidant activities. In this study, we prepared one low-molecular-weight porphyran and its sulfated, acetylated, phosphorylated and benzoylated derivatives. Their antioxidant activities were investigated including scavenging effect of superoxide, hydroxyl and 1,1-diphenyl-2-picrylhydrazyl radicals. The results of chemical analysis and FT-IR spectrums showed the modification was successful. And in addition, we found that certain derivative exhibited stronger antioxidant activity than low-molecular-weight porphyran. The benzoylated derivative showed the most excellent antioxidant activity in three assays, so this derivative needs to be attended to.  相似文献

The crude polysaccharide (LEP) was extracted by hot water from the fruiting bodies of Lentinus edodes, and further purified by DEAE-cellulose and Sepharose CL-6B chromatography, giving three polysaccharide fractions coded as LEPA1, LEPB1 and LEPC1. In this study, their chemical and physical characteristics of polysaccharide fractions and antioxidant capacities, including scavenging activity against hydroxyl radicals, superoxide radicals and Fe2+-chelating ability, were valuated. The results showed that LEPC1 exhibited significantly antioxidant activity at a concentration-dependent manner. Therefore these results indicated that the water-extractable polysaccharide fraction was a potent antioxidant and could be developed to be new health medicine for fighting against various human diseases.  相似文献

In order to study the relationship between chemical structure and properties of modified carrageenans versus antioxidant activity in vitro, kappa-carrageenan oligosaccharides were prepared through mild hydrochloric acid hydrolysis of the polysaccharide, and these were used as starting materials for the partial synthesis of their oversulfated, acetylated, and phosphorylated derivatives. The structure and substitution pattern of the oligosaccharides and their derivatives were studied using FTIR and (13)C NMR spectroscopy, and their in vitro antioxidant activities were investigated. Certain derivatives of the carrageenan oligosaccharides exhibited higher antioxidant activity than the polysaccharides and oligosaccharides in certain antioxidant systems. The oversulfated and acetylated derivatives, which scavenge superoxide radicals, the phosphorylated and low-DS acetylated derivatives, which scavenge hydroxyl radicals, and the phosphorylated derivatives, which scavenge DPPH radicals, all exhibited significant antioxidant activities in the systems examined. The effect of the molecular weight of the carrageenan on antioxidant activities, however, is not obvious from these studies.  相似文献

Porphyran extracted from red algae Porphyra haitanensis is a sulfated polysaccharide, which possesses excellent antioxidant activities. In this study, we prepared the acetylated, phosphorylated and benzoylated derivatives of porphyran. And then the antioxidant activities of all the samples were investigated including scavenging effects of superoxide and hydroxyl radicals and reducing power. The results of chemical analysis and FT-IR spectrum showed the modifications of porphyran were successful. And in addition, we found that certain derivative exhibited stronger antioxidant activity than raw material. And the mechanism of the structure–function relationship of these derivatives needs to be attended to.  相似文献

Ultrasonic circulating extraction technology was applied for the polysaccharide extraction from Asparagus officinalis. The crude polysaccharides were deproteinized by Sevag method and three main polysaccharide fractions, AOP-4, AOP-6 and AOP-8 were obtained by fractional precipitation with gradient concentrations of ethanol (40%, 60% and 80%). The in vitro antitumor and antioxidant activities of the polysaccharide fractions were evaluated by MTT assay and free radical-scavenging assay, respectively. Deproteinized AOPs showed higher antioxidant and antitumor activities than crude AOP. AOP-4 with molecular weight 5.75 × 104 Da showed significant function of scavenging hydroxyl radical. Three AOP fractions had significant antitumor activity against HeLa and BEL-7404 cells in a dose dependent manner. Furthermore, the inhibit activity of AOP-4 against HeLa cells was higher than those of other AOPs and the inhibition rate reached 83.96% at the concentration of 10 mg/mL. These results indicated that the AOP might be useful for developing natural safe antitumor drugs or health food.  相似文献

Sulfated polysaccharides (F1, F2) from seaweed Corallina officinalis were isolated through anion-exchange column chromatography. Their chemical characteristics were determined by GC, HPLC, FT-IR and UV spectra. F1 and F2 contained only two monosaccharides, namely galactose and xylose. The antioxidant activities of F1, F2 and the de-sulfated polysaccharides (DF-1, DF-2) in vitro were investigated, including hydroxyl radicals scavenging effect, superoxide radical scavenging capacity, DPPH radical activity and reducing power. As expected, antioxidant assay showed that the two sulfated polysaccharide fractions (F1, F2) possessed considerable antioxidant properties and had more excellent abilities than de-sulfated polysaccharides (DF-1, DF-2).  相似文献

Tricholoma matsutake, a high-class edible mushroom in China, has been regarded as famous foods and biopharmaceutical materials with a great deal of interest. In the previous investigations, researchers believe the water-soluble polysaccharide β-glucan is the major active component of T. matsutake, which displays various biological activities. In the present study, two novel alkali-extracted polysaccharide fractions, TM-APS-1 and TM-APS-2, were isolated from the fruit bodies of T. matsutake by DEAE-cellulose and Sepharose CL-6B columns on ÄTKA explorer chromatography system. Their chemical and physical characteristics and radical scavenging capacity were valuated, including chemical methods, GC, HPLC, scavenging activity against DPPH radicals, superoxide radicals, hydroxyl radicals, and chelating ability. The results showed that TM-APS-1 and TM-APS-2 exhibited significantly antioxidant activity at a concentration-dependent manner. The alkali-extracted polysaccharide fractions from T. matsutake can be developed to be novel functional food or pharmaceutical products with antioxidant effects.  相似文献

The differentiation-inducing factor-1 (DIF-1) is a lipophilic signal molecule (chlorinated alkylphenone) that induces stalk cell differentiation in the cellular slime mold Dictyostelium discoideum. In addition, DIF-1 and its derivatives have been shown to possess anti-leukemic activity and glucose consumption-promoting activity in vitro in mammalian cells. In this study, to assess the chemical structure-effect relationship of DIF-1, we synthesized eight derivatives of DIF-1 and investigated their stalk cell-inducing activity in Dictyostelium cells and pharmacological activities in mammalian cells. Of the derivatives, two amide derivatives of DIF-1, whose hydrophobic indexes are close to that of DIF-1, induced stalk cell differentiation as strongly as DIF-1 in Dictyostelium cells. It was also found that some derivatives suppressed cell growth in human K562 leukemia cells and promoted glucose consumption in mouse 3T3-L1 cells. These results give us valuable information as to the chemical structure-effect relationship of DIF-1.  相似文献

Two kinds of xyloglucan derivatives (xyloglucan selenious ester and sulfated xyloglucan) were prepared and evaluated on antioxidant activity and antitumor activity. Compared with xyloglucan, xyloglucan derivatives have new bioactivity against oxidative damage and tumor. Furthermore, xyloglucan selenious ester is more potent than sulfated xyloglucan at antioxidant activity and antitumor activity in vitro. The current data suggest for the first time that selenition of xyloglucan significantly increases its bioactivity and the chemical modification of polysaccharide may allow the preparation of derivatives with new properties and a variety of applications.  相似文献

High-efficiency ultrasonic treatment was used to extract the polysaccharides of Psidium guajava (PPG) and Psidium littorale (PPL). The aims of this study were to compare polysaccharide activities from these two guavas, as well as to investigate the relationship between ultrasonic conditions and anti-glycated activity. A mathematical model of anti-glycated activity was constructed with the artificial neural network (ANN) toolbox of MATLAB software. Response surface plots showed the correlation between ultrasonic conditions and bioactivity. The optimal ultrasonic conditions of PPL for the highest anti-glycated activity were predicted to be 256 W, 60 °C, and 12 min, and the predicted activity was 42.2%. The predicted highest anti-glycated activity of PPG was 27.2% under its optimal predicted ultrasonic condition. The experimental result showed that PPG and PPL possessed anti-glycated and antioxidant activities, and those of PPL were greater. The experimental data also indicated that ANN had good prediction and optimization capability.  相似文献

Three sulfated polysaccharide derivatives (phosphorylated and aminated fucoidan) were synthesized, and their potential antioxidant activities were investigated employing various established in vitro systems. Two methods were used in phosphorylation fucoidan: polyphosphoric acid and POCl3 method. Aminated fucoidan was prepared using the epichlorohydrin and ammonia water. All fucoidan derivatives possessed considerable antioxidant activity, and exhibited stronger antioxidant ability than fucoidan in certain tests. The phosphorylated fucoidan showed stronger hydroxyl radical and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and reducing power. The mechanism on influence the antioxidant activity of samples of phosphate and amino group was indicated.  相似文献

The purpose of the present study was to evaluate the in vivo efficacy of two cinnamic acid synthetic derivatives (allyl 3-[4-hydroxyphenyl]propanoate; HPP304, 1-naphthyl-methyl 3-[4-hydroxyphenyl]propanoate; HPP305) in high-cholesterol fed rats and compare their actions to that of cinnamic acid. Cinnamic acid and its synthetic derivatives were supplemented with a high-cholesterol diet for 42 days at a dose of 0.135 mmol/100 g of diet. The supplementation of HPP304 and HPP305 significantly lowered cholesterol and triglyceride levels in the plasma and liver with a simultaneous increase in the HDL-cholesterol concentration, whereas cinnamic acid only lowered the plasma cholesterol concentration. Cinnamic acid lowered hepatic HMG-CoA reductase activity in high-cholesterol fed rats, however, its synthetic derivatives (HPP304 and HPP305) did not affect HMG-CoA reductase activity compared to the control group. Instead, the HPP304 and HPP305 supplements significantly lowered hepatic acyl coenzyme A:cholesterol acyltransferase activity and increased the fecal bile acid. The SOD activity of the erythrocytes and liver was not different between the groups, however, the activities of CAT and GSH-Px, and the level of GSH in the erythrocytes were significantly higher in the HPP304 and HPP305 groups than in the control group. On the other hand, the activities of CAT and GSH-Px, and the level of malondialdehyde in the liver were significantly lower in the HPP304 and HPP305 groups. The antioxidant activities of these cinnamic acid synthetic derivatives were similar to the cinnamic acid in the high-cholesterol fed rats. In addition, HPP304 and HPP305 lowered amniotransferase activity in the plasma. These results suggest that two cinnamic acid synthetic derivatives (HPP304 and HPP305) exert lipid-lowering action and antioxidant properties without hepatotoxicity in high-cholesterol fed rats.  相似文献

以海洋红酵母为材料, 通过化学抽提法得到多糖, 用经典的Sevag 法进行脱蛋白处理, 经多级沉淀得到纯糖并采用硫酸-蒽酮法测得其中葡萄糖含量; 考马斯亮蓝法分析蛋白质含量。以定量海洋红酵母多糖人工注射日本蟳, 注射等量生理盐水为对照, 定时测定其血清中部分免疫活性因子的活性; 实验表明: 提取多糖为蛋白多糖, 其中葡萄糖含量为3.6%, 蛋白质含量1.9%, 含有多种氨基酸, 其中天冬氨酸含量最多; 注射后12 h 日本蟳血清中总超氧化物歧化酶(T-SOD) 活力达到最高, 酸性磷酸酶(ACP) 活力在注射后24 h 达到最高, 碱性磷酸酶(AKP) 48 h 达到最高, 过氧化氢酶(CAT) 48 h 达到最高, 溶菌酶(LZM) 12 h 即达到最高, 最高点分别高于对照组24%、43%、25%、35%、95%; 72 h 后都恢复至对照组水平。结论: 海洋红酵母多糖注射48 h 内日本蟳体内免疫活性因子均有不同程度的提高, 对日本蟳有较强免疫刺激作用。  相似文献

Four polysaccharides were extracted from Salvia miltiorrhiza Bunge using hot water, ultrasonic, alkali, and enzyme methods. Preliminary structural characterization was conducted by physicochemical property, Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM) analyses. Antioxidant activities against 2,2-diphenyl-1-picryl-hydrazyl (DPPH), hydroxyl, and superoxide radicals were also evaluated. The physicochemical property analysis indicated the identicalness of the polysaccharide indices obtained by the hot water and ultrasonic methods. The indices obtained by the alkali and enzyme methods were significantly different. The FTIR spectra revealed the general characteristic absorption peaks of the four polysaccharides. The SEM images demonstrated significant differences in the surface features of the different polysaccharides. The antioxidant activity assay revealed the significant antioxidant activities of three polysaccharides. Overall, the polysaccharides from S. miltiorrhiza Bunge may have potential applications in the medical and food industries.  相似文献

Sulfated derivatives of polysaccharide from Momordica charantia L. (MCPS) with different degree of sulfation (DS) were synthesized by chlorosulfonic acid method with ionic liquids as solvent. Fourier transform infrared spectra and 13C nuclear magnetic resonance spectra indicated that C‐6 substitution was predominant in MCPS compared with the C‐2 position. Compared with the native polysaccharide from Momordica charantia L. (MCP), MCPS exhibited more excellent antioxidant activities in vitro, which indicated that sulfated modification could enhance antioxidant activities of MCP. The aim of this study was to investigate the inhibitory effect of Sargassum polysaccharide on oxidative stress induced by infectious bursa disease virus (IBDV) in chicken bursal lymphocytes. The levels of IL-1β, IL-8, IL-10, TNF-α, MCP-1, reduced glutathione and reactive oxygen species in chicken bursal lymphocytes treated with IBDV or both IBDV and Sargassum polysaccharide were measured, and the activities of superoxide dimutase and glutathione peroxidase were evaluated. Our results showed that oxidative stress appeared when chicken bursal lymphocytes were incubated with IBDV for 8 h at 100 TCID50. Sargassum polysaccharide inhibited oxidative stress by increasing the amount of reduced glutathione, promoting the activities of superoxide dimutase and glutathione peroxidase and reducing the level of reactive oxygen species. The polysaccharide also raised IL-1β, IL-8, IL-10 and TNF-α levels in cells infected with IBDV. These findings suggest that Sargassum polysaccharide acts against infection by elevating antioxidant capacity and cytokine levels in chicken bursal lymphocytes.  相似文献

This study analyzed sulfated polysaccharides, such as fucans, from the brown alga Lobophora variegata to verify their antioxidant activity in vitro, antitumoral effect on human colon adenocarcinoma cell line HT-29, and anti-inflammatory activity. Sulfated polysaccharide fractions containing fucans were obtained after fractionation with increasing volumes (v) of acetone (0.3–2.0 v). The polysaccharide was eluted with 1.5 v of acetone and named F1.5. The results showed that F1.5 contained a high yield. Chemical and structure analyses were performed by infrared spectroscopy, electrophoresis in agarose gel, and chemical dosages (sugar, protein, phenolic compounds, and sulfate). We observed that this sulfated polysaccharide had antioxidant activity and antitumoral effect. Anti-inflammatory activity in vivo of F1.5 was observed in the croton oil mouse-ear model at 75 mg kg-1. The results were correlated with histopathological analysis.  相似文献

The chemical carboxymethylated polysaccharide (CMAAP), which is derived from the poorly water-soluble, crude polysaccharide from Auricularia auricula (AAP), was prepared. The degree of substitution of CMAAP was 0.857, and its solubility in water reached 0.6 mg/mL, much higher than that of AAP (0.1 mg/mL). One fraction, CMAAP22, was purified. Chemical analysis indicated that CMAAP22 was composed of mannose and glucose in a ratio of 1.06:1 with a molecular weight of 3.4 × 106 Da. IR spectra and 13C NMR spectra indicated that the C2, C4 and C6 positions were partially substituted by -CH2COOH. AFM revealed that CMAAP22 was spherical particle structure with a diameter of 80-300 nm. Moreover, the antioxidant activity of CMAAP22 was improved, nearly twice as much as that of the AAP, especially in the scavenging of DPPH and ABTS+.  相似文献

A water-soluble crude polysaccharide, obtained from fern Pteridium aquilinum, was fractionated by DEAE-Sepharose Fast-Flow column chromatography, and purified by Sephacryl S-400 HR column chromatography. The average molecular weight (Mw) of the purified polysaccharide (PLP) is 458,000 Da. The monosaccharide components of PLP were characterized by gas chromatography (GC), and the majority of the monosaccharide components was glucose (relative mass 58.1%) with low levels of galactose, mannose, rhamnose, and arabinose (relative mass 18.7%, 6.8%, 10.2%, and 6.1%, respectively). The Fourier-transform infrared spectra (FTIR) of PLP revealed typical characteristics of polysaccharides. On the basis of the ferric-reducing antioxidant power assay (FRAP), DPPH radical-scavenging, the superoxide radical assay, and self-oxidation of 1,2,3-phentriol assay, the antioxidant activities of PLP were investigated. The purified polysaccharide was demonstrated to have strong reductive power (FRAP value: 827.6 μmol/L), moderate scavenging activities against DPPH radicals (83.1%) and superoxide radicals (60.5%), and moderate inhibiting power for self-oxidation of 1,2,3-phentriol (52.4%).  相似文献
