Somatic embryos from immature cotyledons in peanut (Arachis hypogaea) were initiated on media supplemented with 2,4-dichlorophenoxyacetic acid (2,4-d). Over 90% primary embryogenesis and 41–46% repetitive embryogenesis were obtained 12 weeks after initiation by maintaining embryogenic cultures on medium containing 20 mg 1-1 2,4-d. Maintenance of cultures on medium with 30 or 40 mg I-1 2,4-d resulted in lower primary and secondary embryogenesis, and proliferation of nonembryogenic callus. Transfer of embryogenic cultures to a secondary medium with 10 or 20 mg I-1 2,4-d significantly enhanced secondary embryogenesis compared to basal medium without the growth regulator. The use of Murashige & Skoog versus Finer's media had no significant effect on embryogenesis (85–95%), repetitive embryogenesis (11–37%) or mean embryo number. Summary Embryogenic masses were obtained from immature leaves of peanut (Arachis hypogaea L.) cultured on a medium containing 20 mg/l 2,4-D. Somatic embryos developed from these masses following transfer to a medium containing 3 mg/l 2,4-D. The embryo morphology was quite variable. Following transfer to hormone-free medium, these embryos germinated. Shoot elongation was obtained in 25% of the embryos following transfer to a medium supplemented with 0.5 mg/l each of BAP and Kn. Summary Immature zygotic embryos from open-pollinated and selfed Carica papaya L. fruits, 90 to 114 days post-anthesis, produced 2 to 20 somatic embryos on apical domes, cotyledonary nodes, and radicle meristems after culture for three weeks on half-strength Murashige and Skoog (MS) medium supplemented with 0.1 to 25 mg l–1 2,4-dichlorophenoxyacetic acid (2,4-D), 400 mg l–1 glutamine, and 6% sucrose. After six weeks of culture, about 40 to 50% of the zygotic embryos had become embryogenic, and each embryogenic embryo yielded hundreds of somatic embryos within five months of culture on media supplemented with 2,4-D. Somatic embryos matured on half-strength MS medium, germinated on MS medium containing 5 mg l–1 kinetin, and grew large enough for greenhouse culture on MS medium. Statistical analyses of the data revealed very significant differences in androgenesis induction ofA. carnea Hayne anther culture depending on the bud length, nutrient medium composition and age of the parental tree. Significant mutual influence of all these factors was also observed. The highest number of androgenic anthers was obtained when 4 mm long buds were used. Older trees (60 and 100 yrs) gave a higher number of androgenic anthers than the younger ones (20 and 40 yrs). MS medium supplemented with 2,4-d and Kin (1 mg l–1, each) was the most favourable for androgenesis induction. Pollen embryos (haploids and aneuploids) were formed by the division of uninuclear microspores.The highest percentage of germinated embryos and further synchronous development of the shoot and root was achieved in MS medium supplemented with IAA, GA3 (1 mg l–1) and activated charcoal (1%). Somatic embryos were induced from immature cotyledons and immature embryonal axis ofArachis hypogaea L. on L-6 basal medium supplemented with NAA, picloram or 2,4-D at 5–50 mg 1-1. Immature embryonal axis produced a higher number of somatic embryos in comparison with immature cotyledons. The highest number of responding cultures was produced on medium supplemented with NAA (50 mg 1-1), while the highest average number of somatic embryos per culture was produced on medium with 2,4-D (10 or 20 mg 1-1) and picloram (30 mg 1-1) from cotyledons. The somatic embryos developed into plants on basal medium supplemented with activated charcoal and about 100 plants were successfully transferred to the field. Summary Goldenseal (Hydrastis canadensis L.), a popular native American medicinal plant, is currently listed as endangered or threatened in over one-third of the states in which it is listed. The objective of this study was to develop an in vitro culture protocol for Goldenseal. Excise embryos were grown on Gamborg's B-5 medium with 0,1 or 10 μM gibberellic acid (GA3), and supplemented with 30 gl−1 sucrose and 8 gl−1 agar. Germinated embryos provided explants (leaf and root tissue) that were subsequently cultured on various media with combinations of naphthleneacetic acid (NAA) and benzyladenine (BA). All NAA/BA combinations produced multiple shoots, roots, and callus. Leaf explants cultured on medium with 1∶10 μM NAA:BA and root explants on medium with 1∶1 μM NAA:BA could be successfully used for mieropropagation.  相似文献

Organogenic callus cultures of seashore mallow, Kosteletzkya virginica (L.) Presl., originated from excised mature embryos or stem sections of aseptically germinated plants initially cultured on Murashige & Skoog minimal organics medium containing 30000 mg l-1 glucose, 2.0 mg l-1 indoleacetic acid and 1.0 mg l-1 kinetin. Plants were regenerated via shoots and roots from callus cultures following transfer through a series of media with different cytokinin/auxin ratios and changes in carbohydrate source. Meristematic regions, shoot and root primordia were observed during histological examination of the tissues. Somatic embryos were not found.  相似文献

Callus was induced from seeds of Puccinellia distans (L.) Parl. on MS medium supplemented with 2 mgl-1 2,4-dichlorophenoxyacetic acid and 0.5 mgl-1 kinetin. Morphogenesis initiation was achieved during subculture on medium containing 0.1 mgl-1 2,4-D. From the point of morphogenetic capacity, 3 types of callus were selected. High frequency of plant regeneration was obtained by selection of embryogenic type of callus, and culture on N6 medium and N6 medium supplemented with kinetin (5–10 mgl-1), or kinetin (2 mgl-1) and IAA (0.5 mgl-1). Callus cultures were obtained from immature and mature embryos of Hordeum marnium on MS media containing 0.5 mg l-1 parachlorophenoxyacetic acid or 2 mg l-1 2,4-dichlorophenoxyacetic acid. Regeneration occurred after transferring calli to MS either devoid of hormones or supplemented with 1 mg l-1 indole-3-acetic acid and 1 mg l-1 zeatin. The regeneration capacity of the immature embryo derived calli (94%) was about 5 times higher than that of mature embryo derived calli (17%). A total of 30 and 964 plantlets were obtained from 21 mature and 59 immature embryo derived calli, respectively. Low frequency (less than 1%) of albino plantlets was obtained from both explants after 3–9 months in culture. Plants expressing transient chlorophyll deficiency were produced from immature embryo derived cultures at a frequency of 10%. Petioles from in vitro grown plants of interspecific grapevine hybrids cvs `Bianca', `Podarok Magaracha' and `Intervitis Magaracha' were cultured on solid NN medium supplemented with 2,4-D and BA at various concentrations. The callus developed was cultured in liquid NN medium supplemented with 0.5 mg l–1 BA to induce formation of somatic embryos. Somatic embryos of globular and heart-stage developed in suspensions of `Podarok Magaracha' and `Intervitis Magaracha'. In contrast, `Bianca' did not undergo embryogenesis beyond globular stage. This made it necessary to perform subculture of the suspensions to HTE liquid medium supplemented with 0.2 mg l–1 BA for the development of globular embryos into heart stage. Heart-stage embryos developed into torpedo-stage after subculturing suspensions of all three cultivars to liquid HTE medium supplemented with 0.1 mg l–1 IAA and 30 mg l–1 sodium hummate. Torpedo-stage embryo suspensions were subcultured in liquid HTE medium supplemented with 0.5 mg l–1 BA, 0.5 mg l–1 GA3 and 0.5 mg l–1 GA3 + 0.2 mg l–1 BA. After 12 days of incubation, plantlets were cultured on solid M2MS medium: without growth regulators and with 0.5 mg l–1 BA. Plantlets that developed in liquid HTE media with 0.5 mg l–1 GA3 or 0.5 mg l–1 GA3 + 0.2 mg l–1 BA produced 82–90% shoots on solid M2MS medium with 0.5 mg l–1 BA after 50 days of culture.  相似文献

Summary Plant regeneration via somatic embryogenesis was obtained in peanut (Arachis hypogaea L.) from axes of mature zygotic embryos. The area of greatest embryogenic activity was a 2-mm region adjacent to and encircling the epicotyl. Somatic embryogenesis was evaluated on Murashige and Skoog media supplemented with a variety of auxin treatments. Maximum production occurred on medium supplemented with 3 mg · liter−1 4-amino-3,5,6-trichloropicolinic acid. Explant cultures were transferred to half-strength medium supplemented with 1 mg · liter−1 gibberellic acid for somatic embryo germination and early plantlet growth. Plantlets, transferred to soil, were placed in a greenhouse and grown to maturity.  相似文献

A protoplast-to-plant regeneration system has been established for sweet potato (Ipomoea batatas (L.) Lam.) and its wild relative, I. lacunosa L. Viable protoplasts, isolated from preplasmolyzed stems and petioles of in vitro-grown plants, were cultured on liquid MS (Murashige & Skoog 1962) medium that supported cell division and colony formation. Embryogenic calli of sweet potato were induced on agar-solidified MS medium supplemented with 3% (w/v) sucrose, 50 mg l-1 casamino acids, 0.2–0.5 mg l-1 2,4-d, 1.0 mg l-1 kinetin and 1.0 mg l-1 ABA. On average, 3 plants were regenerated from a single sweet potato callus subcultured on semi-solid MS medium containing 3% (w/v) sucrose, 800 mg l-1 glutamine, 2.0 mg l-1 BA or 1.0 mg l-1 kinetin and 1.0 mg l-1 GA3. Embryogenic calli of I. lacunosa L. were initiated on semi-solid MS medium containing 0.2–0.5 mg l-1 IAA and 1.0–2.0 mg l-1 BA. Protoplasts isolated from embryogenic callus of Citropsis schweinfurthii (Engl.) Swing. & M. Kell were cultured in MT (Murashige and Tucker 1969) basal medium containing 5% sucrose supplemented with 0.0, 0.001, 0.01, 0.1 or 1.0 mg l–1 BA, 0, 300, 600 or 900 mg l–1 malt extract and 0.6 M sorbitol. The highest plating efficiency was obtained on MT basal medium containing 5% sucrose supplemented with 0.01 mg l–1 BA and 600 mg l–1 malt extract. MT basal medium containing 5% sucrose and supplemented with 0.01 mg l–1 kinetin was found to be a medium suitable for the development of globular somatic embryos derived from protoplasts into heart-shaped somatic embryos with cotyledon-like structures. The highest percentage of shoot formation was obtained using 0.1 mg l–1 GA3. Summary Haploid protoplasts isolated from prothallus (i.e. female gametophyte) of Ginkgo biloba, at densities ranging from 5×104 to 105 protoplasts per milliliter, were able to divide and form microclones which directly evolved into embryos, when they were cultured in two different liquid media. These were: the Murashige and Tucker medium (1969) modified by omitting ammonium ions and supplementing with glutamine, benzyladenine and various levels of naphthaleneacetic acid; or the Bourgin and Nitsch medium (1967) without growth regulators, supplemented with coconut milk. Three months later, the number of embryos ranged from 165 to 1900 embryos ml–1 depending on the culture medium. Plant regeneration via direct somatic embryogenesis from cotyledons, hypocotyls and leaves in seabuckthorn (Hippophae rhamnoides L.) was achieved. The influences of basal media, carbon sources, plant growth regulators (PGRs) with different concentrations and combinations on embryogenesis capacity of explants were studied. The highest frequency of somatic embryos production and germination was obtained on Schenk and Hildebrandt medium (SH) supplemented with 1.0 mg dm−3 kinetin and 0.2, 0.5 mg dm−3 indole-3-acetic acid. Granulated sugar was the optimal carbon source. The embryo-derived plantlets with well-developed roots and shoots were transferred successfully to the greenhouse with a maximum survival rate of 55 %. Histological observation revealed that the somatic embryos were similar to those of zygotic embryos.  相似文献

A system for somatic embryogenesis and plant regeneration of spinach from hypocotyl segments has been established. Callus was induced on solid media supplemented with 8.5–15.0 mg.l−1 of indole-3-acetic acid and 3.46–34.64 mg.l−1 gibberellic acid. Callus was then subcultured on different media (solid or liquid) with or without IAA, or continuously maintained on the initiating media. Somatic embryos were obtained in subcultures on IAA-containing media as well as in long-term cultures on initiating media. The best results were achieved in liquid subcultures. About 60% of plantlets survived after transplanting in pots.  相似文献

A highly reproducible system for efficient somatic embryogenesis was developed to regenerate plantlets from cotton (Gossypium hirsutum L.) cultivars (Nazilli M-503 and Nazilli 143). Shoot apices, hypocotyls and nodes of 10-d-old seedlings were used as explants. High frequency (100 %) embryogenic calli was initiated from all tested explants on Murashige and Skoog (1962) (MS) media supplemented with 1 g dm–3 polyvinylpyrrolidone (PVP), 1 mg dm–3 6-benzylaminopurine (BAP), 0.5 mg dm–3 kinetin for Nazilli M-503 and 1 g dm–3 PVP, 2 mg dm–3 BAP, 0.5 mg dm–3 kinetin for Nazilli-143. Globular stage somatic embryos were produced 4 months after transfer to hormone-free MS medium supplemented with 1 g dm–3 PVP. Subsequent subculture of globular embryos every 3 weeks on hormone-free MS medium led to the development of torpedo and cotyledonary stage embryos with the frequency of 75 and 83.2 % from hypocotyl explants of Nazilli M-503 and Nazilli-143, respectively. Afterwards, mature somatic embryos were isolated and cultured on hormone-free MS medium for germination and development into plantlets. The highest germination frequency (42.9 %) for Nazilli M-503 somatic embryos were obtained on hormone-free MS medium after 5 months with hypocotyl explants, whereas germination frequencies of Nazilli-143 embryos from hypocotyl, node and apex explants varied between 22 – 30 %.  相似文献

Calli from young embryos of Cocos nucifera L. were induced on B5 medium supplemented with IAA-conjugates (IAA-asp or IAA-ala) at a concentration of 2.0 mg/1 and callusing was increased by about 10% if both IAA-conjugates, IAA-asp and IAA-ala were added together. Differentiation of shoots and roots was achieved by transferring calli to B5 medium supplemented with either IAA-asp (2.0 mg/1)+Kn(2.0 mg/1) or NAA (2.0 mg/1). Summary A somatic embryogenesis protocol was developed for Eschscholzia californica Chan. (California poppy) using embryogenic cell suspensions and optimized media conditions. Rapidly-growing, finely-dispersed embryogenic cell suspension cultures were established from embryogenic callus and maintained in B5 liquid media supplemented with 0.5 mg 1−1 (2.26 μM) 2,4-dichlorophenoxyacetic acid. Culture conditions were optimized by investigating the effect of basal media composition, gyratory shaker speed, various carbon sources, different cytokinins, and AgNO3 on the efficiency of somatic embryogenesis. After 40 d in culture, the somatic embryos that formed were counted and their overall growth expressed as pecked cell volume. The selected media consisted of either Gamborg (B5) or Murashige and Skoog (MS) salts and vitamins supplemented with 40 g 1−1 (117 mM) sucrose, 0.05 mg 1−1 (0.22 μM) 6-benzylaminopurine, and 10 mg l−1 (58.8 μM) AgNO3. Somatic embryo production was substantially reduced at shaker speeds above 40 rpm. Glucose and snerose were the most effective carbon sources, whereas fructose, galactose, and maltose resulted in a reduced yield and growth of somatic embryos. The development of somatic embryos was promoted by AgNO3 at concentrations below 10 mg l−1 (58.8 μM). A semi-solid medium containing 1.5 g l−1 Gel-rite produced the highest frequency of somatic embryo conversion, and promoted the efficient growth of plantlets. Using the reported protocol, over 500 viable somatic embryos were produced per 25 ml of embryogenic cell suspension culture.  相似文献

Plants were obtained via somatic embryogenesis in callus derived from in vitro raised leaf and petiole explants of Aconitum heterophyllum Wall. Callus was induced on a Murashige-Skoog medium supplemented with either 2,4-dichlorophenoxy acetic acid (2,4-d 1 mg l-1) and kinetin (KN 0.5 mg l-1) with coconut water (CW 10% v/v) or naphthalene acetic acid (NAA 5 mg l-1) and benzylaminopurine (BAP 1 mg l-1). Somatic embryos appeared after 2–3 months or 2 subculture passages when 2,4-d or NAA induced source of the callus was transferred to a MS medium containing BAP (1 mg l-1) and NAA (0.1 mg l-1). For successful plantlet formation, the somatic embryos were transferred to a medium containing 1/4 strength MS nutrient with indole-3-butyric acid (IBA 1 mg l-1). Alternatively, the somatic embryos were dipped in a concentrated solution of IBA for 5 min and placed on a hormone free medium.
