盐胁迫对盐生植物黄花补血草种子萌发和幼苗生长的影响   总被引:2,自引:0,他引:2

盐生植物黄花补血草广泛分布于我国西北地区、东北西部以及华北北部,对改良盐碱土壤具有重要的生态作用。以黄花补血草(Limonium aureum(L.)Hill)为材料,研究分析了不同浓度NaCl胁迫对其种子萌发和幼苗生长产生的抑制效应及作用机制。结果表明:低浓度NaCl(25 mmol/L和50 mmol/L)处理不影响黄花补血草种子萌发和幼苗生长,25 mmol/L NaCl甚至促进了根生长,而高浓度NaCl(100 mmol/L和150 mmol/L)处理明显抑制种子萌发及幼苗生长。利用荧光探针的检测结果表明,NaCl处理的幼苗根中过氧化氢(H2O2)和一氧化氮(NO)含量明显高于对照水平。碘化丙啶(PI)染色结合激光共聚焦显微镜观察及检测相对电导率结果显示,高浓度NaCl处理抑制了幼苗根尖伸长区细胞的伸长生长,增加了细胞膜的通透性,对根细胞造成了明显的伤害。此外,高浓度NaCl处理诱导叶片丙二醛(MDA)含量显著升高。以上结果说明,黄花补血草对低浓度的盐具有一定的耐盐性,但高浓度盐降低了种子的萌发率,使幼苗根中H2O2产生增加,抑制根尖伸长区细胞的伸长生长,对根、叶造成明显氧化损伤,从而抑制黄花补血草幼苗的生长。  相似文献

本文以二色补血草(Limonium bicolor)为实验材料,用Hoagland营养液和200mmol·L—NaCl、NaBr、NaNO3溶液分别处理12h,测定二色补血草盐腺的Na+分泌速率、叶片Na+含量和MDA(丙二醛)含量以及质膜透性,并利用非损伤微测技术探索可能与盐腺相关的转运蛋白,以探讨不同阴离子对二色补血草盐腺分泌Na+的作用及其可能原因。结果表明:在NaCl处理12h时二色补血草叶片Na+分泌速率达到最大,然后逐渐下降;不同钠盐处理下叶片Na+分泌速率为NaCl〉NaBr=NaNO3〉Hoagland,而叶片Na+含量NaBr〉NaCl〉NaNO3〉Hoagland;不同盐处理下叶片质膜透性和MDA含量无显著性差异;利用Na—K—C1共转运体专一性抑制剂bumetanide处理发现Na+分泌速率显著降低。这些结果表明Na—K—Cl共转运体可能参与盐腺分泌Na+。  相似文献

用不同浓度的NaCl处理盐生植物黄花补血草和大叶补血草愈伤组织,研究其中H2O2、MDA及渗透性调节物质等生理指标含量的变化,从细胞水平分析比较2种补血草愈伤组织对盐环境的适应机制.结果表明:随NaCl胁迫浓度的升高,2种补血草愈伤组织的H2O2和MDA含量逐渐显著增加;愈伤组织中脯氨酸含量增加幅度在75和150mmol/LNaCl下明显表现为黄花补血草大于大叶补血草,而其中的可溶性糖含量增加程度于相同NaCl浓度下明显呈现出黄花补血草小于大叶补血草的趋势;可溶性蛋白含量在黄花补血草中随NaCl浓度升高而逐渐增加,但在大叶补血草中却表现为低浓度(75mmol/L)比对照升高,而高浓度(150和300mmol/L)比对照明显减少的趋势.研究发现,2种补血草愈伤组织中脯氨酸、可溶性糖和可溶性蛋白这些有机渗透性调节物质对盐胁迫的反应特性存在差异,并与其耐盐性有关.  相似文献

耐盐植物的选育是盐渍土地生物改良的关键。通过对13个NaCl盐度梯度下,真盐生植物囊果碱蓬、盐地碱蓬、高碱蓬、盐角草、盐爪爪,泌盐植物大叶补血草、耳叶补血草、黄花补血草8种盐生植物的萌发试验,测定其种子发芽率、发芽势、发芽指数和相对盐害率等指标,研究不同NaCl盐度胁迫对其萌发特性的影响。结果表明:低盐度(50 mmol/L)能促进8种盐生植物的萌发;随着盐度的上升,盐角草、盐地碱蓬、囊果碱蓬、高碱蓬表现出较强的萌发耐盐性,发芽势和发芽指数优于其他植物;盐角草的萌发耐盐性最佳,在1000 mmol/L盐度下萌发率仍能达到54.0%。进一步通过S型生长曲线和线性模型分析得出,8种植物为对抗逆境大致分为"快速型"和"缓慢型"两种萌发策略;随盐度的升高,初始萌发时间和萌发高峰时间均不同程度的向后推迟。盐度≤200 mmol/L时,囊果碱蓬的萌发占据优势,其萌发速率大于其他植物,且差异显著;中盐度400 mmol/L左右时,盐地碱蓬和盐角草萌发最快,二者无显著差异;盐度≥600 mmol/L时,盐角草萌发速率较快,相比其他植物差异显著。泌盐植物的萌发耐盐适宜浓度和耐盐极限浓度均低于真盐生植物。8种植物均有作为氯化物为主盐渍土地区生物改良材料的潜力。  相似文献

利用叶表皮离析、扫描电镜以及常规石蜡切片法对烟台补血草[ Limonium franchetii( Debx.) Kuntze]叶片盐腺的分布和结构特征及其发育过程进行了观察.结果表明:烟台补血草叶片上、下表皮均有盐腺分布,上、下表皮的盐腺密度分别为7.57和8.09 mm-2,同一时期叶片上表皮盐腺密度略小于下表皮.烟台补血草的成熟盐腺是由20个细胞构成的复合结构,其中,中央有4个分泌细胞,每个分泌细胞外侧均伴有1个长方形的毗邻细胞,再向外依次包围着4个内杯状细胞和4个外杯状细胞,靠近叶肉细胞处还有4个收集细胞,收集细胞与盐腺的其他细胞同源;在4个分泌细胞顶端的角质层中央各有1个小孔,称为泌盐孔.烟台补血草盐腺发育过程依次经历原表皮细胞期、2细胞期、4细胞期、8细胞期、16细胞期和20细胞期6个阶段,由1个单独的原表皮细胞发育而成,发育过程中细胞均进行垂周分裂;另外,同一时期的叶片存在盐腺发育不同步的现象.根据观察结果推测烟台补血草主要通过泌盐孔L向外泌盐.  相似文献

通过水培试验,研究了100 mmol/L NaCl盐浓度下,0.25 mmol/L Spd浸种处理对两个番茄品种白果强丰(耐盐基因型)和江蔬14号(盐敏感基因型)植株干重、根冠比(R/T)、幼苗叶片和根系抗氧化酶活性及活性氧含量的影响.具体试验处理如下:(a) 对照(蒸馏水浸种+ 0 mmol/L NaCl),(b) NaCl (蒸馏水浸种+ 100 mmol/L NaCl),(c) Spd(0.25 mmol/L Spd浸种 +100 mmol/L NaCl).结果表明,在盐胁迫下,两个番茄品种幼苗叶片和根系内超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性升高,H2O2含量和O·-2产生速率增高,幼苗生长受到抑制,幼苗地上部、地下部干重均明显低于对照,R/T增大,且江蔬14号的变化幅度大于白果强丰.Spd浸种处理降低了盐胁迫下番茄幼苗叶片和根系内O·-2产生速率和H2O2含量,进一步提高了SOD、POD和CAT活性,促进幼苗干重增加,缓解了盐胁迫对植株的伤害.与耐盐基因型番茄品种白果强丰相比,Spd浸种处理对盐敏感基因型番茄品种江蔬14号的作用效果更为明显.总之,Spd浸种处理通过提高盐胁迫下植株体内抗氧化酶活性,降低ROS水平来缓解盐胁迫对番茄幼苗的伤害,提高幼苗耐盐能力.  相似文献

以黄河三角洲贝壳堤岛贝壳沙为基质,及该岛上生长的优势草本植物二色补血草幼苗为材料,用不同浓度的NaCl溶液(0,50,100,200,300mmol/L)模拟盐胁迫处理30d,探讨二色补血草光合生理生态特性对盐胁迫的响应特征。结果表明:二色补血草对盐胁迫和光合有效辐射(PAR)具有较强的适应能力,在盐浓度为50~100mmol/L条件下,其净光合速率(Pn)、瞬时水分利用效率(WUE)、表观光能利用效率(LUE)、羧化效率(CE)、光饱和点(LSP)、表观量子效率(AQY)均达到最高值,光补偿点(LCP)最低,表明其在此盐分范围内,利用光的能力较强,光照生态幅最宽。当盐分浓度为300mmol/L时,其Pn、WUE、LUE、CE、LSP和AQY显著下降,LCP升高,Pn、Tr、Gs、WUE、LUE、CE对PAR的响应曲线与对照差异较大,即高盐胁迫加重了二色补血草对光的敏感性;WUE、LUE、CE的最优PAR和盐分范围存在一定的差异,低PAR下有利于光能的利用,中PAR下有利于水分的利用,高PAR下有利于CO2的利用;WUE对盐分的适应范围最大,CE其次,LUE最小。  相似文献

濒危植物长叶红砂适应盐胁迫的生理生化机制研究   总被引:2,自引:0,他引:2  
以濒危盐生植物长叶红砂(Reaumuria trigyna)幼苗为材料,研究了不同浓度NaCl溶液(0、100、200、300和400mmol/L)处理30d对其生长和生理生化指标的影响,以分析长叶红砂的耐盐生理机制。结果表明:(1)100和200mmol/L NaCl处理时,长叶红砂鲜重和干重均显著增加,但随着盐浓度继续增加,长叶红砂幼苗生长受到抑制,且地上部受到的抑制大于根部,显示长叶红砂适宜生长的NaCl浓度是200mmol/L。(2)随NaCl胁迫浓度的升高,长叶红砂的净光合速率(Pn)、蒸腾速率(Tr)和气孔导度(Gs)呈下降趋势,胞间CO2浓度(Ci)呈上升趋势,说明光合速率的下降使利用CO2的能力下降,胞间积累了大量的CO2,且盐处理浓度越高量越大。(3)随NaCl胁迫浓度的升高,长叶红砂幼苗Na+、Cl-含量增加,可溶性糖、脯氨酸、游离氨基酸及可溶性蛋白等有机渗透调节物质的合成增加。研究认为,长叶红砂是通过调节叶片Na+、Cl-以及有机渗透调节物质含量来提高其耐盐能力。  相似文献

以冬小麦(Triticum aestivum L.)品种周麦18(Zhoumai18)和豫麦49(Yumai49)为材料,采用盆栽培养,研究100、250、350 mmol/L和450 mmol/L NaCl 浓度胁迫下,小麦幼苗干物质分配、根系特征、叶绿素含量、游离脯氨酸含量和根系活力变化规律.结果表明,随着盐分浓度的增加,两个品种小麦的叶面积、地上干重以及根的长度显著减小;根系干重、根直径、根表面积、根体积、根系活力以及叶绿素含量呈先上升后下降趋势,在250 mmol/L NaCl处理下达最大.叶绿素a/b随NaCl浓度升高而上升.随盐分浓度变化周麦18叶片游离脯氨酸含量高而变化幅度大,450mmol/L处理组的含量高于对照组1.5倍.供试品种冬小麦耐盐阈值为250~350 mmol/L.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Fluid collected from hatching eggs of Haemonchus contortus contained a lipase which hydrolysed 2-naphthyl laurate (about 0·7 μmol naphthol freed /h/106 eggs). The fluid also hydrolysed l-leucinamide (about 2·3 μmol leucine freed/h/106 eggs). The fluid when added to normal or heated eggs caused ‘hatching’. ‘Hatching’ also occurred in exsheathing fluid from infective juveniles and in a preparation of pancreatic lipase containing leucine aminopeptidase. A purified mammalian leucine aminopeptidase in combination with several different lipases did not attack egg shells.The ‘spontaneous’ hatching of eggs of H. contortus was strongly inhibited by 1,10-phenanthroline, 10?3M, and this inhibition was reversed by Zn2+. However, the inhibition of ‘hatching’ of eggs in externally applied hatching fluid, or the hydrolysis of leucinamide in hatching fluid was generally less marked.  相似文献
