采用自行设计式及仿制圆筒式诱捕器,以人工合成性信息素(Z,E)-9,12-tetradecadienyl acetate (简称TDA) 为引诱源在实验室条件下对影响诱捕器诱捕印度谷螟Plodia interpunctella效果的几个因素进行分析测定。多元线性回归分析结果表明:日平均温度在18.5~26.2℃,人工合成性信息素TDA散发日数9~37天,温度(X1)、TDA散发日数(X2)、当日释放蛾(雄)量(X3)、累计2日释放蛾量(X4)、累计3日释放蛾量(X5)等5个因素与每日诱捕蛾量间存在着相关关系。对5个因素进行逐步回归分析和筛选,得出线性回归方程:Y=-27.31+1.37X1+0.28X3,回归系数R(0.90)>R0.01(n-2,0.63)。统计分析结果表明:日平均温度(X1)、当日释放蛾量(X3)与诱蛾量(Y)之间呈显著线性相关关系。卡方测验表明预测值与实测值之间差异不显著。  相似文献

通过蚊虫酯酶蛋白的淀粉凝胶电泳分析和基因组DNA的限制性酶切片段长度多态性(RFLPs)比较, 对尖音库蚊Culex pipiens、三带喙库蚊Culex tritaeniorhynchus和中华按蚊Anopheles sinensis有机磷杀虫药剂敏感种群的酯酶蛋白和结构基因的多态性进行分析。发现在蛋白质水平上,三带喙库蚊敏感种群(n=54)在酯酶α和β位点分别存在2个和3个等位基因,在DNA水平上有2.9%的个体具有与酯酶β11基因1.3 kb Cdna片段同源的1.3 kb单拷贝带存在。发现中华按蚊敏感种群 (n= 50)中具有低活性的非特异性酯酶存在,在蛋白质水平上,酯酶α和β位点各有一个等位基因;在DNA水平上,通过对单个蚊虫基因组DNA的研究未发现有与酯酶β11基因同源的酯酶编码基因的存在。对尖音库蚊北京敏感种群(n= 64)的研究发现,在酯酶α和β位点都存在5个等位基因,在DNA水平上,使用一个限制性内切酶(EcoRI),15只蚊虫的样本在酯酶β位点发现了5个等位基因,说明在尖音库蚊北京敏感种群的酯酶β基因周围存在着较大的中性多态性,在有机磷杀虫剂的选择下,这些中性多态性可能会成为基因扩增的潜在因素。  相似文献

西藏环状病毒(Tibet orbivirus,TIBOV)是环状病毒属中的新成员,首次分离自中国西藏自治区的圆斑按蚊,本研究首次对从三带喙库蚊标本中分离到的西藏环状病毒(HN11121株)进行全基因组序列测定和分子遗传进化分析。结果显示除第2节段以外,三带喙库蚊分离的HN11121病毒株与从圆斑按蚊分离的西藏环状病毒原始分离株(XZ0906)第1~10节段核苷酸和其编码的氨基酸序列长度相同,核苷酸和氨基酸同源性分别为70.3%(S6)~98.7%(S5)和79.8%(S6)~99.8%(S10);而HN11121病毒株与XZ0906病毒株第2节段核苷酸长度分别为2 850bp和2 838bp,分别编码950和946个氨基酸,核苷酸和氨基酸同源性分别为55.8%和47.1%。病毒VP2蛋白氨基酸位点差异结果显示从三带喙库蚊分离的HN11121病毒与从圆斑按蚊分离的XZ0906相比存在390个氨基酸差异位点。基于HN11121病毒株VP1基因和VP3基因核苷酸序列的系统进化分析结果显示HN11121属于环状病毒属西藏环状病毒,但是从三带喙库蚊分离的HN11121病毒与在圆斑按蚊分离的西藏环状病毒原始分离株(XZ0906)处在不同的进化分支。以上结果提示,从三带喙库蚊分离的西藏环状病毒(HN11121)与从圆斑按蚊分离的西藏环状病毒在病毒基因组和病毒基因组分子遗传进化均存在较大差异,鉴于三带喙库蚊是我国乙脑病毒的主要传播媒介,同时该蚊种也是我国南方广泛存在的蚊虫种类,因此加强三带喙库蚊中西藏环状病毒监测对于了解西藏环状病毒的生物多态性具有重要意义。  相似文献

研究了环境因子对角倍蚜Schlechtendalia chinensis (Bell) 秋迁蚜生殖和雌性蚜发育的影响。温、湿度单因子试验表明,秋迁蚜在26℃和80%RH条件下有最大生殖量;温、湿度对秋迁蚜生殖量的影响均符合开口向下的二次抛物线变化趋势,极端温、湿度会导致生殖量的下降。采用三元一次正交组合设计,研究了环境温度(X1)、湿度(X2)和光照强度(X3)三因子不同水平组合对雌性蚜发育的影响,表明温度是影响发育历期的主要因子,其次是光照强度,最后是湿度。因此,适当高温、强光照条件可以加快雌性蚜发育;而适当高湿条件可以降低雌性蚜的发育速率而延长其发育历期。在人工培育角倍蚜生产中,创造有利于秋迁蚜生殖的温、湿度条件可以使秋迁蚜产下较多的越冬侨蚜;在适当降低温度、增加湿度的阴暗条件下贮留雌性蚜可以适当延长其发育,以使角倍蚜与盐肤木在物候上达到最佳吻合。  相似文献

利用莎车县农业气象试验站2008—2013年巴旦姆物候期和同期气象观测资料,分析了巴旦姆物候期的变化特征以及气温、日照时数对物候期的影响.结果表明:巴旦姆花期之前的物候期始日之间均呈正相关,与花期之后的物候期始日的相关性大多较小,花芽膨大早迟与休眠期、生长期天数分别呈极显著的正相关和负相关.以果实成熟期为界,气温对之前、之后物候期间隔日数的影响分别为负相关和正相关,日照时数与物候期间隔日数大多为正相关.果实成熟 叶变色始期间隔日数对平均最高气温以及花序出现 开花末期、叶变色始期 落叶末期间隔日数对日照时数存在明显的响应.当巴旦姆休眠期符合日平均气温-3.0~-7.5 ℃的天数满30 d后,经过17~28 d将进入花芽萌动期.花芽萌动期、开花始期、叶变色始期和落叶末期的始日分别与首个候平均气温≥4 ℃且候平均最高气温≥12 ℃、春季侯平均气温≥14 ℃且侯平均最高气温≥22 ℃、秋季首个侯平均气温≤10 ℃且侯平均最高气温≤18 ℃和冬季首个侯平均气温≤1.9 ℃的候序一一对应.利用偏最小二乘法回归分析,建立巴旦姆盛花期始日预测模型,经过检验模型效果较好.  相似文献

球形芽孢杆菌TS-1丙酮粉对七种蚊虫的毒力测定   总被引:1,自引:0,他引:1  
用乳糖-丙酮沉淀法制备的球形芽孢杆菌Bacillus sphaericus,TS-1菌粉对七种蚊虫幼虫进行了毒力测定,以淡色库蚊,致倦库蚊最为敏感,其LC50分别为8.1μg/L(1.79×102cfu/ml),8.9μg/L(1.96×102cfu/ml),三带喙库蚊敏感性较淡色库蚊差13倍,2龄白纹伊蚊和中华按蚊分别差20和600倍;黄斑伊蚊,二株埃及伊蚊剂量高达105—106cfu/ml相差1,000—10,000倍以上几乎无效.本文结合B.t.I.标准生物测定方法,对毒力测定中检查时间、数据、虫龄、虫数、饲料等问题进行了讨论.  相似文献

为掌握云南省西双版纳州勐海县蚊媒病毒分布状况,用诱蚊灯采集蚊虫标本,用细胞培养法分离病毒,对阳性分离物进行分子生物学鉴定。2012年7月,在勐海县打洛镇采集蚊虫32批共1 468只,其中三带喙库蚊(Culex tritaeniorhynchus)28批1 383只,致倦库蚊(Culex quinquefascitatus)2批66只,按蚊(Anopheles)2批19只。用金黄地鼠肾细胞(Golden hamster kidney cell,简称BHK-21)和白纹伊蚊细胞(Aedes albopictus cell,简称C6/36)进行病毒分离,结果有2批三带喙库蚊标本(BNDL1205和BNDL1227)能引起C6/36细胞产生聚合病变,而不能引起BHK-21细胞病变。用黄病毒属特异性引物对这2株阳性分离物进行RT-PCR,扩增得到800bp左右的条带并进行核苷酸序列测定。对核苷酸序列的系统进化分析表明,这2株病毒与分离自越南的广平病毒(Quang Binh virus,QBV)VN180株亲缘性最近,处在同一进化分支;与VN180株的核苷酸同源性分别为83.4%和82.9%,而与既往国内外分离的18株蚊虫黄病毒的核苷酸差异较大。结果表明BNDL1205和BNDL1227病毒为类似QBV,此为国内首次报道。  相似文献

利用1971—2010年天山北坡经济带棉区11个代表站的气象资料,采用气候倾向率、t 检验和IDW插值方法,分析了棉区3月下旬至4月下旬各旬平均气温和≥12 ℃初日的时空变化对棉花播期的影响.结果表明: 研究期间,天山北坡经济带 3月下旬、4月上、中、下旬平均气温分别以0.8、0.5、0.1和0.5 ℃·(10 a)-1的倾向率呈增加趋势,但是≥12 ℃初日却以-0.5 d·(10 a)-1的倾向率呈提前趋势.各气象要素均在20世纪90年代发生突变,突变年后3月下旬、4月上、中、下旬平均气温依次增加2.5、1.9、1.1和1.5 ℃,分别达到7.2、10.0、13.2和15.6 ℃.3月下旬至4月下旬各旬平均气温的高值区普遍位于天山北坡东部棉花主产区的乌苏市、沙湾县和玛纳斯县站点周围,而低值区基本在东部的乌鲁木齐市附近.≥12 ℃初日的空间分布具有地区差异性,研究期间,初日日序较早的区域由精河县向西扩展到棉花主产区玛纳斯县周围,而日序较晚的区域从乌鲁木齐市以南的大部区域缩小至乌鲁木齐市周围.随着突变年后≥12 ℃初日逐年提前,大部县市棉花适宜播期在4月22—28日,采用地膜播种可使适宜播期提前至4月15—21日.  相似文献

1984年我们对北京地区的乙型脑炎病毒和M14病毒进行了监测,观察到三带喙库蚊季节消长高峰在8月中旬,较发病率较高的1980年和1981年晚3周左右:乙型脑炎病毒检出率(批)为16.1%,最低现场感染率为1:303,检出高峰在8月中旬(44.0%),较1981年晚2周以上;到8月初才发现所感染的猪,明显晚于1980年和1981年,受上述众因素的影响,本年度本市乙型脑炎发病率仍较低(0.8/10万),在M14病毒的监测中,发现三带喙库蚊环病毒M14病毒检出率为9.32%,最低现场感染率为1:526,当年最高检出率(32%)和最低现场感染率高峰(1:156),高峰出现在8月30日,与乙脑病毒自然感染率出现的高蜂相一致;猪感染M14病毒与感染乙脑病毒的时间基本相同,表明这两种病毒在自然界传播中不存在明显的互相干扰关系,乙脑低流行年的出现并未受到M14病毒的影响。  相似文献

Large turves from a ryegrass/white clover based pasture wereexposed to 350 or 700 µl l-1 CO2 for a period of 217 din controlled environment rooms. The temperature was increasedduring the experiment from 10/4 °C day/night to 16/10 °Cand finally to 22/16 °C. The turves were cut to a heightof 2 cm at intervals and growth rates calculated from the regrowth. Growth rates over the duration of the experiment were 8% higherat elevated CO2; the difference between CO2 treatments beingstatistically significant only at the highest temperature. Speciescomposition of the turves at 350 µl l-1 CO2 showed seasonalchanges similar to those measured in the field. The effect ofCO2 was to exaggerate the normal decline of ryegrass at warmertemperatures and increase the proportion of white clover. About30% of the total growth rate was from other species (notablyBromus hordeaceus L. and Poa trivialis L.) and this fractionwas similar between CO2 levels. Root mass was measured at theend of the experiment and was 50% higher at elevated CO2. The modest above-ground response to CO2 was a result of CO2stimulation occurring only at the higher temperature. Becauseof the CO2 x temperature interaction, the effect of CO2 in temperateregions will be seasonal. When this is matched with seasonalgrowth patterns of herbage species, a complex response of pasturecommunities to CO2 is possible. In our case, white clover wasgrowing most strongly during the period of greatest CO2 stimulationand consequently its growth was enhanced more than that of ryegrass;however, the cooler season growth of ryegrass gives it a temporalniche which is little affected by CO2 and this may be importantfor ryegrass stability if it is an inherently poor responderto CO2. The Effect of Temperature on Leaf Appearance in Rice   总被引:13,自引:3,他引:10  
Temperature is the principal environmental determinant of cropleaf appearance. The objective of this study is to analyse whetherthere are different effects of day temperature (TD) and nighttemperature (TN) on main-stem leaf appearance in rice (OryzasativaL.). Plants of 12 rice cultivars were grown at five constant temperatures(22, 24, 26, 28 and 32 °C) and four diurnally fluctuatingtemperatures (TD/TN: 26 /22, 30 /22, 22 /26 and 22 /30 °C)with a constant photoperiod of 12hd-1. The leaf appearance onthe main stem was measured. A constant change in leaf appearance rate was observed duringontogeny. The relation between the number of emerged leavesand days from seedling emergence was described by a power-lawequation with only one cultivar-specific parameter. Values forthis parameter were estimated for the five constant temperaturetreatments, and the relation between this parameter and temperaturewas quantified by a nonlinear model. Leaf appearance for thefour fluctuating temperature treatments could be accuratelypredicted on the basis of these relations in each cultivar.This indicated that there were no specific effects ofTDandTNonleaf appearance in rice, in contrast with phenological developmentto flowering. The optimum temperature for leaf development wasfound to be substantially higher than for development to flowering. The final main-stem leaf number differed with diurnal temperatureconditions. When a diurnal temperature delayed flowering, itincreased the leaf number as well. This might explain whyTDandTNhada different effect on development to flowering but not on leafdevelopment. 为了研究寄主营养对桔小实蝇耐寒性的作用,测定了以15种果蔬饲养的桔小实蝇1日龄蛹的过冷却点(supercooling points,SCP); 再选取南瓜、西红柿、柑桔、番石榴和杨桃等5种果蔬,测定了桔小实蝇3龄老熟幼虫、1日龄蛹、3日龄蛹、5日龄蛹、7日龄蛹和雌雄成虫的过冷却点,并观察了1日龄蛹的低温存活力。结果表明:(1)15种果蔬饲养所得的桔小实蝇1日龄蛹SCP均值在-11.03℃~-13.17℃,不同寄主发育的桔小实蝇SCP值存在显著性差异,其中以取食蒲桃的最高,为-11.03℃,取食苦瓜的最低,为-13.17℃。(2)5种果蔬饲养所得的桔小实蝇各虫态的SCP均值存在极显著差异(F(4,863)=35.6,P<0.01); 同一寄主上的桔小实蝇不同虫态之间SCP均值也达到极显著性差异(F(6,863)=392.9,P<0.01); 且寄主和发育龄期之间存在着极显著的交互作用(F(24,863)=9.4,P<0.01)。(3)桔小实蝇各发育阶段,SCP值表现一定变化: 老熟幼虫发育至1日龄蛹,SCP值变化不大; 蛹发育至3、5和7天过冷却能力明显增强,降至-20℃左右,但他们之间没有明显区别; 羽化后3~5天的成虫SCP值又升高至-10℃左右。老熟幼虫、1日龄蛹和2~3日龄成虫与3日龄、5日龄和7日龄蛹的SCP值之间有显著性差异。(4)将5种果蔬饲养所得的桔小实蝇1日龄蛹置于6℃和-3℃下进行较长时间(1~8天)和较短时间(1~8 h)的低温处理,发现番石榴、杨桃和南瓜发育的蛹经低温处理后的校正羽化率较西红柿和柑桔发育的蛹高; 同样在0℃、3℃、6℃和9℃处理(2天)的实验中,得出相似的结果。因此,本实验结果表明桔小实蝇幼虫由于生活寄主的不同使得其下一代蛹的耐寒性产生了差异,引起其差异的原因值得进一步研究。  相似文献

Gas exchange and organic acid accumulation of the C3-CAM intermediateClusia minor L. were investigated in response to various day/nighttemperatures and two light regimes (low and high PAR). For bothlight levels equal day/night temperatures between 20°C and30°C caused a typical C3 gas exchange pattern with all CO2uptake occurring during daylight hours. A day/ night temperatureof 15°C caused a negative CO2 balance over a 24 h periodfor low-PAR-grown plants while high-PAR-grown plants showeda CAM gas exchange pattern with most CO2 uptake taking placeduring the dark period. However, there was always a considerablenight-time accumulation of malic acid which increased when thenight-time temperature was lowered and had its maximum (54 mmolm–2) at day/night temperature of 30/15°C. A significantamount of malic acid accumulation (23 mmol m–2) in low-PAR-grownplants was observed only at 30/15°C. Recycling of respiratoryCO2 in terms of malic acid accumulation reached between 2·0and 21·5 mmol m–2 for high-PAR-grown plants whilethere was no significant recycling for low-PAR-grown plants.Both low and high-PAR-grown plants showed considerable night-timeaccumulation of citric acid. Indeed under several temperatureregimes low-PAR-grown plants showed day/night changes in citricacid levels whereas malic acid levels remained approximatelyconstant or slightly decreased. It is hypothesized that lowand high-PAR-grown plants have different requirements for citrate.In high-PAR-grown plants, the breakdown of citrate preventsphotoinhibition by increasing internal CO2 levels, whereas inlow-PAR-grown plants the night-time accumulation of citric acidmay function as an energy and carbon saving mechanism.  植物群落季相阶段的划分,对于诊断地方、区域和全球尺度上生态系统对气候变化的快速响应和进行遥感植被生长季节的地面检验,具有重要 的科学意义。该文利用物候累积频率拟合法对我国东部温带地区7个站点1982~1996年的植物群落季相阶段进行划分,并分析了植物群落季相的 空间差异和年际变化及其与气候因子的关系。结果表明:1)各站点多年平均变绿期和旺盛光合期初日随纬度的升高而推迟,凋落期和休眠期初 日随纬度的升高而提前;多年平均变绿期、旺盛光合期和凋落期长度随纬度的变化不甚明显,而休眠期则随纬度的升高明显延长;2)在研究期 间内,站点平均变绿期初日以0.6 d&;#8226;a-1的平均速率显著提前,且长度以0.7 d&;#8226;a-1的平均速率显著延长;旺盛光合期初日呈不显著推迟,长 度呈不显著缩短;凋落期初日呈微弱提前,长度呈微弱延长;休眠期初日呈微弱提前,但长度却以0.9 d&;#8226;a-1的平均速率显著缩短;3)站点平 均变绿期初日与当月平均气温的负相关显著,平均气温每升高1 ℃,初日提前约4.3 d;站点平均旺盛光合期初日与初日前第二个月到初日当月 平均气温的负相关显著,平均气温每升高1 ℃,初日提前约4.4 d;站点平均凋落期和休眠期初日与气温的相关均不显著。  相似文献

三带喙库蚊体内猪繁殖与呼吸综合征病毒的分离与鉴定   总被引:1,自引:0,他引:1  
【目的】调查猪场蚊虫是否能携带猪繁殖与呼吸综合征(PRRS)病毒。【方法】采集发生PRRS疫情的3个养猪场蚊虫样本,采用RT-PCR方法检测PRRS病毒核酸,取阳性蚊虫样本接种Marc-145细胞进行病毒的分离培养,以间接免疫荧光抗体技术和分子克隆技术进行病毒的鉴定。【结果】 养猪场内的蚊虫主要有三带喙库蚊Culex tritaeniorhychus、凶小库蚊Culex modestus、中华按蚊Anopheles sinensis和骚扰阿蚊Armigeres obturbans,其中三带喙库蚊占86.76%;以PRRS病毒N基因引物进行扩增,三带喙库蚊样本呈现阳性反应,而其他蚊种均为阴性。在蚊虫接种的Marc-145细胞中可见细胞融合和空泡形成等细胞病变效应;用抗PRRS病毒N蛋白抗体和羊抗猪IgG(H+L)-FITC进行间接免疫荧光染色,感染细胞呈现黄绿色荧光;以NSP2基因引物进行RT-PCR扩增、克隆与测序,发现库蚊源病毒与相应猪场猪源病毒中相应基因的序列具有较高同源性。【结论】 三带喙库蚊为猪舍优势蚊种,并能携带猪繁殖与呼吸综合征病毒。  相似文献

Starting in 1996, individual trees of Scots pine (Pinus sylvestrisL.) aged 30 years, were grown in closed-top chambers and exposedto either normal ambient conditions (CON), elevated CO2(approx.700 µmol mol-1; Elev. C), elevated temperature (approx.2 °C and approx. 6 °C above the outside ambient temperatureduring the ‘growing season’ and ‘off season’,respectively; Elev. T) or a combination of elevated CO2and warmertemperature (Elev. CT). Sap flow was monitored simultaneouslyby the constant-power heat balance method in a total of 16 trees,four for each treatment, over a 32 d period in summer 1998 (afterthe completion of needle expansion and branch elongation). Toquantify the contributions of crown and physical environmentalvariables to total crown transpiration, a ‘sun/shade model’was developed and used to partition the changes in transpirationto different sources. The results of the sap flow measurementsindicate that (1) total daily sap flow (Etree.d) varied from0.15–3.41 kg per tree; (2) the treatment effect on Etree.ddependedgreatly on the weather conditions; (3) the cumulative Etree.dforthe 32 d dropped significantly by 22% relative to CON (P =0.038)under Elev. C and increased significantly by 21% (P =0.043)and 16% (P =0.048) under Elev. T and Elev. CT, respectively.In general, the modelled transpiration gave good agreement withthe sap flow results. The model computations showed that, ona typical sunny day in summer, the effect of treatment on crownstomatal conductance was responsible for approx. 80% of thechange inEtree.d , while the increase in needle area and theeffect on total radiation absorption contributed only a smallpercentage. Furthermore, sunlit needles were responsible forover 60% of change in transpiration. The effect of the treatmentson Etree.dwas larger at high temperature and vapour pressuredeficit but was not sensitive to incident daily radiation. Maximum daily consumption was estimated for Mysis relicta fedad libitum rations of Daphnia pulex at 4,10,15 and 18°C.Gut-residence time was also evaluated for M.relicta fed clado-ceranprey at 4, 10 and 157deg;C. Mean daily consumption (g dry weightof Daphnia g–1 dry weight of Mysis day–1) rangedfrom 6% at 4%C to 12% at 10°. At 18°C, Mysis feedingrate declined to 9% day1. Mean, weight-adjusted consumptionrates exhibited a ‘dome-shaped’ response in relationto water temperature. Consumption rate was highest at 10°Cand lowest at 4°C. Estimated Q10 was more sensitive from4 to 10°C (Q10= 3) than from 10 to 15°C (Q10=1.2). Gut-residencetime for Mysis was inversely related to water temperature, implyingthat evacuation rate increases linearly with water temperature.Feeding and gut-evacuation rates become disassociated at watertemperatures >10°C. As water temperature increased above1°C, relative evacuation rate increased, whereas feedingrate declined.
