The prevalence of glucose-6-phosphate dehydrogenase (G6PD) deficiency and its gene mutations were studied in the Achang population from Lianghe County in Southwestern China. We found that 7.31% (19 of 260) males and 4.35% (10 of 230) females had G6PD deficiency. The molecular analysis of G6PD gene exons 2―13 was performed by a PCR-DHPLC-Sequencing or PCR-Sequencing. Sixteen inde-pendent subjects with G6PD Mahidol (487G>A) and the new polymorphism IVS5-612 (G>C), which combined into a novel haplotype, were identified accounting for 84.2% (16/19). And 100% Achang G6PD Mahidol were linked to the IVS5-612 C. The percentage of G6PD Mahidol in the Achang group is close to that in the Myanmar population (91.3% 73/80), which implies that there are some gene flows between Achang and Myanmar populations. Interestingly, G6PD Canton (1376G>T) and G6PD Kaiping (1388G>A), which were the most common G6PD variants from other ethnic groups in China, were not found in this Achang group, suggesting that there are different G6PD mutation profiles in the Achang group and other ethnic groups in China. Our findings appear to be the first documented report on the G6PD genetics of the AChang people, which will provide important clues to the Achang ethnic group origin and will help prevention and treatment of malaria in this area.  相似文献

This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2) of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes. Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. By producing the diploid eggs as a unique reproductive way, the diploid gyno- genetic progeny of allotetraploid hybrids of red crucian carp × common carp had important signifi- cances in both biological evolution and production application.  相似文献

Satellite RNAs(satRNAs)are molecular parasites that interfere with the pathogenesis of thehelper viruses.In this study,the relative accumulation of cucumber mosaic virus(CMV)-Fny genomicRNAs with or without satRNAs were quantitatively analyzed by real-time RT-PCR.The results showed thatsatRs apparently attenuated the symptoms of CMV-Fny on Nicotiana tabacum by depressing the accumu-lation of CMV-Fny genomic RNAs,tested as open reading frames.The accumulation of CMV-Fny 1a,2a,2b,3a,and CP genes was much higher than that of CMV-Fny with satRs added(CMV-Fsat),at differentinoculation times.CMV-FnyΔ2b,in which the complete 2b gene and 41 amino acids at the C-terminal of the2a gene were deleted,caused only a slight mosaic effect on N.tabacum seedlings,similar to that of CMV-Fsat,but the addition of satRs to CMV-FnyΔ2b showed further decrease in the accumulation of CMV-FnyΔ2b genomic RNAs.Our results indicated that the attenuation of CMV,by adding satRs or deleting the2b gene,was due to the low accumulation of CMV genomic RNAs,and that satRNA-mediated reduction ofCMV genomic RNAs accumulation in N.tabacum was possibly related to the 2b gene.  相似文献

A preliminary study on the interaction of G protein (guanine triphosphate binding pro- tein) β1γ2 subunits and their coupled components in cell signal transduction was conducted in vitro. The insect cell lines, Sf9 (Spodoptera frugiperda) and H5 (Trichoplusia ni) were used to express the recombinant protein Gβ1γ2. The cell membrane containing Gβ1γ2 was isolated through affinity chromatography column with Ni-NTA agarose by FPLC method, and the highly purified protein was obtained. The adenylyl cyclase 2 (AC2) activity assay showed that the purified Gβ1γ2 could significantly stimulate AC2 activity. The interaction of β1γ2 subunits of G protein with the cytoplasmic tail of various mammalian adenylyl cyclases was monitored by BIAcore technology using NTA sensor chip, which relies on the phenomenon of surface plasmon resonance (SPR). The experiments showed the direct binding of Gβ1γ2 to the cytoplasmic tail C2 domain of AC2. The specific binding domain of AC2 with Gβ1γ2 was the same as AC2 activity domain which was stimulated by β1γ2.  相似文献

With three different methods, the total flavonoids from the roots of cultivated Gtycyrrhiza pallidiflora Maxim. were determined and compared by ultraviolet-visible spectrophotometry. The results showed that the extract yield of total flavonoids in crude extracts was 4.54% by direct determination method, 4.27% by AI( NO3)3 colorimetry method and 2.09% by AlCl3 colorimetry method. The content of total flavonoids in roots of cultivated G. pallidiflora was only 0.05%. The cultivated G. pallidiflora in Lanzhou is not unfit for the medicine source of flavonoids.  相似文献

In plants, triose phosphate/phosphate translocator (TPT) is the first regulation point forpartitioning of photosynthate between source and sink. Studies on the characteristic of TPT and itsregulation on the distribution of assimilates are critical for improving the utilization rate of photosyntheticassimilates. Chloroplasts with intactness of more than 91% and high purity were isolated from wheat( Triticurn aestivurn L. cv. Jing 411) leaves. Analysis of SDS-PAGE and labeling with an irreversible specificinhibitor, [H3]2^-DIDS (4, 4‘-diisothiocyano-2, 2‘-stilbenedisulfonate, DIDS) demonstrated that wheat TPTwas a chloroplast membrane protein with a 35 kD molecular weight, which comprised about 15% of the totalmembrane proteins of chloroplasts. Western blotting analysis showed that wheat TPT is uniquelydistributed in the envelope membrane of chloroplasts, but not detected in the membranes of vacuoles andmitochondria. The silicone-oil-layer centrifugation system was employed to study the kinetic properties ofTPT. The results showed that the maximal transport activity of TPT was the highest for dihydroxyacetonephosphate (DHAP)/inorganic phosphate (Pi), then for phosphoenolpyruvate (PEP)/Pi and glucose-6-phosphate (G6P)/Pi. The Km value of TPT was the lowest for DHAP, followed by Pi, PEP and G6P,therefore the most preferred substrate of TPT is DHAP. The transport of wheat TPT to DHAP was stronglyinhibited by DIDS with a degree of 95%. Inhibition of TPT transport activity led to an obvious accumulationof starch in chloroplasts, therefore the TPT protein of wheat controls the export of TP out of chloroplastsinto cytosol. Except for the need of participating in the Calvin cycle, the ratio of TP exported out ofchloroplast to the one used for synthesizing starch was at least 93.6:6.4. The TPT protein from wheat hasmuch high transport efficiency, which plays an important role in the regulation of the distribution ofassimilates in wheat chloroplasts.  相似文献

The C_3 halophyte Suaeda salsa was used to investigate the roles of Ca~(2 ),Ca~(2 )channels,and calmodulin(CAM)in betacyaninmetabolism.Seeds of S.salsa were cultured in both the dark and light for 3 days.The fresh weight and betacyanin contentwere much higher in S.salsa seedlings formed in the dark than in seedlings formed in the light.The addition of Ca~(2 )tothe half-strength MS nutrient solution promoted betacyanin accumulation in the dark,whereas Ca~(2 )depletion by EGTAsuppressed the dark-induced betacyanin accumulation in shoots of S.salsa.The Ca~(2 )channel blocker LaCl_3 also inhibiteddark-induced betacyanin accumulation.The highest activity of CaM and the maximum betacyanin content decreased by51% and 45%,respectively,in shoots of S.salsa seedlings treated with the potent CaM antagonist chlorpromazine in thedark.Furthermore,the other CaM antagonist N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide(W-7)also inhibited theactivity of CaM and dark-dependent betacyanin accumulation,whereas its less active structural analog N-(6-aminohexyl)-1-naphthalenesulfonamide(W-5)had little effect on the responses to dark of S.salsa seedlings.These results suggest thatCa~(2 ),Ca~(2 )-regulated ion channels,and CaM play an important role in dark-induced betacyanin accumulation in the shootsof the C_3 halophyte S.salsa.  相似文献

本研究旨在探讨宫颈癌前病变和宫颈癌的发生发展与人乳头瘤病毒及协同因子(HSV,CMV)的关系。 对81例不同宫颈病变组织进行HPV16/18和HPV6/11原位杂交,同时对103例不同宫颈病变组织用DNA扩增法检测HPV、HSV 和CMV。结果表明病毒DNA原位杂交信号的分布与HE染色中挖空细胞的分布一致。HPV16/18与不同宫颈病变组织原位杂交阳性率平均为51%,HPV6/11的则为64%。经PCR检测,HPV16/18、HPV6/11、HSV、 CMV在不同宫颈病变组织中的阳性率分别为21%、4%、23%和0%。HSV可协同HPV16/18恶性转化宫颈上皮细胞,并对其协同机制进行了细胞及分子生物学的探讨。 Abstract:The carcinogenesis of the human cervical precancerous lesion,cervical carcinoma is known closely associated with human papillomavirus (HPV).The purpose of this article is to identify whether HSV and CMV play as co factor role in the carcinogenesis.Eighty one cases of various cervical lesions were analyzed by HPV6/11,HPV16/18 in situ hybridization.Meanwhile,HPV,HSV and CMV were determined in 103 cases of various cervical lesions.The results show that the distribution of positive hybridization signal was consistent with the distribution of Koilocytic cells in HE stain.Of these cervical specimens investigated,the positive rates of HPV16/18 and HPV6/11 using ISH were 51% and 64%,respectively,the infection rates of HPV16/18,HPV6/11,HSV and CMV using PCR were 21%,4% 23% and 0%,respectively.The co operation effect of HPV and HSV occurred in the oncogenesis of human cervical carcinoma,and moreover,the cellular and molecular biological mechanisms were discussed.  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献
