Six sequence‐characterized amplified region (SCAR) markers linked to the apple scab resistance gene Vf were evaluated for their utility in marker‐assisted selection (MAS) in apple breeding. Of the six SCARs used in this study, ACS‐6 was located left of the Vf gene, ACS‐7 and ACS‐9 co‐segregated with Vf, and ACS‐8, ACS‐4, ACS‐5 were located right of the Vf gene. Three families derived from crosses between scab‐resistant and scab‐susceptible cultivars, including ‘Liberty’ × ‘Deljub’, ‘Liberty’ × ‘Delcorf’, and ‘Florina’ בDelcorf’, previously screened for scab resistance following greenhouse inoculation with the fungal pathogen Venturia inaequalis, were genotyped and compared with phenotypic reactions to scab infection in the field. For each family, a subset progeny of 30 seedlings (propagated onto Malling 9 rootstock and of 7 years old) was selected based on fungal sporulation according to the following scheme. Ten seedlings with no visible scab sporulation on leaves were given phenotypic scores of 0 (deemed resistant); 10 seedlings with moderate scab sporulation were given phenotypic scores of 1.0 (deemed moderately resistant); and 10 seedlings with heavy sporulation were given phenotypic scores of 2.0 (deemed susceptible). DNA was isolated from leaf tissue collected from all 90 seedlings, parents and Malus floribunda 821, the original source of the Vf gene, and screened with all six SCARs. All six SCARs were present in the two scab‐resistant parents, ‘Liberty’ and ‘Florina’, and M. floribunda 821; while, the two scab‐susceptible parents, ‘Deljub’ and ‘Delcorf’, lacked all SCARs. All SCARs were either present or absent in varying numbers of seedlings in each progeny with phenotypic ratings of either 0 (resistant) or 1.0 (moderately resistant); while all seedlings with phenotypic ratings of 2.0 (susceptible) lacked all SCARs. The inconsistencies between phenotypic scab ratings and SCAR marker data are discussed.  相似文献

Untrastructural interactions between Venturia inaequalis and callus cultures from scab susceptible and resistant apple varieties, were similar. Host cell wall changes, appositions, and invagination of host plasmamembrane at sites of close contact with fungal hyphae were regularly observed. The ultrastructural observations are described and discussed. The host cell alterations as well as many fungal structures corresponded to those known in young leaves of susceptible apple varieties.  相似文献

Recently, a epidemic of apple proliferation (AP) in an orchard area of Trentino (North Italy) occurred. The most affected cultivars were Golden Delicious, Florina and Renetta Canada grafted on different rootstocks. In this area the known or supposed vectors of the disease were not present. At the same time as the AP symptoms appeared, a notable increase of the presence of psyllids was observed on apple trees so a correlation between these insects and the AP was hypothesized. Four different psyllid species were found in the orchard: Cacopsylla melanoneura (Förster 1848), Cacopsylla costalis (Flor 1861), Cacopsylla mali (Schmidberber 1836) and Trioza urticae (Linnaeus 1758). The first two were more frequent in spring at the adult stage. In 1997 C. costalis was particularly numerous and was used to plan AP transmission trials. The transmission from infected AP apple trees to the insect in field and from the psyllids to non-infected Golden Delicious and Florina plants in the greenhouse were conducted. For the control of phytoplasma presence in the psyllids and in the apple plants polymerase chain reaction (PCR), nested PCR and restriction fragment length polymorphism analysis were used. In autumn 1997, some inoculated plants showed symptoms of AP. Molecular results were consistent with the presence of phytoplasma in C. costalis and in inoculated apple cultivars.  相似文献

Using resistance genes from a crossable donor to obtain cultivars resistant to diseases and the use of such cultivars in production appears an economically and environmentally advantageous approach. In apple, introgression of resistance genes by classical breeding results in new cultivars, while introducing cisgenes by biotechnological methods maintains the original cultivar characteristics. Recently, plants of the popular apple ‘Gala’ were genetically modified by inserting the apple scab resistance gene Rvi6 (formerly HcrVf2) under control of its own regulatory sequences. This gene is derived from the scab‐resistant apple ‘Florina’ (originally from the wild apple accession Malus floribunda 821). The vector used for genetic modification allowed a postselection marker gene elimination to achieve cisgenesis. In this work, three cisgenic lines were analysed to assess copy number, integration site, expression level and resistance to apple scab. For two of these lines, a single insertion was observed and, despite a very low expression of 0.07‐ and 0.002‐fold compared with the natural expression of ‘Florina’, this was sufficient to induce plant reaction and reduce fungal growth by 80% compared with the scab‐susceptible ‘Gala’. Similar results for resistance and expression analysis were obtained also for the third line, although it was impossible to determine the copy number and TDNA integration site–such molecular characterization is requested by the (EC) Regulation No. 1829/2003, but may become unnecessary if cisgenic crops become exempt from GMO regulation.  相似文献

本研究对9个赤霉病抗性不同小麦品种采用赤霉病菌分生孢子悬浮液以单花针注法进行了田间和温室抗病性鉴定;测定了各品种的胚性愈伤组织和盛花期麦穗分别经赤霉病菌毒素和分生孢子接种前后SOD活性的变化。结果表明,各品种SOD活性与其对赤霉病抗性呈极显著的正相关。接种后寄主的SOD活性均有提高,抗病品种比感病品种提高幅度大,且有新的同工酶带出现。抗病品种望水白比感病品种Alondra“S”多出两条SOD同工酶谱带。SOD在小麦抗赤霉病上可能起积极作用,其活性有可能作为鉴定小麦抗赤霉病的一种生理生化指标。  相似文献

抗赤霉病小麦品种苏麦3号、繁9能转化镰刀菌毒素脱氧雪腐镰刀菌烯醇(DON)成产物X,而感病小麦品种宁麦6号、徐州21无转化能力。产物X对小麦黄化芽鞘的伸长生长无抑制作用,而对禾谷镰刀菌分生孢子的萌发有明显抑制。说明抗性小麦品种对赤霉病菌毒素的脱毒是小麦重要的抗赤霉病机制。  相似文献

Phenolic Compounds of Apple and their Relationship to Scab Resistance   总被引:4,自引:0,他引:4  
The contents of 15 different phenolic compounds in apple leaves and fruit skin from various orchards were determined by HPLC. High concentrations of flavan-3-ols were found in those orchards which were not infected by Venturia inaequalis . This observation corroborates earlier findings on different cultivars. The hypothesis that these compounds could be involved in the resistance of apple to scab was tested by inhibiting the key enzyme of the phenol biosynthesis, the phenylalanine-ammonia-lyase, in young shoots of the resistant genotype 'Sir Prize'. After inoculation of the inhibitor-treated leaves, severe symptoms of the disease occurred. The fungal infection was confirmed by histological studies. No flavonal-accumulation could be observed at the infection site which was the case in the non-inhibited but inoculated controls.  相似文献

采用RAPD技术,用524个随机引物对3个抗病品种和4个感病品种的基因组DNA进行扩增,发现3个引物能在抗、感品种间检测到稳定的多态性。这3个引物及它们产生的多态片段是S347 1220bp、S372 872bp和S375 1360bp,其中S347 1220bp和S372 875bp在感病品种上呈显性扩增,在抗病品种上无扩增,相反S375 1360bp在感病品种上无扩增,在抗病品种上呈显性扩增。初步判断这3个特异带与小麦赤霉病抗性有关。  相似文献

Energy flow in orchards can be used to determine first the best management practices and second the possibility of using those which have best environmental advantages. Conventional and organic peach and kiwi orchards were selected in order to (a) determine energy flow of the farming systems, and (b) reveal the importance of energy inputs in crop and farming system replacement. Fifteen farms (four conventional and three organic kiwi orchards; four conventional and four organic peach orchards) were selected with proportional stratified random sampling during the years 2010–2013. The Hierarchical Cluster Analysis (HCA) method was applied using nine production coefficients' variables (fertilizers, fungicides, insecticides, weed control, diesel, labor, irrigation, branches shoring, and machinery) and revealed three groups of the studied orchards. The highest contributors in cluster formation were weed control, branches shoring, labor, and machinery. The effect of the production coefficients on the grouping of the studied orchards reveals their importance for these crops and farming systems. Most of the production coefficients showed their lowest values in organic kiwi orchards (Group 3), so it could be said that they can play a key role in the replacement of the peaches, and conventional kiwi orchards. It seems that production coefficients can be used as a “tool” for decision makers who are seeking for crops and farming systems with low energy inputs and best environmental advantages in order to use them in crop replacement in agro-ecosystems.  相似文献

以5个赤霉病抗性水平不同的小麦品种为材料,研究了禾谷镰刀菌粗毒素对小麦成熟胚脱分化与再分化的影响.结果表明,小麦成熟胚愈伤组织的诱导率和分化率在品种间、毒素浓度间、毒素浓度×品种间差异极显著.低浓度(5 g/L)禾谷镰刀菌粗毒素能够促进抗性较强品种的出愈速度,高浓度禾谷镰刀菌粗毒素(10 g/L和15 g/L)对成熟胚愈伤组织的出愈速度起到抑制作用,随着浓度提高,抑制作用增强.成熟胚愈伤组织的诱导率和分化率随着禾谷镰刀菌粗毒素浓度升高而下降,品种的抗性越差下降幅度越大.10 g/L毒素浓度,5个品种成熟胚愈伤组织的SOD活性均比对照(0 g/L)增加;20 g/L毒素浓度,5个品种成熟胚愈伤组织的SOD活性均有所下降,抗性品种苏麦3号和郑9023的SOD活性仍高于对照,其它3个感性品种低于对照,SOD活性与小麦品种的赤霉病抗性水平存在一定关系.研究结果为小麦赤霉病抗性育种提供了一定的技术支撑.  相似文献

 Apple scab, caused by the fungus Venturia inaequalis (Cke.) Wint., is an important disease in commercial apple production. A mapping population of 155 individuals, derived from a cross between the apple varieties ‘Prima’ (resistant)בFiesta’ (susceptible), was scored for response to the disease in replicated field and glasshouse trials throughout Europe. Twenty data sets were selected and cluster analysis was used to form a consensus score for the population fitting a 1 : 1 segregation ratio of resistance:susceptibility. The progeny were scored with molecular markers. A detailed map covering 54 cM of the ‘Prima’ linkage group containing the Vf gene for scab resistance was constructed using 24 molecular markers linked to the resistance gene. One isoenzyme marker (Pgm-1), six RFLP markers and 17 RAPD markers formed a linkage group with the consensus measure of resistance to scab. Four marker bridges were established with the corresponding ‘Fiesta’ linkage group with additional markers (one isozyme, one RFLP, three RAPD and one AFLP). A low chi-square value indicated a good fit of the marker ordering, which was in close agreement with previously reported linkage positions for some of the markers and Vf. Differences were observed in the ability of different scoring methods to resolve susceptible and resistant classes. The results obtained for the consensus classification of resistance to scab for the population may suggest the presence of virulent inocula at some sites, which could overcome the Vf gene for resistance. The consequences of relying on individual scoring occasions for studying Vf scab resistance are discussed in the context of linkage analysis, conventional breeding selection, and marker-assisted selection. Abstract: Changes in population densities of the apple sawfly Hoplocampa testudinea (Klug) and its parasitoid Lathrolestes ensator (Brauns) were monitored in 15 apple orchards for a period of up to 4 years. The parasitoid species was found in all orchards except one, and was more numerous in plantings on sandier soils. Post‐bloom insecticide applications against other pests, and carbaryl for fruit thinning, often decimated both host and parasitoid in integrated pest management orchards. In organic orchards, where synthetic pesticides are banned, and Quassia is the only remedy against sawfly, the pest is more problematic. The low levels of parasitism in all organic orchards, except one, were possibly due to the application of wettable sulphur during the parasitoid flight period. The sawfly usually recovers more quickly than its parasitoid when chemical control is discontinued, because propagation of L. ensator is limited in various ways. The parasitoid is time limited, because suitable second instar host larvae are rarely available for more than a week on a single apple variety. Moreover, parasitism levels stay moderate because the parasitoid females do not avoid superparasitism. Finally, relatively more sawflies than parasitoids emerge after 10 months underground, because the incidence of prolonged diapause is more elevated in L. ensator than in the sawfly. It is suggested that both the high incidence of prolonged diapause and the inability to avoid superparasitism are useful in reducing the risk of local extinction. Elevated sawfly attack in a single early apple variety would reduce exploitation of suitable host larvae in other nearby varieties, in as far as the parasitoid is not able to distinguish fruitlets with accessible second instar host larvae from those with inaccessible older larvae. Although the former are available for a limited time, the latter may keep the parasitoids from moving to the less abundant second instar larvae in late(r) varieties. Reduction of elevated host density in such an early variety by a properly timed application of a short‐lived pesticide, such as Quassia, increases parasitism levels proportionally, and is expected to promote parasitoid movement to host larvae in other varieties nearby.  相似文献

Organic farming aims to be environmentally sound, but the question as to whether organic cropping brings more nutritional benefits to farmers and consumers than the conventional cropping needs still to be answered. To gain insights into the molecular effects of organic farming we used proteome analysis to analyze cabbage (Brassica oleracea L. var 'capitata') and carrot (Daucus carota var. 'sativus') Our aim was to identify the metabolic pathways that are affected by different cropping regimes and thus, may have an effect on quality, storability and pathogen resistance of crops. By means of two dimensional gel electrophoresis and MALDI tandem mass spectrometry we compared proteomes of cabbage and carrot root, obtained in the first growth season, cropped under three different schemes. These included a conventional scheme (C) and two organic schemes, O1, in which nutrients were delivered in a form of slurry, in accordance to regulations of organic farming and O2, in which nutrient supply was based mainly on autumn green manures. Proteins were extracted from lyophilized plant tissues into a buffer containing high concentrations of urea/thiourea, two detergents and reducing agent. This approach allowed short handling times of fresh plant materials. In the case of cabbage samples, the abundance levels of 58 out of more than 1300 quantified protein spots varied significantly between conventional farming and any of the organic cropping systems. Proteome profiles were also very similar between carrot root samples, where 68 out of 1800 resolved protein spots varied significantly. Proteins of the glycolytic pathway and Krebs cycle as well as several proteins related to amino acid and protein metabolism were overexpressed in organically farmed cabbage. Proteins related to detoxification processes were overexpressed in conventionally grown cabbage. Proteins involved in metabolism of carbohydrates, polypeptides and secondary metabolites were affected by different cropping regimes in carrots. The proteomes of conventionally grown vegetables varied from organically grown vegetables to a larger extent than the two organic cropping schemes varied from each other. In conclusion, this proteomics platform is suitable and useful for systematic studies of the effects of organic and conventional farming techniques on plant metabolism.  相似文献

Host plant resistance is the most efficient and easy way to manage chickpea blight caused by Ascochyta rabiei (Pass.) Lab. For this purpose, 374 chickpea lines/varieties from various research organisations were evaluated in plastic tunnels. None of the line showed immune response against the blight; however, one line (K-01005) was found to be highly resistant. Moreover, 15 entries were resistant, 136 exhibited moderate resistant reaction, 150 were susceptible and 72 showed highly susceptible response. The genotypes found that resistance against blight can serve as a source of resistance for breeding programmes, and they could be released for commercial production directly.  相似文献

The cultivated apple is susceptible to several pests including the rosy apple aphid (RAA; Dysaphis plantaginea Passerini), control of which is mainly based on chemical treatments. A few cases of resistance to aphids have been described in apple germplasm resources, laying the basis for the development of new resistant cultivars by breeding. The cultivar ‘Florina’ is resistant to RAA, and recently, the Dp-fl locus responsible for its resistance was mapped on linkage group 8 of the apple genome. In this paper, a chromosome walking approach was performed by using a ‘Florina’ bacterial artificial chromosome (BAC) library. The walking started from the available tightly linked molecular markers flanking the resistance region. Various walking steps were performed in order to identify the minimum tiling path of BAC clones covering the Dp-fl region from both the “resistant” and “susceptible” chromosomes of ‘Florina’. A genomic region of about 279 Kb encompassing the Dp-fl resistance locus was fully sequenced by the PacBio technology. Through the development of new polymorphic markers, the mapping interval around the resistance locus was narrowed down to a physical region of 95 Kb. The annotation of this sequence resulted in the identification of four candidate genes putatively involved in the RAA resistance response.  相似文献

Abstract:  Resistance of the codling moth Cydia pomonella (L.) (Lep., Tortricidae) to the organophosphorus compound (OP) azinphosmethyl was observed in apple orchards in Israel. The level of resistance varied with the pest control strategy. Compared with a sensitive laboratory population, the resistance level was highest in insects from the preventative pest control strategy, intermediate in integrated pest management (IPM) orchards, and relatively low in the organic orchards. The level of azinphosmethyl resistance in larvae (but not in adults) exposed for 17 generations in the laboratory to a pesticide-free diet was reduced by 50%. Codling moth larvae resistant to azinphosmethyl were also resistant to various insect growth regulators (IGRs). The IGRs include three chitin synthesis inhibitors (diflubenzuron, novaluron and teflubenzuron), two juvenile hormone mimics (pyriproxyfen and fenoxycarb) and one ecdysone agonist (methoxyfenozide). Codling moth resistant to azinphosmethyl was tolerant to methoxyfenozide and novaluron without previous history of application in apple orchards, indicating the possibility of cross-resistance. According to this study, managing resistance programs in apple orchards should be based on IPM principles with minimum use of conventional neuroactive pesticides.  相似文献

Abstract 1 The presence and abundance of arthropods were compared in three olive orchards under organic, integrated and conventional management regimes. In each olive orchard, trees were sampled in the canopy by beating branches and soil arthropods by placing pitfall traps. Contrary to expectations, the highest abundance of arthropods occurred in the integrated management orchard. The most abundant groups were Formicidae and the species Euphyllura olivinae (Homoptera: Psyllidae). 2 Canopies and the soil under the tree canopy (interior soil) were selected as the most informative sites for sampling. The months with the strongest differences were May, June and July, especially June. In the canopy, Araneae, Coleoptera, Diptera, Heteroptera, Hymenoptera, Homoptera, Lepidoptera, Neuroptera and Thysanoptera were the most abundant, and showed significant differences in abundance among orchards with different management regimes. Moreover, in the canopies, Coleoptera and Lepidoptera showed a seasonal pattern of abundance and consistent significant differences between the organic orchard vs. the integrated and conventional ones in both years of study. In the soil, 12 orders showed significant differences in abundance among management regimes at some point of the sampling season. 3 In a search for biological indicators that could help to distinguish between management regimes, a discriminant analysis applied to the data indicated that only the samples from the canopy were classified according to their management regime in a consistent way over time. The groups selected by the analysis to establish differences among management regimes were Coleoptera, Diptera, Heteroptera, Lepidoptera and Thysanoptera. The analysis applied to compare organic vs. non‐organic olive orchards, again identified Coleoptera and Lepidoptera as suitable groups. The results suggest that these two orders are potential bioindicators to distinguish, in a simple way, organic olive orchards from non‐organic ones.  相似文献

The development of resistant crop varieties depends on the reliability of the method of screening for resistance. One of the techniques widely used in evaluating field resistance is the field screening method. However, most cowpea varieties regarded as having field resistance are often found to be susceptible in farmers’ fields, where the inoculum density can be higher. This study rescreened 96 cowpea varieties, evaluated earlier in breeders’ fields as resistant to Xanthomonas campestris pv. vignicola, in field and greenhouse under high inoculum pressure. There were significant differences (P ≤ 0.05) in the reactions of cowpea varieties to bacterial blight in terms of disease incidence and severity. Results from the field screening showed that there were 69 susceptible, 25 moderately susceptible, and 2 resistant varieties. In artificial inoculation in the greenhouse IT81D‐1228‐14, IT82E‐16, IT93K‐2271‐2‐2, TVu 1235, and TVu 4630, which were moderately susceptible in the field showed a susceptible reaction. Tvu 12349 and Tvu 15549 gave consistent reactions in the field and in the greenhouse and are therefore good sources of stable resistance to bacterial blight pathogen. Stem canker incidence also varied significantly (P ≤ 0.05) among the cowpea varieties. Seventeen varieties did not manifest canker symptoms.  相似文献
