为探讨Janus蛋白酪氨酸激酶2-转导及转录激活因子5(JAK2-STAT5)途径在介导血管紧张素Ⅱ(AngⅡ)诱导的血管平滑肌细胞(VSMC)血管舒-缩肽表达调节的作用及分子机制, 以Ang Ⅱ为诱发因素刺激培养的大鼠VSMC, 用免疫共沉淀、Western印迹分析和激光共聚焦显微镜观察STAT5磷酸化及其核转位, 用电泳迁移率改变分析(EMSA)确定STAT5与血管活性肽基因调控区顺式调控元件的结合活性. 结果显示, STAT5磷酸化水平分别于Ang Ⅱ刺激10 min和12 h出现两个高峰, 增加的磷酸化STAT5主要分布在细胞核内. Ang Ⅱ诱导的STAT5活化与核转位可被JAK2的特异抑制剂AG490所抑制. EMSA结果显示, 用Ang Ⅱ刺激VSMC后, 核蛋白与含有血管紧张素原基因启动子STAT5识别序列的探针结合活性显著升高,而核蛋白与含有心钠素(ANF)基因启动子STAT5识别序列的探针结合活性则呈下降趋势, 核蛋白与两种探针的结合活性均可被JAK2抑制剂AG490所消除, 并且加入抗STAT5抗体后均可出现滞后的超迁移带. 结果提示, Ang Ⅱ通过激活JAK2-STAT5介导信号向胞核内传递, STAT5与相应的顺式元件结合是启动血管紧张素原和心钠素基因表达所必需的转录调控机制之一.  相似文献

AP-1在AngⅡ正反馈调节其前体基因表达中的作用   总被引:1,自引:0,他引:1  
血管紧张素Ⅱ(AngⅡ)可诱导其前体基因在血管平滑肌细胞(vascularsmoothmusclecells,VSMC)中进行表达,其作用机制与促进转录激活蛋白-1(activatingprotein-1,AP-1)的基因调控区中存在的AP-1位点结合有关.为进一步明确AngⅡ调节AP-1结合活性的分子机制,用放线菌酮(cycloheximide,CHX)作为c-Jun磷酸化抑制剂,经DNA-蛋白质相互作用和蛋白质印迹实验,探讨AngⅡ对AP-1结合活性的影响并探讨其分子机制.结果表明,受AngⅡ刺激的VSMC,其核蛋白中AP-1的组成亚基之一c-Jun水平明显升高.免疫细胞化学染色显示,在被AngⅡ处理的细胞中,c-Jun主要定位于细胞核,胞浆中几乎检测不出该转录激活蛋白的存在.用丝氨酸磷酸化抗体检测证实,AngⅡ可诱导c-Jun磷酸化.电泳迁移率改变分析(electrophoreticmobilityshiftassay,EMSA)显示,c-Jun的磷酸化水平与AP-1结合血管紧张素原基因顺式元件的活性,和对该基因的转录激活作用呈正相关关系,CHX通过阻断c-Jun磷酸化抑制AngⅡ诱导的AP-1结合活性,但是不影响c-Jun的表达水平.上述结果提示,AP-1的磷酸化活化是AngⅡ正反馈调节其前体基因表达的重要机制之一,首次发现CHX是c-Jun磷酸化的抑制剂.  相似文献

为研究AP-1和STAT5协同调节血管紧张素原基因表达的作用机制,用凝胶电泳迁移率改变分析(EMSA)和染色质免疫沉淀分析(ChIP assay)检测血管紧张素Ⅱ(AngⅡ)诱导的AP-1和STAT5的DNA结合活性;用凝胶超迁移分析和免疫共沉淀方法观察AP-1和STAT5的相互作用. 结果显示,AngⅡ可分别促进AP-1和STAT5与血管紧张素原基因调控区顺式元件的结合以及二者之间的相互作用;核蛋白与含AP-1结合位点的寡核苷酸探针结合形成的复合物可与抗c-Jun抗体和抗STAT5抗体形成超迁移电泳区带;而用抗c-Jun抗体也可从STAT5 染色质免疫沉淀复合物洗脱液中检测到AP-1的存在. 而且,AP-1和STAT5的相互作用程度及其二者与DNA的结合活性与血管紧张素原表达活性具有一致关系,该效应可被JAK2特异抑制剂AG490所抑制. 上述结果提示,与顺式元件结合的AP-1和STAT5通过相互作用而形成四元复合物协同调节血管紧张素原基因的表达,JAK2对该过程具有诱导活化作用.  相似文献

目的:研究血管紧张素Ⅱ(AngⅡ)对血管平滑肌细胞血小板源生长因子(PDGF)受体表达的影响.方法:采用大鼠主动脉球囊内皮剥脱术制备主动脉再狭窄模型,观察形态学变化;放免法测定主动脉AngⅡ含量;免疫印迹法测定主动脉PDGF-β受体含量,并与假手术组相比较.培养大鼠主动脉血管平滑肌细胞(VSMC),AngⅡ刺激正常培养的与洛沙坦预处理过的VSMC 6 h,测定PDGF-β受体含量.结果:球囊内皮剥脱术后14 d,主动脉中层VSMC大量增殖,内膜显著增厚,AngⅡ含量显著升高(P<0.05),PDGF-β受体表达显著增强(P<0.05).AngⅡ诱导VSMC PDGF-β受体表达显著增强(P<0.01),AngⅡ受体拮抗剂洛沙坦完全抑制AngⅡ对PDGF-β受体上调的诱导作用.结论:AngⅡ可通过其Ⅰ型受体诱导血管平滑肌细胞PDGF受体上调,这可能是AngⅡ促VSMC发生增殖的一个重要机制.  相似文献

KLF5(Krüppel-like factor 5)是KLF家族转录因子中的成员,参与多种与增 殖相关基因的转录调节.通过体外培养大鼠血管平滑肌细胞(vascular smooth muscle cells, VSMC),以Ang Ⅱ为增殖诱导因素,研究KLF5介导VSMC增殖的机制. 研究发现,Ang Ⅱ可剂量依赖性诱导VSMC增殖,并伴有KLF5和c-Jun蛋白表达水平的 升高.为了证实KLF5参与Ang Ⅱ诱导的细胞增殖过程,用siRNA敲低内源性KLF5的表达后,发现细胞增殖活力受到明显的抑制;交互免疫沉淀和GST pull down分析结果显示,KLF5与c-Jun在体内和体外存在物理学上的相互作用,并且Ang Ⅱ可诱导二者之间的相互缔合.这些结果表明,KLF5通过与c-Jun相互作用进而介导Ang Ⅱ诱导的VSMC 增殖.  相似文献

为了探讨血管平滑肌细胞 ( VSMC)基质金属蛋白酶 - 2 ( MMP- 2 )基因的表达调控机制 ,利用Northern印迹杂交和 MMP- 2活性酶图分析检查 b FGF、TNF- α和 IL- 1 β对 VSMC MMP- 2基因表达的影响 ,应用电泳迁移率改变实验 ( EMSA)和 CAT分析对其作用机制进行研究 .结果证实 ,3种细胞因子均能显著诱导 MMP- 2基因表达 ,其作用强度依次为 b FGF>TNF-α>IL - 1β.将 MMP-2基因 5′侧翼 - 61 9~ 1 9bp调控序列克隆进携带报告基因的重组质粒 p SV0 - CAT后 ,经转染VSMC及 CAT分析显示 ,在上述 3种细胞因子的作用下 ,该调控序列可激活 cat基因表达 ,三者促进 cat表达的活性与其诱导 VSMC表达 MMP- 2的结果相一致 ;EMSA结果显示 ,被 b FGF和TNF- α刺激的 VSMC中产生与该基因调控区序列特异结合的转录调控因子 .提示细胞因子除可激活 VSMC细胞周期调节基因表达外 ,还可通过诱导 MMP- 2表达而发挥其对细胞外基质代谢的调节作用及参与 VSMC迁移的启动过程 ;细胞因子对 VSMC MMP- 2基因表达的诱导作用是通过促进转录调控因子的合成或活化而实现的 .  相似文献

目的:RNA干扰(RNAi)是一种新的高效特异地阻断基因表达的基因阻断技术.本研究旨在探讨血管紧张素Ⅱ1型受体(AT1R)的短发夹环RNA质粒(pAT1R-shRNA)对大鼠血管平滑肌细胞(VSMC)增殖的影响.方法:构建pAT1R-shRNA质粒,并转染入鼠VSMC中,应用RT-PCR及Western blot检测血管平滑肌细胞AT1R的mRNA和蛋白表达的变化,验证pAT1R-shRNA是否具有RNAi作用;应用台盼蓝染色法和MTT法检测VSMC增殖情况.结果:构建的质粒pAT1R-shRNA经测序鉴定验证与预期相符.转染组AT1R的mRNA和蛋白表达明显减少,与对照组相比有显著性差异(P<0.01);转染质粒同时加AngⅡ刺激的VSMC增生明显受到抑制,与对照组相比有显著性差异(P<0.01).结论:构建的pAT1R-shRNA质粒具有RNAi作用,并对AngⅡ所诱导的VSMC增殖有抑制作用.  相似文献

利用转录因子“诱骗”策略 ,阻断血管平滑肌细胞 (VSMC)表型特异基因和增殖相关基因的反式激活 ,揭示VSMC表型转化和增殖之间的关系 .电泳迁移率改变分析结果表明 ,相当于分化型VSMC特异表达基因共有顺式元件CArG和细胞增殖相关基因共有顺式元件E2F的双股寡核苷酸(ODNs)可分别与从分化型和去分化型VSMC中提取的核蛋白特异性结合 ,形成DNA 蛋白质复合物 .Northern杂交结果显示 ,导入VSMC中的CArGODN可使平滑肌α肌动蛋白 (α actin)表达活性降低 ,肌丝数量减少 ,明显抑制转染细胞的再分化过程 .去分化型VSMC被E2FODN转染后 ,增殖相关基因c myc表达受到抑制 ,细胞增殖速率减慢 ,去分化表型特征减弱 .结果提示 ,E2F和CArG调控元件分别对VSMC增殖和分化起重要调节作用 ,并证实VSMC表型转化与增殖是两个密切相关但不完全相同的细胞事件 .  相似文献

体外培养的分化型血管平滑肌细胞 (vascularsmoothmusclecells ,VSMC)以特异性标志基因表达、长梭形外观及对兴奋剂刺激产生收缩反应为其表型特征 .以血清饥饿法培养处于超汇合 (overconfluence)状态的人VSMC ,观察其分化型标志基因表达活性及其与细胞形态特征和收缩反应性之间的关系 ,探讨细胞生存环境对VSMC基因表达及表型的影响 .研究显示 ,生长至超汇合的VSMC由含血清培养转为血清饥饿后 ,收缩蛋白如SMα肌动蛋白 (SMα actin)、SM2 2α、h1 calponin、肌球蛋白重链 (MHC)SM1和SM2亚型的表达活性明显上调 ,证实血清饥饿诱导的收缩蛋白基因表达和血清应答因子 (serumresponsefactor ,SRF)与CArG顺式元件结合活性的增强有关 .同时 ,血清饥饿还可激活参与VSMC分化调节的转录调控因子SmLIM、Gax和分化相关蛋白HRG 1基因的转录 .随着血清饥饿培养时间的延长 ,VSMC逐渐形成多层、束状、成极性排列的形式 ,对兴奋剂刺激产生的收缩反应明显增强 .结果表明 ,超汇合状态的去分化型VSMC脱离血清刺激后 ,可以再分化成熟并重新获得收缩能力  相似文献

采用Westernblot、氚 胸腺嘧啶 ( 3H TdR)和氚 亮氨酸 ( 3H Leu )掺入等技术和方法 ,用血管紧张素Ⅱ(AngⅡ )和血管紧张素 ( 1 7) [Ang ( 1 7) ]刺激大鼠血管平滑肌细胞 (VSMCs) ,观察和分析Ang ( 1 7)对VSMCs增殖及蛋白激酶C (PKC)和胞外调节蛋白激酶 (ERK)表达的影响。Ang ( 1 7)能明显抑制基础和AngⅡ刺激下的VSMCsPKC ζ和ERK1/ 2蛋白表达 (P <0 0 1或P <0 0 5 ) ,减少3H TdR和3H Leu掺入量 (P <0 0 1或P <0 0 5 )。结果提示 ,Ang ( 1 7)对VSMCs增殖有抑制作用 ,这可能与影响PKC ζ和ERK1/ 2蛋白表达有关。  相似文献

Primary culture of vascular smooth muscle cells (VSMC) from rat aorta was used for the study of the effect of different peptides derived from proadrenomedullin on the expression of adrenomedullin (ADM) gene. ADM and preproADM(22-41) (PAMP) secreted by VSMC were measured by radioimmunoassay, and ADM mRNA in VSMC was determined by quantitative RT-PCR. After the incubation of VSMC in 10(-7)M ADM for 24h, PAMP in the medium and ADM mRNA in the VSMC were decreased by 34 and 41.3%, respectively, and cAMP concentration in the VSMC was increased by 385%. After the incubation of VSMC in 10(-7)M PAMP for 24h, ADM in the medium and ADM mRNA in the VSMC were decreased by 12.2 and 39.1%, respectively, and cAMP concentration in the VSMC was increased by 67%. The decreased ADM mRNA in VSMC induced by the ADM and PAMP treatment was completely reversed by the pre-treatment of the cells in 10(-7)M protein kinase inhibitor for 30 min. After the incubation of VSMC in 10(-7)M preproADM(153-185) (ADT) for 24h, however, ADM in the medium and ADM mRNA in the VSMC were increased by 21 and 35.2%. The increased ADM mRNA in VSMC induced by the ADT treatment was partially blocked by the co-incubation in ADM and ADT, and was totally blocked by the co-incubation in PAMP+ADM and ADT, but was not blocked by the co-incubation in PAMP and ADT. Our results suggest that the four peptides derived from proadrenomedullin may have different effects, possibly through a cAMP-dependent pathway, on the expression of ADM gene.  相似文献

The newly formed corpus luteum (CL) develops rapidly and has the features of active vascularization and mitosis of steroidogenic cells. Such local mechanisms must be strictly regulated by the complex relationship between angiogenic growth factors and vasoactive peptides such as angiotensin (Ang) II, atrial natriuretic peptide (ANP), and endothelin (ET)-1. Thus, the objective of the present study was to determine 1) the changes in vasoactive peptides and progesterone (P) concentrations within the developing CL, along with the changes in concentration in ovarian venous plasma (OVP) and jugular venous plasma (JVP) in the cow, 2) the effects of CL exposure to vasoactive peptides on Ang II and P secretion, and 3) the expression of mRNA for ANP type C receptor in the bovine CL and endothelial cells (ETC) from bovine developing CL. A microdialysis system (MDS) was surgically implanted into multiple CL of six cows on Day 3 after a GnRH injection that induced superovulation, and a catheter was simultaneously inserted into the ovarian vein. The Ang II concentration in OVP was higher than that in JVP throughout the experiment, while the intraluteal release of Ang II was stable. During the experimental period, the concentrations of other vasoactive peptides (ANP and ET-1) showed no clear changes in plasma and were below detectable levels in the MDS perfusate. Exposure of CL to Ang II using the MDS stimulated P release, while exposure to ANP enhanced Ang II release within the developing CL. However, ET-1 had no effect on either P or Ang II release. The expression of mRNA for ANP type C receptor was mainly observed in early CL and ETC. The results suggest that the ET-Ang-ANP system in the preovulatory follicle switches to an Ang-ANP system to enhance both the angiogenesis and steroidogenesis that are actively occurring in developing CL.  相似文献

We investigated whether vascular smooth muscle cells (VSMC)-derived from human produce angiotensin (Ang) II upon change from the contractile phenotype to the synthetic phenotype by incubation with fibronectin (FN). Expression of alpha-smooth muscle (SM) actin, apparent in the contractile phenotype, was decreased by FN. Expressions of matrix Gla and osteopontin, apparent in the synthetic phenotype, were increased by FN. Ang II measured by radioimmunoassay (RIA) was significantly increased in human VSMC by FN. Expression of mRNAs for Ang II-generating proteases cathepsin D, cathepsin G, ACE, and chymase was increased by FN. Expressions of cathepsin D and cathepsin G proteins were also increased by FN. Ang I-generating activity, which was inhibited by an aspartyl protease inhibitor pepstatin A, was readily detected in the conditioned medium from human VSMC. Antisense oligodeoxynucleotides (ODNs) that hybridize with cathepsin D and cathepsin G significantly inhibited FN-increased Ang II in conditioned medium and cell extracts. In VSMC conditioned medium, FN-induced elevation of Ang II was significantly inhibited by temocapril but not by chymostatin. Ang II type 1 receptor antagonist CV11974 completely, and antisense cathepsin D and cathepsin G ODNs partially inhibited the FN-stimulated growth of human VSMC. These results indicate that the change of homogeneous cultures of human VSMC from the contractile to the synthetic phenotype sequentially increases expression of proteases cathepsin D, cathepsin G, and ACE, production of Ang II and productions of growth factors, culminating in VSMC proliferation. These findings implicate a new mechanism for the pathogenesis of human vascular proliferative diseases.  相似文献

观察血管紧张素Ⅱ(AngⅡ)对心肌肌浆网Ca2+,Mg2+-ATPase基因(SERCA2a)转录调节的影响,评价DMP811对此效应的干预作用.6周龄雄性SD大鼠随机分为3组,每组6只.组1:生理盐水输注;组2:AngⅡ输注+DMP811管饲(3mg·d-1·kg-1);组3:AngⅡ输注(200ng·min-1·kg-1.1周后称其体重,取心脏并称重,提取心脏总RNA后采用Northernblot的方法检测SER-CA2a的转录水平,采用RT-PCR检测AngⅡ1型受体(AT1)mRNA水平.实验后,组3心重(CW)、心重/体重(C/B)、AT1受体转录水平均高于组1(分别增加4.7±0.4%,4.9±0.9%和24.7±3.5%;P<0.01),而SERCA2a基因转录水平显著低于组1(降低20.1±3.0%,P<0.01),并且SERCA2amRNA水平与AT1受体mRNA水平呈负相关(r=-0.74,P<0.01).AngⅡ导致的上述改变能被DMP811完全阻断.AngⅡ通过其Ⅰ型受体的介导,诱导了SERCA2a的转录下调  相似文献

Specific binding site for atrial natriuretic factor (ANF), a potent natriuretic and vasorelaxant polypeptide recently isolated from mammalian atria, was studied in cultured vascular smooth muscle cells (VSMC) of the rat aorta. Binding studies of 125I-labeled-synthetic alpha-human natriuretic peptide (alpha-hANP) revealed the presence of a non-interacting, single class of high affinity binding sites for alpha-hANP on VSMC in culture: the apparent dissociation constant (Kd) was approximately 1-2 X 10(-9)M and the number of maximal binding sites was approximately 200,000-300,000 sites/cell. A variety of vasoactive substances and other polypeptide hormones did not affect the binding of 125I-labeled-alpha-hANP to its binding sites. alpha-hANP significantly increased the concentrations of intracellular cyclic GMP in VSMC in a dose-dependent manner (3.2 X 10(-9)-1.6 X 10(-7)M). These data indicate that the specific receptor for ANF is present in VSMC and suggest that intracellular cyclic GMP may be involved in its vasorelaxant effect.  相似文献

Endothelin-1 (ET1) is a vasoactive peptide that stimulates hypertrophy of vascular smooth muscle cells (VSMC) through diverse signaling pathways mediated by G(q)/G(i)/G(13) heterotrimeric G proteins. We have found that ET1 stimulates the activity of cAMP-dependent protein kinase (PKA) in VSMC as profoundly as the G(s)-linked beta-adrenergic agonist, isoproterenol (ISO), but in a transient manner. PKA activation by ET1 was mediated by type-A ET1 receptors (ETA) and recruited an autocrine signaling mechanism distinct from that of ISO, involving G(i)-coupled betagamma subunits of heterotrimeric G proteins, extracellular signal-regulated kinases ERK1/2, cyclooxygenase COX-1 (but not COX-2) and prostacyclin receptors. In the functional studies, inhibition of PKA or COX-1 attenuated ET1-induced VSMC hypertrophy, suggesting the positive role of PKA in this response to ET1. Furthermore, we found that ET1 stimulates a Gbetagamma-mediated, PKA-dependent phosphorylation and inactivation of glycogen synthase kinase-3 (GSK3), an enzyme that regulates cell growth. Together, this study describes that (i) PKA can be transiently activated by G(i)-coupled agonists such as ET1 by an autocrine mechanism involving Gbetagamma/calcium/ERK/COX-1/prostacyclin signaling, and (ii) this PKA activation promotes VSMC hypertrophy, at least in part, through PKA-dependent phosphorylation and inhibition of GSK3.  相似文献

利用体外培养的心肌细胞,观察血管紧张素Ⅱ(AngⅡ)和蛋白激酶C(PKC)在诱导AngⅡ1型受体(AT1)基因表达及蛋白质代谢中的作用.研究结果表明:AngⅡ可诱导AT1mRNA水平一过性下调,呈时间及剂量依赖性,10nmol/LAngⅡ刺激细胞6h,引起AT1mRNA水平降低幅度最大,降至对照的51.6%±9.5%,然后逐渐回升,24h恢复至对照水平.30μmol/LH-7(PKC抑制剂)能阻断AngⅡ诱导的AT1mRNA水平的下调.0.3μmol/L的PMA(PKC激活剂)单独应用可诱导AT1mRNA水平下调达对照的43%±8%,加入AT1拮抗剂DMP811及Dup753均可阻断AngⅡ诱导的AT1mRNA水平的下调.10nmol/L的AngⅡ刺激心肌细胞96h可使蛋白含量降低至对照的73.4%±5.6%,而加药持续刺激144h可使蛋白含量较对照增加33.8%±6.3%,H-7不能阻断AngⅡ诱导的蛋白含量降低,但可有效地抑制蛋白含量的增加.以上结果提示:AngⅡ对心肌细胞AT1基因的转录和细胞的蛋白代谢有调节作用,而PKC则参与了AngⅡ的这种调节作用  相似文献

目的探讨RNA干扰血管生成素样蛋白7 (Angptl7)基因对血管紧张素Ⅱ(AngⅡ)诱导的血管平滑肌细胞(VSMC)炎症因子的影响及其作用机制。 方法体外培养人VSMC,分为常规F12K培养基培养(对照)和1 μg/mL AngII培养24 h。VSMC用AngⅡ(1 μg/mL)处理24 h后,采用siRNA-Angptl7和阴性对照siRNA-NC在Lipofectamine 2000介导下转染VSMC。RT-qPCR检测mRNA表达水平;Griess反应测定一氧化氮(NO)含量;蛋白免疫印记法检测相关蛋白的改变;酶联免疫吸附法检测VSMC中炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素-1β (IL-1β)和IL-6水平。多组间比较采用单因素方差分析,组间两两比较采用LSD-t检验,两组间比较采用独立样本t检验。 结果与对照比较,1 μg/mL AngⅡ处理可促进VSMC中Angptl7 mRNA (0.97±0.06比3.05±0.21)和蛋白表达(1.01±0.12比1.61±0.14),亦可促进VSMC中IL-1β[(45.21±8.10)比(126.17±11.77) pg/mL]、IL-6[(50.50±7.51)比(108.50±9.51)pg/mL]和TNF-α的表达[(60.77±9.58)比(185.67±17.35)pg/mL],差异有统计学意义(P均< 0.01)。与对照和转染siRNA-NC相比,转染siRNA-Angptl7下调Angptl7蛋白表达(0.99±0.12,0.98±0.12比0.44±0.14,P < 0.01)。与AngⅡ干预组相比,siRNA-Angptl7降低AngⅡ介导的VSMC炎症反应相关蛋白TNF-α、IL-6和IL-1β的表达,核因子κB (NF-κB)/诱导型一氧化氮合酶(iNOS)/环氧化酶2 (COX-2)信号通路相关蛋白NF-κB、iNOS和COX-2表达及NO含量亦降低,差异有统计学意义(P均< 0.01)。与siRNA-NC相比,siRNA-Angptl7组AngⅡ诱导的VSMC炎症反应相关蛋白TNF-α (0.99±0.13比0.51±0.12)、IL-6 (1.00±0.12比0.38±0.05)和IL-1β的表达(0.99±0.14比0.48±0.11),NF-κB (1.00±0.10比0.42±0.08)、iNOS (1.02±0.12比0.42±0.10)和COX-2表达(1.00±0.11比0.52±0.12)均降低,NO含量[(54.78±2.76)比(18.08±3.61)μmol/L]亦降低,差异有统计学意义(P均< 0.01)。 结论AngⅡ可通过Angptl7促进VSMC炎症反应,下调Angptl7蛋白表达可以抑制VSMC的炎症反应,其作用机制可能与抑制NF-κB/iNOS-COX-2信号通路有关。  相似文献

目的:研究肾上腺髓质素2(ADM2)拮抗血管紧张素Ⅱ(AngⅡ)发挥舒张血管的作用及机制。方法:将18只180~200 g雄性SD大鼠随机分为3组(n=6):对照组、AngⅡ(150 ng/(kg·min))组和AngⅡ(150 ng/(kg·min))+ADM2(500 ng/(kg·h))组,采用皮下埋植微量渗透泵的方法给药。2周后颈动脉插管法测量大鼠血压,测定血浆一氧化氮(NO)含量和内皮型一氧化氮合酶(eNOS)活性。DHE染色法检测大鼠动脉壁活性氧产生。制备大鼠离体血管环,观察ADM2的舒血管作用。培养人脐静脉内皮细胞系EA.hy 926,用DCFH-DA荧光探针检测AngⅡ和ADM2对血管内皮细胞活性氧释放的影响。结果:与AngⅡ组相比,ADM2显著降低了大鼠血压,血浆中eNOS活性提高、NO含量增加,血管壁活性氧产生减少。ADM2呈浓度依赖性和内皮依赖性舒张血管环,并明显抑制了AngⅡ引起的血管内皮细胞活性氧产生。结论:ADM2可能通过拮抗AngⅡ诱导的血管内皮氧化应激效应,改善内皮功能,发挥舒张血管、降低血压的作用。  相似文献
