利用从126个RAPD(RandomAmplifiedPolymorphicDNAs)随机引物中筛选到的可扩增出清晰条带、主带明显、稳定的8条引物,对采集自江苏、安徽和江西不同寄主的45个Phytophthoraboehmeriae菌株进行全基因组DNARAPD标记遗传多样性分析。选用引物共标出DNA指纹图带68条,其中多态性条带20条,多态性检测率为29.4%,表明该种内不同地区和寄主来源的菌株间变异较小。利用Popgene软件计算供试菌株间的遗传距离并绘制聚类树状图,供试菌株被划分为2个遗传聚类组。菌株间的遗传相似性与菌株的寄主来源有一定的相关性,来自江苏、江西和安徽棉花上的27个菌株被划分在同一遗传聚类组内,而分离自构树、枫杨和苎麻的18个菌株被划分在另一个遗传聚类组。结果还表明菌株间遗传相似性与其地区来源无直接相关性。  相似文献

Genetic diversity of Histoplasma capsulatum strains in Brazil   总被引:1,自引:0,他引:1  
This study establishes the genetic relatedness among Brazilian Histoplasma capsulatum samples obtained from different sources. A PCR-based random amplified polymorphic DNA (RAPD) assay was used to delineate polymorphisms among isolates in geographically diverse regions in Brazil. RAPD fingerprints revealed distinct DNA profiles and provided a high level of discrimination among H. capsulatum strains from different locations. Cluster I was composed of H. capsulatum isolates from the northeast region. The majority of strains from southeast and south were categorized as major cluster II. The strain 84564 from Rio de Janeiro State showed no genetic correlation to any of the isolates from the same state. The RAPD patterns of H. capsulatum isolates from Goias (Cluster III) were unrelated to DNA fingerprints observed among the other H. capsulatum strains (48% similarity). This study is the first report that stratifies the clusters of H. capsulatum strains from Brazil by molecular typing and associates them with the geographical origin.  相似文献

采用RAPD方法对来源于栗疫病菌8个不同子囊壳的子囊孢子后代和无性生殖的对照群体各23个菌株进行了群体结构的比较。从RAPD随机引物中筛选出扩增多态性丰富的4条引物,共扩增出条带73条,多态性检测率为100%。研究结果表明,在8个子囊壳和无性生殖群体中的基因多样性,64.27%由群体内部引起,只有35.73%的多样性由群体之间的基因差异引起。各子囊壳群体间存在的基因流动很小(Nm=0.8994)。有性群体和无性群体之间的遗传距离为0.1389,基因流动值为3.4212,说明子囊壳群体和无性生殖群体之间存在一定的系统关系。分析表明栗疫病菌子囊孢子后代在自然界的传播对自然界的病菌的多样性起重要的作用。  相似文献

用筛选出的12个随机引物,对链格孢属Alternaria13个小孢子种和作为对照的3个大孢子种共55个分离系(isolates)进行RAPD分析。大孢子种Alternariasolani、A.porri和形态独特的A.leucanthemi在树状聚类图上遗传距离0.44处与所有供试小孢子种区分开,它们彼此之间在遗传距离0.25处相区分,表明所采用的RAPD分析方法适于链格孢种间亲缘关系的研究。所有供试的链格孢小孢子种在遗传距离0.31水平上被聚在一起,表明它们之间的亲缘关系较近。A.infectoria与其它链格孢小孢子种之间遗传距离较远;A.longipes的3个分离系和A.brassicicola的7个分离系在较低的遗传距离上被聚在一起,表明它们是独立的种。其它供试链格孢小孢子种的不同分离系在树状聚类图上未显示明确区分。  相似文献

Random amplified polymorphic DNA (RAPD) was used for identification and assessment of genetic diversity between isolates of Streptomyces from soil. Genomic DNA from 18 Streptomyces isolates and 2 reference strains were amplified using four different 10-mer primers. Different DNA fingerprinting patterns were obtained for all the isolates. Electrophoretic and cluster analysis of the amplification products revealed incidence of polymorphism among the isolates and none of them was identical to the reference strains although there were some common amplification bands. Two highly divergent groups were determined among the isolates. The results indicate that RAPD is an efficient method for discriminating and studying genetic diversity of Streptomyces isolates.  相似文献

[目的]建立鼠李糖乳杆菌(Lactobacillus rhamnosus,Lr)菌株之间的分子鉴别方法并分析不同分离株之间的遗传多样性.[方法]从56份采集自中国新疆和田和广西巴马瑶族自治县的长寿老人粪便样本中分离得到的乳酸菌中,经生理生化分析和API 50CHL试验条鉴定,获得10株Lr.对10株Lr分离株和1株Lr标准株ATCC7469进行了随机扩增多态DNA分析,从50条随机引物中筛选到5条在菌株水平上具有鉴别力的引物P14、OPG28、OPG25、P7和P4并建立和优化了Lr菌株RAPD指纹图谱扩增方法.根据RAPD结果计算菌株间的遗传相似系数并进行聚类分析.[结果]获得了清晰稳定的DNA指纹图谱,扩增产物大小在100~2000bp之间,菌株间呈现显著的DNA多态性,不同来源的Lr分离株的遗传相似系数在0.581~0.935之间,在相似系数0.80水平上可以将11株Lr菌株分为5个类群,其中分离自新疆和田的Lr菌株归在类群B和类群C,而分离自广西巴马瑶族自治县的Lr菌株归在类群D和类群E.[结论]应用RAPD方法对Lr菌株进行分子鉴别是可行的,不同来源的Lr之间存在着较大的种内遗传多态性和不同的亲缘关系.  相似文献

Genetic relationships among 132 strains of Vibrio vulnificus (clinical, environmental, and diseased-eel isolates from different geographic origins, as well as seawater and shellfish isolates from the western Mediterranean coast, including reference strains) were analyzed by random amplified polymorphic DNA (RAPD) PCR. Results were validated by ribotyping. For ribotyping, DNAs were digested with KpnI and hybridized with an oligonucleotide probe complementary to a highly conserved sequence in the 23S rRNA gene. Random amplification of DNA was performed with M13 and T3 universal primers. The comparison between ribotyping and RAPD PCR revealed an overall agreement regarding the high level of homogeneity of diseased-eel isolates in contrast to the genetic heterogeneity of Mediterranean isolates. The latter suggests the existence of autochthonous clones present in Mediterranean coastal waters. Both techniques have revealed a genetic proximity among Spanish fish farm isolates and a close relationship between four Spanish eel farm isolates and some Mediterranean isolates. Whereas the differentiation within diseased-eel isolates was only possible by ribotyping, RAPD PCR was able to differentiate phenotypically atypical isolates of V. vulnificus. On the basis of our results, RAPD PCR is proposed as a better technique than ribotyping for rapid typing in the routine analysis of new V. vulnificus isolates.  相似文献

利用12个随机引物对来自云南省不同地理环境的立枯丝核菌RhizoctoniasolaniKühn第一融合群(AG-1)的13个菌株进行遗传分化关系的研究。结果表明受试菌株被标记的DNA谱带多态性检测率为100%,即受试菌株间几乎无同质的DNA谱带。利用UPGMA法构建分子系统树分析发现,遗传距离0.5将其划分为9个遗传聚类组,遗传距离0.86将其划分为6个遗传聚类组,而遗传距离0.95可将其划分为3个遗传聚类组。结果表明受试AG1融合群的13个菌株具明显的遗传分化。  相似文献

RAPD fingerprinting was used for strain identification and the assessment of genetic diversity within a field population of Bradyrhizobium japonicum . Total genomic DNAs from 13 field isolates and two inoculant strains were amplified using six different 10-mer primers. Different and informative band patterns were obtained for all strains analysed. Cluster analysis unexpectedly revealed that none of the field isolates was identical to inoculant strains which were regularly used for soybean inoculation. Among field isolates two highly divergent groups were determined. The results indicate that RAPD is a very discriminative and efficient method for differentiating and studying genetic diversity of B. japonicum strains.  相似文献

应用体细胞不亲和性反应、交配型因子分析和基因组DNA的RAPD分析,研究了一个分布于方圆约1km的6根倒木上的18个香菇野生菌株间的遗传差异。结果表明,该群体大多数菌株间配对(80.4%)体细胞不亲和,而同一倒木菌株间配对的体细胞亲和率达 62.5%。不同倒木菌株间未发现体细胞亲和的配对。该群体存在 11个特异的A因子和7个特异的B因子。同一倒木的菌株有的交配型因子相同,有的则不同。不同倒木的菌株大多数交配型因子不同,未发现交配型因子完全相同的菌株。RAPD分析显示,体细胞亲和的菌株,交配型因子完全相同的菌株,在基于DNA相似系数的遗传相关聚类中,首先聚为小类。总起来看,在自然群体中,香菇个体间的遗传差异与其空间分布之间存在一定的联系,随着空间距离的增大,菌株间的异质性相应增高。  相似文献

Serologic and genetic typing with RAPD (Random Amplified Polymorphic DNA) method was used for epidemiologic analysis of GBS. 125 strains isolated from various clinical samples from adult patients were tested. In serologic typing seven serotypes have been found. Serotypes III and R were the most often encountered, containing 37,6% and 20,8% of samples. There was no dependence between serologic type and disease process. Optimalisation of RAPD reaction parameters was based on the standard strains of GBS. In the group of strains tested with the use of RAPD method, eleven genetic profiles were found, with prevalence of profile B (25,8%). Five other profiles occured with similar frequency (8,8% - 12,8%). Among streptococci isolated from patients with the infection of genitourinary tract, great differentiation in the genetic profiles of the strains has been found. Each serologic type contained various genetic profiles. Genetic variety showed by RAPD method indicates the raised ability of this technique to find differences among isolates of GBS.  相似文献

从四个不同银耳段木栽培场地采集杂菌样本,经初步鉴定后纯化得到48个菌株。通过对子实体、孢子及菌丝等的形态观察确认从银耳栽培段木上分离的48株杂菌隶属于炭团菌属,在形态上与截头炭团菌Annulohypoxylon annulatum最为相似。为了进一步确认从银耳栽培段木上分离的48株杂菌的分类地位,对其中14个供试菌株进行了ITS系统发育分析,结果表明:14个供试菌株之间的遗传距离为0.036时可以形成两个明显分枝,且分枝与地理来源之间并无明显相关性;供试菌株在分类地位上属于炭团菌属,与形态学鉴定结果一致,并与截头炭团菌的关系最为接近,遗传距离在0.2左右;与银耳伴生菌香灰菌的遗传距离为0.29左右。从75条引物中筛选出13条引物对所有48个供试菌株进行ISSR扩增,扩增共获得103条带,其中多态性条带72条,占条带总数的69.9%。聚类分析结果表明:在相似水平为75%时,48个菌株可以划分为5个类群,且类群的划分与地理来源之间并无明显相关性;多数供试菌株之间的遗传相似性较低,这表明供试菌株在DNA水平上存在比较显著的遗传变异,具有较丰富的遗传多样性。  相似文献

基于菌落和菌体的形态特征以及ITS序列分析结果,将分离自中国海南省海口市东寨港红树林保护区红海榄根际土壤的菌株F7鉴定为曲霉属黄绿组的一种真菌。在确定和优化菌株F7培养基的基础上,初步确定了适合其分泌活性代谢产物的改良查氏培养基组成:3%乳糖,1%蛋白胨,0.3%NaNO3,0.05%KCl,0.05%MgSO4·7H2O,0.001%FeSO4,0.1%K2HPO4,pH7.0。将菌株F7接种于该培养基中,28℃下160r/min振荡培养7d后收获发酵液,经乙酸乙酯和正丁醇萃取后获得了乙酸乙酯萃取物(EAE)、正丁醇萃取物(BE)以及水萃取物(WSE),其中EAE和BE对金黄色葡萄球菌、藤黄八叠球菌和枯草芽孢杆菌的生长显示出明显的抑制活性,最低抑菌浓度(MIC)介于0.625-5.0mg/mL之间。同时,上述3种萃取物对人肝癌细胞株HepG2的增殖也表现出一定的抑制活性,IC50约为120μg/mL左右。  相似文献

The randomly amplified polymorphic DNA (RAPD) method was used to investigate the genetic diversity in Xanthomonas cynarae, which causes bacterial bract spot disease of artichoke. This RAPD analysis was also intended to identify molecular markers characteristic of this species, in order to develop PCR-based markers which can be used to detect this pathogenic bacterium in artichoke fields. Among the 340 RAPD primers tested, 40 were selected on their ability to produce reproducible and reliable fingerprints in our genetic background. These 40 primers produced almost similar patterns for the 37 X. cynarae strains studied, different from the fingerprints obtained for other Xanthomonas species and other xanthomonad-like bacteria isolated from artichoke leaves. Therefore, X. cynarae strains form a homogeneous genetic group. However, a little DNA polymorphism within this species was observed and the collection of X. cynarae isolates was divided into two groups (one containing three strains, the second one including all other strains). Out of seven RAPD markers characteristic of X. cynarae that were cloned, four did not hybridize to the genomic DNA of strains belonging to other Xanthomonas species. These four RAPD markers were converted into PCR markers (specific characterized amplified regions [SCARs]); they were sequenced, and a PCR primer pair was designed for each of them. Three derived SCARs are good candidates to develop PCR-based tests to detect X. cynarae in artichoke fields.  相似文献

ABSTRACT: BACKGROUND: Haemophilus parasuis is the causative agent of Glasser's disease and is a pathogen of swine in high-health status herds. Reports on serotyping of field strains from outbreaks describe that approximately 30% of them are nontypeable and therefore cannot be traced. Molecular typing methods have been used as alternatives to serotyping. This study was done to compare random amplified polymorphic DNA (RAPD) profiles and whole cell protein (WCP) lysate profiles as methods for distinguishing H. parasuis reference strains and field isolates. RESULTS: The DNA and WCP lysate profiles of 15 reference strains and 31 field isolates of H. parasuis were analyzed using the Dice and neighbor joining algorithms. The results revealed unique and reproducible DNA and protein profiles among the reference strains and field isolates studied. Simpson's index of diversity showed significant discrimination between isolates when three 10mer primers were combined for the RAPD method and also when both the RAPD and WCP lysate typing methods were combined. CONCLUSIONS: The RAPD profiles seen among the reference strains and field isolates did not appear to change over time which may reflect a lack of DNA mutations in the genes of the samples. The recent field isolates had different WCP lysate profiles than the reference strains, possibly because the number of passages of the type strains may affect their protein expression.  相似文献

采用酯酶同工酶及RAPD技术对香菇3个亲本双核体(苏香、野生46#、野生80#)及其10个单核体、以及它们的10个杂交后代进行了遗传差异和亲缘关系的研究,同时针对亲本单核体酯酶同工酶标记和RAPD标记遗传距离、以及杂交子和亲本单核体RAPD标记遗传距离与香菇产量超亲优势进行了相关性分析。结果表明,酯酶同工酶和RAPD技术都可进行香菇杂种优势群的划分研究,但RAPD标记检测的多态性要远远高于酯酶同工酶标记。相关分析结果表明,亲本单核体酯酶同工酶标记遗传距离与香菇产量超亲优势无相关性,而RAPD标记遗传距离与其存在极显著正相关;杂交子和以苏香为来源的单核体亲本之间RAPD标记遗传距离与香菇产量超亲优势也存在极显著正相关,而杂交子和以野生46#、野生80#为来源的单核体亲本之间RAPD标记遗传距离与其相关不显著。  相似文献
