不同生殖期鳜肝脏超微结构变化的观察   总被引:16,自引:0,他引:16  
应用透射电镜对生殖季节与非生殖季节鳜肝脏超微结构的变化进行了观察。鳜肝细胞含有单个卵圆形的核,核仁清楚;细胞质内含有粗面内质网、线粒体、糖原颗粒和脂滴等细胞器和内含物。胆小管由相邻的数个肝细胞质膜凹陷围成,而肝血窦则由内皮细胞的胞质构成。还发现了贮脂细胞、枯否氏细胞和成纤维细胞。胆小管腔和窦周隙内浸润许多由肝细胞发出的微绒毛结构。鳜肝细胞的超微结构在产卵前后呈现明显变化:产卵前的肝细胞内富含线粒体、糖原颗粒和脂滴,粗面内质网发达;而产卵后的肝细胞内核仁发生迁移,部分细胞核囊泡化,糖原颗粒和脂滴排空,少数肝细胞具双核结构。非生殖期多数肝细胞核含有双核仁结构,胞质内溶酶体数量增多。  相似文献

应用光镜和透射电镜对繁殖期间唐鱼Tanichthys albonubes肝脏组织的显微和超微结构进行了观察。结果显示,唐鱼肝细胞具单核,中央核仁显著;细胞质内分布着粗面内质网、线粒体、糖原颗粒和脂滴等细胞器和内含物。胆小管由2~3个相邻肝细胞质膜凹陷围成,而肝血窦则由内皮细胞的胞质、成纤维细胞等参与构成。肝细胞与周边细胞通过3种不同方式进行联系:肝细胞之间的紧密连接;与血窦的间接连接;与胆小管的邻接。这些联系方式显示了肝脏具有内分泌腺和外分泌腺功能的特点。研究还发现雌性唐鱼肝脏具有"暗"细胞和"淡"细胞两种类型。本文还讨论了唐鱼肝脏与其他硬骨鱼类肝脏一般组织结构和超微结构的异同点。  相似文献

The ultrastructure of rat liver cells after running exercise was investigated. When rats were trained for a month and sacrificed immediately after the last exercise it was revealed that the number of liver cells mitochondria increased, but many of them had alterations: mitochondria became swollen, had lucid matrix. There were some variations in degree of alterations between different mitochondria: a) in the same hepatocyte, b) in different hepatocytes of the same animal, that was connected with individual sensitivity of organelles on the levels of the cell and of the organ. Rough endoplasmic reticulum bore few ribosomes. Glycogen was absent. There were abundant vesicles of smooth endoplasmic reticulum, autophagic vacuoles and peroxisomes in the liver cell cytoplasm. Adaptation of rat liver to the exercise programme becomes evident by 1.5 month of exercise. Mitochondria and rough endoplasmic reticulum were numerous and of normal structure. There were many peroxisomes and glycogen granules in the cytoplasm of hepatocyte. The presence of large autophagic vacuoles in the cytoplasm of some hepatocytes were obviously connected with more rapid destruction of some organelles, than in control.  相似文献

The ultrastructure of liver cells was studied in rooks (Corvus frugilegus) living in radioactive and chemical contamination areas. The ultrastructure of liver cells from rook as well as jackdaw (Corvus monedula) and hooded crow (Corvus cornix) (Corvidae family) from a conventionally clean area was studied as control. Control hepatocytes proved to contain a great number of mitochondria, many of which were swollen and had clear matrix and disorganized cristae. The cristae nearly lacked glycogen and had abundant lipid droplets, which often tightly contacted mitochondria. The cytoplasm of hepatocytes in birds from both ecologically unfavorable areas had numerous mitochondria with the same ultrastructure. In contrast to control, the hepatocyte cytoplasm: (1) contained a lot of glycogen; (2) there were many lipid droplets, which directly contacted glycogen granules; and (3) had more abundant peroxisomes. In addition to normal erythrocytes, the sinusoids contained erythrocytes with mitochondria, vesicles, and lipid droplets in their cytoplasm. Analysis of many micrographs of lipid droplets contacting glycogen granules, mitochondria, peroxisomes, and cisterns of smooth endoplasmic reticulum allowed us to propose that glycogen is synthesized via gluconeogenesis from glycerol and products of fatty acid oxidation in the liver cell cytoplasm of rooks from ecologically unfavorable areas as distinct from control.  相似文献

Yearling coho salmon were fed either a commercial trout (TC) diet or a diet consisting of homogenized adult Pacific Ocean coho salmon (PS), supplemented with vitamins and minerals. The hepatosomatic index of the TC-fed fish killed 12 h after the last feed or after 4 or 8 weeks of fasting were significantly larger ( P 0.05) than comparable PS-fed groups.
'Light' and'dark' hepatocyte cell types were evident in all groups although the dark cells were less numerous in the fasted fish. Hepatocytes of fasted fish contained markedly more endoplasmic reticulum than fed fish. Hepatocytic glycogen content of the TC-fed fish was higher than in PS-fed fish. Whereas in the TC-fed group hepatic glycogen was still evident in fish fasted for 8 weeks, the hepatocytes of the fasted PS-fed group contained little or no glycogen. The suitability of commercial fish diets for salmonids is discussed.  相似文献

The ultrastructure of rat liver cells was studied during a 6 months' furacilline (5-nitro-2-furtfurilsemicarbazon) feeding in dose of 40 mg per day, and also during the next 8 months after the treatment cessation. An irregular swelling of membranous structure in addition to disorganization and partial reduction of the granular endoplasmic reticulum were found in the hepatocyte cytoplasm after the prolonged furacilline feeding as well as glycogen depletion and the tendency of the agranular endoplasmic reticulum to enlargement. These changes rarely reached the essential intensity with the transition to necrobiosis. They disappeared already a month after the cessation of furacilline treatment. In all the terms of experiment, the nucleoli were hypertrophied and retained their loose nucleoloneme structure. No sings of furacilline carcinogenic activity were found in rat liver during the 14 months of investigation.  相似文献

Histochemical and ultrastructural aspects of adipocyte differentiation in subcutaneous tissue of fetal pigs were analyzed in a longitudinal study. A matrix of collagen fibers surrounding adipocytes developed after the establishment of a distinct and continuous PAS-positive basement membrane. The degree of plasma membrane invagination and specialization was positively correlated with the extent of basement membrane and collagen matrix formation. Close spatial relationships between narrow, smooth endoplasmic reticulum, plasma membrane invaginations, the surface of lipid droplets and mitochondria were observed in differentiating adipocytes. Histochemical and ultrastructural criteria for the identification of preadipocytes are: (1) perivascular location; (2) mitochondria localized in the Golgi zone; (3) cytosolic glycogen; (4) rough endoplasmic reticulum with cisternae uniformly and approximately 600 A wide; (5) free ribosomes and few polysomes, and (6) lipid droplets encased by microfilaments. These criteria permitted clear distinction from obvious fibroblasts and macrophages. Other stromal cells were morphologically abnormal. Occasionally, adipocytes and perivascular cells exhibited close intercellular contacts that were morphologically distinct from intercellular contacts between contiguous endothelial cells.  相似文献

The simple, high columnar gallbladder epithclia of the rainbow trout (Salmo gairdneri, Rich.) and the tench (Tinca linca, L.) are composed of light and dark cells. Within the dark cells two types must be distinguished: less hydrated, but normal epithelial cells and degenerating cells. The latter seem to be eliminated from the epithelium through phagocytosis by monocytes and macrophages, which possibly correspond with the ‘basal regenerative cells’ of earlier investigations. Since they are mainly found near the basement membrane, an hypothesis is put forth that they do not enter the gallbladder lumen, but migrate back into the subepithelial connective tissue after phagocytosis. Light and less hydrated dark cells are specialized for absorption of water and dissolved bile salts and for secretion of mucus, which mainly consists of acid glycoproteins. Interspecific differences are seen with regard to both functions. So, in the tench only single microvilli arc developed apically, whereas a true brush border is seen in the rainbow trout. Thus, a higher absorptive rate can be assumed in the rainbow trout. Or, with respect to mucus secretion, in the rainbow trout the granular endoplasmic reticulum predominates, whereas in the tench the smooth endoplasmic reticulum, often associated with glycogen and even forming ‘glycogen bodies’, is better developed. In the rainbow trout fibrillar granules of highly condensed acid glycoproteins are seen to be secreted. In the tench only vesicles of flocculent content occur. Perhaps, these morphological and subsequent metabolic differences may be related to the various feeding habits of the predatory rainbow trout and the omnivorous tench. Seasonal variations as expected especially in the tench after winter rest could not be established.  相似文献

Summary The administration of chorionic gonadotrophin to prepuberal mice results in precocious maturation of the testicular interstitial cells. The cytoplasm of the nine-day-old cells is characterized by abundant lipid droplets, large numbers of glycogen particles and mitochondria. By contrast, the membranous organelles are poorly developed.Human chorionic gonadotrophin brings about mobilization of lipid droplets and glycogen particles, and differentiation of large areas of agranular endoplasmic reticulum.The present observations are in agreement with the reports that human chorionic gonadotrophin increases the secretion of testosterone and that the agranular endoplasmic reticulum is the site of storage of steroid and of the enzymes involved in the biosynthesis of androgens.  相似文献

The effect of ethanol on hepatocytes from pregnant minipigs and their half-term fetuses was studied with the aid of morphometric methods. In the pregnant minipigs the hepatocytes of the ethanol-treated animals showed a significant increase in the volume density of mitochondria, autophagic vacuoles, Golgi complexes and smooth endoplasmic reticulum, and a significant decrease of glycogen. In the half-term fetuses the hepatocytes of ethanol-exposed animals showed no significant change in the volume density of mitochondria, peroxisomes, autophagic vacuoles, Golgi complexes, smooth endoplasmic reticulum or glycogen, and no significant change in the surface density of granular endoplasmic cisternae. The present investigation indicates that in the maternal hepatocyte certain cytoplasmic components are quantitatively changed by ethanol, whereas the volume and surface densities of identical components in the fetal hepatocyte are unaffected. The significance of these results is discussed.  相似文献

Livers of 45 fingerling grey mullets ( Liza sp. 5-15 cm long) were subjected to light microscopy, transmission electron microscopy and enzyme cytochemical examination. The hepatic structure was found to be comparable in all three Liza species. As in other fish species, Liza liver parenchyma is arranged in cords (or dual-plated laminae, in sections parallel to the sinusoidal axis). Hepatocytes showed numerous microvilli in their perisinusoidal and canalicular areas. They contained a centrally located nucleus, fairly large areas of rough endoplasmic reticulum, consisting mainly of parallel cisternae, and pools of glycogen. A few small lipid droplets were evenly distributed in the cytoplasm. The Golgi fields, between the canaliculus and the nucleus, consist mainly of dictyosomes, lysosomes and autophagic vacuoles. Acid phosphatase activity was cytochemically localized to reaction products in secondary lysosomes and lipofuscin granules. A comparison between grey mullet liver and that of other teleosts was made.  相似文献

T he effects of environmental pollutants from two small streams in south-west Germany on the liver of brown trout (S almo trutta f. Fario ) were studied as biomarkers by means of quantitative and semi-quantitative electron microscopy, and quantitatively by morphometrical measurements. Cellular damage was assessed semi-quantitatively based on a classification of ultrastructural responses. Both methods revealed more severe cellular effects in the liver of trout which had been exposed to the highly polluted stream than in those exposed to the lightly polluted river. Morphometrical studies showed a significant reduction of glycogen storage and a significant increase in number of mitochondria, peroxisomes and cisternae of the endoplasmic reticulum. The biomarker responses of this study were correlated with the results obtained by limnological and analytical investigations, and reflect the levels of pollution in each stream.  相似文献

Summary Heparin-binding fibroblast growth factors (HBGF) have been implicated in the regeneration of both parenchymal and nonparenchymal cells of the liver. The response to and phenotype of hepatocyte receptors for HBGF-1 (acidic fibroblast growth factor) and HBGF-2 (basic fibroblast growth factor) were compared to keratinocytes, fibroblasts, and endothelial cells. HBGF-1 stimulated DNA synthesis in hepatocytes, keratinocytes, fibroblasts, and endothelial cells whereas activity of HBGF-2 was limited to fibroblasts and endothelial cells. HBGF-2 antagonized the mitogenic activity of HBGF-1 for hepatocytes and keratinocytes. Hepatocytes and keratinocytes exhibited both high- and low-affinity, nonmatrix receptor sites for HBGF-1, but only low-affinity sites for HBGF-2. The mesenchymal cells displayed only high-affinity sites for both HBGF-1 and HBGF-2. Northern blot and immunochemical analysis revealed that the expression of HBGF receptor genesbek andflg are partitioned between normal hepatocytes and nonparenchymal cells, respectively. Expression of epithelial cell-specific, mesenchymal cell-derived HBGF-7 (keratinocyte growth factor) mRNA in regenerating liver tissue was undetectable relative to HBGF-1. The results support a multifunctional role of HBGF-1 acting through different receptor phenotypes in hepatocyte and nonparenchymal cells during liver regeneration.  相似文献

Liver tissue was sampled from flounder (Pleuronectes americanus) throughout the year with the intention of documenting changes in the ultrastructure coincident with the production and secretion of antifreeze proteins. In the winter, hepatocytes are dedicated to the production of these proteins and, in the female, also reproductive proteins. In both sexes, liver cells in the summer contain abundant lipid and glycogen stores. In the female, there is a conspicuous hepatocyte transformation from a fat-filled cell in the summer to one with well-developed rough endoplasmic reticulum in the winter. Large amounts of rough endoplasmic reticulum (11.2 mg/gm) were recovered after subcellular fractionation of female wintertime liver. The increased appearance of secretory organelles and the high number of nucleolar profiles observed in winter animals is consistent with the elevated demand for protein secretion and synthesis in both sexes. The fractional volumes occupied by lipid droplets and mitochondria were different when comparisons were made between sex and season. Females contained a greater volume of lipid than did males, and summer animals contained more lipid than those in winter.  相似文献

Some characteristics of the mitochondria of hepatocytes and of three hepatoma cell lines have been compared. By means of stereologic analysis of electron micrographs of cross-sections through cells the volume of mitochondria per unit volume of cell cytoplasm and the surface areas of the mitochondrial envelope and cristae membranes have been measured. The relative mitochondrial volume in the cytoplasm decreases with increasing growth rate but the surface area of outer and cristae membranes per unit volume of mitochondria is not altered. The internal organization of hepatoma mitochondria, however, differs distinctly from that of normal liver mitochondria as evident from electron micrographs; the hepatoma cells contain mitochondria in which parallel cristae appear to cross the whole mitochondrial profile unlike the irregular, short cristae seen in normal liver mitochondria. Furthermore, in the fast-growing hepatoma cells the mitochondrial matrix appears less dense than in the hepatocyte. Hepatoma cells contain less organized rough endoplasmic reticulum than normal liver cells and the spatial relationship of the mitochondria to the rough cisternae, seen in the hepatocyte, is absent in the fast-growing hepatoma cell lines. It is concluded that hepatoma cells have fewer mitochondria than normal liver cells, but that the organelles have a normal content of inner membranes.  相似文献

The recirculating perfusion of adult rat liver with a Ca-++-free Hanks' solution produces a release of the adhesiveness of cells and a cleaving of the desmosomes. The addition of collagenase and hyaluronidase to the perfusion medium leads to complete dissociation of the liver tissue into a mixture of isolated cells and cell cords in which the hepatocytes remain connected with specific junctional differentiations, namely the gap and tight junctions. Individual cells are released by submitting the suspension of cell trabeculae to a gentle rolling. The gap junctions are ruptured at least in one of the two adjacent cells and remain generally attached to the other cell taking with them a small portion of cytoplasm. This technique of isolation of hepatocytes yields about 60-65% of the parenchymal cells contained in a liver; endothelial cells and other cells of the connective tissue are not recovered. The ultrastructural preservation of the isolated hepatocytes is excellent and the glucose-6-phosphatase activity, confined to the endoplasmic reticulum, appears unaltered in most cells. Protein, DNA and RNA recovery in the preparations of isolated hepatocytes is satisfactory, amounting to 70% of that found in liver homogenate; glycogen, the most labile component examined, is partly lost or degraded during the manipulations. Cell diameters measured by different methods confirm the preservation of the original volume of the in situ hepatocytes and the presence of more than one type of parenchymal cell. By submitting this heterogeneous cell population to an isopycnic density gradient centrifugation, two types of hepatocytes can be distinguished: the light hepatocytes, with a mean diameter of 20.5 mum and a mean density of 1.10, are characterized by an extended smooth- walled endoplasmic reticulum entrapping dispersed alpha-glycogen particles; the heavy hepatocytes, with a mean diameter of 19.0 mum and a mean density of 1.14, present a relatively reduced compartment of smooth endoplasmic reticulum, but large accumulations of glycogen. It is suggested that the cell fraction of low density is enriched in centrolobular cells and the high density fraction in perilobular hepatocytes.  相似文献

Peroxidase-positive endothelial cells in sinusoids of the mouse liver   总被引:3,自引:0,他引:3  
The cytochemical localization of endogenous peroxidase activity in sinus lining cells of mouse liver has been investigated. Kupffer cells, as identified by their exclusive ability to phagocytize large (0.8 micron) latex particles, exhibited strong peroxidase activity in nuclear envelope and endoplasmic reticulum. In addition, weak to moderate peroxidase activity was found in 57% of all endothelial cells. The enzyme in endothelial cells was also localized in nuclear envelope and endoplasmic reticulum, with a negative reaction in the Golgi apparatus. These observations indicate that peroxidase staining, as a marker for identification of Kupffer cells in mouse liver, is only of limited value and should be used in conjunction with other methods (e.g., latex phagocytosis).  相似文献

Summary Rats were fed on a 25% casein diet or the same diet supplemented with desiccated thyroid. The rats were killed after 16 days. Histological sections of the livers of the control rats show coarse, basophilic inclusions and abundance of glycogen in the cytoplasm. In the thyroid-fed rats there is a diffuse, cytoplasmic basophilia with basophilic rods and no or almost no glycogen. Under the electron microscope large areas of glycogen are to be seen in the cytoplasm of the control animals. Mitochondria and rough-surfaced endoplasmic membranes, often in large stacks, are found together. The liver cells of the thyroid-fed rats have little or no glycogen in their cytoplasm. Mitochondria, endoplasmic reticulum, and free ribosomes and polysomes are evenly distributed all over the cytoplasm. There seems to be an increase in the ratio of free to membrane-bound ribosomes and polysomes in the thyroid-fed rats.
