Aim The analysis of diversity across multiple scales is hampered by methodological difficulties resulting from the use of different sampling methods at different scales and by the application of different definitions of the communities to be sampled at different scales. It is our aim to analyse diversity in a nested hierarchy of scales by applying a formalized sampling concept used in population ecology when analysing population structure. This concept involved a precise definition of the sampled vegetation type by the presence of a target species, in our case Hornungia petraea. We compared separate indices of inventory diversity (i.e. number of species) and differentiation diversity (i.e. extent of change in species composition or dissimilarity) with indices derived from species accumulation curves and related diversity patterns to topographical plot characteristics such as area and distance. Location Ten plots were established systematically over a distance of 100 km each in the distribution centre of H. petraea in Italy (i.e. Marche and Umbria) and in a peripheral exclave in Germany (i.e. Thuringia and Saxony‐Anhalt). Methods We used a nested sampling design of 10 random subplots within plots and 10 systematically placed plots within regions. Internal α‐diversity (species richness) and internal β‐diversity (dissimilarity) were calculated on the basis of subplots, α‐, β‐ and γ‐diversity on the basis of plots in Italy and Germany. In addition, indices of inventory diversity and differentiation diversity were derived by fitting species accumulation curves to the Michaelis–Menten equation. Results There was no significant difference in the internal α‐diversity between German and Italian plots but the α‐ and γ‐diversity were higher in Italy than in Germany. In Germany, the internal β‐diversity and β‐diversity were lower than in Italy. The differentiation diversity increased with increasing scale from subplots over plots to regions. The same results were obtained by calculating species accumulation curves. Significant positive correlations were encountered between the internal α‐diversity and α‐diversity in both countries, while the internal β‐diversity and internal α‐diversity showed a correlation only for the Italian plots. Similarity decay was found for German plots with respect to inter‐plot distance and for Italian plots with respect to altitudinal difference and to a smaller degree to distance between plots. Main conclusions The design chosen and the consistent analysis of species accumulation curves by the Michaelis–Menten equation yielded consistent results over different scales. The specific therophyte vegetation type in this study reflected diversity patterns also observed in other studies, e.g. a greater differentiation diversity in central than in peripheral habitats and a trend of increasing species richness towards lower altitudes. No asymptotic saturation of species richness between different scales was observed. Indications were found that the absolute level of inventory diversity at a particular scale and the completeness of the sampling procedure are the main clues for explaining the relationship between inventory and differentiation diversity at this particular scale.  相似文献

基因型多样性作为遗传多样性的重要组成部分,不仅在群落水平,而且在种群水平均可以发挥重要生态作用。以羊草(Leymus chinensis)为研究对象,设置5种基因型多样性梯度试验控制小区,进行基因型多样性对羊草种群年龄结构的影响研究。结果表明:随基因型多样性梯度的逐步增加,2a分蘖株生物量和数量均显著增加(P<0.05);1-4龄级羊草根茎生物量、长度和干物质积累量多数呈极显著性增加的变化趋势(P<0.01)。在1、2、4、8和12种基因型多样性梯度时,分蘖株生物量及数量年龄结构均以1a或2a分蘖株所占比例为最高,年龄结构表现为增长型或稳定型;而羊草种群根茎生物量和长度也以2a根茎所占比例为最高,年龄结构表现为稳定型。随基因型多样性水平的增加,羊草种群分蘖株数量和生物量的年龄结构从增长型向稳定型过渡,但根茎长度和生物量的年龄结构均为稳定型。12基因型多样性水平的4龄级分蘖株和根茎的特征高于其他基因型多样性水平。因此,在不同基因型多样性水平羊草种群分蘖株和根茎具有增长型或稳定型的年龄结构,适当的基因型多样性数量显著促进了各龄级羊草分蘖株和根茎的生物量和数量特征的升高,但高度的基因型多样性水平会增加羊草种群中高龄级的分蘖株和根茎所占的比例,种群发展表现出衰退信号。  相似文献

The extreme polymorphism of MHC class I has been argued to be driven by balancing selection from pathogens, with the prediction that populations exposed to a wider variety of diseases should have higher diversity. We assembled a global database of allotype frequencies for MHC class I genes and investigated possible drivers of genetic diversity, measured in different ways. We first looked for a decline in diversity with distance from Africa (a consequence of drift during human expansions) and then investigated the link with pathogen richness once the effect of drift had been corrected for. Using heterozygosity, we recovered a clear decline in diversity from Africa and confirmed the positive relationship between genetic diversity and pathogen richness for all three classical MHC class I genes. However, when we considered a sequence-based measure of genetic diversity, the correlation with geographic distance from Africa vanished for HLA-C, and the correlations with pathogen richness for the three MHC class I genes were much weaker. HLA-C is known to consist of two functional classes of allotypes (classified with respect to the 80th residue), which interact with different KIR receptors. While this separation provided some improvement in the fit between genetic diversity and distance from Africa for one class, much clearer and consistent patterns were recovered when we used the 90th residue to separate HLA-C allotypes into two new classes. This suggests that this residue, which is also involved in the binding of KIR, might have had an important evolutionary role that has been overlooked.  相似文献

在山西吕梁山系按照纬度从北向南依次选取马伦草原、荷叶坪、云顶山作为研究区,于2015、2016、2017年7月下旬进行亚高山草甸植被群落多样性调查,研究吕梁山亚高山草甸物种多样性不同年份、不同纬度的时空变化格局。结果表明:(1)吕梁山亚高山草甸群落中矮嵩草(Kobresia humilis)和珠芽蓼(Polygonum viviparum)、线叶嵩草(Kobresia capillifolia)和珠芽蓼、车前草(Plantago asiatica)和披碱草(Elymus dahuricus)分别为马伦草原、荷叶坪、云顶山的优势种,不同山地植物优势种和次优势种均以菊科(Asteraceae)、蔷薇科(Rosaceae)、莎草科(Cyperaceae)、禾本科(Gramineae)植物为主。(2)吕梁山亚高山草甸物种多样性在不同年份分布较为稳定,Pielou指数在连续3年内均无较大变化; Shannon-Wiener指数、Simpson指数和Patrick指数在2016年最高,同时该年份的降水天数也是最多,表明吕梁山亚高山草甸物种多样性在时间上呈现出受降水条件影响较大的变化格局。(3)吕梁山偏南部的云顶山多样性指数高于偏北部的马伦草原和荷叶坪,表明吕梁山亚高山草甸物种多样性在空间上表现出由北向南逐渐升高的变化格局。因此,吕梁山亚高山草甸物种多样性在时空上呈现出受降水条件影响较大,且由北向南逐渐升高的变化格局,降水条件和纬度梯度对该山地亚高山草甸物种多样性的时空变化格局产生了重要影响。  相似文献

Recent developments of molecular tools have revolutionized our knowledge of microbial biodiversity by allowing detailed exploration of its different facets and generating unprecedented amount of data. One key issue with such large datasets is the development of diversity measures that cope with different data outputs and allow comparison of biodiversity across different scales. Diversity has indeed three components: local (α), regional (γ) and the overall difference between local communities (β). Current measures of microbial diversity, derived from several approaches, provide complementary but different views. They only capture the β component of diversity, compare communities in a pairwise way, consider all species as equivalent or lack a mathematically explicit relationship among the α, β and γ components. We propose a unified quantitative framework based on the Rao quadratic entropy, to obtain an additive decomposition of diversity (γ = α + β), so the three components can be compared, and that integrate the relationship (phylogenetic or functional) among Microbial Diversity Units that compose a microbial community. We show how this framework is adapted to all types of molecular data, and we highlight crucial issues in microbial ecology that would benefit from this framework and propose ready‐to‐use R‐functions to easily set up our approach.  相似文献

A previously characterized compound microsatellite locus ARMS1, containing penta- and decameric repeat units, has been reported to reveal genetic diversity in Ascochyta rabiei (Pass.) Labr. isolates. Therefore, 37 isolates of Ascochyta rabiei collected from different states of India and 38 isolates from fifteen other countries in the world were examined for their diversity at this locus. Twenty-six alleles on the basis of size (228--451 base pairs) were detected in the world isolates examined, while 15 alleles (287--418 base pairs) were observed in isolates from the Indian subcontinent. To the best of our knowledge, this study is the first to demonstrate diversity in representative Ascochyta rabiei isolates from different parts of the world at the ARMS1 locus.  相似文献

【目的】从南海4个站位的深海沉积物中分离真菌,揭示其多样性并测定抗菌活性。【方法】使用4种培养方法和8种培养基,从12个深海沉积物样本中分离培养真菌,通过菌落形态观察和ITS序列系统发育分析进行鉴定。采用滤纸片扩散法和生长速率法分别测试真菌小量发酵液粗浸膏的抗细菌和抗真菌活性。【结果】共分离到125株纯培养真菌,基于形态和ITS序列分析,排重后得到18个种类型,这些真菌可以划分到12个属,大多数属于子囊菌门(Ascomycota),只有2株属于担子菌门(Basidiomycota)。4个站位可培养真菌多样性具有差异性。抑菌活性筛选显示,大多数真菌具有较好的抑菌活性;链格孢属(Alternaria)、青霉属(Penicillium)、匐柄霉属(Stemphylium)这几个属的真菌表现出对多种指示细菌有抑制作用,尤其是Alternaria tenuissima DN09、Alternaria alternata DN14和Penicillium chrysogenum DN16对G~+和G~–细菌均表现出抑制作用。【结论】本研究揭示了南海深海沉积物可培养真菌多样性和抑菌活性,为进一步利用深海沉积物来源真菌奠定了基础。  相似文献

 Studies of allelic variations at six isozyme loci revealed genetic diversity of 380 East Asian accessions of the Barley Core Collection. Genetic variation was found in both cultivars and landraces in different regions. Allelic variations at the Aco-1 and Aco-2 loci were detected for East Asian barley for the first time. Moreover, the Aco-1 locus displayed the highest genetic diversity among the six loci assayed. Indian cultivars showed the highest diversity, followed by Korean and Chinese cultivars. Landraces from Bhutan and Nepal showed the lowest diversity. Cultivars had generally higher diversity than landraces within as well as among regions. The cluster analysis of genetic identity showed that all landraces from different countries can be placed in one group; the cultivars from Japan, India and Korea each form independent groups. Gpi-1 Gu, Pgd-1 Tj, Aco-1 Si, Ndh-2 D and Aco-2 A were rare alleles found in only a few accessions of 6-rowed barley. The Pgd-2 Tn allele was very rare in East Asian accessions. Octopus minor (Sasaki, 1920) is an important economic fishery resource in China. In order to explore the stock information and the phylogeographic status of O. minor, mitochondrial DNA cytochrome c oxidase subunit I (COI, 565?bp) and 16S rRNA (493?bp) genes were amplified from 11 different sampling locations. Genetic diversity evaluated by haplotypic and nucleotidic diversity implied high diversity in Lianjiang, and relatively low diversity in Rongcheng, which suggests that effective measures to protect the O. minor resource in this area are urgently required. Private haplotypes and remarkable higher pairwise ΦST in Yilan are responsible for the deep genetic divergence between Yilan and the 10 other populations. Haplotypes networks and two clusters’ topological structure also support the distinct subgroups (lineages A and lineages B), which apparently possess smaller genetic variation than mean interspecies distance. Taiwan island and its strait may act as a natural barrier that restricts the gene flow from the mainland. Deep genetic divergence between mainland and Taiwanese east coasts suggests different genetic stock, indicating that different management strategies are required.  相似文献

Colletotrichum truncatum is a fungal species associated with anthracnose disease in many economically important crops within the plant families Fabaceae and Solanaceae. Understanding the degree of genetic diversity within C. truncatum population will provide insights into the ability of this species to evolve in response to environmental conditions, and thus be helpful in designing effective control strategies for this pathogen. In this study, microsatellite markers from 27 loci were used to investigate the genetic diversity and population structure among 99 isolates of C. truncatum from India. All the loci (100%) were polymorphic and a total of 140 different alleles were amplified. Six distinct groups were obtained based on unweighted pair group method with arithmetical average cluster analysis. The isolates belonging to Group V showed the highest level of genetic diversity and a broad host range. Analysis of molecular variance analysis showed that the variation occurs mostly within groups. Microsatellite markers-based genetic diversity estimation revealed high diversity among C. truncatum isolates from India.  相似文献

Assessment of the Bacterial Diversity in Fenvalerate-Treated Soil   总被引:4,自引:0,他引:4  
The impact of the pesticide fenvalerate on the diversity of the bacterial community in soil was investigated in this study. After treatment with 0.1, 0.5 or 1.0 mg fenvalerate g–1 soil in three soils and incubation for a 40-day period, the changes in diversity were monitored by two different methods. The cultivable heterotrophic diversity was investigated by colony morphology on solid LB medium. Genetic diversity was measured as bands on denaturing gradient gel electrophoresis (DGGE) gels by total genomic DNA extraction and purification, PCR-amplification of bacterial 16S rDNA fragments. The Shannon–Wiener index of diversity (H), richness (S) and evenness (E H) were used to measure changes in the bacterial community in the soils. The results of the cultivable heterotrophic diversity and genetic diversity showed that there was an obvious decrease in diversity due to the application of fenvalerate to the soils, and the different amounts added had different impacts on the diversity. Bands appearing to be either enhanced or inhibited as a result of the fenvalerate treatments were excized and sequenced. Sequencing of excized DGGE bands indicated that application of fenvalerate had an obvious impact on several Pseudomonas spp., or Xanthomonas campestrisor Streptomyces avermitilis. This revealed that microbial community changes can occur due to the application of fenvalerate to soil.  相似文献

Studies on the effect of plant-species diversity on various ecological processes has led to the study of the effects of plant-genetic diversity in the context of community genetics. Arthropod diversity can increase with plant-species or plant-genetic diversity (Wimp et al. in Ecol Lett 7:776–780, 2004). Plant diversity effects can be difficult to separate from other ecological processes, for example, complementarity. We asked three basic questions: (1) Are arthropod communities unique on different host-plant genotypes? (2) Is arthropod diversity greater when associated with greater plant-genetic diversity? (3) Are arthropod communities more closely associated with host-plant genetics than the plant neighborhood? We studied canopy arthropods on Populus fremontii trees randomly planted in a common garden. All trees were planted in a homogeneous matrix, which helped to reduce P. fremontii neighborhood effects. One sample was comprised of few P. fremontii genotypes with many clones. A second sample was comprised of many P. fremontii genotypes with few clones. A second data set was used to examine the relationships between the arthropod community with P. fremontii genetic composition and the neighborhood composition of the focal host plant. Unique arthropod communities were associated with different P. fremontii genotypes, and arthropod community diversity was greater in the sample with greater P. fremontii genotypic diversity. Arthropod community similarity was negatively correlated with P. fremontii genetic distance, but arthropod community similarity was not related to the neighborhood of the P. fremontii host plant.  相似文献

多山地区城市扩展过程中,建成区内遗存有大量自然山体,形成城市人工基质"海"中的遗存山体"岛屿生境",这些城市遗存山体长期受到来自城市人工环境的各种间接或直接的干扰。为探索城市基质与城市遗存自然山体植物群落多样性之间的关系,以黔中地区典型的喀斯特多山城市--贵阳为研究区域,按不同规模选择15座城市遗存山体,每座样山按坡向+坡位组合方式设置群落调查样地,共设置143个样地;以样山边界为基准,100 m为步长向外设置20个缓冲区,总宽度为2000 m,计算缓冲区总不透水表面积(PTIA)、植被覆盖度(VC)、破碎化指数(FI)、土地利用(LU)4个城市基质特征指标。运用Pearson相关分析和线性回归模型分析了城市基质特征指标与城市遗存山体植物群落物种多样性之间的相关关系。结果表明:①PTIA对不同规模城市遗存山体整体植物多样性的影响都存在明显空间尺度效应,在2000 m时相关性最高,呈显著正相关;不同规模城市遗存山体整体植物多样性与单个或多个LU类型存在显著相关性,但影响效应存在显著差异;VCFI与城市遗存山体整体植物多样性无显著相关关系;②不同规模城市遗存山体各层次植物多样性与城市基质特征 4个指标在不同的空间尺度上均存在显著相关性;③城市遗存山体植物多样性Shannon-Wiener指数(H'')与每个城市基质特征指标一元线性回归拟合度高,而多元回归R2为0。探明城市基质特征对城市遗存山体植物多样性具有一定的影响,但影响规律不一致且存在空间尺度差异,说明城市人工环境对城市遗存山体植物多样性维持的影响非常复杂,需要从不同的角度开展深入的研究,才有可能揭示城市遗存山体植物多样性的城市化响应机制,进而指导城市遗存山体保护规划与管理。  相似文献

Many ecological studies have used diversity indices to assess the impact of environmental disturbance. In particular, ground beetles have been advocated as a good group for assessing disturbance. Most studies on various organisms have used only one or two indices. For our study of the impact of tillage disturbance on carabid beetles in farm fields in southern Ontario, Canada, we used seven different diversity indices (richness, Shannon–Wiener, Berger–Parker, Q-statistic, Margalef, and evenness). Few studies have used deviation from diversity abundance models as a measure of disturbance; however, we use three that are applicable to our data (geometric, log-normal and log-series). The indices and models were used to test the null hypothesis that there is no change in diversity with increasing tillage disturbance, and that there is no difference in diversity with different crops or years. We were not able to reject the null hypothesis that there is any diversity difference among farms. We also found that there was no single diversity index or model that was better than any other at detecting disturbance. These results are supplemented by a meta-analysis of 45 published data sets for the same taxon but in different habitats. The meta-analysis supports the conclusions from our field research that diversity indices and models are not useful for detecting the possible effect of disturbance on assemblages of carabid beetles.  相似文献

In view of the progressive destruction of the biosphere, biodiversity research plays an important role in the search for factors affecting species diversity and in the analysis of the effects of species diversity on ecosystems. However, research into how species diversity and composition are related to genetic structures is still at the beginning. In this study, we combined population genomics, new correlative approaches, and randomization permutation methods to identify adaptive genetic differentiation in populations of the Norway spruce Picea abies L. that display different levels of species diversity. We detected a significant positive large covariation (C) between species diversity (ν a ) and genotype frequency at a gene locus, resulting from differential selection by species diversity or by environmental factors. The results may contribute to understand the interplay between ecological and evolutionary processes in determining community structure and dynamics.  相似文献

Aim Anthropogenic changes in land use may have major consequences for global biodiversity. However, species diversity is determined by a suite of factors that may affect species differently at different spatial scales. We tested the combined effects of land use and spatial scale on α, β and γ diversity in the tropics using experimental communities of cavity‐nesting bees and waSPS (Hymenoptera: Aculeata). We aimed to determine whether: (1) land‐use intensity negatively affects species richness of cavity‐nesting Hymenoptera, (2) β diversity, both within and between plots, is higher in more natural systems, (3) species richness of flowering herbs correlates positively with species richness of Hymenoptera within and across habitats, (4) richness of cavity‐nesting Hymenoptera in highly modified habitats declines with increasing distance from natural or semi‐natural habitats, (5) the effects of land use, herb diversity and forest distance on Hymenoptera α and β diversity vary at different spatial scales, and (6) bees and waSPS respond to land use in a similar way. Location Manabi, south‐west Ecuador. Methods We examined diversity (species richness) within 48 plots of five habitat types that comprised a gradient of decreasing agricultural intensity from rice and pasture to coffee agroforests, unmanaged abandoned agroforests and forest fragments, using standardized nesting resources for reproducing communities of cavity‐nesting bees and waSPS. Results (1) Land use significantly affected α diversity of trap‐nesting bees and waSPS at the subplot (per trap) scale, but not subplot β diversity or plot‐scale species richness (γ diversity). (2) Beta diversity was surprisingly higher between plots within a land‐use type than between land‐use types. (3) Species richness of bees and waSPS increased with diversity of flowering herbs at the subplot (trap) scale only. (4) Forest distance correlated positively with bee species richness at the plot scale only. (5) Land use, herb diversity and forest distance each showed significant correlations with bee and wasp diversity at only one spatial scale. (6) Despite differences in life history, bees and waSPS responded to land‐use intensity in a similar way. Main conclusions The effects of land use on species richness were highly dependent on spatial scale. Subplot‐scale analyses showed that rice and pasture contained the highest species diversity, whereas plot‐scale analyses showed no significant difference in the diversity of different land‐use types. We emphasize caution in the estimation of biodiversity at only one spatial scale, and highlight the surprisingly large contribution of managed land to the regional biodiversity of these species.  相似文献

Cryptic species are continuously discovered in rotifers using different methods to delineate these units of diversity. DNA taxonomy is the most effective method taxonomists have to untie potential cryptic taxa. Here, we estimate hidden diversity in a genus of bdelloid rotifers, Adineta. We analyse cryptic diversity using a coalescent approach to infer evolutionarily significant units from a phylogenetic tree obtained from cytochrome oxidase I sequences. Cryptic diversity was measured for eight traditional species and from several additional undetermined populations. Taxonomic inflation of up to 36 taxa was found in A. vaga from DNA taxonomy. As observed in other microscopic organisms, cryptic taxa within each traditional species were not geographically isolated, but had significantly narrower ecological niches than expected by chance alone.  相似文献

Intraspecific variation among 84 isolates of the anamorphic fungusChaunopycnis alba from 26 different geographical locations was analyzed by investigating optimal growth temperatures, differences in the production of secondary metabolites and presence or absence of the cyclosporin synthetase gene. The genetic diversity was assessed using random amplified polymorphic DNA (RAPD). Analysis of these data showed high genetic, metabolic and physiological diversity within this species. Isolates from the Antarctic represented the most homogeneous group withinC. alba and together with isolates from the Arctic these polar strains differed from alpine, temperate and tropical strains by low optimal growth temperatures and by low production of secondary metabolites. Isolates from tropical climes were characterized by high optimal growth temperatures and by the production of comparatively diverse metabolite spectra. Most of the isolates that were similar in the combination of their physiological and metabolic characters were also genetically related. Isolates from different geographical origins did not show many similarities, with the exception of the cyclosporin A-producing isolates, and large diversity could be observed even within a single habitat. This leads us to the suggestion that for pharmaceutical screening programs samples should be collected from a diversity of different geographical and climatic locations. For the selection of strains for screening the RAPD assay seems to be the most powerful tool. It reflected the highest intraspecific diversity and the results corresponded well with the other characteristics.  相似文献

The microbiomes associated with bee nests influence colony health through various mechanisms, although it is not yet clear how honeybee congeners differ in microbiome assembly processes, in particular the degrees to which floral visitations and the environment contribute to different aspects of diversity. We used DNA metabarcoding to sequence bacterial 16S rRNA from honey and stored pollen from nests of 4 honeybee species (Apis cerana, A. dorsata, A. florea, and A. laboriosa) sampled throughout Yunnan, China, a global biodiversity hotspot. We developed a computational pipeline integrating multiple databases for quantifying key facets of diversity, including compositional, taxonomic, phylogenetic, and functional ones. Further, we assessed candidate drivers of observed microbiome dissimilarity, particularly differences in floral visitations, habitat disturbance, and other key environmental variables. Analyses revealed that microbiome alpha diversity was broadly equivalent across the study sites and between bee species, apart from functional diversity which was very low in nests of the reclusive A. laboriosa. Turnover in microbiome composition across Yunnan was driven predominantly by pollen composition. Human disturbance negatively impacted both compositional and phylogenetic alpha diversity of nest microbiomes, but did not correlate with microbial turnover. We herein make progress in understanding microbiome diversity associated with key pollinators in a biodiversity hotspot, and provide a model for the use of a comprehensive informatics framework in assessing pattern and drivers of diversity, which enables the inclusion of explanatory variables both subtly and fundamentally different and enables elucidation of emergent or unexpected drivers.  相似文献

应用随机扩增多态性DNA(RAPD)技术对草履蚧保定、石家庄、邯郸16个不同寄主地理种群遗传多样性和种群分化进行研究,结果显示4个RAPD引物共扩增出41个多态性位点,多态位点比率为100%。遗传距离指数在0.701—0.4360,平均为0.2395。其中以邯郸枫杨和邯郸垂丝海棠为寄主的草履蚧种群遗传距离最小(0.0701);以石家庄紫叶李和邯郸木槿为寄主的种群遗传距离最大(0.4360)。遗传一致度系数在0.6466—0.9290。说明草履蚧不同种群遗传多样性丰富并存在遗传差异。聚类分析结果表明草履蚧种群遗传多样性同时受到寄主和地理因素的双重影响,且不同寄主草履蚧种群已产生明显的遗传分化。  相似文献
