Changes in the main parameters of α-and β-adrenergic responses, sensitivity to agonists (EC 50) and maximum response (P m) of hindlimb and small intestinal blood pressure in situ and systemic blood pressure were studied in rabbits adapted to cold for 1–30 days (daily exposures to ?10°C for 6 h). The responses to phenylephrine, noradrenaline, adrenaline, clonidine (α-agonists), and isopropylnoradrenaline (β-agonist) corresponded to the equation p = (P m A n )/(EC 50 n + A n ) (1) with n = 1 and n = 2, respectively. Cold adaptation induced reciprocal changes in the response of both EC 50 and P m to α-agonists and in the response of P m alone to isopropylnoradrenaline. The significant differences of the parameters from control observed during the first 5 days of adaptation gradually decreased by day 30. After 10 days of adaptation, the efficiency (E = P m/2EC 50) of response to α-and β-agonists of adrenoceptors significantly increased.  相似文献

The modulation of I A K+ current by ten trivalent lanthanide (Ln3+) cations spanning the series with ionic radii ranging from 0.99 ? to 1.14 ? was characterized by the whole-cell patch clamp technique in bovine adrenal zona fasciculata (AZF) cells. Each of the ten Ln3+s reduced I A amplitude measured at +20 mV in a concentration-dependent manner. Smaller Ln3+s were the most potent and half-maximally effective concentrations (EC50s) varied inversely with ionic radius for the larger elements. Estimation of EC50s yielded the following potency sequence: Lu3+ (EC50= 3.0 μm) ≈ Yb3+ (EC50= 2.7 μm) > Er3+ (EC50= 3.7 μm) ≥ Dy3+ (EC50= 4.7 μm) > Gd3+ (EC50= 6.7 μm) ≈ Sm3+ (EC50= 6.9 μm) > Nd3+ (EC50= 11.2 μm) > Pr3+ (EC50= 22.3 μm) > Ce3+ (EC50= 28.0 μm) > La3+ (EC50= 33.7 μm). Ln3+s altered selected voltage-dependent gating and kinetic parameters of I A with a potency and order of effectiveness that paralleled the reduction of I A amplitude. Ln3+s markedly slowed activation kinetics and shifted the voltage-dependence of I A gating such that activation and steady-state inactivation occurred at more depolarized potentials. In contrast, Ln3+s did not measurably alter inactivation or deactivation kinetics and only slightly slowed kinetics of inactivated channels returning to the closed state. Replacement of external Ca2+ with Mg2+ had no effect on the concentration-dependent inhibition of I A by Ln3+s. In contrast to their action on I A K+ current, Ln3+s inhibited T-type Ca2+ currents in AZF cells without slowing activation kinetics. These results indicate that Ln3+ modulate I A K+ channels through binding to a site on I A channels located within the electric field but which is not specific for Ca2+. They are consistent with a model where Ln3+ binding to negative charges on the gating apparatus alters the voltage-dependence and kinetics of channel opening. Summary Lung cell culture may be useful as anin vitro alternative to study the susceptibility of the lung to various toxic agents. Lungs from female Wistar rats were enzymatically digested by recirculating perfusion through the pulmonary artery with a sequence of solutions containing deoxyribonuclease, chymopapain, pronase, collagenase, and elastase. Lung tissue was microdissected and resuspended and the cells obtained were washed by centrifugation. By this isolation method, 2×108 cells per rat lung were obtained with an average viability of 97%. Lung cells cultured in medium containing antibiotics and serum maintained a viability of >70% for 5 d. Rat primary lung cells were exposed to various toxic agents and their viability was assessed by formazan production capacity after 18 h of incubation. Compared to rat and mouse hepatocyte cultures (EC50=5.8 mM), rat primary lung cells were much more susceptible to hydrogen peroxide (EC50=0.6 mM). All cell types were equally sensitive to the more potent toxicanttert-butylhydroperoxide (EC50=0.1 mM). Paraquat was more toxic to lung cells (EC50=0.03 mM) than to rat (EC50=2.8 mM) and mouse (EC50=0.2 mM) hepatocytes. In contrast, rat lung cells were less sensitive to sodium nitroprusside (EC50=2.6 mM) compared to rat (EC50=0.2 mM) and mouse (EC50=0.03 mM) hepatocytes. Nitrofurantoin and menadione (at EC50=0.04 mM and 0.006 mM, respectively) were more toxic to rat lung and liver cells than to murine hepatocytes (EC50=0.2 mM and 0.04 mM, respectively). Our findings demonstrate the applicability of this rat primary lung cell culture for studying the effects of lung toxicants. The osmotic pressure equation for nonideal, associating systems of the type nA +mB ? AnBm, has been derived, by using the assumption yA nB m/yA nyB m = 1. This treatment can also be applied to related associations such as nA + mB ? AB + AB2 + A2B + …. From osmotic pressure experiments on the pure reactants it is possible to obtain the molecular weights (MA and MB) of the reactants and also the virial coefficients (BAA and BBB) of the reactants. The osmotic pressure of a nonreacting mixture of A and B can be calculated from these measurements. It can be used along with osmotic pressure measurements on equilibrium mixtures of A and B to obtain expressions containing the equilibrium constant (or constants) and the cross-virial coefficients (BAB and BBA). Several procedures are described for the evaluation of the equilibrium constant (or constants) and the BAB or BBA terms. It appears that this procedure is a general one which is applicable to associations of the type nA + mB ? AB + A2B + AB2 + …. By correcting for nonideal behavior, one should then be able to apply it to any method available for analyzing ideal associations of the types considered here. In addition it is possible, subject to certain restrictions, to analyze associations of the type 3A + B ? A2 + AB.  相似文献

The stroke volume of the left ventricle (SV) was calculated from noninvasive recordings of the arterial pressure using a finger photoplethysmograph and compared to the values obtained by pulsed Doppler echocardiography (PDE). A group of 19 healthy men and 12 women [mean ages: 20.8 (SD 1.6) and 22.2 (SD 1.6) years respectively] were studied at rest in the supine position. The ratio of the area below the ejection phase of the arterial pressure wave (A s) to SV, as obtained by PDE, yielded a calibration factor dimensionally equal to the hydraulic impedance of the system (Z ao =A s ·SV –1). TheZ ao amounted on average to 0.062 (SD 0.018) mmHg · s · cm–3 for the men and to 0.104 (SD 0.024) mmHg · s · cm–3 for the women. TheZ ao was also estimated from the equation:Z ao = a · (d + b ·HR + c ·PP + e ·MAP)–1, whereHR was the heart rate,PP the pulse pressure,MAP the mean arterial pressure and the coefficients of the equation were obtained by an iterating statistical package. The value ofZ ao thus obtained allowed the calculation of SV from measurements derived from the photoplethysmograph only. The mean percentage error between the SV thus obtained and those experimentally determined by PDE amounted to 14.8 and 15.6 for the men and the women, respectively. The error of the estimate was reduced to 12.3 and to 11.1, respectively, if the factorZ ao, experimentally obtained from a given heart beat, was subsequently applied to other beats to obtain SV from theA s measurement in the same subject.  相似文献

Many grass species exist in the oil exploration areas of North Dakota. The objective of this study was to evaluate seed germination of 65 grass species affected by crude oil. Germination of all species was reduced by crude oil, ranging from 4.3 to 100%. Twenty-eight species were tolerant, 29 moderately tolerant, 6 moderately sensitive, and 2 sensitive. Based on the tolerance levels, the following were used to further test the dose response to crude oil: strong creeping red fescue (Festuca rubra L. ssp. rubra), perennial ryegrass (Lolium perenne L.), orchardgrass (Dactylis glomerata L.), buffalograss [Buchloe dactyloides (Nutt.) Engelm.], little bluestem [Schizachyrium scoparium (Michx.) Nash], witchgrass (Panicum capillare L.), sand dropseed [Sporobolus cryptandrus (Torr.) Gray], Johnsongrass [Sorghum halepense (L.) Pers.], and smooth crabgrass [Digitaria ischaemum (Schreb.) Schreb. ex Muhl.]. The EC50 of germination and biomass was effective in ranking the 9 species. Buffalograss, sand dropseed, and orchardgrass were ranked as the most tolerant species with EC50 values of 0.1, > highest concentration tested, 0.05 m3 m?3 (P < 0.05), respectively. Smooth crabgrass and little bluestem were ranked as most sensitive with EC50 values of 0.03 and 0.04 m3 m?3 (P < 0.05), respectively. Abstract— The presynaptic regulation of stimulated dopa-mine release from superfused rat striatal synaptosomes by opioids and γ-aminobutyric acid (GABA) was studied. It was found that in addition to dopamine D2 autoreceptors, calcium-dependent K+-stimulated [3H]dopamine release was inhibited through activation of a homogeneous population of k -opioid receptors in view of the potent inhibitory effect of the k -selective agonist U69.593 (EC50 0.2 nM) and its antagonism by norbinaltorphimine. Neither μ-nor δ-selective receptor agonists affected release of [3H]-dopamine. In addition, GABA potently inhibited the evoked [3H]dopamine release (EC50 0.4 nM) through activation of GABAA receptors in view of the GABA-mimicking effect of muscimol, the sensitivity of its inhibitory effect to picro-toxin and bicuculline, and the absence of an effect of the GABAB receptor agonist baclofen. In the presence of a maximally effective concentration of GABA, U69,593 did not induce an additional release-inhibitory effect, indicating that these receptors and the presynaptic D2 receptor are colocalized on the striatal dopaminergic nerve terminals. The excitatory amino acid agonists N-methyl-d -aspartate and kainate, as well as the cholinergic agonist carbachol, stimulated [3H]dopamine release, which was subject to k -opioid receptor-mediated inhibition. In conclusion, striatal dopamine release is under regulatory control of multiple excitatory and inhibitory neurotransmitter by activation of colocalized presynaptic receptors for excitatory amino acids, acetylcholine, dopamine, dynorphins, and GABA within the dopaminergic nerve terminals. Together, these receptors locally control ongoing dopamine neurotransmission.  相似文献

EC50 and EC95 (the effective concentrations to cause inhibitions by 50 and 95%, respectively) are commonly used to express fungicide potency. Different methods are currently employed to calculate EC50 and EC95 values. In this study, EC50 and EC95 values for fungicide epoxiconazole against 34 isolates of Sclerotinia sclerotiorum were calculated with seven different methods. Results showed that for both EC50 and EC95 calculations, there was no significant difference among three statistical programs IBM spss ®, GraphPad Prism® and dps ® (P  0.066). Methods linear log (linear regression of mycelial growth inhibition vs. logarithmic concentration) and interpolation log (linear interpolation from inhibition and logarithmic concentration data) were not significantly different (P  0.058) from IBM spss in EC50 calculations. These results indicate that among the seven methods, the three statistical programs IBM spss , GraphPad Prism, dps and linear log method are appropriate for EC50 calculations. But for EC95 calculations, only the three statistical programs are recommended, and GraphPad Prism is likely to give a little higher values than spss and dps .  相似文献

  • 1 The adenylate energy charge (ECA) of microbial communities from streambed sediments was measured during three different seasons, under experimental manipulation and in culture.
  • 2 The ECA values of sediments (x±S.E.) in the autumn, winter and spring were low and constant; 0.22±0.03 (n=12), 0.32±0.04 (n= 12) and 0.28±0.03 (n=6) respectively.
  • 3 A 5 h exposure of sediments to an algal lysate at 3.0–4.0°C and a 48 h exposure of sediments to tryptone-yeast extract at 8.0–18.0°C failed to increase ECA even though respiration increased 3.7-fold during the latter exposure.
  • 4 The cellular ECA of a bacterial monoculture, sampled in log phase, was 0.90±0.10 (n=3), but cxtracclluhir AMP depressed the total culture ECA to 0.21±0.01 (n=4).
  • 5 Attempts to isolate extracellular AMP from the interstitial waters of sediments were unsuccessful.
  • 6 The data suggest that under natural conditions ECA is of limited use as a monitor of subtle changes in the physiological state of microbial communities in streambed sediments.

Examination of 28 healthy subjects and 66 patients was performed using laser Doppler flowmetry (LDF) of the skin of fingers in the red and infrared ranges and wavelet analysis of microvascular blood flow oscillations. Formulas were suggested for the total (TF), nutritive (NF), and shunt (SF) local blood flows. TF = MI × Ac × An /(Pm × ), where MI is the microcirculation index in perfusion units (PU), An and Ac are the averaged maximum amplitudes of the neurogenic and cardiac rhythms (in PU), Pm is the mean arterial pressure (in mm Hg), and is the mean square deviation of the amplitude of blood flow oscillations (in PU). NF = TF/SP, where SP is the shunting parameter. SP = An/Am, where Am is the averaged maximum amplitude of the myogenic rhythm (in PU). SF = TF - NF. Significant positive relationships between the skin oxygenation and NF (red), temperature and TF (red), and blood flow measured by hydrogen clearance and TF (infrared) were revealed by comparison of the LDF parameters with polarographic and thermographic data. The advantages of the LDF parameters compared with the MI were shown, especially in skin denervation syndromes. We evoked changes in lower lip blood flow and systemic arterial blood pressure by electrically stimulating the central cut end of the lingual nerve in artificially ventilated, urethane-anesthetized, cervically vago-sympathectomized cats, rats, rabbits, and guinea pig. The systemic arterial blood pressure changes were species-dependent: increases in rat, consistent decreases in rabbit and guinea pig, and variable among individuals in cat. In cat and rabbit, lip blood flow increases, which occurred only ipsilaterally to the stimulated nerve and showed no statistically significant correlation with the systemic arterial blood pressure changes. In rat, the ipsilateral lip blood flow increase was markedly greater than the contralateral one, and although there was a significant correlation between each of them and the systemic arterial blood pressure changes, the ipsilateral increase presumably included an active vasodilatation. In guinea pig, lip blood flow decreased on both sides in proportion to the systemic arterial blood pressure reductions. Endothelins are known to be among the most potent endogenous vasoconstrictors. Vasoconstriction of the spiral modiolar artery, which supplies the cochlea, may be implicated in hearing loss and tinnitus. The purpose of the present study was to determine whether the spiral modiolar artery responds to endothelin, whether a change in the cytosolic Ca2+ concentration ([Ca2+]i) mediates the response and which endothelin receptors are present. The vascular diameter and [Ca2+]i were measured simultaneously by videomicroscopy and microfluorometry in the isolated spiral modiolar artery from the gerbil. ET-1 induced a transient [Ca2+]i increase and a strong and long-lasting vasoconstriction. The transient [Ca2+]i increase underwent rapid desensitization, was independent of extracellular Ca2+ and inhibited by the IP3-receptor blocker (75 μm) 2-aminoethoxydiphenyl borate (2-APB) and by depletion of Ca2+ stores with 10−6 m thapsigargin. In contrast, the vasoconstriction displayed no comparable desensitization. The initial vasoconstriction was independent of extracellular Ca2+ but maintenance of the constriction depended on the presence of extracellular Ca2+. The half-maximal concentration values (EC 50) for the agonists ET-1, ET-3 and sarafotoxin S6c were 0.8 nm, >10 nm and >100 nm, respectively. Affinity constants for the antagonists BQ-123 and BQ-788 were 24 nm and 77 nm, respectively. These observations demonstrate that ET-1 mediates a vasoconstriction of the gerbil spiral modiolar artery via ETA receptors and an IP3 receptor-mediated release of Ca2+ from thapsigargin-sensitive Ca2+ stores. The marked difference in desensitization between Ca2+ mobilization and vasoconstriction suggests that Ca2+ mobilization is not solely responsible for the vasoconstriction and that other signaling mechanisms must be present. The cytomolecular discrimination of the Am- and A-genome chromosomes facilitates the selection of wheat-Triticum monococcum introgression lines. Fluorescence in situ hybridisation (FISH) with the commonly used DNA probes Afa family, 18S rDNA and pSc119.2 showed that the more complex hybridisation pattern obtained in T. monococcum relative to bread wheat made it possible to differentiate the Am and A chromosomes within homoeologous groups 1, 4 and 5. In order to provide additional chromosomal landmarks to discriminate the Am and A chromosomes, the microsatellite repeats (GAA)n, (CAG)n, (CAC)n, (AAC)n, (AGG)n and (ACT)n were tested as FISH probes. These showed that T. monococcum chromosomes have fewer, generally weaker, simple sequence repeat (SSR) signals than the A-genome chromosomes of hexaploid wheat. A differential hybridisation pattern was observed on 6Am and 6A chromosomes with all the SSR probes tested except for the (ACT)n probe. The 2Am and 2A chromosomes were differentiated by the signals given by the (GAA)n, (CAG)n and (AAC)n repeats, while only (GAA)n discriminated the chromosomes 3Am and 3A. Chromosomes 7Am and 7A could be differentiated by the lack of (GAA)n and (AGG)n signals on 7A. As potential landmarks for identifying the Am chromosomes, SSR repeats will facilitate the introgression of T. monococcum chromatin into wheat.  相似文献

Toxicity (1/IGC50) in the Tetrahymena population growth assay and reactivity (1/EC50) with the thiol moiety of the cysteine residue of glutathione (GSH) were determined for a series of aromatic isothiocyanates (NCSs). Comparison of both toxicity and reactivity between the analogues revealed that derivatives with the NCS-moiety attached directly to an aromatic ring (e.g., phenyl derivatives) are less toxic and less reactive than those with the NCS attached to an aliphatic carbon (e.g., benzyl derivatives). These differences in potency are hypothesized to relate to difference in the ease of the Michael reaction, the proposed molecular mechanism. 1,4-Phenylene diisothiocyanate is more toxic and more reactive than its mono-NCS homologue. While there is good predictivity for the phenyl and naphthyl derivatives with the model log(1/IGC50) = 0.545(log K ow) + 16.21A max – 5.91, based on the 1-octanol/water partition coefficient (K ow) and maximum acceptor superdelocalizability (A max), toxicity of the other derivatives, which are outside the structural domain of the model training set, are poorly fitted. Owing to hydrolysis, the benzoyl, and cinnamyl analogues are less toxic than predicted by their thiol reactivity; however, the toxicity of the remaining compounds is modeled by the relationship log(1/IGC50) = 1.77 [log (1/EC50)] + 0.60; n = 12, s = 0.34, r 2 = 0.718, q 2 = 0.629, F = 26.  相似文献

Independent short-term effects of photosynthetic photon flux density (PPFD) of 50–400 μmol m−2 s−1, external CO2 concentration (C a) of 85–850 cm3 m−3, and vapor pressure deficit (VPD) of 0.9–2.2 kPa on net photosynthetic rate (P N), stomatal conductance (g s), leaf internal CO2 concentration (C i), and transpiration rates (E) were investigated in three cacao genotypes. In all these genotypes, increasing PPFD from 50 to 400 μmol m−2 s−1 increased P N by about 50 %, but further increases in PPFD up to 1 500 μmol m−2 s−1 had no effect on P N. Increasing C a significantly increased P N and C i while g s and E decreased more strongly than in most trees that have been studied. In all genotypes, increasing VPD reduced P N, but the slight decrease in g s and the slight increase in C i with increasing VPD were non-significant. Increasing VPD significantly increased E and this may have caused the reduction in P N. The unusually small response of g s to VPD could limit the ability of cacao to grow where VPD is high. There were no significant differences in gas exchange characteristics (g s, C i, E) among the three cacao genotypes under any measurement conditions.  相似文献

A process-based leaf gas exchange model for C3 plants was developed which specifically describes the effects observed along light gradients of shifting nitrogen investment in carboxylation and bioenergetics and modified leaf thickness due to altered stacking of photosynthetic units. The model was parametrized for the late-successional, shade-tolerant deciduous species Acer saccharum Marsh. The specific activity of ribulose-1,5-bisphosphate carboxylase (Rubisco) and the maximum photosynthetic electron transport rate per unit cytochrome f (cyt f) were used as indices that vary proportionally with nitrogen investment in the capacities for carboxylation and electron transport. Rubisco and cyt f per unit leaf area are related in the model to leaf dry mass per area (MA), leaf nitrogen content per unit leaf dry mass (Nm), and partitioning coefficients for leaf nitrogen in Rubisco (PR) and in bioenergetics (PB). These partitioning coefficients are estimated from characteristic response curves of photosynthesis along with information on lear structure and composition. While PR and PB determine the light-saturated value of photosynthesis, the fraction of leaf nitrogen in thylakoid light-harvesting components (PL) and the ratio of leaf chlorophyll to leaf nitrogen invested in light harvesting (CB), which is dependent on thylakoid stoichiometry, determine the initial photosynthetic light utilization efficiency in the model. Carbon loss due to mitochondrial respiration, which also changes along light gradients, was considered to vary in proportion with carboxylation capacity. Key model parameters - Nm, PR, PB, PLCB and stomatal sensitivity with respect to changes in net photosynthesis (Gr) – were examined as a function of MA, which is linearly related to irradiance during growth of the leaves. The results of the analysis applied to A. saccharum indicate that PB and PR increase, and Gf, PL and CB decrease with increasing MA. As a result of these effects of irradiaiice on nitrogen partitioning, the slope of the light-saturated net photosynthesis rate per unit leaf dry mass (Ammax) versus Nm relationship increased with increasing growth irradiance in mid-season. Furthermore, the nitrogen partitioning coefficients as well as the slopes of Ammax versus Nm were independent of season, except during development of the leaf photosynthetic apparatus. Simulations revealed that the acclimation to high light increased Ammax by 40% with respect to the low light regime. However, light-saturated photosynthesis per leaf area (Aamax) varied 3-fold between these habitats, suggesting that the acclimation to high light was dominated by adjustments in leaf anatomy (Aamax=AmmaxMA) rather than in foliar biochemistry. This differed from adaptation to low light, where the alterations in foliar biochemistry were predicted to be at least as important as anatomical modifications. Due to the light-related accumulation of photosynthetic mass per unit area, Aamax depended on MA and leaf nitrogen per unit area (Na). However, Na conceals the variation in both MA and Nm (Na=NmMA), and prevents clear separation of anatomical adjustments in foliage structure and biochemical modifications in foliar composition. Given the large seasonal and site nutrient availability-related variation in Nm, and the influences of growth irradiance on nitrogen partitioning, the relationship between Aamax and Na is universal neither in time nor in space and in natural canopies at mid-season is mostly driven by variability in MA. Thus, we conclude that analyses of the effects of nitrogen investments on potential carbon acquisition should use mass-based rather than area-based expressions.  相似文献

The kinetics of t-[3H]butylbicycloorthobenzoate (TBOB) binding to the convulsant sites of the γ-aminobutyric acidA (GABAA) receptor-ionophore complex were examined in synaptosomal membrane preparations of rat brain. On and off rates of TBOB binding were accelerated by 1 μM GABA and decelerated by 1 μM bicuculline methochloride, a GABAA antagonist. The presence of GABA and bicuculline methochloride created rapid and slow phases of dissociation, respectively. The three groups of rate constants distinguished for the dissociation of 4 nM and 30 nM [3H]TBOB represent multiaffinity states of the convulsant sites depending on the presence of GABA or bicuculline methochloride. Apparent association rate constants do not obey the equation kapp=koff±kon [TBOB] without assuming interconvertibility of the kinetic states during binding. Avermectin B1a (AVM B1a), a chloride channel opening agent, accelerated the association and dissociation of TBOB and resulted in a biphasic effect on TBOB binding, i.e., enhancement at low concentrations (EC50, 7.8 nM) followed by displacement at high concentrations (IC50 6.3 μM) of AVM B1a. AVM B1a resulted in similar biphasic effects on t- [35S]butylbicyclophosphorothionate binding. DIDS, an isothiocyanatostilbene derivative with irreversible anion channel blocking effect, selectively inhibited basal [3H]TBOB binding (IC50 125 μM DIDS) leaving the enhancement by AVM B1a unaffected.  相似文献

Exposure to ethanol during human embryonic period has severe teratogenic effects on the cardiovascular system. In our study, we demonstrated that ethanol of gradient concentrations can interfere with the establishment of circulatory system in embryonic zebrafish. The effective concentration to cause 50% malformations (EC50) was 182.5 mmol/L. The ethanol pulse exposure experiment displayed that dome stage during embryogenesis is the sensitive time window to ethanol. It is found that 400 mmol/L ethanol pulse exposure can induce circulatory defects in 43% treated embryos. We ruled out the possibility that ethanol can interfere with the process of hematopoiesis in zebrafish. By employing in situ hybridization with endothelial biomarker (Flk-1), we revealed that ethanol disrupts the establishment of trunk axial vasculature, but has no effect on cranial vessels. Combined with the results of semi-thin histological sections, the in situ hybridization experiments with arterial and venous biomarkers (ephrinB2, ephB4) suggested that ethanol mainly interrupts the development of dorsal aorta while has little effect on axial vein. Further study indicated the negative influence of ethanol on the development of hypochord in zebrafish. The consequent lack of vasculogenic factors including Radar and Ang- 1 partly explains the defects in formation and integrity of dorsal aorta. These results provide important clues to the study of adverse effects of ethanol on the cardiovascular development in human fetus.  相似文献

RNA oligomers with the sequence An-U-G-Um, n = 7–9, m = 5–10, have been synthesized and found to form hairpin loops in 21 mM or 101 mM Na+. The hairpin loops displayed melting temperatures 13°–29°C greater than that of the hairpin loop A6-C8-U6 in the same solvent. The increased stability of these hairpin loops is attributed to the presence of the trinucleotide A-U-G in the loop. Circular dichroism (CD) spectra were taken of the hairpin loops A7-U-G-U6,7,8, A8-U-G-U6,7,8, and A9-U-G-U6 in 21 mM Na+, and compared with circular dichroism spectra of A6-U6 in 1 M Na+. Difference spectra were calculated between each An-U-G-Um and 11.5 mM (nucleotide) A6-U6 at similar temperatures and identical singlestrand fractions to give the “experimental” CD spectrum of the unbase-paired nucleotides in the loop, assuming, five, six, or seven base pairs. CD spectra were calculated for each of the assumed unbase-paired sequences using the measured CD spectra of ApA, A-U-G-, A, and U, and compared with the experimental spectra. The best agreement was found for hairpin-loop models containing five base pairs and five to eight unbase-paired nucleotides in the loop.  相似文献
