Modification of the main parameters of cholinergic reactions in rabbits, i.e., of the specific sensitivity to agonists (EC 50), maximal reaction values (P m ) of arterial pressure in the hind limb vessels and small intestine in situ, and systemic arterial pressure after adaptation to cold for 1, 5, 10, and 30 days has been investigated. The depressor reaction to acetylcholine (muscarinic cholinergic receptors agonist) was established to correspond to the model p = (P m A n )/(EC 50 n + A n ) with n =1. In the control EC 50 was equal to 0.85, 1.01, and 1.21 nmol/kg, while P m equaled 100, 32.6, and 61.2 mm Hg for the hind limb vessels, small intestine in situ, and systemic arterial pressure, respectively.  相似文献

Leaf-level net photosynthesis (An) estimates and associated photosynthetic parameters are crucial for accurately parameterizing photosynthesis models. For tropical forests, such data are poorly available and collected at variable light conditions. To avoid over- or underestimation of modeled photosynthesis, it is critical to know at which photosynthetic photon flux density (PPFD) photosynthesis becomes light-saturated. We studied the dependence of An on PPFD in two tropical forests in French Guiana. We estimated the light saturation range, including the lowest PPFD level at which Asat (An at light saturation) is reached, as well as the PPFD range at which Asat remained unaltered. The light saturation range was derived from photosynthetic light-response curves, and within-canopy and interspecific differences were studied. We observed wide light saturation ranges of An. Light saturation ranges differed among canopy heights, but a PPFD level of 1,000 µmol m−2 s−1 was common across all heights, except for pioneer trees species that did not reach light saturation below 2,000 µmol m−2 s−1. A light intensity of 1,000 µmol m−2 s−1 sufficed for measuring Asat of climax species at our study sites, independent of the species or the canopy height. Because of the wide light saturation ranges, results from studies measuring Asat at higher PPFD levels (for upper canopy leaves up to 1,600 µmol m−2 s−1) are comparable with studies measuring at 1,000 µmol m−2 s−1.  相似文献

The stability of RNA hairpin loops containing A-U-G: An-U-G-Um   总被引:5,自引:0,他引:5

RNA oligomers with the sequence An-U-G-Um, n = 7–9, m = 5–10, have been synthesized and found to form hairpin loops in 21 mM or 101 mM Na+. The hairpin loops displayed melting temperatures 13°–29°C greater than that of the hairpin loop A6-C8-U6 in the same solvent. The increased stability of these hairpin loops is attributed to the presence of the trinucleotide A-U-G in the loop. Circular dichroism (CD) spectra were taken of the hairpin loops A7-U-G-U6,7,8, A8-U-G-U6,7,8, and A9-U-G-U6 in 21 mM Na+, and compared with circular dichroism spectra of A6-U6 in 1 M Na+. Difference spectra were calculated between each An-U-G-Um and 11.5 mM (nucleotide) A6-U6 at similar temperatures and identical singlestrand fractions to give the “experimental” CD spectrum of the unbase-paired nucleotides in the loop, assuming, five, six, or seven base pairs. CD spectra were calculated for each of the assumed unbase-paired sequences using the measured CD spectra of ApA, A-U-G-, A, and U, and compared with the experimental spectra. The best agreement was found for hairpin-loop models containing five base pairs and five to eight unbase-paired nucleotides in the loop.  相似文献

Studies were conducted to examine the influence of the H+-ATPase inhibitor bafilomycin A1 on cultured rabbit nonpigmented ciliary epithelial cells (NPE). Cytoplasmic pH and sodium concentrations were measured by digital fluorescence microscopy using BCECF and SBFI respectively. In some experiments, cell sodium content was measured by atomic absorption spectroscopy. Added alone, bafilomycin A1 (100 nm) failed to change cytoplasmic pH but it caused an increase of cytoplasmic sodium concentration which occurred within 10 min. It is likely that the rise of cytoplasmic sodium concentration was responsible for the stimulation of active sodium-potassium transport which occurred in bafilomycin A1-treated cells as judged by a 50% increase of ouabain sensitive potassium (86Rb) uptake. In bafilomycin A1-treated cells, but not in control cells, dimethylamiloride (DMA) inhibited ouabain-sensitive potassium (86Rb) uptake in a dose-dependent manner with an IC50 of ∼2 μm. DMA (10 μm) also prevented the increase of cytoplasmic sodium caused by bafilomycin A1. Added alone, DMA (10 μm) failed to change cytoplasmic sodium content but reduced cytoplasmic pH by ∼0.4 pH units. In cells that first received 10 μm DMA, the subsequent addition of bafilomycin A1 (100 nm) caused a further cytoplasmic pH reduction of ∼0.3 pH units. Taken together, the results suggest H+-ATPase might contribute to the regulation of basal cytoplasmic pH in cultured NPE. In the presence of bafilomycin A1, Na-H exchanger activity appears to be stimulated, so stabilizing cytoplasmic pH but resulting in an increase of cytoplasmic sodium concentration and consequent stimulation of active sodium-potassium transport. A purified peptide from Androctonus australis Garzoni venom (AaG) affects selectively a K+-current recorded from cerebellum granular cells. This current is characterized by fast activating and inactivating kinetics similar to an IA-type current. Addition of 2 μm peptide Aa1 (from Androctonus australis, toxin 1) to the external side of the channel suppressed completely and in a selective manner the IA-type current, with an IC50 value of 130 nm, whereas in the same conditions, the other potassium current, identified as delayed rectifier (Id), was not affected. Additionally, we show that another partially purified peptide (III-12) from the same venom was able to block reversibly both K+-currents. The effect of l-arginine on transepithelial ion transport was examined in cultured M-1 mouse renal cortical collecting duct (CCD) cells using continuous short circuit current (I SC ) measurements in HCO3 /CO2 buffered solution. Steady state I SC averaged 73.8 ± 3.2 μA/cm2 (n= 126) and was reduced by 94 ± 0.6% (n= 16) by the apical addition of 100 μm amiloride. This confirms that the predominant electrogenic ion transport in M-1 cells is Na+ absorption via the epithelial sodium channel (ENaC). Experiments using the cationic amino acid l-lysine (radiolabeled) as a stable arginine analogue show that the combined activity of an apical system y+ and a basal amino acid transport system y+L are responsible for most cationic amino acid transport across M-1 cells. Together they generate net absorptive cationic amino acid flux. Application of l-arginine (10 mm) either apically or basolaterally induced a transient peak increase in I SC averaging 36.6 ± 5.4 μA/cm2 (n= 19) and 32.0 ± 7.2 μA/cm2 (n= 8), respectively. The response was preserved in the absence of bath Cl (n= 4), but was abolished either in the absence of apical Na+ (n= 4) or by apical addition of 100 μm amiloride (n= 6). l-lysine, which cannot serve as a precursor of NO, caused a response similar to that of l-arginine (n= 4); neither L-NMMA (100 μm; n= 3) nor L-NAME (1 mm; n= 4) (both NO-synthase inhibitors) affected the I SC response to l-arginine. The effects of arginine or lysine were replicated by alkalinization that mimicked the transient alkalinization of the bath solution upon addition of these amino acids. We conclude that in M-1 cells l-arginine stimulates Na+ absorption via a pH-dependent, but NO-independent mechanism. The observed net cationic amino acid absorption will counteract passive cationic amino acid leak into the CCD in the presence of electrogenic Na+ transport, consistent with reports of stimulated expression of Na+ and cationic amino acid transporters by aldosterone. We here report on studies on the frog skin epithelium to identify the nature of its excretory H+ pump by comparing transport studies, using inhibitors highly specific for V-ATPases, with results from immunocytochemistry using V-ATPase-directed antibodies. Bafilomycin A1 (10 μm) blocked H+ excretion (69 ± 8% inhibition) and therefore Na+ absorption (61 ± 17% inhibition after 60 min application, n= 6) in open-circuited skins bathed on their apical side with a 1 mm Na2SO4 solution, ``low-Na+ conditions' under which H+ and Na+ fluxes are coupled 1:1. The electrogenic outward H+ current measured in absence of Na+ transport (in the presence of 50 μm amiloride) was also blocked by 10 μm bafilomycin A1 or 5 μm concanamycin A. In contrast, no effects were found on the large and dominant Na+ transport (short-circuit current), which develops with apical solutions containing 115 mm Na+ (``high-Na+ conditions'), demonstrating a specific action on H+ transport. In immunocytochemistry, V-ATPase-like immunoreactivity to the monoclonal antibody E11 directed to the 31-kDa subunit E of the bovine renal V-ATPase was localized only in mitochondria-rich cells (i) in their apical region which corresponds to apical plasma membrane infoldings, and (ii) intracellularly in their neck region and apically around the nucleus. In membrane extracts of the isolated frog skin epithelium, the selectivity of the antibody binding was tested with immunoblots. The antibody labeled exclusively a band of about 31 kDa, very likely the corresponding subunit E of the frog V-ATPase. Our investigations now deliver conclusive evidence that H+ excretion is mediated by a V-ATPase being the electrogenic H+ pump in frog skin. Received: 21 May 1996/Revised: 24 December 1996  相似文献

The effect of high irradiance (HI, photosynthetically active photon flux density of 1 300 μmol m−2 s−1) on net photosynthetic rate (P N), chlorophyll fluorescence parameters, and xanthophyll cycle components were studied in fruit tree bayberry leaves. HI induced the photoinhibition and inactivation of photosystem 2 (PS2) reaction centres (RCs), which was characterized by decreased P N, maximum yield of fluorescence after dark adaptation (Fm), photochemical efficiency of PS2 (Fv/Fm) and quantum yield of PS2 (ΦPS2), and increased reduction state of QA (1-qP) and non-photochemical quenching (NPQ). Initial fluorescence (F0) showed a decrease after the first 2 h, and subsequently increased from the third hour exposure to HI. Furthermore, a greater increase in the ratio (Fi-F0)/(Fp-F0) which is an expression of the proportion of the QB non-reducing PS2 centres, whereas a remarked decrease in the slope of Fi to Fp which represents the rate of QA reduction was observed in leaves after HI exposure. Additionally, HI caused an increase in the pool size of the xanthophyll cycle pigments and sustained elevated contents of zeaxanthin (Z), antheraxanthin (A), and de-epoxidation state (DES) at the end of the irradiation period. During HI, decreased Fm, Fv/Fm, ΦPS2, NPQ, slope of Fi to Fp, V+A+Z, and DES, and increased F0, 1-qP, ratio (Fi-F0)/(Fp-F0), and V were observed in dithiothreitol (DTT)-fed leaves compared to control ones under the same conditions. Hence photoinhibition caused by HI in bayberry was probably attributed to inactivation of PS2 RCs, and photoprotection from photodamage were mainly related to the xanthophyll cycle-dependent heat dissipation in excess photons.  相似文献

The osmotic pressure equation for nonideal, associating systems of the type nA +mB ? AnBm, has been derived, by using the assumption yA nB m/yA nyB m = 1. This treatment can also be applied to related associations such as nA + mB ? AB + AB2 + A2B + …. From osmotic pressure experiments on the pure reactants it is possible to obtain the molecular weights (MA and MB) of the reactants and also the virial coefficients (BAA and BBB) of the reactants. The osmotic pressure of a nonreacting mixture of A and B can be calculated from these measurements. It can be used along with osmotic pressure measurements on equilibrium mixtures of A and B to obtain expressions containing the equilibrium constant (or constants) and the cross-virial coefficients (BAB and BBA). Several procedures are described for the evaluation of the equilibrium constant (or constants) and the BAB or BBA terms. It appears that this procedure is a general one which is applicable to associations of the type nA + mB ? AB + A2B + AB2 + …. By correcting for nonideal behavior, one should then be able to apply it to any method available for analyzing ideal associations of the types considered here. In addition it is possible, subject to certain restrictions, to analyze associations of the type 3A + B ? A2 + AB.  相似文献

Smooth muscle cells isolated from the secondary and tertiary branches of the rabbit mesenteric artery contain large Ca2+-dependent channels. In excised patches with symmetrical (140 mm) K+ solutions, these channels had an average slope conductance of 235 ± 3 pS, and reversed in direction at −6.1 ± 0.4 mV. The channel showed K+ selectivity and its open probability (P o ) was voltage-dependent. Iberiotoxin (50 nm) reversibly decreased P o , whereas tetraethylammonium (TEA, at 1 mm) reduced the unitary current amplitude. Apamin (200 nm) had no effect. The channel displayed sublevels around 1/3 and 1/2 of the mainstate level. The effect of [Ca2+] on P o was studied and data fitted to Boltzmann relationships. In 0.1, 0.3, 1.0 and 10 μm Ca2+, V 1/2 was 77.1 ± 5.3 (n= 18), 71.2 ± 4.8 (n= 16), 47.3 ± 10.1 (n= 11) and −14.9 ± 10.1 mV (n= 6), respectively. Values of k obtained in 1 and 10 μm [Ca2+] were significantly larger than that observed in 0.1 μm [Ca2+]. With 30 μm NS 1619 (a BKCa channel activator), V 1/2 values were shifted by 39 mV to the left (hyperpolarizing direction) and k values were not affected. TEA applied intracellularly, reduced the unitary current amplitude with a K d of 59 mm. In summary, BKCa channels show a particularly weak sensitivity to intracellular TEA and they also display large variation in V 1/2 and k. Summary 1. The purpose of this study was (a) to identify if astrocytes show a similar non-Nernstian depolarization in low K+ or low Ca2+ solutions as previously found in human glial and glioma cells, and (b) to analyze the influence of the K+ conductance on the membrane potential of astrocytes.2. The membrane potential (Em) and the ionic conductance were studied with whole-cell patch-clamp technique in neonatal rat astrocytes (5–9 days in culture) and in human glioma cells (U-251MG).3. In 3.0 mM K+, Em was –75 ± 1.0 mV (mean ± SEM,n=39) in rat astrocytes and –79 ± 0.7 mV (n=5) in U-251MG cells. In both cell types Em changed linearly to the logarithm of [K+]0 between 3.0 and 160 mM K+. K+ free medium caused astrocytes to hyperpolarize to –93 ± 2.7 mV (n=21) and U-251MG cells to depolarize to –27 ± 2.1 mV (n=3).4. The I-E curve did not show inward rectification in astrocytes at this developmental stage. The slope conductance (g) exhibited only a small decrease (–19%) in K+ free solution and no significant change in 160 mM K+.5. Ba2+ (1.0 mM) depolarized astrocytes to –45 ± 2.9 mV (n=11), decreasing the slope conductance (g) by 42.4 ± 8.3% (n=11). Ca2+ free solution depolarized astrocytes to –53 ± 3.4 mV (n=12) and resulted in a positive shift of the I-E curve, increasing g by 15.3 ± 8.2% (n=8).6. Calculations indicated that a block of K+ channels explains the depolarizing effect of Ba2+. The effects of K+ free or Ca2+ free solutions on Em can be explained by a transformation of K+ channels to non-specific leakage channels. That astrocytes show a different reaction to low K+ than glioma cells can be related to the lack of inwardly rectifying K+ channels in astrocytes at this developmental stage.  相似文献

Large Conductance Ca2+-Activated K+ Channels in Human Meningioma Cells   总被引:2,自引:0,他引:2  
Cells from ten human meningiomas were electrophysiologically characterized in both living tissue slices and primary cultures. In whole cells, depolarization to voltages higher than +80 mV evoked a large K+ outward current, which could be blocked by iberiotoxin (100 nm) and TEA (half blocking concentration IC50= 5.3 mm). Raising the internal Ca2+ from 10 nm to 2 mm shifted the voltage of half-maximum activation (V 1/2) of the K+ current from +106 to +4 mV. Respective inside-out patch recordings showed a voltage- and Ca2+-activated (BK Ca ) K+ channel with a conductance of 296 pS (130 mm K+ at both sides of the patch). V 1/2 of single-channel currents was +6, −12, −46, and −68 mV in the presence of 1, 10, 100, and 1000 μm Ca2+, respectively, at the internal face of the patch. In cell-attached patches the open probability (P o ) of BK Ca channels was nearly zero at potentials below +80 mV, matching the activation threshold for whole-cell K+ currents with 10 nm Ca2+ in the pipette. Application of 20 μm cytochalasin D increased P o of BK Ca channels in cell-attached patches within minutes. These data suggest that the activation of BK Ca channels in meningioma cells does not only depend on voltage and internal Ca2+ but is also controlled by the cytoskeleton. Recombinant frequency was compared with nucleotide distance in crosses involving markers in either the PRM or the cy region of phage λ. For each pair of markers, we performed reciprocal four-factor crosses of the following types: (I) A+m1 +m2-B- x A-m1 -m2+B+; and (II) A+m1 -m2+B- x A-m1 +m2-B+. In crosses of type I, the frequency of A+m1 +m2+B+ recombinants among total (selected) A+B+ progeny was directly proportional to nucleotide distance between m1 and m2 in the range from 3 to 160 nucleotides. When less than three nucleotides separated m1 and m2, the measured yields of m1+m2+ recombinants were significantly depressed.

We also found that the frequency of A+m1 +m2+B+ recombinants among total A+B+ progeny was significantly lower (about 10-fold on the average) in crosses of type II than in the corresponding crosses of type I. Since mismatch correction should yield A+m1 +m2+B+ recombinants with approximately equal frequencies in type I and II crosses, we suggest: (1) that most m1+m2+ recombinants produced in type I crosses must arise from the formation of heteroduplex structures with a discontinuity (in the source of genetic information) between sites m1 and m2, and (2) that mismatch correction is not a major pathway for production of recombinants for close markers in normal λ infection.

The outer sulcus epithelium was recently shown to absorb cations from the lumen of the gerbil cochlea. Patch clamp recordings of excised apical membrane were made to investigate ion channels that participate in this reabsorptive flux. Three types of channel were observed: (i) a nonselective cation (NSC) channel, (ii) a BK (large conductance, maxi K or K Ca ) channel and (iii) a small K+ channel which could not be fully characterized. The NSC channel found in excised insideout patch recordings displayed a linear current-voltage (I-V) relationship (27 pS) and was equally conductive for Na+ and K+, but not permeable to Cl or N-methyl-d-glucamine. Channel activity required the presence of Ca2+ at the cytosolic face, but was detected at Ca2+ concentrations as low as 10−7 m (open probability (P o ) = 0.11 ± 0.03, n= 8). Gadolinium decreased P o of the NSC channel from both the external and cytosolic side (IC50∼ 0.6 μm). NSC currents were decreased by amiloride (10 μm− 1 mm) and flufenamic acid (0.1 mm). The BK channel was also frequently (38%) observed in excised patches. In symmetrical 150 mm KCl conditions, the I-V relationship was linear with a conductance of 268 pS. The Goldman-Hodgkin-Katz equation for current carried solely by K+ could be fitted to the I-V relationship in asymmetrical K+ and Na+ solutions. The channel was impermeable to Cl and N-methyl-d-glucamine. P o of the BK channel increased with depolarization of the membrane potential and with increasing cytosolic Ca2+. TEA (20 mm), charybdotoxin (100 nm) and Ba2+ (1 mm) but not amiloride (1 mm) reduced P o from the extracellular side. In contrast, external flufenamic acid (100 μm) increased P o and this effect was inhibited by charybdotoxin (100 nm). Flufenamic acid inhibited the inward short-circuit current measured by the vibrating probe and caused a transient outward current. We conclude that the NSC channel is Ca2+ activated, voltage-insensitive and involved in both constitutive K+ and Na+ reabsorption from endolymph while the BK channel might participate in the K+ pathway under stimulated conditions that produce an elevated intracellular Ca2+ or depolarized membrane potential. A microsomal (Na++ K++ Mg2+)ATPase preparation from sugar beet roots was used. The activation by simultaneous addition of Na+ and K+ at different levels was examined in terms of steady state kinetics. The observed data can be summarized in the following way: 1. The apparent affinity between the enzyme and the substrate MgATP depends on the ratio between Na+ and K+. At low Na+ concentration (below 5 mM), the apparent Km decreases with increasing concentrations of K+ (1–20 mM). At 5 mM Na+, the K+ level does not change the apparent Km, while at Na+ levels above 10 mM, the apparent Km between enzyme and substrate increases with increasing concentration of K+. 2. When the MgATP concentration is kept constant, homotropic cooperativity (concerning one type of ligand) and heterotropic cooperativity (concerning different types of ligands) exist in the activation by Na+ and K+. The Na+ binding is cooperative with different Km values and Hill coefficients (n) in the presence of low and high concentration of K+. At low Na+ level (< 5 mM). a negative cooperativity exists for Na+ (nNa < 1) which is more pronounced in the presence of high [K+]. When the concentration of Na+ is raised the negative cooperativity disappears and turns into a positive one (nNa > 1). Only K+ binding in the presence of low [Na+] shows cooperativity with a Hill coefficient that reflects changes from negative to positive homotropic cooperativity with increasing concentrations of K+ (nK < 1 → nK > 1). In the presence of [Na+] > 10 mM, the changes in nk are insignificant. 3. A model is proposed in which one or two different K sites and one or two Na sites control the catalytic activity, with multiple interactions between Na+, K+ and MgATP. 4. In the presence of Na+ (< 10 mM), K+ is probably bound to two K sites, one of which translocates K+ through the membrane by an antiport Na+/K+ mechanism. Organic spacer cations in layered 2D (A1)2(A2)n?1BnX3n+1 (where A1 is an organic cation acting as a spacer between the perovskite layers, A2 is a monovalent cation, e.g., Cs+,CH3NH3+, CH(NH2)2+) perovskite materials improve the long‐term stability of the resulting solar cells, but hamper their power conversion efficiency due to poor carrier generation/transportation. Rational guidelines are thus required to enable the design of organic spacer cations. Herein, mixed A1 cations are employed in layered 2D perovskites to investigate the interplay between alkylamine cations and unsaturated alkylamine cations. It is revealed that alkylamine spacer cations are able to facilitate precursor assembly, which results in the orientated growth of perovskite crystals. Unsaturated alkylamine cations further lead to reduced exciton binding energy, which improves carrier pathway in the 2D perovskites. By mixing both cations, substantially improved open circuit voltage is observed in the resultant photovoltaic cells with the efficiency of 15.46%, one of the highest one based on (A1)2(A2)3Pb4I13 layered 2D perovskites. The generality of the design principle is further extended to other cation combinations.  相似文献

This paper introduces a new type of Cayley graphs for building large-scale interconnection networks, namely WGn2m\mathit{WG}_{n}^{2m}, whose vertex degree is m+3 when n≥3 and is m+2 when n=2. A routing algorithm for the proposed graph is also presented, and the upper bound of the diameter is deduced as ⌊5n/2⌋. Moreover, the embedding properties and maximal fault tolerance are also analyzed. Finally, we compare the proposed networks with some other similar network topologies. It is found that WGn2m\mathit{WG}_{n}^{2m} is superior to other interconnection networks because it helps to construct large-scale networks with lower cost.  相似文献
