应用电镜技术研究了东方扁虾(Thenus orientalis)精子发生的全过程,精原细胞呈椭圆形,其染色质分布较均匀,线粒体集中于细胞一端形成“线粒体区”。初级精母细胞较大,染色质凝聚成块,次级精母细胞核质间常出现大的囊泡,胞质内囊泡丰富而线粒体数量却明显减少,早期精细胞核发生极化、解聚,部分胞质被抛弃。中期精细胞外观呈金字塔形,分为三区;正在形成的顶体位于塔顶,核位于塔基部,居间的细胞质基质内富含膜复合物,后期精细胞顶体进一步分化。形成顶体帽和内、外顶体物质等三个结构组份。成熟精子核呈盘状或碗状,具有5-6条内部充满微管的辐射臂。  相似文献

Testicular samples were collected to describe the ultrastructure of spermiogenisis in Alligator mississipiensis (American Alligator). Spermiogenesis commences with an acrosome vesicle forming from Golgi transport vesicles. An acrosome granule forms during vesicle contact with the nucleus, and remains posterior until mid to late elongation when it diffuses uniformly throughout the acrosomal lumen. The nucleus has uniform diffuse chromatin with small indices of heterochromatin, and the condensation of DNA is granular. The subacrosome space develops early, enlarges during elongation, and accumulates a thick layer of dark staining granules. Once the acrosome has completed its development, the nucleus of the early elongating spermatid becomes associated with the cell membrane flattening the acrosome vesicle on the apical surface of the nucleus, which aids in the migration of the acrosomal shoulders laterally. One endonuclear canal is present where the perforatorium resides. A prominent longitudinal manchette is associated with the nuclei of late elongating spermatids, and less numerous circular microtubules are observed close to the acrosome complex. The microtubule doublets of the midpiece axoneme are surrounded by a layer of dense staining granular material. The mitochondria of the midpiece abut the proximal centriole resulting in a very short neck region, and possess tubular cristae internally and concentric layers of cristae superficially. A fibrous sheath surrounds only the axoneme of the principal piece. Characters not previously described during spermiogenesis in any other amniote are observed and include (1) an endoplasmic reticulum cap during early acrosome development, (2) a concentric ring of endoplasmic reticulum around the nucleus of early to middle elongating spermatids, (3) a band of endoplasmic reticulum around the acrosome complex of late developing elongate spermatids, and (4) midpiece mitochondria that have both tubular and concentric layers of cristae. Spermiogenesis, in particular the head differentiation of Diplometopon zarudnyi, was studied at the ultrastructural level by Transmission Electron Microscope (TEM). The process includes acrosomal vesicle development, nuclear elongation, chromatin condensation and exclusion of excess cytoplasm. In stage I, the proacrosomal vesicle occurs next to a shallow fossa of the nucleus, and a dense acrosomal granule forms beneath it. This step commences with an acrosome vesicle forming from Golgi transport vesicles; simultaneously, the nucleus begins to move eccentrically. In stage II, the round proacrosomal vesicle is flattened by projection of the nuclear fossa, and the dense acrosomal granule diffuses into the vesicle as the fibrous layer forms the subacrosomal cone. Circular manchettes surrounded by mitochondria develop around the nucleus, and the chromatin coagulates into small granules. The movement of the nucleus causes rearrangement of the cytoplasm. The nucleus has uniform diffuse chromatin with small indices of heterochromatin. The subacrosome space develops early, enlarges during elongation, and accumulates a thick layer of dark staining granules. In stage III, the front of the elongating nucleus protrudes out of the spermatid and is covered by the flat acrosome; coarse granules replace the small ones within the nucleus. One endonuclear canal is present where the perforatorium resides. In stage IV, the chromatin concentrates to dense homogeneous phase. The circular manchette is reorganized longitudinally. Summary

In Cerastoderma glaucum, Sertoli cells are rich in lipids, glycogen and lysosomes, and premeiotic cells exhibited nuage, a prominent Golgi complex and endoplasmic reticulum cisternae encircling the nucleus. The Golgi complex gives rise to proacrosomal vesicles during mid-spermiogenesis, and the round acrosomal vesicle, with a dense fibrillar core, migrates laterally while linked to the plasma membrane as it develops the subacrosomal material. In its final position, the vesicle becomes cap-shaped (0.6 μm) and differentiates into apical light and basal dense regions. The elongated and helicoidal nucleus (8–9.9 μm) has a thin tip (0.3 μm) that invades the subacrosomal space, and in the midpiece (0.8 μm) two of the four mitochondria extend laterally to the nucleus (1.5–2.1 μm). In Spisula subtruncata, Sertoli cells are rich in lipids, glycogen and phagocytosed sperm. Premeiotic cells exhibit nuage, a prominent Golgi complex that gives rise to proacrosomal vesicles from the leptotene stage and a flagellimi that is extruded at zygotene. The acrosomal vesicle forms during the round spermatid stage and differentiates into a large and dense basal region and an apical light region. It then migrates while linked to the plasma membrane by its apical pole. Development of the subacrosomal perforatorium is associated with nuage materials and endoplasmic reticulum vesicles. The mature cap-shaped (0.6 μm) acrosomal vesicle exhibits a large apical and irregular region with floccular contents and a basal dense region. The round nucleus becomes barrel-shaped (1.5 μm) and the midpiece (0.8 μm), with four mitochondria, contains a few glycogen particles.  相似文献

The Golgi complex of the spermatid consists of a central aggregation of minute vacuoles, partially surrounded by a lamellar arrangement of flattened vesicles. In the formation of the acrosome, one or more moderately dense homogeneous granules arise within vacuoles of the Golgi complex. The coalescence of these vacuoles and their contained granules gives rise to a single acrosomal granule within a sizable membrane-limited vacuole, termed the acrosomal vesicle. This adheres to the nuclear membrane and later becomes closely applied to the anterior two-thirds of the elongating nucleus to form a closed bilaminar head cap. The substance of the acrosomal granule occupies the narrow cleft between the membranous layers of the cap. The caudal sheath is comprised of many straight filaments extending backward from a ring which encircles the nucleus at the posterior margin of the head cap. Sperm from the crayfish, Pacifastacus leniusculus, resemble other reptantian sperm in that they are composed of an acrosome, subacrosomal region, nucleus, membrane lamellar complex, and spikes which radiate from the nuclear compartment. The acrosome (PAS positive vesicle) can be subdivided into three regions: the apical cap, crystalline inner acrosomal material, and outer acrosomal material which is homogeneous except for a peripheral electron dense band. The nucleus contains uncondensed chromatin and bundles of microtubules which project into the spikes. The orientation of the microtubule bundles relative to the nuclear envelope near the base of the subacrosomal region suggests that the nuclear envelope may function in the organization of the spike microtubules.  相似文献

The ultrastructural features of spermatogenesis were investigated in the hermaphroditic sea star Asterina minor. The primordial germ cells in the genital rachis contain small clusters of electron-dense material (nuage material) and a stack of annulate lamellae. They also have a flagellum and basal body complex situated close to the Golgi complex. After the development of the genital rachis into the ovotestis, spermatogenic cells increase in number and differentiation begins. Nuage material is observed in spermatogonia, but it gradually disappears in spermatocytes. The annulate lamellae do not exist beyond the early spermatogonial stage. By contrast, a flagellum and basal body complex are found throughout spermatogenesis. The Golgi-derived proacrosomal vesicles appear in the spermatocyte and coalesce to form an acrosomal vesicle in the early spermatid. The process of acrosome formation is as follows: (1) a lamella of endoplasmic reticulum (ER) continuous with the outer nuclear membrane encloses the posterior portion of the acrosomal vesicle; (2) the vesicle attaches to the cell membrane with its anterior portion; (3) periacrosomal material accumulates in the space between the acrosomal vesicle and the ER; (4) the nucleus proper changes its features to surround the acrosome; (5) amorphous, electron-dense material is deposited under the electron-dense disk; and (6) the nucleus forms a hollow opposite the electron-dense material.  相似文献

The spermatozoon of Chiton marginatus is a long uniflagellate cell displaying structural features of “modified sperm.” The nucleus presents a conical shape with a long apical cylindrical extension. The chromatin is homogeneously dense. Scattered inside the condensed nucleus, a few nuclear lacunae are visible. The acrosomal complex is lacking. Some mitochondria are located in a laterofrontal structure side by side with the nucleus. The typical midpiece is absent. The cytoplasm forms a thin layer around the nucleus and the mitochondria. The proximal centriole is in a basal nuclear indent. The distal centriole serves to form the axoneme tail with the usual microtubular pattern. During nuclear maturation, the early spermatid nucleus is spherical and contains fine granular chromatin patches. The nuclear envelope shows a deposit of dense material at the base of the nucleus, forming a semicircular invagination occupied by a flocculent mass. In middle spermatid stage, the chromatin gets organized in filaments, coiled as a hank, attached over the inner surface of the basal thickening of the nuclear envelope. The nucleus starts to elongate anteroposteriorly. At the pointed apical portion of the spermatid, a group of microtubules is observed seeming to impose external pressure to the nucleus giving rise to the long apical nuclear point. The mitochondria have a basal position. Late spermatids have an elongated conical nucleus. The chromatin filaments are further condensed, and lacunae appear inside the nucleus. Sperm of the prawn Sicyonia ingentis were studied cytochemically and ultrastructurally. Striking cytological differences were noted between these natantian sperm and previously studied reptantian sperm. In general, the S. ingentis sperm are composed of a spherical main body that is partially encompassed by a morphologically diverse cap region, from which extends a single appendage or spike. The main body houses an uncondensed, Feulgen-positive nuclear region that is partially surrounded by a cytoplasmic band. A single layer of small, 600 Å, vesicles lines the periphery of the cytoplasmic band. Large membranous vesicles extend from the inner surface of the cytoplasmic band into the nuclear region. The nucleus is separated from the cap or acrosomal complex by a dense plate and a highly organized crystalline lattice, which is composed of geometric squares that are approximately 350 Å in dimension. The cap region also contains convoluted membrane pouches; a central granular core; spherical bodies; an electron-dense, saucer-shaped plate; and a large anterior granule. The convoluted membrane pouches and anterior granule are periodic acid-Schiff (PAS) positive. The anterior granule also demonstrates RNAase-stable red fluorescence with acridine orange staining. A spiralled spike, approximately 6 μm long, extends from the anterior end of the cap. The cap and spike are bound by a double membrane, which results from the fusion of the plasma membrane and the convoluted pouch membrane. The sperm's acrosome is thought to be composed of the two PAS-positive cap components and the spike.  相似文献

Spermiogenesis in the aplysiid, Aplysia kurodai (Gastropoda, Opisthobranchia) was studied by transmission electron microscopy, with special attention to acrosome formation and the helical organization of the nucleus and the other sperm components. In the early spermatid, the periphery of the nucleus differentiates into three characteristics parts. The first part is that electron-dense deposits accumulate on the outer nuclear envelope. This part is destined to be the anterior side of the sperm because a tiny acrosome is organized on its mid-region at the succeeding stage of spermiogenesis. The second part, in which electron-dense material attaches closely to the inner side of the nuclear envelope, is the presumptive posterior side. A centriolar fossa is formed in this part and the axoneme of the flagellum extends from the fossa. A number of lamellar vesicles derived from mitochondria assemble around the axoneme and form the flagellum complex. The third part is recognized by the chromatin which condenses locally along the inner nuclear envelope. During development of the spermatid, this part extends to form a spiral nucleus accompanied by chromatin condensation and formation of microtubular lamellae outside the extending nucleus.
Finally, in the mature sperm, a tiny, spherical acrosomal vesicle is detected at the apex. The slender nucleus, overlapping both the primary and secondary helices which are composed of different structural elements, winds around the flagellum axoneme.  相似文献

A comparative ultrastructural study of spermatogenesis was performed in the bivalve molluscs Pitar rudis and Chamelea gallina (Veneridae) from Turkey. Sertoli cells appeared to be rich in glycogen, lipid droplets and germ-cell phagolysosomes. Premeiotic cells exhibited nuage and a flagellum, with the Golgi complex and the rough endoplasmic reticulum originating proacrosomal vesicles during the pachytene stage. In round spermatids, the acrosomal vesicle migrated linked to the plasma membrane. In P. rudis, the acrosomal vesicle base formed a thin expansion that attached to the nuclear apex and was associated with development of the perforatorium. The cap-shaped acrosomal vesicle then differentiated into external and internal regions, and also into a small apical light region, although some cells exhibited an apical extension of the external component. On the contrary, two lateroapical light pouches developed in C. gallina. During spermiogenesis, chromatin became fibrillar and then condensed while the nucleus turned conical shaped in P. rudis or slightly curved in C. gallina. In P. rudis, the midpiece contained glycogen and four mitochondria, although five mitochondria were sometimes observed, whereas in C. gallina the midpiece contained four mitochondria. Comparison with other members of Veneroida shows a common ectaquasperm type, but novel findings in acrosome biogenesis.  相似文献

The course of acrosomal differentiation observed during spermiogenesis in two starfishes shows that the central components of the mature acrosome are produced by Golgi activity. In the early spermatid, small Golgi-derived vesicles enter the hydrated acrosomal mass and appear to contribute their membrane constituents to the acrosomal-membrane precursor elements. A single lamella of smooth endoplasmic reticulum and fine-fibrillar material associated with it surround the membraneprecursor complex. In a drastic reorganization by which the spermatid acquires antero-posterior symmetry, the acrosome becomes embedded in the anterior part of the nucleus directly beneath the plasma membrane. All the other organelles congregate in the posterior cytoplasm; a thin layer of cytoplasm persists around the sides of the nucleus. During late spermiogenesis two additional acrosomal components become increasingly conspicuous. One is the layer of fine-fibrillar material associated with the smooth endoplasmic reticular vesicles surrounding the Golgi-derived elements. This material is finally pushed towards the center of the sperm head by a late accretion of fibrous product which appears to be synthesized throughout spermiogenesis by the ribosomes, and accumulates around the anterior part of the acrosome as the cytoplasmic matrix diminishes.  相似文献

Summary Some aspects of spermiogenesis have been studied in the testis of the teiid lizard Cnemidophorus lemniscatus lemniscatus by electron microscopy. Shortly after the acrosomal vesicle is lodged in a nuclear concavity of the spermatid, a dense granule differentiates in the center of the subacrosomal space. It is cone-shaped and shows a longitudinal striation. Its base applies to the acrosomal membrane and, through this, to the acrosomal granule. Its rounded vertex causes a depression of the nuclear membranes which, initially juxtaposed, separates at this point to form a vesicle. The granule develops and becomes a rod when spermiogenesis is advanced and the subacrosomal space has taken the form of a secondary cap. The rod is cylindrical, retains its original striation and has a convex acrosomal end. It encloses the vesicle formed by the nuclear envelope in its base and follows the apex of the nucleus. Meanwhile, the acrosomal granule loses its identity and the acrosomal cap is filled with a dense substance, in which a fringe of translucent material differentiates. This fringe lies in the dorsal and apical margins of the acrosome and is incompletely divided by longitudinal crests of the dense acrosomal substance. A projection of the Sertoli cell forms an accessory cap which envelops the acrosome and is in turn covered by the cytoplasm of the spermatid, constituting an intricate association. Two reflex membranes underlie the plasmalemma in the outer surface of the projection of the Sertoli cell. They are continuous with one another at their ends and with the cell membrane in the edge of pores. In the peripheral cytoplasm of the spermatid facing the accessory cap, numerous microtubules run longitudinally.
