Nuclear polyhedrosis in tracheal cells caused by the hypertrophy strain of a nuclear polyhedrosis virus (HNPV) has many morphological and developmental features that distinguish it from that caused by the typical strain (TNPV). The most obvious difference is the morphogenic sequence due to the relatively slow virogenic development in HNPV-infected cells, in which are found extensive membranous profiles similar to viral envelopes, electron dense granules, large fibrous bodies, and microtubules. These structures also occur in TNPV-infected cells but are far less abundant and conspicuous. Fibrous bodies found in the cytoplasm and nucleus appear to be morphologically identical. Cellular distortion in a hypertrophied tracheal cell is seen as tearing of mestracheon folds between cells, separation of septate desmosomes, and attenuation in the cellular sheath.  相似文献

A strain of a cytoplasmic-polyhedrosis virus causes the formation of crystalline inclusions almost entirely in the nucleus, and only rarely in the cytoplasm, of the midgut epithelial cells of the silkworm Bombyx mori. It also differs from the typical strain in causing the hypertrophy of the nucleoli and the formation of dense reticulum and spherical bodies in the nucleus. The virus particles and the virogenic stromata of the new strain resemble those of the typical strain. The cytoplasmic inclusions contain virus particles, while the nuclear inclusions do not. When the infected larvae are kept at 30°C for 15 hr or at 35°C for 3–15 hr, the nuclear inclusions break up into particles of 70–250 nm in diameter. The particles are dispersed in the cells but not present in the spaces previously occupied by the decomposed inclusions.  相似文献

Ultrastructural responses of soybean looper cells of various tissues infected with Pseudoplusia includens icosahedral virus (PIIV), a newly characterized RNA virus [Y. C. Chao, H. A. Scott, and S. Y. Young (1983)J. Gen. Virol.64, 1835–1838], were studied in situ. Most cells of fat body and epidermis consistently contained virus particles and associated cytopathic structures. Virus particles, corresponding to those of purified PIIV in morphology and size, always occurred in the cytoplasm either in membrane-bound virogenic stroma and/or freely in the ground cytoplasm. Virogenic stroma, which appeared to be distinct from those induced by other insect viruses, consisted of electron-dense matrix material, in which virus particles were embedded, and membranous vesicles, 70 or 80 nm in diameter, containing nucleic acid-like fibrils. Virus particles in virogenic stroma occurred as hexagonally arranged crystalline arrays made up primarily of homogeneously dense particles, while those in the ground cytoplasm were dispersed randomly and had an electron-lucent central core. Extremely large numbers of virus particles were also located in noncellular cuticle layers of the integument.  相似文献

The ultrastructure, development, and histopathology of a new cytoplasmic polyhedrosis virus of Aedes cantator are described. Virus particles measure 70 nm in diameter, are icosahedral in shape, and consist of a central electron-dense core surrounded by a capsid with six projections. Occlusion bodies are irregular in size (0.5–3.0 μm) and shape and contain several virus particles. Virus particles are assembled within an interconnecting network of fine filaments and are occluded by the deposition of a proteinaceous crystal around groups of mature virus particles within a virogenic stroma. Infections are confined to cells of the cardia, gastric ceca, and posterior portion of the midgut, which hypertrophy and frequently lyse. Infected larvae die during the fourth larval instar or as pupae. The prevalence of infection in natural field populations is less than 1%.  相似文献

BmMNPV, a Nucleopolyhedrovirus isolated from infected Bombyx mori (L.) larvae in Paraná State--Brazil, was used to inoculate healthy 5th-instar B. mori larvae and examine the infection on central nervous system (CNS) cells. Samples of nervous tissue were removed from the infected insects, at different sampling times, and processed for cytopathology studies by light and transmission electron microscopy using routine techniques. The experiment included both inoculated and non-inoculated larvae (control). BmMNPV infection was detected on the 5th day after inoculation in CNS cells. Initially, infection was characterized by nuclear hypertrophy and the presence of virogenic stroma, in which the progeny virions were produced. Virions are enveloped and occluded into protein crystal, the polyhedra. Lyses of infected CNS cells were undetected; however, free mature polyhedra were seen in spaces inside the CNS. These polyhedra possibly came from trachea that penetrate the CNS and its cells, which are susceptible to BmMNPV and lyses after infection. We conclude that the tracheal system is responsible for disseminating BmMNPV infection in B. mori CNS and that the tracheal branches allow non-occluded virions to pass through the blood-brain barrier.  相似文献

The pathology of a Baculovirus (BV) in Aedes triseriatus was studied. The virus infected the cardia, gastric caeca, and the entire stomach of larval midgut epithelium. The progression of the disease was similar to that of other Baculoviruses of the nuclear polyhedrosis virus (NPV) type. Rodshaped nucleocapsids were formed within a Feulgen-positive virogenic stroma and along the nuclear envelope. These nucleocapsids were enveloped by a membranous material and occluded randomly in small irregular and polyhedral proteinic inclusions. The disease differed from other BVs of the NPV type in that the small proteinic inclusions gradually coalesced as they grew, forming large fusiform inclusions.  相似文献

The larva ofPseudaletia unipuncta (Haworth) is susceptible to 2 nuclear polyhedrosis viruses (NPV), the typical (TNPV) and the hypertrophy (HNPV) strains. The interference between the 2 viruses was studied primarily in the tracheal and fat tissues. When both viruses were microfed simultaneously, no larva had mixed virus infections and most were infected with TNPV. In order to obtain 50% infection by each virus the inoculum of HNPV had to be increased by 1000 polyhedra/ml at the LD50 value of TNPV. Mixed infection resulted when TNPV was inoculated 1 and especially 2 days after HNPV inoculation. In mixed infection, each virus attacked certain areas of the tracheae instead of random cells. These results indicated an interference or antagonism between the 2 viruses. The possible cause for the interference is discussed.
Résumé La larve dePseudaletia unipuncta (Haworth) est sensible à 2 virus à polyèdres nucléaires (NPV), la souche typique (TNPV) et la souche causant une hypertrophie (HNPV). L'interférence entre ces 2 virus a été étudiée essentiellement dans les trachées et le corps adipeux. Si les deux virus sont inoculés simultanément par micro ingestion aucune larve ne présente d'infection mixte et la plupart d'entre elles est infectée par le TNPV. Pour obtenir 50% d'infection par chacun des virus, l'inoculum de HNPV doit être accru de 1000 polyèdres par ml par rapport à l'inoculum provoquant la DL 50 du TNPV. Une infection mixte est enregistrée quand le TNPV est inoculé 1 jour et de préférence 2 jours après l'introduction du HNPV. Dans ce cas d'infection mixte chaque virus attaque certaines régions des trachées et non pas des cellules réparties au hasard. Ces résultats prouvent une interférence ou un antagonisme entre les 2 virus, dont la cause est discutée.

Euglena gracilis strain (Z) cells were synchronized under photoautotrophic conditions using a 14 hour light:10 hour dark regimen. The cells grew during the light period (growth phase) and divided during the following 10 hour period either in the dark or in the light (division phase). Changes in morphology of the pyrenoid and in the distribution of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) within the chloroplasts were followed by immunoelectron microscopy during the growth and division phases of Euglena cells. Epon-embedded sections were labeled with an antibody to the holoenzyme followed by protein A-gold. The immunoreactive proteins were concentrated in the pyrenoid, and less densely distributed in the stroma during the growth phase. During the division phase, the pyrenoid could not be detected and the gold particles were dispersed throughout the stroma. Toward the end of the division phase, the pyrenoid began to form in the center of a chloroplast, and the immunoreactive proteins started to concentrate over that rudimentary pyrenoid. During the growth phase, small areas rich in gold particles, called `satellite pyrenoid,' were observed, in addition to the main pyrenoid. From a comparison of photosynthetic CO2-fixation with the total carboxylase activity of Rubisco extracted from Euglena cells in the growth phase, it is suggested that the carboxylase in the pyrenoid functions in CO2-fixation in photosynthesis.  相似文献

For a better understanding of virus x host interactions, transmission electron microscopy was used to characterize the intrahaemocoelic infection of Anticarsia gemmatalis larval haemocytes by A. gemmatalis M nucleopolyhedrovirus (AgMNPV). At 12 h post-infection (h p.i.), we observed nuclear hypertrophy, budded virus assembling, and protrusion towards the cytoplasm, virion envelopment, and accumulation of fibrillar aggregates in the cytoplasm. Around 24 h p.i., fibrillar aggregates also appeared inside nuclei of infected cells. By 48 h p.i., virogenic stroma and polyhedra were visualised in nuclei and at 72 h p.i., widespread infection in haemocytes was observed. Cell remnants and free polyhedra were phagocytosed by granular haemocyte 1 and plasmatocytes. Entire cells were phagocytosed only by plasmatocytes. Necrosis of infected cells was quite common, suggesting a putative cytotoxic response. Granular haemocyte 1 presented a more exuberant protrusion of budded viruses in comparison to other haemocytes. All types of haemocytes were shown to be infected, and the intense virus replication in some of these cells reveals the importance of haemolymph for AgMNPV spread in its natural host, a critical factor for permissiveness.  相似文献

Recently, we showed that the Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) VP80 protein is essential for the formation of both virion types, budded virus (BV) and occlusion-derived virus (ODV). Deletion of the vp80 gene did not affect assembly of nucleocapsids. However, these nucleocapsids were not able to migrate from the virogenic stroma to the nuclear periphery. In the current paper, we constructed a baculovirus recombinant with enhanced-green fluorescent protein (EGFP)-tagged VP80, allowing visualization of the VP80 distribution pattern during infection. In baculovirus-infected cells, the EGFP-VP80 protein is entirely localized in nuclei, adjacent to the virus-triggered F-actin scaffold that forms a highly organized three-dimensional network connecting the virogenic stroma physically with the nuclear envelope. Interaction between VP80 and host actin was confirmed by coimmunoprecipitation. We further showed that VP80 is associated with the nucleocapsid fraction of both BVs and ODVs, typically at one end of the nucleocapsids. In addition, the presence of sequence motifs with homology to invertebrate paramyosin proteins strongly supports a role for VP80 in the polar transport of nucleocapsids to the periphery of the nucleus on their way to the plasma membrane to form BVs and for assembly in the nuclear periphery to form ODVs for embedding in viral occlusion bodies.  相似文献

在昆虫种群中常常发生杆状病毒持续性感染,持续性感染可转变为增殖性感染而引发病毒流行病。本研究拟建立一个病毒-细胞模型,用于探讨杆状病毒持续性感染分子机制。将甜菜夜蛾核多角体病毒(Spodoptera exigua nucleopolyhedrovirus,SeMNPV)在其宿主细胞Se301中无稀释连续传代以弱化病毒毒力,用传至第8代的SeMNPV感染Se301细胞后,虽然大部分细胞因病毒感染而裂解,但仍有少量细胞存活并可传代培养,该传代细胞株命名为P8-Se301。P8-Se301细胞在对数生长期的群体生长倍增时间为58~65 h,慢于Se301细胞的生长速度。光镜和电镜观察表明,少部分P8-Se301细胞具有病毒发生基质、病毒粒子、多角体等病毒感染特征。终点稀释法和感染中心测定表明,4.14%±0.99%的P8-Se301细胞可持续释放感染性的子代病毒,但该子代病毒在Se301细胞中的复制速度较野生型SeMNPV慢。结果表明,P8-Se301细胞呈现典型的持续性感染特征。  相似文献

Subcellular responses to infection by Race 3 of Heterodera glycines in susceptible (''Lee'') and resistant (''Forrest'' and ''Bedford'') soybean cultivars were compared. Syncytial formation, initiated in susceptible as well as resistant soybean cultivars, was characterized by wall perforations, dense cytoplasm, and increased endoplasmic reticulum, In susceptible plants, syncytia developed continuously until nematode maturity. This included hypertrophy of nuclei, increase of rough endoplasmic reticulum in early stages of infection, and formation of wall ingrowths at a late stage of infection. In the resistant reaction in Forrest, a necrotic layer surrounded syncytium component cells demarcating them from surrounding normal cells and leading to syncytial necrosis. Wall appositions were prominently formed near the necrotic layer, and the cytoplasm of the syncytium component cells was extremely condensed. The whole syncytium became necrotic at a late stage of infection. Bedford had nuclear degeneration prior to cytoplasmic degradation. Chromatin was often scattered throughout the syncytial cytoplasm. Finally the whole syncytium became degenerated with plasmalemma completely detached from the syncytial cell walls. The differences in resistant responses reflect a difference in genetic composition of the soybean cultivars tested.  相似文献

Tumor–stroma interactions are referred to as essential events in tumor progression. There has been growing attention that bone marrow-derived mesenchymal stem cells (BMSCs) can travel to tumor stroma, where they differentiate into tumor-associated fibroblast (TAF)-like cells, a predominant tumor-promoting stromal cell. However, little is definitively known about the contributors for this transition. Here, using an in vitro direct co-culture model of colon cancer cells and BMSCs, we identify that colon cancer cells can induce adjoining BMSCs to exhibit the typical characteristic of TAFs, with increased expression of α-smooth muscle actin (α-SMA). Importantly, the present data also reveals that activated Notch signaling mediates transformation of BMSCs to TAFs through the downstream TGF-β/Smad signaling pathway.  相似文献

Evidence that aromatase expression in tumor-associated breast stroma is elevated, provides a rationale for use of aromatase inhibitors (AIs) in breast cancer treatment. However, regulation of local aromatase expression in cancer-free breast stroma is poorly understood. Recent clinical work indicates that stromal cells in dense breast tissue tend to express higher levels of aromatase than their counterpart from non-dense tissue. Consistent with the clinical observation, our cell culture-based study indicated that cell density, cell shape, and extracellular matrix (ECM) significantly induced stromal aromatase expression via a distinct signal transduction pathway. In addition, we identified a number of cell surface markers that are commonly associated with aromatase-expressing stromal cells. As mammographic density is one of the strongest and most prevalent risk factors for breast cancer, these findings provide a potential mechanistic link between alterations in tissue composition of dense breast tissue and increased stromal aromatase expression. Further exploration of the in vitro model system may advance understanding of an important problem in breast cancer biology.  相似文献
