本文用高效液谱方法,检测了正常成年大鼠子宫内膜胞浆中游离酪氨酸(Tyr)含量,发现其浓度随动情周期的不同而发生波动,在动情前期最低,动情后期最高。同时检测了子宫内膜胞浆中孕激素受体(PR)含量的变化,以动情前期最高,动情后期最低。同样呈有规律的变化。表明酪氨酸与孕激素受体含量在子宫内膜胞浆中浓度的变化呈负相关,实验进一步证实,当向子宫腔内局部注射酪氨酸时,酪氨酸明显减少动情前期、动情期、间情期大鼠子宫内膜胞浆孕激素受体含量,但对动情后期孕激素受体含量无显著影响,这些结果提示酪氨酸可能影响孕激素受体的含量。  相似文献

用ELISA法测定大鼠下丘脑、卵巢、子宫在动情周期中钙调素(Calmodulin)的含量变化,并用免疫组化法(Immunohistochemical technique)研究了大鼠动情周期中子宫内钙调素的分布。结果为:(1)处在动情期的大鼠,其子宫、卵巢、下丘脑中钙调素的含量明显高于间情期、动情前期及动情后期的含量。其顺序为动情期>动情前期>动情后期>间情期。其中以下丘脑、卵巢的含量变化较急骤。提出钙调素含量变化与雌激素的含量一致,由此推测钙调素对维持卵巢正常功能,子宫生长发育有密切关系。(2)经免疫组化定位,不论处在间情期或动情期的子宫,钙调素的分布在肌层与内膜层均有,以内膜层较多,特别是在动情期的分布更高。此与钙调素含量测定的结果一致。  相似文献

在假妊大鼠,口服酪氨酸可使卵巢静脉血中孕酮含量减少,子宫静脉血中前列腺素含量增加,卵巢对~(125)I-hCG 的结合力降低。在切除一侧卵巢的大鼠,口服酪氨酸可使对侧卵巢代偿性增生及子宫重量增加受抑制。同时也明显地抑制卵巢 hCG 受体功能,使卵巢对~(125)I-hCG 的结合力,hCG 受体数目和亲和力降低。本实验结果提示,酪氨酸可直接或间接作用于卵巢,抑制黄体功能,使孕酮分泌减少,从而可能具有抗生育作用。  相似文献

本工作应用氚标记配基交换法测定了早孕大鼠子宫细胞核雌二醇与孕酮-受体复合物水平,同时也观察了外周血浆这两种甾体激素浓度的变化。实验结果表明,早孕期间子宫细胞核中雌二醇和孕酮受体水平发生了显著的改变,分别在受孕第3天和第4天出现雌二醇和孕酮受体高峰(均为~20,000结合位点/细胞);同时也分别在这两天观测到血浆雌二醇和孕酮浓度高峰(0.08毫微克/毫升和29毫微克/毫升)。我们得出的结论与其他研究工作者报道的一致,即血循环中卵巢甾体激素水平的升高导致子宫细胞核大量集聚激素-受体复合物,从而加速基因表达,刺激子宫的生长和细胞分化,为胚泡的着床造成适当的子宫环境。  相似文献

用免疫细胞化学LSAB法,兔抗人孕酮受体抗体(适用于小鼠)显示小鼠卵巢、输卵管、子宫和阴道中的孕酮受体。结果显示:未注射RU486的动物的子宫与阴道内孕酮受体丰富,输卵管中含量较少,卵巢中未见阳性反应物质。RU486注射4天后,子宫与阴道中孕酮受体明显减少;注射后7天,输卵管、子宫和阴道都为阴性反应。该结果显示了孕酮受体在雌性小鼠生殖系统内的分布特点,提示RU486封闭孕酮受体的最佳时间为用药后第7天。  相似文献

为了验证雌激素作用存在于完整的和去卵巢兔胚泡着床过程中,本文用~3H—雌二醇交换法测定了早期妊娠 D_3、D_5、D_7子宫组织的雌二醇受体浓度,并与去卵巢后仅补充孕酮的 D_7子宫作比较。结果显示:在 D_3、D_5、D_7的子宫细胞浆和细胞核中都存在雌二醇受体。妊娠7天去卵巢兔的子宫细胞核中雌二醇受体量与妊娠7天卵巢完整的兔作比较,二者无明显差异。这说明去卵巢后子宫细胞核中雌激素受体浓度并未受影响,因此雌激素作用依然存在。关于胚泡是否有雌激素受体的问题,本工作在胚龄5、6、7天胚泡中未测得雌二醇受体。据此,胚泡着床时,雌激素的作用可能主要通过母体子宫组织而发挥其生理效应。  相似文献

内皮素—1对大鼠排卵前卵泡颗粒细胞产生孕酮的影响   总被引:7,自引:1,他引:6  
本文用离体细胞体外孵育法研究了内皮素-1(ET)对大鼠排卵前卵泡颗粒细胞孕酮生成的影响及其作用机理。结果发现,ET能显著抑制hCG刺激下的孕酮产生,抑制作用在浓度为10-8mol/L时,即有显著意义(P<0.05,n=6),至10-7mol/L时则有非常显著的意义(P<0.01,n=6);不同浓度ET(10-7—10-7mol/L),对颗粒细胞基础孕酮的产生无明显影响。进一步研究表明,ET对hCG刺激下孕酮生成的抑制作用,在用免抗人内皮素抗血清(ET-A)1:1000及cAMP后能明显被逆转。实验中还观察到,ET使颗粒细胞LH/hCG受体数下降,亲和力降低。本文结果提示,ET可能为卵巢内的一种局部调节肽,通过作用于ET受体,干扰LH/hCG受体功能和cAMP生成而抑制颗粒细胞孕酮的产生。  相似文献

本文研究了大豆黄酮对大鼠乳腺发育及其对垂体生长激素(GH)和催乳素(PRL)分泌的影响,结果表明:1)胃饲或皮下注射大豆黄酮能显著提高去卵巢大鼠或正常处女大鼠乳腺重量和乳腺DNA、RNA含量;2)胃饲或皮下注射大豆黄酮能显著提高正常处女大鼠血清GH和PRL含量,3)大豆黄酮能显著提高大鼠乳腺,垂体和下丘脑胞浆雌二醇受体的数目,并显著提高乳腺孕酮受体的数目,体外受体竞争结合试验结果表明:大豆黄酮与大  相似文献

Analysis of rat uterine cytosol by means of immobilized antibody discloses the presence of two distinct high affinity, low capacity estradiol binding components. One of these is readily saturable by the antiestrogen tamoxifen (TS = tamoxifen sensitive), the other is tamoxifen insensitive (TI). Only TS-estradiol binding shows positive cooperativity at low ligand concentration. TS but not TI is lost when frozen tissue is thawed at 4°C then refrozen and stored for an additional 12 hours. Experiments with ovariectomized rats show that TI is formed only in the presence of estradiol. In the estrus cycle TS increases in the order: metestrus, diestrus, proestrus and estrus. The quantity of TI is the same in metestrus and diestrus and also the same but fractionally higher in proestrus and estrus.  相似文献

An oral administration of antifertility drug Lyndral (17 alpha-ethinyl estradiol) at doses 5 micrograms/rat/a week for 3 consecutive weeks altered the internal biochemical milieu of uterus, showing a cyclic variation in both acid and alkaline phosphatase of the uterine fluid with high levels being present at proestrus and estrus in control rats. In Lyndral treated rats acid phosphatase showed a tendency to increase in estrus and metestrus rats, whereas alkaline phosphatase increased significantly during proestrus and estrus stages of the cycle. All these altered phosphatase levels in uterine fluid, after Lyndral treatment, gives some insight into the hormonal sensitiveness of these enzymes.  相似文献

The percentage of labeled cells in the uterine luminal epithelium of cycling mice showed the different zonal distributions at each stage of estrous cycle after cumulative labeling with 3H-thymidine for 36 hr. It was estimated that the proliferating fraction in the epithelium at proestrus, estrus, metestrus, and diestrus was 100%, 100%, 40% and 5%, respectively. The percentage of labeled cells in the uterine luminal epithelium of cycling mice treated with progesterone remained below 10% level for at least 20 hr after injections of progesterone. Total labeling was attained in the uterine epithelium of castrated mice by the administration of estradiol-17beta. On the other hand, the cell proliferation in the uterine epithelium of castrated mice treated with estradiol and progesterone was markedly suppressed and the percentage of labeled cells remained approximately at 35%. The remaining cell population, however, still showed the mitotic potency when mice received estradiol. It is suggested from this study that the effect of progesterone is to suppress the epithelial cell proliferation and transfer cells into resting cell fraction which is still evoked to proliferate as the effect of estradiol and that a key factor controlling epithelial proliferation in mouse uterus during the estrous cycle is proliferating fraction rather than cell cycle time.  相似文献

Cyclic changes in estrogen receptor expression in the uterine tissue of 60 female dogs were evaluated, using an immunohistochemical technique on formalin-fixed paraffin-embedded sections. The expression of estrogen receptors in the uterine horns, body and cervix was quantified by means of an immunohistochemical score. A negative correlation was found between staining scores in the uterine horns and serum progesterone levels. Generally, staining scores in the uterine horns were highest during proestrus, declined during estrus and were lowest during early metestrus. During anestrus high staining scores for estrogen receptors were observed, indicating sensitivity for estrogens in a sexual quiescence stage. Compared with the uterine horns, high staining scores were found in the uterine body and cervix during estrus and metestrus. No positive staining for estrogen receptors was detected in 1 pregnant uterus. Fluctuations in estrogen receptors were more pronounced in endometrial stroma cells than in epithelial cells of the uterine horns. The importance of stromal cells in the sexual cyclicity of the canine uterus should not be underestimated when studying uterine endocrinology and pathology.  相似文献

Previous work has shown that the immature rat uterus contains epidermal growth factor (EGF) receptors and that tissue levels of this receptor are increased by the administration of exogenous estrogens. This study was undertaken to determine if estrogen administration also elevated EGF receptor levels in the mature animal and if the growth factor receptor levels varied in concert with endogenous estrogens throughout the estrous cycle. In the mature, castrate rat administration of estradiol, but not non-estrogenic steroids, causes a 2-3-fold elevation of uterine EGF receptors as judged by ligand binding. This increase is maximum in 18 h and is due to an increase in the number of binding sites. In cycling animals EGF receptor levels are low at metestrus, rise at diestrus, reach a maximum (approximately twice metestrus values) at proestrus, and then return at estrus to metestrus levels. These changes in EGF receptor levels parallel changes in plasma estrogens and occupied nuclear estrogen receptor reported by other workers. These results indicate that uterine EGF receptors are increased by exogenous estrogens in both mature and immature animals, and support a physiological role for estrogens in the regulation of this growth factor receptor.  相似文献

Treating mature rat uterine cytosol with dextran coated charcoal (DCC) for 2 hrs at 0-4C in the absence of ligand causes the subsequently formed receptor-estradiol complex to be stable at 37C. Receptor binding is increased by the DCC treatment for uteri excised at metestrus or diestrus but remains nearly unchanged for uteri obtained at proestrus or estrus. Results suggest that the DCC removes or inactivates factor(s) present in the cytosol which render the receptor complex thermolabile.  相似文献

Analysis of rat uterine cytosol for Tamoxifen binding reveals that the saturable binding sites are only partially inhibited by estradiol-17β. Partial thermal denaturation of the cytosol at 30° C for 2 h 30 allows the characterization of a high affinity (Kd = 3.3 × 10?9M) saturable Tamoxifen class of binding sites insensitive to estradiol-17β while remaining sensitive to the antiestrogens CI628 and Nafoxidine. The uterine concentration of these binding sites is lower in the uterus of immature or castrated animals, increases from metestrus to proestrus and reaches a peak on the day of estrus.  相似文献

Serum androgen levels in the bitch increase during proestrus and remain elevated until metestrus. To find out whether androgens can have a direct impact on the canine uterus, androgen receptors (AR) were identified immunohistochemically in uterine tissue. Androgen receptor distribution in the uterine horns, body and cervix was described during different cycle stages, during pregnancy and in the postpartum period. Nuclear staining for AR was observed in cells of the surface epithelium, glandular ducts, basal glands and stroma of the endometrium, and in myometrial smooth muscle cells. In addition, cytoplasmic staining was observed in epithelial cells from proestrus to early metestrus, when the cells were secretory active, and in stroma cells during pregnancy, suggesting a role for androgens in decidualization. During pregnancy and in the postpartum period nuclear staining for AR was nearly absent. During the estrus cycle stroma cells stained with higher intensities for AR than epithelial cells, supporting the idea that stroma cells mediate some effects of steroid hormones on epithelial cells in the genital tract. In contrast with earlier findings on estrogen receptor-alpha and progesterone receptors, no significant changes in androgen receptor expression were observed during the estrus cycle. Few correlations were found between the staining for AR and serum levels of the sex steroids. The present findings suggest that there is a basal expression of AR in the canine uterus throughout the estrus cycle that may not be influenced by sex steroid hormones.  相似文献

Progesterone and estradiol play an important role in the regulation of lung functions such as ventilation and vasoconstriction. The genomic actions of progesterone and estradiol are mediated by their nuclear receptors: progesterone receptors (PR) and estrogen receptors (ER). These actions are linked to interactions between steroid receptors and some cofactors that function as coactivators or corepressors. In this work we determined the content of PR isoforms, ER-beta, one coactivator (SRC-1), and one corepressor (SMRT) in the lung of both female rats during the estrous cycle and intact males by Western blot. The rat lung presented a higher content of PR-A than that of PR-B during the estrous cycle. The highest content of both PR isoforms was observed on the day of proestrus whereas the lowest one was found on the day of estrus. In contrast, the content of ER-beta was the lowest on the day of proestrus and it increased at estrus. The content of SRC-1 and SMRT increased on the day of diestrus. SRC-1 content decreased at proestrus and estrus, while SMRT content decreased at proestrus and increased again at estrus. In the lung of adult male rats the content of PR isoforms, ER-beta and SMRT was lower than in that of females, whereas the content of SRC-1 was similar in both sexes. Our results suggest a sexual dimorphism in the content of PR, ER-beta, and SMRT in the rat lung as well as a relation of their content to the physiological levels of progesterone and estradiol.  相似文献
