Binding of the cationic tetra(tributylammoniomethyl)-substituted hydroxoaluminum phthalocyanine (AlPcN4) to bilayer lipid membranes was studied by fluorescence correlation spectroscopy (FCS) and intramembrane field compensation (IFC) methods. With neutral phosphatidylcholine membranes, AlPcN4 appeared to bind more effectively than the negatively charged tetrasulfonated aluminum phthalocyanine (AlPcS4), which was attributed to the enhancement of the coordination interaction of aluminum with the phosphate moiety of phosphatidylcholine by the electric field created by positively charged groups of AlPcN4. The inhibitory effect of fluoride ions on the membrane binding of both AlPcN4 and AlPcS4 supported the essential role of aluminum-phosphate coordination in the interaction of these phthalocyanines with phospholipids. The presence of negative or positive charges on the surface of lipid membranes modulated the binding of AlPcN4 and AlPcS4 in accord with the character (attraction or repulsion) of the electrostatic interaction, thus showing the significant contribution of the latter to the phthalocyanine adsorption on lipid bilayers. The data on the photodynamic activity of AlPcN4 and AlPcS4 as measured by sensitized photoinactivation of gramicidin channels in bilayer lipid membranes correlated well with the binding data obtained by FCS and IFC techniques. The reduced photodynamic activity of AlPcN4 with neutral membranes violating this correlation was attributed to the concentration quenching of singlet excited states as proved by the data on the AlPcN4 fluorescence quenching.  相似文献

Summary Characteristics of inorganic carbon assimilation by photosynthesis in seawater were investigated in six species of the Fucales (five Fucaceae, one Cystoseiraceae) and four species of the Laminariales (three Laminariaceae, one Alariaceae) from Arbroath, Scotland. All of the algae tested could photosynthesise faster at high external pH values than the uncatalysed conversion of HCO 3 - to CO2 can occur, i.e. can use external HCO 3 - . They all had detectable extracellular carbonic anhydrase activity, suggesting that HCO 3 - use could involve catalysis of external CO2 production, a view supported to some extent by experiments with an inhibitor of carbonic anhydrase. All of the algae tested had CO2 compensation concentrations at pH 8 which were lower than would be expected from diffusive entry of CO2 supplying RUBISCO as the initial carboxylase, consistent with the operation of energized entry of HCO 3 - and / or CO2 acting as a CO2 concentrating mechanism. Quantitative differences among the algae examined were noted with respect to characteristics of inorganic C assimilation. The most obvious distinction was between the eulittoral Fucaceae, which are emersed for part of, or most of, the tidal cycle, and the other three families (Cystoseiraceae, Laminariaceae, Alariaceae) whose representatives are essentially continually submersed. The Fucaceae examined are able to photosynthesise at high pH values, and have lower CO2 compensation concentrations, and lower K1/2 values for inorganic C use in photosynthesis, at pH 8, than the other algae tested. Furthermore, the Fucaceae are essentially saturated with inorganic C for photosynthesis at the normal seawater concentration at pH 8 and 10°C. These characteristics are consistent with the dominant role of a CO2 concentrating mechanism in CO2 acquisition by these plants. Other species tested have characteristcs which suggest a less effective HCO 3 - use and CO2 concentrating mechanism, with the Laminariaceae being the least effective; unlike the Fucaceae, photosynthesis by these algae is not saturated with inorganic C in normal seawater. Taxonomic and ecological implications of these results are considered in relation to related data in the literature.  相似文献

Summary The use of the locust (Schistocerca americana) hindleg in postural control was examined in animals that stood on a repeatedly swayed vertical substrate. Myograms were recorded from leg muscles and the angle of the femoro-tibial joint was monitored photographically. Two discrete strategies were observed,; in compensatory reactions the hindleg was held in place, while in flexion reactions, the leg was moved, most often to complete flexion of the femoro-tibial joint. Tightly coupled, rhythmic bursting occurred in the flexor and levator muscles of the leg during compensatory reactions. Bursting was initiated repeatedly when the substrate was being pulled away from the animal. Bursting was correlated with subsequent decreases in the rate of change of the femorotibial joint angle. Compensatory and flexion reactions occurred preferentially in different ranges of joint angles: most often, compensatory reactions occurred in the midrange, while flexion reactions were elicited in the extremes of joint angle. These differences may be due to the mechanical advantages of the tibial muscles and the leg may be moved to full flexion because of a locking mechanism of the flexor muscle tendon. These reactions are compared with known reflexes of hindleg proprioceptors and contrasted with similar responses of vertebrates.  相似文献

A relation between gene dosage and UDP-glucose:flavonoid 3-O-glucosyl-transferase (UFGT) activity was found in homozygous dominant and recessive parental lines and their F1 progeny for both of the genes An1 and An2. In both F2 crosses, progeny plants could be classified as belonging to groups showing either a low or a medium to high UFGT activity. Test crosses showed that heterozygous and homozygous dominant plants were present throughout the medium- to high-active group. The dosage relation in F2 plants is most probably confounded by the segregation of modifiers. Thermal inactivation experiments indicated that structurally different UFGT enzymes are formed in homozygous dominant lines as well as in lines homozygous recessive for either An1 or An2. Lines homozygous recessive for the gene An4 contain a UFGT with a half-life time at 55° C of less than 8 min, whereas UFGTs from lines homozygous dominant for An4 show a half-life time of 25 min or above, with one exception. This relation was confirmed in the F2 progeny; heterozygotes for An4 showed an intermediate half-life time. It is concluded that An4 might be the structural gene for the enzyme; An1 and An2 are both regulatory genes. UFGT activity in flowerbuds of An4/An4 plants seems to be lower than in an4/an4 plants. Anthers of flowers of an4/an4 lines, however, are virtually devoid of UFGT activity.  相似文献

ABSTRACT. The detailed behavioural mechanisms underlying an instance of compensation for changes in dietary nutrients are described for the first time in an insect. Nymphs of Locusta migratoria L. were given one of four artificial diets on the third day of the fifth instar, and their feeding patterns recorded in detail for 12 h. The diets represented combinations of two protein and two digestible carbohydrate levels (28% and 14% dry weight) presented in an otherwise complete nutrient mix. At the nutrient levels used, locusts regulated their intake of food with respect to protein but not digestible carbohydrate in the diet. They ate more of the lower protein diets by eating the same sized meals more frequently than insects fed on the higher protein diets. Compensation was not complete over the 12 h observation period: insects on the lower-protein diets ingested and absorbed 72% as much nitrogen as those insects fed on the higher-protein diets. Possible physiological mechanisms underlying the behavioural mechanisms are discussed.  相似文献

Vegetative plants of Poa pratensis L. cv. Holt (origin 69°N) raised in short days gave large and significant increases in plant dry weight, plant height and leaf area upon exposure to continuous light, compared with 8-h short days, at essentially identical daily inputs of radiant energy (8-h summer daylight ± low intensity extension). For example, by the fourth harvest (after 26, 34 and 46 days at 21, 15 and 9°C, respectively), the dry weights of plants in long days were 81, 163 and 195% greater than those of the corresponding short-day controls at the respective temperatures. Plant leaf areas in long days were between two and four times as large as control values by the end of the experiment. This was mainly due to increased leaf length caused by long-day stimulation of cell extension and division. However, the photoperiod did not affect the partitioning of assimilates amongst leaves, culms and stolons. Most of these effects could also be brought about by exogenous gibberellin application to plants in short days. However, in contrast to the effect of long days, gibberellin treatment also induced stem internode elongation even in these vegetative plants. Examination by standard growth analysis procedures revealed that the observed increases in relative growth rate were due primarily to increased net assimilation rate followed, several days later, by increases in leaf area ratio when newly-emerged leaves began to constitute a significant proportion of the leaf area. It is concluded that these reactions are of great adaptive significance for growth at the marginal temperatures prevailing at high latitudes.  相似文献

Experiments were conducted to compare the growth and growth compensation of three strains of juvenile Oreochromis niloticus . Ten full sib families (10 replicates) per strain were split and grown under crowded and uncrowded conditions for 3 weeks (the treatment). Both treatments were then grown an additional 12 weeks under less crowded conditions (the compensation). Standard length measurements were made at the end of crowding and the end of compensation. Each replicate included a size-matched control population of a fourth (red) reference strain. ANCOVA with the reference strain used as a cofactor revealed significant strain effects on specific growth throughout the experiment. The reference strain removed most of the random among-replicate error variance as shown by an increase of r 2 from 0.06 to 0.91 when it was included in the statistical models. If the reference fish had not been used, approximately 450 replicate families would have been needed to achieve the sensitivity of the present experiment (a difference of 7% among strains significant at P =0.05). We conclude that the CLSU strain grows significantly more slowly than the Israel and NIFI strains under the experimental conditions, that the crowding effect was essentially eliminated after 12 weeks of compensation, and that the reference strain greatly improved the resolution of the strain-testing experiment.  相似文献

A gene-dosage population was obtained by crossing two genotypes that were duplex for the GBSS allele. Nulliplex, simplex, duplex or triplex/quadruplex plants could be identified by monitoring the segregation of red and blue microspores after staining with iodine. GBSS activity was significantly different for all groups and showed an almost linear dosage effect for the wildtype GBSS gene. A dosage effect was found for amylose content that was not linear. The amylose content was similar for both the duplex and triplex/quadruplex group. Within the simplex group, differences in amylose content were found, which might be due to a different genetic background. There was no linear correlation between GBSS activity and amylose content. A certain level of GBSS activity led to a maximum amount of amylose, and further increase in GBSS activity did not result in a further increase in amylose content. The presence of one or more wildtype GBSS allele(s), and therefore the presence of amylose in the starch granules, had a great influence on the physico-chemical properties of the starch suspensions.  相似文献
