Reproductive behaviour of the catfish,Silurus asotus was studied in temporary waters around paddy fields. Spawning occurred nocturnally during the first week from the initiation of irrigation. In reproductive activities, a male first energetically pursued a female with its head near to the female’s belly (chasing) and then began to cling to the female’s body from the side, bending its tail or head (clinging). Finally the male enfolded the female’s body, with its anus near to the female’s (enfolding). In some cases, 2–4 males pursued a single female and two males enfolded a female at the same time. Although no aggressive behaviour was evident between males, it was always the largest male that could most frequently approach and enfold the female. The mating pair moved a long distance in a ditch, paddy field and/or creek, performing reproductive activities. It is thought that the spawning site and period of spawning of the fish enable the larvae to avoid the danger of predation and to efficiently feed, firstly on plankton and later on larvae of other fishes which become abundant during the irrigation period. Although some eggs and larvae may die due to the drying out or high water temperatures of such unstable temporary waters, scattering eggs may reduce the incidence of the annihilation of the young.  相似文献

The three intra- and extracellular polysaccharide fractions were isolated during the growth phase of Dictyostelium discoideum NC-4, and the change in content of component sugars of four fractions during the culture period was examined. Myxamoebae most extensively contain a polysaccharide fraction extracted with phenol-water (polysaccharide fraction I) in a quantity of about 15–23% per dry cell. After 15 h the uronic acid formed in the polysaccharide fraction I, and the cell, could be aggregated. The glucosamine content in the polysaccharide fraction I reached a maximum as the myxamoebae entered the exponential phase, and a large amount of galactose was produced as the cell entered the stationary phase. The phenol-water extract from the cells of the stationary phase was reacted with concanavalin-A.  相似文献

A sequence-specific antibody that recognizes a portion of the prosegment of human renin precursor was raised and used to provide direct evidence that plasma inactive renin contains the prosequence of renal renin and is therefore probably prorenin rather than an inactivated form of previously active renin. Acute subdural hematoma (ASDH) sometimes occurred in judo because of the bridging veins’ failure by rotation of the brain in the skull. However, the relationship between intracranial behaviour and the motion of the body on occiput impact has not yet been clarified. In this study, we developed an intracranial mechanical model based on multibody dynamics and compared it with experimental results. The results show the importance of modelling bridging veins and cerebral spinal fluid to the relative displacement between brain and skull. The proposed model will contribute to accident analyses or the optimum design of supporting devices.  相似文献

Hepatitis C virus (HCV) is a positive-strand RNA virus that frequently causes persistent infections and is uniquely associated with the development of hepatocellular carcinoma. While the mechanism(s) by which the virus promotes cancer are poorly defined, previous studies indicate that the HCV RNA-dependent RNA polymerase, nonstructural protein 5B (NS5B), forms a complex with the retinoblastoma tumor suppressor protein (pRb), targeting it for degradation, activating E2F-responsive promoters, and stimulating cellular proliferation. Here, we describe the mechanism underlying pRb regulation by HCV and its relevance to HCV infection. We show that the abundance of pRb is strongly downregulated, and its normal nuclear localization altered to include a major cytoplasmic component, following infection of cultured hepatoma cells with either genotype 1a or 2a HCV. We further demonstrate that this is due to NS5B-dependent ubiquitination of pRb and its subsequent degradation via the proteasome. The NS5B-dependent ubiquitination of pRb requires the ubiquitin ligase activity of E6-associated protein (E6AP), as pRb abundance was restored by siRNA knockdown of E6AP or overexpression of a dominant-negative E6AP mutant in cells containing HCV RNA replicons. E6AP also forms a complex with pRb in an NS5B-dependent manner. These findings suggest a novel mechanism for the regulation of pRb in which the HCV NS5B protein traps pRb in the cytoplasm, and subsequently recruits E6AP to this complex in a process that leads to the ubiquitination of pRb. The disruption of pRb/E2F regulatory pathways in cells infected with HCV is likely to promote hepatocellular proliferation and chromosomal instability, factors important for the development of liver cancer.  相似文献

Acidic extracellular pH is a common feature of tumor tissues. We have reported that culturing cells at acidic pH (5.4-6.5) induced matrix metalloproteinase-9 expression through phospholipase D, extracellular signal regulated kinase 1/2 and p38 mitogen-activated protein kinases and nuclear factor-kappaB. Here, we show that acidic extracellular pH signaling involves both pathways of phospholipase D triggered by Ca2+ influx and acidic sphingomyelinase in mouse B16 melanoma cells. We found that BAPTA-AM [1,2-bis(2-aminophenoxy)-ethane-N,N,N',N'-tetraacetic acid tetrakis (acetoxymethyl) ester], a chelator of intracellular free calcium, and the voltage dependent Ca2+ channel blockers, mibefradil (for T-type) and nimodipine (for L-type), dose-dependently inhibited acidic extracellular pH-induced matrix metalloproteinase-9 expression. Intracellular free calcium concentration ([Ca2+]i) was transiently elevated by acidic extracellular pH, and this [Ca2+]i elevation was repressed by EGTA and the voltage dependent Ca2+ channel blockers but not by phospholipase C inhibitor, suggesting that acidic extracellular pH increased [Ca2+]i through voltage dependent Ca2+ channel. In contrast, SR33557, an L-type voltage dependent Ca2+ channel blocker and acidic sphingomyelinase inhibitor, attenuated matrix metalloproteinase-9 induction but did not affect calcium influx. We found that acidic sphingomyelinase activity was induced by acidic extracellular pH and that the specific acidic sphingomyelinase inhibitors (perhexiline and desipramine) and siRNA targeting aSMase/smpd1 could inhibit acidic extracellular pH-induced matrix metalloproteinase-9 expression. BAPTA-AM reduced acidic extracellular pH-induced phospholipase D but not acidic sphingomyelinase acitivity. The acidic sphingomyelinase inhibitors did not affect the phosphorylation of extracellular signal regulated kinase 1/2 and p38, but they suppressed nuclear factor-kappaB activity. These data suggest that the calcium influx-triggered phospholipase D and acidic sphingomyelinase pathways of acidic extracellular pH induced matrix metalloproteinase-9 expression, at least in part, through nuclear factor-kappaB activation.  相似文献
