A total of 2063 mid- to late-instar larval black flies were collected from 64 stream sites in South Carolina and screened for the presence of the trichomycete fungus Harpella melusinae. Sixteen of 18 host species were colonized by H. melusinae on at least one occasion. Prevalence of H. melusinae in larvae of Simulium tuberosum cytospecies "A" was highest in acidic streams with low conductivity, whereas H. melusinae colonized larvae of Simulium verecundum most frequently in slower-moving streams. Ecological conditions, therefore, can serve as predictors of the prevalence of H. melusinae. Prevalence in host larvae was significantly lower in the Piedmont ecoregion than in the Mountain ecoregion. Prevalence did not differ in the host species S. verecundum across ecoregions, suggesting that different prevalences among host species might indicate some host preference. The prevalence of H. melusinae differed significantly between two univoltine host species (Simulium venustum and Prosimulium magnum) at the same site but not between two multivoltine host species (S. tuberosum cytospecies "FG" and S. tuberosum cytospecies "CDE"), suggesting that host life history could be important in determining fungal prevalence.  相似文献

Actinomycetes could be isolated efficiently on a defatted wood powder medium from the guts of various species of termites. The actinomycete flora in the termites' guts depended largely on the area in which the termites naturally occur. In termites from the same area, the actinomycete flora changed depending on the taxonomic difference between termite species. Some actinomycetes isolated from termites' guts grew satisfactorily on lignin-related media, and the others grew on cellulose-related media.  相似文献

Trichomycetes are an ecological group of fungi and protists that colonize the gut lining of invertebrates in aquatic and moist terrestrial habitats. The diversity of this group appears to be high with many new species discovered each year. A new genus of fungal trichomycete, Trifoliellum (Harpellales), is described here with the type species T. bioblitzii. This genus is characterized by having unique, trefoil-shaped asexual spores (trichospores). Another new species, Legeriosimilis halifaxensis, also is described from the same mayfly host, Eurylophella temporalis, collected from the same site near Halifax, Nova Scotia.  相似文献

Unusual microbial xylanases from insect guts   总被引:2,自引:0,他引:2  
Recombinant DNA technologies enable the direct isolation and expression of novel genes from biotopes containing complex consortia of uncultured microorganisms. In this study, genomic libraries were constructed from microbial DNA isolated from insect intestinal tracts from the orders Isoptera (termites) and Lepidoptera (moths). Using a targeted functional assay, these environmental DNA libraries were screened for genes that encode proteins with xylanase activity. Several novel xylanase enzymes with unusual primary sequences and novel domains of unknown function were discovered. Phylogenetic analysis demonstrated remarkable distance between the sequences of these enzymes and other known xylanases. Biochemical analysis confirmed that these enzymes are true xylanases, which catalyze the hydrolysis of a variety of substituted beta-1,4-linked xylose oligomeric and polymeric substrates and produce unique hydrolysis products. From detailed polyacrylamide carbohydrate electrophoresis analysis of substrate cleavage patterns, the xylan polymer binding sites of these enzymes are proposed.  相似文献

Summary Exudate from the phloem of Ricinus communis L. was negatively stained, examined in the electron microscope, and the filamentous components compared with those in fixed, sectioned material. In the exudate, two main fibrillar components were observed. One component has a diameter of 20±0.35 (standard error) nm, the other of 14.1±0.34 nm. This second compoent has projections along its length measuring 5 by 14 nm and spaced at intervals of 6.5–10 nm. Fibrils have been found possessing characteristics of both fibril types, suggesting some structural relationship between the two, possibly an interconvertibility. Several other types of fibrils occurred less frequently in the exudate. The exudate also contains torus-shaped structures measuring 13.5–15 nm in diameter. Sections of mature sieve elements of Ricinus and Acer rubrum L. contain fibrils structurally similar to the 14-nm fibrils from the exudate of Ricinus. Ricinus exudate was also fixed and pelleted in the ultracentrifuge. Thin sections of the pellet afforded cross-sectional views of the 20-nm fibrils, and showed that these fibrils apparently have a solid core. Possible models for the structure of the 20-nm filaments are described.  相似文献

Effective (nitrogen-fixing) root nodules of Oxytropis maydelliana Trautv., O. arctobia Bunge and Astragulus alpinus L. were collected in the high Arctic tundra and subsequently processed for structural studies. The cylindrically-shaped perennial nodules consisted of the following tissues: nodule cortex, nodule meristem, nodular vascular bundles, an active central region with uninfected and infected cells at various stages of development, and a proximal region of senescent cells. The active central region was dark red-coloured due to the presence of the pigment leghemoglobin. The host cells became infected by the growth of infection threads into cells recently derived from the nodule meristem and the subsequent endocytotic release of rhizobia from unwalled membrane-bound regions of the infection thread. The host plasma membrane adjacent to the unwalled regions of infection thread gave rise to the peribacteroid membrane which surrounded the released bacteria. Thus, nodule development and the basic tissue arrangement of the arctic nodules was similar to that of cylindrically-shaped nodules formed on temperate species of legumes.
The arctic legume nodules are unique in having large numbers of lipid droplets present in the cytoplasm of the nodule cortex and uninfected cells of the central active region. Newly infected cells also have lipid droplets. More developed infected cells lack lipid droplets but often contain amyloplasts. Mature differentiated bacteria were spherically-shaped and contained electron-dense inclusions. Electron-dense material was also present in vesicles formed from dilated endoplasmic reticulum and in the peribacteroid space. The lipid droplets present in the host cytoplasm of the nodule cortex and uninfected cells of the central tissue may be storage products which are used to support nitrogen-fixation in nodules growing under cool temperatures of this harsh environment.  相似文献

Summary The ultrastructure of the seminiferous tubules was studied in rats that had been subjected to whole body irradiation on the 19th day of gestation. The seminiferous tubules from 3 months-old irradiated animals are devoid of germ cells and contain only Sertoli cells. Compared with controls of the same age, the seminiferous tubule basal membrane is thickened and multilayered and several alterations are observed in the Sertoli cells. The most characteristic of these alterations are: (a) an abnormal number of nuclear heterochromatin clumps, (b) the presence of numerous cytoplasmic vacuoles and various sized lipid droplets, (c) elaborate interdigitations and junctions between adjacent cells, and (d) the presence of anomalous ectoplasmic specializations disposed perpendicularly to the Sertoli cell membrane.  相似文献

The aim of this study was to determine the fine structure of amylopectin from grain amaranth. Amaranthus amylopectin was hydrolyzed with α-amylase, and single clusters and a group of clusters (domain) were isolated by methanol precipitation. The domain and the clusters were treated with phosphorylase a and then β-amylase to remove all external chains, whereby the internal structure was obtained. The ,β-limit dextrins were analyzed on Sepharose CL 6B. The average DP (degree of polymerization) and peak-DP values of fractions of clusters were 57 and 82, respectively; the values of the domain were 137 and 309, respectively. The unit chain length profiles were analyzed by high-performance anion-exchange chromatography with pulsed amperometric detector (HPAEC–PAD). The results showed that the domain fraction contained 2.2 clusters, and single clusters were composed of 13 chains. The ,β-limit dextrins of the clusters were further hydrolyzed with α-amylase to characterize their building block composition. The average DP of the branched blocks was 11 and they contained on average 2.5 chains. Their average chain length, internal chain length, and degree of branching were approximately 4.3, 2.8, and 14, respectively. A cluster consisted of 6 branched blocks, and the internal chain length between the blocks was 6.8.  相似文献

Z-discs from the dorsal longitudinal indirect flight muscles of the honeybee (Apis mellifera) are perforated with hundreds of triangular-shaped tubes ordered into an hexagonal array. Each tube is surrounded by 80 Å thick rims which incorporate six thin filaments, three from each bordering sarcomere. Although the triangular rims of the tubes are oriented identically in any plane perpendicular to the fibril axis, this orientation changes as the tubes cross the Z-line. The tubes rotate approximately 60 ° about an axis parallel to that of the fibril in passing from one I-Z junction to another.On the basis of filament counting in the A (overlap zone) and I bands of stretched myofibrils, it is concluded that the primary filaments are physically continuous with the Z-lines by material which appears to participate both in the formation of Z-rim substance and the surrounding matrix.Finally, evidence is presented to support the view that filament lattices of adjacent sarcomeres are displaced from one another, so that each thick filament faces the trigonal position of three thick filaments on the other side of the Z-disc.  相似文献
