FJAT-4748 is a bacterial strain isolated from forest soil samples taken from Dongba Valley, Lijiang, Kunming, Yunnan Province, PR China. This strain was identified as Lysinibacillus sp. based on a 16S rRNA gene sequence analysis. FJAT-4748 has been shown to possess antifungal activity against different fungi, including Colletotrichum acutatum, Aspergillus niger, Fusarium solani, Fusarium moniliforme and Fusarium oxysporum. The results of the present study indicate that this antifungal activity results from volatile organic compounds (VOCs) produced by this strain. The observed inhibition rates of VOCs from FJAT-4748 against these fungi were 100%, 100%, 37.20%, 18.94% and 7.64%, respectively. GC-MS analysis identified 24 VOCs from FJAT-4748, which included different categories of compounds, such as aldehydes, ketones, alcohols, aromatic hydrocarbons and alkanes. Of these 24 VOCs, the most abundant compound was 2-ethyl-1-hexanol, which constituted 36.24% of the total VOCs based on the relative peak area. In the in vitro C. acutatum mycelial growth assay, 2-ethyl-1-hexanol exhibited the strongest activity, with an inhibitory rate of 100% using 10?µL/plate of this VOC. The activity of benzaldehyde was lower. 2-decanone showed the weakest activity among the compounds tested. The inhibitory activity of an artificial mixture of three VOCs against the C. acutatum increased with the amount of artificial mixture used. The inhibition rate reached 100% using 30?µL/plate of this artificial mixture in the plate test. Taken together, these results show that the antifungal VOCs produced by Lysinibacillus sp. FJAT-4748 are potentially useful as agents for controlling anthracnose caused by Colletotrichum acutatum.  相似文献

Two antifungal aliphatic compounds, SPM5C-1 and SPM5C-2 with a lactone and ketone carbonyl unit, respectively obtained from Streptomyces sp. PM5 were evaluated under in vitro and in vivo conditions against major rice pathogens, Pyricularia oryzae and Rhizoctonia solani. These compounds were dissolved in distilled water/medium to get the required concentrations. The well diffusion bioassay indicated that the of SPM5C-1 remarkably inhibited the mycelial growth of P. oryzae and R. solani in comparison to SPM5C-2. Though SPM5C-2 showed low antifungal activity against P. oryzae, it was not active against R. solani. Further, SPM5C-1 completely inhibited the growth of P. oryzae and R. solani at concentrations of 25, 50, 75 and 100 μg/ml. Greenhouse experiments revealed that spraying of SPM5C-1 at 500 μg/ml on rice significantly decreased blast and sheath blight development by 76.1% and 82.3%, respectively, as compared to the control with a corresponding increase in rice grain yield.  相似文献

【背景】禾谷镰刀菌(Fusarium graminearum)是一种危害小麦生产的重要病原真菌。【目的】筛选对禾谷镰刀菌具有拮抗活性的链霉菌菌株,为该病原的生物防治提供理论基础。【方法】采用稀释涂布法分离链霉菌,利用平板对峙法筛选高活性拮抗菌株;通过形态学、生理生化特征和16S rRNA基因序列分析确定其分类地位;采用生长速率法分析其发酵条件及无菌发酵液的稳定性;并测定该菌株的防病效果和抑菌谱。【结果】筛选到一株对禾谷镰刀菌具有较强抑制作用的链霉菌菌株21-6,抑菌率为75.2%±2.1%。根据形态学、生理生化特征及16S rRNA基因序列分析,将其鉴定为Streptomyces stelliscabiei。菌株21-6在pH为中性条件下的PDB培养基中培养5 d能够产生更好的抑菌效果。无菌发酵液能够抑制禾谷镰刀菌的菌丝生长和孢子萌发过程。无菌发酵液不受高温、胃蛋白酶、胰蛋白酶及蛋白酶K的影响,耐酸但对碱性条件敏感。发酵液对禾谷镰刀菌侵染小麦胚芽鞘具有抑制效果。菌株21-6具有聚酮合酶pks-pks-基因。此外,该菌株对5种植物病原真菌均具有抑制效果。【结论】链霉菌菌株21-6对禾谷镰刀菌具有较好的生防潜力。  相似文献

由微生物介导的吡啶降解技术是解决高盐吡啶环境污染的经济有效方法之一,开发具有吡啶降解性能且能够耐受高盐分的微生物是该类研究的重要前提。本研究从山西太原钢铁公司焦化废水处理厂活性污泥中分离培养了一株耐盐吡啶降解菌,通过菌落形态和16S rDNA基因系统发育分析,鉴定其为红球菌属(Rhodococcus sp.)的细菌。耐盐性实验结果表明,菌株LV4能够在0%–6%盐度范围内生长,并完全降解初始浓度为500 mg/L的吡啶;但当盐度高于4%时,菌株LV4因其生长变缓而导致吡啶完全降解时间明显延长。扫描电镜结果显示,高盐环境会使菌株LV4的菌体细胞分裂变慢,诱导细胞表面分泌更多的颗粒状胞外聚合物(extracellular polymeric substance, EPS)。当盐度不高于4%时菌株LV4主要依靠EPS中蛋白含量的增加来响应高盐环境的冲击。单因素实验优化发现,菌株LV4在盐度为4%的高盐环境中降解吡啶的最佳条件为温度30℃、pH 7.0、转速为120 r/min (DO 10.30 mg/L)。最优条件下菌株LV4对于初始浓度为500 mg/L的吡啶,在经过12 h的适应期后,...  相似文献

A bioflocculant-producing bacterium isolated from soil was identified as Bacillus sp. and the bioflocculant produced was named MBFF19. Effects of physico-chemical conditions including pH, carbon sources and nitrogen sources on MBFF19 production were studied. Chemical analyses of the purified bioflocculant MBFF19 indicated that it was a sugar-protein derivative, composed of neutral sugar (3.6%, w/w), uronic acid (37.0%, w/w), amino sugars (0.5%, w/w) and protein (16.4%, w/w). The two neutral sugar components were mannose and glucose and the molar ratio was 1.2:1. Infrared spectrophotometry analysis revealed that MBFF19 contained carboxyl, hydroxyl and methoxyl groups in its structural. Flocculating properties of bioflocculant MBFF19 was examined using kaolin, activated carbon and fly coal suspension. Cation supplement had no positive effects on the flocculating activity whereas the presence of Fe3+ inhibited flocculation. Penicillin V acylase was produced, both intracellularly and extracellularly, by Fusarium sp. SKF 235 grown in submerged fermentation. When neopeptone was added to the medium, >95% of the penicillin V acylase was extracellular. In the absence of a complex organic nitrogen source, the fungus produced low levels of totally intracellular penicillin V acylase. MgSO4 was essential for synthesis of the enzyme, which was induced by phenoxyacetic acid and penicillin V. The maximum yield of penicillin V acylase was 430 IU/g dry cell wt. The optimum pH value and temperature for the penicillin V acylase were 6.5 and 55°C, respectively.  相似文献

Interactions between an isolate of the ectomycorrhizal fungus Pisolithus sp. and Afzelia africana Sm. seedlings were studied at the structural and ultrastructural levels. Several different conditions were tested with or without sugar and in a sterile or nonsterile medium. In the growth cabinet, the A. africana/Pisolithus sp. interactions did not produce ectomycorrhizas. A fungal sheath was formed but no Hartig net, and an unusual host epidermal cell wall was observed. Hyphae of Pisolithus sp. induced modifications of epidermal cells of 15-day-old A. africana seedlings indicative of non-mycorrhizal interactions, such as wall thickening, wall ingrowth, papillae formation, degraded host wall material and the presence of intracellular hyphae. Wall ingrowth consisted of depositions of host cell wall materials giving a positive reaction for polysaccharides; however, wall thickenings and papillae showed no homogeneous reactions for polysaccharides. In glasshouse conditions, inocula of Pisolithus sp. in the form of spores or mycelia entrapped in peat-vermiculite added to sterilized soil produced typical ectomycorrhizae only with 6-month-old A. africana seedlings. Under these conditions, no conspicuous cell wall reactions occurred on A. africana roots. The results demonstrate that the establishment of an association between an ectomycorrhizal fungus and a potential host plant is strongly influenced by seedling age and/or environmental conditions. Therefore, in vitro synthesis is not a conclusive demonstration of a symbiotic relationship.  相似文献

Olea europaea preparations are traditionally employed in a variety of troubles, including skin infections. Olive extracts and some of their pure compounds have shown antimicrobial activity in vitro. The present study deals with the antifungal activity of some aliphatic aldehydes from olive fruit [hexanal, nonanal, (E)-2-hexenal, (E)-2-heptenal, (E)-2-octenal, (E)-2-nonenal] against Tricophyton mentagrophytes (6 strains), Microsporum canis (1 strains) and Candida spp. (7 strains). The capability of these substances to inhibit elastase, a virulence factor essential for the dermatophytes colonization, and their cytotoxicity on cultures of reconstructed human epidermis, are also described. Aldehydes tested, inhibited the growth of T. mentagrophytes and M. canis in the range of concentration between <1.9 and 125 microg/ml; the unsaturated aldehydes showed the most broad spectrum of activity in that inhibited all strains tested. None of the aldehydes exhibited activity against Candida spp. strains. (E)-2-octenal and (E)-2-nonenal inhibited the elastase activity in a concentration-dependent manner; the anti-elastase activity suggests an additional target of the antimicrobial activity of these compounds. Aldehydes were devoid of cytotoxicity on cultures of human reconstructed epidermis. The antifungal activity of the aldehydes from olive fruit here reported, substantiates the use of olive and olive oil in skin diseases and suggests that these natural compounds could be useful agents in the topical treatment of fungal cutaneous infections.  相似文献

Nannochloropsis sp. was grown under mixotrophic conditions with 30 mM glucose as carbon source, reaching 507 mg dry wt l(-1) after 8 d. This was 1.4-fold of that obtained under photoautotrophic conditions. Under mixotrophic and photoautotrophic cultivations, the net photosynthetic rate of Nannochloropsis sp. did not change but the respiratory rate increased in the mixotrophic cultivation. The yield of eicosapentaenoic acid was 22 mg l(-1) in the mixotrophic cultivation and 20 mg l(-1) under the photoautotrophic cultivation.  相似文献

【目的】筛选一株海藻酸裂解酶高产菌株,并通过优化产酶条件提高海藻酸裂解酶活性。【方法】以海藻酸钠为唯一碳源的培养基,对福建漳州滨海土壤中的微生物进行筛选和分离,获得海藻酸裂解酶高产菌株;依据形态、生理生化特征及16S rDNA序列分析对目的菌株进行鉴定;然后通过单因素和正交试验对其产酶条件进行优化。【结果】十六烷基吡啶(CPC)染色得到4株透明圈与菌落直径比值(D/d)3的菌株;DNS法测定4菌株发酵液中海藻酸裂解酶活力,其中菌株SH-1的海藻酸裂解酶活性最高,达到315.52 U/mL;经形态、生理生化和16S rDNA测序鉴定,将其命名为Microbulbifer sp. SH-1;通过单因素和正交试验优化,确定该菌株最适产酶培养基为:海藻酸钠10 g/L,NaCl 5 g/L,(NH_4)_2SO_45g/L,MgSO_40.2g/L,K_2HPO_41g/L,FeSO_40.02g/L。对培养条件的进一步优化结果发现,在初始pH 7.5、温度32°C条件下,以1%的接种量将SH-1菌株接入50 mL优化培养基中,240 r/min转速下振荡培养24 h,SH-1菌株产酶最大活性可达757.90 U/mL,比优化前提高了2.4倍。【结论】SH-1最佳产酶条件的建立,为海藻酸裂解酶的大规模制备以及更深层次研究提供了试验基础和理论依据。  相似文献

Two furanoditerpenes, 2α,3α-epoxy-2,3,7,8α-tetrahydropenianthic acid methyl ester (1) and 2α,3α-epoxy-2,3-dihydropenianthic acid methyl ester (2) were isolated and identified from the root of Arcangelisia flava (L.) Merr. The configuration of 1 was determined by X-ray crystallographic analysis and two-dimensional NMR. Fibraurin (3), fibleucin (4), 2β, 3α-dihydroxy-2,3,7,8α-tetrahydropenianthic acid-2,17-lactone (5), p-hydroxybenzaldehyde and vanillin were also isolated and identified by NMR and EI-MS or FAB-MS. The 2β, 3α-dihydroxy-2,3,7,8α-tetrahydropenianthic acid-2,17-lactone (5) showed the highest antifungal activity of the isolated five furanoditerpenes against a white-rot fungus (Trametes versicolor) and a brown-rot fungus (Fomitopsis palustris).  相似文献

Nodulation abilities of bacteria in the subclasses Gammaproteobacteria and Betaproteobacteria on black locust (Robinia pseudoacacia) were tested. Pseudomonas sp., Burkholderia sp., Klebsiella sp., and Paenibacillus sp. were isolated from surface-sterilized black locust nodules, but their nodulation ability is unknown. The aims of this study were to determine if these bacteria are symbiotic. The species and genera of the strains were determined by RFLP analysis and DNA sequencing of 16S rRNA gene. Inoculation tests and histological studies revealed that Pseudomonas sp. and Burkholderia sp. formed nodules on black locust and also developed differentiated nodule tissue. Furthermore, a phylogenetic analysis of nodA and a BLASTN analysis of the nodC, nifH, and nifHD genes revealed that these symbiotic genes of Pseudomonas sp. and Burkholderia sp. have high similarities with those of rhizobial species, indicating that the strains acquired the symbiotic genes from rhizobial species in the soil. Therefore, in an actual rhizosphere, bacterial diversity of nodulating legumes may be broader than expected in the Alpha-, Beta-, and Gammaproteobacteria subclasses. The results indicate the importance of horizontal gene transfer for establishing symbiotic interactions in the rhizosphere.  相似文献

Hydrogen and a bioflocculant could be produced simultaneously by anaerobic culture of Enterobacter sp. BY-29. For production of hydrogen and the bioflocculant by cell culture of the bacterium in batch cultures, cultivation at 37 °C in a medium containing glucose as a carbon source and Polypepton as a nitrogen source was found to be suitable. In continuous production of hydrogen and the bioflocculant by cell culture or immobilized cells of the bacterium, the hydrogen production rate and hydrogen yield by the immobilized cells on porous glass beads in stirred and column reactors were higher than those by the cell culture in a stirred reactor. However, production of the bioflocculant by the cell culture was superior to that by the immobilized cells in continuous production.  相似文献

【目的】从冬季活性污泥和河水底泥中分离筛选得到耐低温好氧反硝化菌,并对影响脱氮的关键因子进行优化,以提高低温条件下的脱氮效果。【方法】采用富集纯化法分离筛选耐低温好氧反硝化菌株。通过形态观察和16S rRNA基因系统发育分析等方法对筛选菌株进行菌种鉴定。以NO3–-N去除率为响应目标,采用Box-Behnken试验设计及响应面回归分析法优化影响脱氮效果的关键因子(C/N、温度、pH和摇床转速),确定最佳培养条件。【结果】从东北寒冷地区冬季河水底泥样品中分离得到的一株耐低温好氧反硝化菌Z6,菌落呈白色半透明圆形,菌体细胞为短杆状,大小为(0.8–1.6)μm×(0.6–0.8)μm,革兰氏染色阴性。与气单胞菌属(Aeromonas)的16S rRNA基因序列高度同源,鉴定该菌为气单胞菌。采用响应面分析方法得到菌株Aeromonas sp. Z6的最佳脱氮条件为:C/N 5.9,温度12°C,pH 6.8,摇床转速155 r/min,在此条件下对NO3-N的去除率为89.72%,与预测值(90.34%)无显著差别。【结论】首次报道气单胞菌(Ae...  相似文献

The performance of a polyurethane (PU) biofilter inoculated with Rhodococcus sp. EH831 was evaluated under different transient loading conditions, such as shutdown, intermittent and fluctuating loading. A mixture of benzene and toluene vapors was employed as model pollutants. When the biofilter was restarted after a 2 week-shutdown, during which neither clean air nor water was supplied, the benzene and toluene removal capacities were rapidly restored after a re-adaptation period of only 1 day. A comparison of the removal capacity under continuous and intermittent loading revealed that constant and periodic loading (8 h on/16 h off per day) and a 2 day-shutdown did not significantly influence the biofilter performance, although the removals of benzene and toluene were relatively unstable and lower under intermittent loading during the initial week. The result of quantitative real-time PCR showed that Rhodococcus sp. EH831 could be maintained during transient loading periods (1010–1011 CFU/g-dry PU) irrespective of the different operating conditions.  相似文献

Three novel compounds with spiro-5, 6-lactone ring skeleton has been isolated from the fermentation broth of Massrison sp. which could be isolated repeatedly from wild Rehmannia glutinosa. Psetariae oryza P-2b was applied to guide fractionation of bioactive compounds produced by Massrison sp. The molecular structures were established by a variety of one- and two-dimensional NMR experiments and the compounds with similar skeleton were reported for the first time from endophytic fungi of terraneous plant. Antifungal and cytotoxic activities of the compounds were tested, compounds 2 and 3 displayed stronger antifungal and cytotoxic activities. The compounds have the potential to be antibiotic against fungal pathogens and tumor cells.  相似文献

A Citrobacter sp., isolated from soil at an effluent treatment plant of a textile and dyeing industry, decolorized several recalcitrant dyes except Bromophenol Blue. More than 90% of Crystal Violet and Methyl Red at 100 M were reduced within 1 h. Gentian Violet, Malachite Green and Brilliant Green lost over 80% of their colors in the same condition, but the percentage decolorization of Basic Fuchsin and Congo Red were less than the others, 66 and 26%, respectively. Decolorization of Congo Red was mainly due to adsorption to cells. Color removal was optimal at pH 7–9 and 35–40 °C. Decolorization of dyes was also observed with extracellular culture filtrate, indicating the color removal by enzymatic biodegradation.  相似文献

Cell-free extracts of Streptomyces sp. DRS I converted ampicillin to cephalexin, presumably due to the activity of the enzyme, expandase. The extract was fractionated and characterized by colorimetric and chromatographic measurements coupled with disc-agar diffusion bioassay against an ampicillin-resistant, cephalexin-sensitive E. coli strain. Though expandase could not be identified, the presence of a hitherto unreported expandase in Streptomyces sp. DRS I is suggested.  相似文献

探讨了不同环境条件对小球藻(Chlorella sp.)叶绿素荧光动力学参数以及净光合放氧速率的影响,确定了以L1海水培养基为基础,以8.8 mmol/L浓度的(NH2)2CO为氮源、0.145 mmol/L NaH2PO4 · H2O浓度为磷源,在150 μmol · m-2 · s-1光照强度、培养温度为18 ℃的小球藻最优培养条件。在此条件下,明显加快了小球藻细胞的生长速度,促进了油脂和脂肪酸的积累,细胞密度增加24%,油脂和脂肪酸含量分别增加了16.8%和66.6%。在培养液中添加外源柠檬酸(最适浓度以0.06 mmol · L-1 · d-1为宜)可以明显提高小球藻的生长速度,促进其脂肪酸的积累。同时也可看出,筛选的小球藻藻种具有生长快、易培养、产油高的优点,可作为生物能源研究的良好材料,为海洋微藻的开发利用奠定了基础。  相似文献
