PSAG12-ipt嵌合基因转化樱桃番茄的研究

摘    要:利用根癌农杆菌(Agrobacterium tumefacienst)介导法将嵌合基因P SAG12-ipt导人樱桃番茄,经双重PCR检测和基因组总DNA Southern杂交测验,共获得17株转基因植株。田间生长实验观察结果显示,这17株转基因植株生长发育及形态正常,但其中的14株长势明显好于对照。对14株中的2株进行叶片叶绿素、细胞分裂素检测结果表明,转基因植株基本定型叶下部,不同叶位叶片叶绿素、细胞分裂素含量明显高于对照,基本定型叶上部叶片与对照基本一致,同一叶位叶片比对照延缓衰老15~20d。

Abstract:Meiated with Agrobacterium tumefaciens,the gomphsis gene P_(SAG12)-ipt was transferred into cherry tomato plants and then 17 transgenic plants of cherry tomato were obtained by double PCR and Southern blotting.The field test of the transgenic plants showed that the seventeen transgenic plants normally grew and developed presenting a normal morphology but fourteen of them demonstrated an obviously stronger vigor than the control plants did.The chlorophyll and cytokinin determinations of the leaves of two plants of the fourteen plants indicated that chlorophyll and cytokinin contents of the full-extended leaves at lower positions and the leaves of the transgenic plants at different positions were remarkably higher than those of the control plants,chlorophyll and cytokinin contents of the full-extended leaves at upper positions were approximately the same as those of the control plants,and the leaves of the transgenic plants began to age 15~20 days later than those of the control plants at the same positions.
