猪血药用物质基础研究(Ⅰ)——脱色条件考察

利用粉末活性炭、羧甲基淀粉钠(CMS)、羧甲基纤维素钠(CMC)吸附作用进行中药猪血的脱色剂初步筛选,发现活性炭脱色效果较好;选取胃蛋白酶、胰蛋白酶、1394酶、木瓜蛋白酶4种酶酶解猪血蛋白,综合考虑胃蛋白酶酶解效果最好;进一步利用45均匀设计法得到猪血最佳酶解脱色条件为加酶量8%、温度45℃、酶解时间0.5 h、底物浓度18%,吸附剂为活性炭,脱色液呈浅黄色,无腥味,蛋白质含量较高,并且酶解后猪血蛋白分解为多肽,具有多功能生物活性。

Study on the Conditions of Decolorization of Porcine Blood

To screen the best decolorant of the traditional Chinese meidicine porcine blood,activated charcoal,CMS and CMC were used,and found thar the best effect was produced by using actirated charcoal.Pepsin,trypsin,1394 enzyme and papain enzyme were selected as the enzymes for hydrolyzing the poricne blood proteins,and the optimum was pepsin.Furthermore,the best hydrolysis and decolorization conditions were the following: volume of enzyme of 8%,temperatureat at 45 ℃,enzymolysis time of 0.5h and substrate concentration of 18% by using 45 uniform design.And the decolorized solution of porcine blood was light-yellow,smellless,high protein concentration and multi-functional biological active peptides which were the enzyme-hydrolyzed products.
