结肠癌中NMNAT2、p53的表达及三者相关性研究

摘    要:目的:检测结肠癌中烟酰胺核苷酸腺苷转移酶2 (Nicotinamide nucleotide adenosine transferase 2, NMNAT2)、p53的表达并分析三者之间的关系。方法:免疫组化S-ABC法(4分为阴性、≥4分为阳性)检测结肠癌标本(48例)、癌旁正常组织标本(40例)中NMNAT2、p53的表达,分析其与结肠癌年龄、性别、病理类型、肿瘤形态、分化程度、浸润深度、TNM分期、淋巴结转移等临床参数的关系及二者的关系。结果:(1) NMNAT2、p53表达于结肠癌组织细胞质、胞核,呈棕黄或棕褐色,在癌旁正常组织中表达较低或缺失。结肠癌中NMNAT2、p53表达阳性者占83.33%、70.83%,癌旁正常组织中表达阳性者占7.50%、0.00%,其差异有统计学意义(P0.05)。(2) NMNAT2、p53阳性率,在不同年龄、性别、病理类型、肿瘤形态、分化程度患者中的差异无统计学意义(P0.05);(T3-T4)期者为96.51%、84.62%,高于(T1-T2)期的68.18%、54.55%(P0.05);(Ⅲ-Ⅳ)期者为95.00%、85.00%,高于(Ⅰ-Ⅱ)期的78.57%、57.14%(P0.05);有淋巴结转移者为100.00%、84.21%,高于无淋巴结转移者的72.41%、62.07%(P0.05)。(3)结肠癌组织中NMNAT2与p53表达呈正相关(rs=0.809, P0.05)。结论:NMNAT2、p53与结肠癌发展相关,且二者呈正相关,因此,联合检测有助于对结肠癌诊治与病情评估。

Abstract:ABSTRACT Objective: To detect the expression of nicotinamide nucleotide adenosine transferase 2(Nicotinamide nucleotide adenosine transferase 2) and p53 in colon cancer and analyze the relationship between the three. Methods: S-ABC immunohistochemical method (<4 = 4, divided into negative into positive) detection of colorectal cancer (48 cases) and adjacent normal tissues (40 cases) the expression of NMNAT2, p53, and the analysis of colon cancer age and gender, pathological type, tumor morphology, differentiation, and Jin Runshen TNM staging, lymph node metastasis and other clinical parameters of the relationship and the relationship between the two. Results: NMNAT2 and p53 expression in colon cancer tissue cytoplasm and nucleus, with Buffy or brown, expression is low or absent in normal tissues. The positive rate of NMNAT2 and p53 expression in colon cancer was 83.33% and 70.83%, respectively. The positive expression rate in 7.50% normal tissues was 7.50% and 0%, respectively, and the difference was statistically significant(P<0.05). The positive rate of p53 and NMNAT2, there was no significant difference in different age, gender, pathological type, tumor size and differentiation degree in patients (P>0.05); (T3-T4) period was 96.51%, 84.62% higher than the 68.18% period (T1-T2), 54.55%(P<0.05); (III - IV) period was 95% 85%, higher than (I-II) period of 78.57%, 57.14% (P<0.05); lymph node metastasis was 100%, 84.21% higher than those without lymph node metastasis, 72.41%, 62.07% (P<0.05). The NMNAT2 and p53 expression in colorectal cancer tissue was positively correlated (rs=0.809, P<0.05). Conclusion: NMNAT2 and p53 are related to the development of colon cancer, and there is a positive correlation between the two. The joint detection can be considered to assist the diagnosis and treatment of colon cancer and the evaluation of the disease.
