Mineralogical segregation of sand grains distinguishes the trace fossil Macaronichnus segregatis, which is composed of a felsic burrow infill with a mafic-and mica-rich burrow mantle. This study focuses on determining the mechanism by which M. segregatis trace-makers segregated mineral grains during deposit feeding. A modern opheliid polychaete, Euzonus mucronata, from Pachena Bay, Vancouver Island (Canada), was examined to explain the activities of their ancient counterparts. Microscopic videotaping of deposit feeding allowed for collection of data on ingestion and excretion through visual grain counts of felsic, mafic, and shell components. Normalization of these grain counts to the composition of the host sediment illustrates preferential ingestion of felsic grains over mafic. Shell fragments were generally avoided and visually mantled the burrows, obscuring the paucity of mafic grains in burrow infills. The avoidance of shell fragments is potentially a function of the large grain size, angular shape, surface texture, and/or associated low nutritive value. The preferential ingestion of felsic grains is attributed to en masse feeding in felsic-rich locales identified through sediment probing. This form of mineral segregation likely reflects the specific nature of the sediment and worm population. Accordingly, en mass deposit feeding in selected felsic-rich localities is one possible mechanism used in the construction of Macaronichnus segregatis and M. segregatis-like structures.  相似文献

Inhibition of the autophagic pathway has recently revealed promising results in increasing pro-death activity of multiple cancer therapeutics. Here, we discuss our findings regarding the autophagy-blocking and anti-neoplastic effects of a synthetic sphingosine analog, FTY720, in mantle cell lymphoma (MCL). We also emphasize how FTY720 enhances the pro-death activity of the fully humanized monoclonal antibody milatuzumab by inhibiting the autophagy-lysosome dependent degradation of its therapeutic target, CD74. Our results provide justification for further evaluation of FTY720 and milatuzumab as a combination therapy for this aggressive B-cell malignancy.  相似文献

The patients with mantle cell lymphoma (MCL) have translocation t(11;14) associated with cyclin D1 overexpression. We observed that iron (an essential cofactor of dioxygenases including prolyl hydroxylases [PHDs]) depletion by deferoxamine blocked MCL cells’ proliferation, increased expression of DNA damage marker γH2AX, induced cell cycle arrest and decreased cyclin D1 level. Treatment of MCL cell lines with dimethyloxalylglycine, which blocks dioxygenases involving PHDs by competing with their substrate 2‐oxoglutarate, leads to their decreased proliferation and the decrease of cyclin D1 level. We then postulated that loss of EGLN2/PHD1 in MCL cells may lead to down‐regulation of cyclin D1 by blocking the degradation of FOXO3A, a cyclin D1 suppressor. However, the CRISPR/Cas9‐based loss‐of‐function of EGLN2/PHD1 did not affect cyclin D1 expression and the loss of FOXO3A did not restore cyclin D1 levels after iron chelation. These data suggest that expression of cyclin D1 in MCL is not controlled by ENGL2/PHD1‐FOXO3A pathway and that chelation‐ and 2‐oxoglutarate competition‐mediated down‐regulation of cyclin D1 in MCL cells is driven by yet unknown mechanism involving iron‐ and 2‐oxoglutarate‐dependent dioxygenases other than PHD1. These data support further exploration of the use of iron chelation and 2‐oxoglutarate‐dependent dioxygenase inhibitors as a novel therapy of MCL.  相似文献

鸟氨酸-尿素循环(OUC)是生物新陈代谢过程中的重要循环过程,但在贝类中尚缺乏相关研究。为此,以厚壳贻贝为研究对象,分别采用氨基酸分析仪和荧光定量PCR研究了其外套膜和后闭壳肌组织中的鸟氨酸-尿素循环途径的主要代谢物和关键基因的含量及其表达量;进一步测试了在精氨酸注射条件下,各主要代谢物和关键基因的含量及表达量变化,以及13C标记尿素注射贻贝后,其贝壳中δ13C比值(13C/12C)变化。结果表明,厚壳贻贝外套膜和后闭壳肌均含有较高浓度的尿素;精氨酸注射导致其两种组织中尿素浓度显著上升(P<0.01),以及瓜氨酸浓度显著下降(P<0.01),但鸟氨酸浓度维持相对稳定的水平。精氨酸注射显著上调了两种组织中的脲酶基因的表达量(P<0.01),但其他基因表达量的变化在两种组织中存在差异,显示出鸟氨酸-尿素循环途径在其两种组织中具有复杂而不同的调控过程。13C标记尿素注射贻贝显著上调了贝壳中δ13C的比值(P <0.01),表明尿素分子可能参与了贻贝贝壳的生物矿化过程。上述研究为深入了解贻贝鸟氨酸-尿素途径与生物矿化之间的关联,以及探讨贻贝对海水酸化耐受性的内在分子机制奠定了基础。  相似文献

p27Kip1 is a cyclin-dependent kinase inhibitor that plays a critical role in regulating G1/S transition, and whose activity is, in part, regulated through interactions with D-type cyclins. We have generated the BD1-9 cell line, a BaF3 pro-B cells derivative in which cyclin D1 can be induced rapidly and reversibly by ponasterone A. The induction of cyclin D1 expression leads to a targeted p27Kip1 accumulation in both cytoplasmic and nuclear compartments. But, only the p27Kip1 form phosphorylated on serine 10 (pSer10-p27Kip1) accumulates in BD1-9 cells. We found that the binding of cyclin D1 and pSer10-p27Kip1 prevents p27Kip1 degradation by the cytoplasmic Kip1 ubiquitylation-promoting complex (KPC) proteosomic pathway. Importantly, the nuclear CDK2 activity which is crucial for G1/S transition is not altered by p27Kip1 increase. Using siRNA techniques, we revealed that p27Kip1 inhibition does not affect the distribution of BD1-9 cells in the different phases of the cell cycle. Our study demonstrates that aberrant cyclin D1 expression acts as a p27Kip1 trap in B lymphocytes but does not induce p27Kip1 relocation from the nucleus to the cytoplasm and does not modulate the G1/S transition. Since our cellular model mimics what observed in aggressive lymphomas, our data bring new insights into the understanding of their physiopathology.  相似文献

Cannabinoid receptors 1 (CB1) and/or 2 (CB2) are overexpressed in many types of human malignancies including mantle cell lymphoma (MCL). Agonists to CB1 and CB2 promote ceramide de novo synthesis, p38–mitogen-activated protein kinase-dependent activation of caspase-3 and apoptotic cell death in most MCLs. However, in this report we describe that in some MCLs the response to treatment with cannabinoids decreased cell viability as assessed by metabolic activity but did not involve the caspase-3 cascade or loss of plasma membrane integrity. Both primary cells from one MCL patient and the MCL cell line Granta519 responded to treatment with cannabinoids by formation of cycloheximide-sensitive cytoplasmic vacuoles, but did not enter apoptosis. The persistent expression of mammalian homolog of Atg8 with microtubule-associated protein-1 light chain-3 II (LC3 II) and p62, as well as the lack of protection from chloroquine, indicates that lysosomal degradation is not involved in this cytoplasmic vacuolation process, distinguishing from classical autophagy. Transmission electron microscopy images and immunofluorescence staining of endoplasmic reticulum (ER) chaperone calreticulin showed that the vacuoles were of ER origin and that chromatin remained normal. These features resemble paraptosis-like cell death—a third type of a programmed cell death not previously described in response to cannabinoids.  相似文献

Early studies on the outer mantle epithelium (OME) cells of the freshwater bivalve Anodonta cygnea (Linnaeus, 1758) revealed high ionic calcium concentrations by electrophysiological methods and subsequently a high tendency to reach an intracellular toxic condition. This toxicity could be neutralized by specific mechanisms in the cytosol of OME cells of A. cygnea. The present immunocytochemistry studies of OME cells by light and transmission electron microscopy (TEM) clearly showed a positive reaction of an antibody directed against the human plasma membrane Ca2+-ATPase 1 (PMCA-1) in the cytoplasm of OME cells. Also, western blot analysis of different fractions of OME cells with anti human PMCA-1 and C28R2 antibodies confirmed the presence of a PMCA-like protein with an unusual topographical localization and a molecular weight of only 70–80 kDa. These results lead us to speculate that this PMCA-like protein is distributed either in the plasma membrane or in the entire cytosol, where it eventually regulates intracellular calcium levels. Interestingly, the antibody reactions showed seasonal variations, being highest in OME samples prepared during summer when A. cygnea live under natural acidosis and absent in samples taken in winter conditions, which is in accordance with the seasonal variation of shell calcification rates. During winter, PMCA-1 antibody reaction was also detected in OME cells of animals kept only under experimentally induced acidosis conditions. Therefore, we assume that a functional role for this PMCA-like protein in the intracellular calcium regulation of OME cells during the mineralization of the shells of A. cygnea can be speculated.  相似文献
