In our previous studies, we observed the biological control effect of lactic acid bacteria strains (LABs) KLF01, KLC02 and KPD03 against different plant pathogenic bacteria in vitro against Ralstonia solanacearum, and strains KLF01 and KLC02 against Pectobacterium carotovorum under greenhouse and field experiments, respectively. In this study, we observed the efficacy of these bacteria against bacterial spot pathogen (Xanthomonas campestris pv. vesicatoria) and their plant growth-promoting activities in pepper (Capsicum annuum L. var. annuum), under greenhouse and field conditions. LABs significantly (P < 0.05) reduced bacterial spot on pepper plants in comparison to untreated plants in both the greenhouse and the field experiments. The plant growth-promoting effect of LABs on pepper varied; some strains had a significant effect on growth promotion (P < 0.05) compared with untreated plants, while some showed no significant effect in the greenhouse and field experiments. Additionally, LABs were able to colonise roots, produce indole-3-acetic acid (IAA), siderophores and solubilise phosphate. These findings indicate that application of LABs could provide a promising alternative for the management of bacterial spot disease in pepper plants and could therefore be used as a healthy plant growth-promoting agent.  相似文献

A procedure for the quantitative analysis of oryzalides and oryzalic acids in leaves of rice cultivars was established by using GC-MS-selective ion monitoring (GC-SIM), 17-deuterated oryzalide B being used as an internal reference. The total contents of four compounds of oryzalides and oryzalic acids in the leaves at three different growth stages of Norin-27, i.e., seedling, tillering, and ripening, showed values of 5.3, 17.9, and 37.9 μg/g, respectively. The contents of oryzalides and oryzalic acids in each organ, i.e., leaf, stem, ear, and root, showed values of 35.0 and 1.7 μg/g for the former two, respectively, and were not detectable for the latter two. A preliminary analysis of Norin-27 and Rantai emas leaves inoculated with an incompatible and compatible strain of Xanthomonas campestris pv. oryzae showed a greater accumulation of oryzalides and oryzalic acids, especially in the lesion area, than the figures for healthy leaves. Mechanical injury by a needle-bundle also increased the total amounts.  相似文献

Xanthomonas translucens pv. undulosa, (Xtu.), causal agent of Bacterial leaf streak (BLS) of wheat, was characterised through pathogencity, hypersensitivity, biochemical and molecular assays. Fifty symptomatic leaves of wheat were collected from eight agro-ecological zones of Punjab out of which 25 were isolated and purified. Maximum incidence and severity in Faisalabad were followed by Multan and Rahim Yar Khan. The pathogen isolated from diseased leaves was identified on the basis of colonies pattern, colour, biochemical and pathogencity test as X. translucens pv. undulosa and confirmed its pathogencity through pathogencity test. For molecular characterization, the bacterial 16S–23S rDNA spacer fragments were amplified by PCR with conserved primers (C1 and C2) and then in combination with specific primers (T1 & T2). 300?bp product amplified by C1 and C2 primer pair confirmed the presence of Xanthomonas, while specific primers T1 and T2 amplified a product of 200?bp, confirmed the presence of X. translucens pv. undulosa. This work will be quite helpful for wheat pathologist and breeders for future management strategy for this disease.  相似文献

Xanthomonas campestris pv. campestris (Xcc), the causal agent of black rot in crucifers, produces a membrane-bound yellow pigment called xanthomonadin to protect against photobiological and peroxidative damage, and uses a quorum-sensing mechanism mediated by the diffusible signal factor (DSF) family signals to regulate virulence factors production. The Xcc gene XCC4003, annotated as Xcc fabG3, is located in the pig cluster, which may be responsible for xanthomonadin synthesis. We report that fabG3 expression restored the growth of the Escherichia coli fabG temperature-sensitive mutant CL104 under non-permissive conditions. In vitro assays demonstrated that FabG3 catalyses the reduction of 3-oxoacyl-acyl carrier protein (ACP) intermediates in fatty acid synthetic reactions, although FabG3 had a lower activity than FabG1. Moreover, the fabG3 deletion did not affect growth or fatty acid composition. These results indicate that Xcc fabG3 encodes a 3-oxoacyl-ACP reductase, but is not essential for growth or fatty acid synthesis. However, the Xcc fabG3 knock-out mutant abolished xanthomonadin production, which could be only restored by wild-type fabG3, but not by other 3-oxoacyl-ACP reductase-encoding genes, indicating that Xcc FabG3 is specifically involved in xanthomonadin biosynthesis. Additionally, our study also shows that the Xcc fabG3-disrupted mutant affects Xcc virulence in host plants.  相似文献

以白菜型油菜‘陇油6号’和‘天油2号’为试验材料,经MAPK抑制剂U0126、H_2O_2清除剂DMTU、NADPH氧化酶抑制剂DPI和IMD预处理后再分别进行盐胁迫、PEG-6000模拟干旱胁迫,研究其对两种油菜幼苗活性氧、抗氧化酶活性和RbohC、RbohF基因表达的影响.结果表明:盐胁迫和PEG-6000模拟干旱胁迫下,两种白菜型油菜中H_2O_2积累量上升,O_2积累量下降,抗氧化酶(超氧化物歧化酶SOD、过氧化氢酶CAT、抗坏血酸过氧化物酶APX和谷胱甘肽还原酶GR)活性和RbohC、RbohF基因表达均升高.与单独胁迫处理相比,两种油菜O_2积累、抗氧化酶活性和RbohC、RbohF基因的表达量均明显降低,经DMTU、DPI和IMD预处理后再分别进行盐和干旱胁迫,H_2O_2积累量下降,但U0126预处理后再进行胁迫处理,H_2O_2积累量上升.说明NADPH氧化酶、MAP激酶级联途径、H_2O_2参与了盐、干旱胁迫下活性氧产生、抗氧化酶活性变化和RbohC、RbohF基因表达的调控.  相似文献

The velvet longhorned beetle, Trichoferus campestris (Coleoptera: Cerambycidae) is a serious wood‐boring pest that is a major threat to the phytosanitary condition of forests and orchards. Its worldwide expansion is a major concern for plant health. We have collected all bibliographical references, phytosanitary reports and authentic photographic evidence from entomological websites to determine the worldwide distribution of T. campestris. The theoretical directions of the spreading and actually occupied area of this arthropod pest were determined over the whole Holarctic range. Furthermore, the potential distribution area was calculated using cumulated temperature in the growing season averaged over 15 years both in the Palearctic and Nearctic regions. Holarctic expansion of the species, including the main parts of North America and Eurasia, is clearly indicated. Its populations occur in 29 countries to date, supported by documentation from 64 publications and 30 online forums. Its spread is continuously westward in the Palearctic; in the Nearctic, the spread was first southward from the Great Lakes region then eastward from Utah. The species has excellently adapted to circumstances of freight by ship and plane, as wood is often used, ensuring optimal conditions for the pest. In addition, the active spreading achieved by flying is an important factor contributing to its expansion. The primary criterion for controlling the species would be the introduction of a monitoring system in affected and exposed areas.  相似文献

以1/4园试营养液为对照,以腐熟发酵的农业秸秆(苋菜和番茄植株残体)浸提液模拟富含N、P的水体进行不结球白菜(Brassica campestris ssp.chinensis Makino)的漂浮栽培,对茎叶的单株鲜质量和干质量及叶片的VC、可溶性蛋白质、蔗糖、果糖、葡萄糖含量及可溶性糖总含量进行了比较分析。结果表明:在24 d的栽培周期内,随生长时间的延长,处理组和对照组茎叶的单株鲜质量和干质量均呈增加趋势,但从第10天开始,处理组茎叶的单株鲜质量和干质量均显著低于对照组。栽培初期叶片VC含量均最高且处理组叶片VC含量显著低于对照组;从第10天开始,叶片VC含量降低,但处理组叶片VC含量与对照组无显著差异。处理组与对照组叶片可溶性蛋白质含量均呈波动的变化趋势,但总体差异不显著。处理组叶片蔗糖含量总体上与对照组差异不显著,仅在第24天显著低于对照组。处理组叶片的果糖含量均高于对照组,特别是在第24天为对照组的1.81倍,差异显著。叶片葡萄糖含量均呈先升高后降低的趋势,可溶性糖总含量则呈波动的变化趋势,对照组叶片葡萄糖含量和可溶性糖总含量总体上高于处理组,但差异不显著。研究结果显示:用秸秆浸提液漂浮栽培,虽然不结球白菜的生长量(即产量)有所减少,但相关的品质指标变化不大,对果糖合成和积累还有一定的促进作用,因而,此种秸秆浸提液可用于栽培商品不结球白菜。  相似文献

在克隆花粉发育相关基因BcMF13时,用RACE技术进行3’端扩增,得到了两个不同长度的3’转录本。与EST库比对,发现与其相似性高的序列都来源于十字花科芸薹属、萝卜属等植物的生殖器官,并且在这些近源属中也出现不同转录本。这些不同转录本的存在说明BcMF13存在剪切异构体,调控过程灵活复杂,反映出该基因具有重要功能。同时,对BcMF13基因的RT-PCR表达研究表明,它只在可育株系中表达,在不育株系中不表达,且主要集中在大花蕾、雄蕊中表达。分析BcMF13推导的蛋白结构,发现其编码的蛋白包含多个生物活性位点。以上结果均表明BcMF13与花粉育性相关,多个剪切异构体的发现说明BcMF13在执行生理功能时较为活跃。  相似文献

Human land-use changes are particularly extensive in tropical regions, representing one of the greatest threats to terrestrial biodiversity and a key research topic in conservation. However, studies considering the effects of different types of anthropogenic disturbance on the functional dimension of biodiversity in human-modified landscapes are rare. Here, we obtained data through an extensive review of peer-reviewed articles and compared 30 Neotropical bat assemblages in well-preserved primary forest and four different human-disturbed habitats in terms of their functional and taxonomic diversity. We found that disturbed habitats that are structurally less similar to primary forest (pasture, cropland, and early-stage secondary forest) were characterized by a lower functional and taxonomic diversity, as well as community-level functional uniqueness. These habitats generally retained fewer species that perform different ecological functions compared to higher-quality landscape matrices, such as agroforestry. According to functional trait composition, different bat ensembles respond differently to landscape change, negatively affecting mainly gleaning insectivorous bats in pasture, narrow-range species in cropland, and heavier animalivorous bats in secondary forest. Although our results highlight the importance of higher-quality matrix habitats to support elevated functional and taxonomic bat diversity, the conservation of bat species that perform different ecological functions in the mosaic of human-modified habitats also depends on the irreplaceable conservation value of well-preserved primary forests. Our study based on a pooled analysis of individual studies provides novel insights into the effects of different human-modified habitats on Neotropical bat assemblages.  相似文献

The crystal structure of XC1028 from Xanthomonas campestris has been determined to a resolution of 2.15 Å using the multiple anomalous dispersion approach. It bears significant sequence identity and similarity values of 64.10% and 70.09%, respectively, with PA2960, a protein indispensable for type IV pilus‐mediated twitching motility, after which the PilZ motif was first named. However, both XC1028 and PA2960 lack detectable c‐di‐GMP binding capability. Although XC1028 adopts a structure comprising a five‐stranded β‐barrel core similar to other canonical PilZ domains with robust c‐di‐GMP binding ability, considerable differences are observed in the N‐terminal motif; XC1028 assumes a compact five‐stranded β‐barrel without an extra long N‐terminal motif, whereas other canonical PilZ domains contain a long N‐terminal sequence embedded with an essential “c‐di‐GMP switch” motif. In addition, a β‐strand (β1) in the N‐terminal motif, running in exactly opposite polarity to that of XC1028, is found inserted into the parallel β3/β1′ strands, forming a completely antiparallel β4↓β3↑β1↓β1′↑ sheet in the canonical PilZ domains. Such dramatic structural differences at the N‐terminus may account for the diminished c‐di‐GMP binding capability of XC1028, and suggest that interactions with additional proteins are necessary to bind c‐di‐GMP for type IV fimbriae assembly.
