Iridoid glycosides are plant defence compounds that are deterrent and/or toxic for unadapted herbivores but are readily sequestered by dietary specialists of different insect orders. Hydrolysis of iridoid glycosides by β‐glucosidase leads to protein denaturation. Insect digestive β‐glucosidases thus have the potential to mediate plant–insect interactions. In the present study, mechanisms associated with iridoid glycoside tolerance are investigated in two closely‐related leaf beetle species (Coleoptera: Chrysomelidae) that feed on iridoid glycoside containing host plants. The polyphagous Longitarsus luridus Scopoli does not sequester iridoid glycosides, whereas the specialist Longitarsus tabidus Fabricius sequesters these compounds from its host plants. To study whether the biochemical properties of their β‐glucosidases correspond to the differences in feeding specialization, the number of β‐glucosidase isoforms and their kinetic properties are compared between the two beetle species. To examine the impact of iridoid glycosides on the β‐glucosidase activity of the generalist, L. luridus beetles are kept on host plants with or without iridoid glycosides. Furthermore, β‐glucosidase activities of both species are examined using an artificial β‐glucosidase substrate and the iridoid glycoside aucubin present in their host plants. Both species have one or two β‐glucosidases with different substrate affinities. Interestingly, host plant use does not influence the specific β‐glucosidase activities of the generalist. Both species hydrolyse aucubin with a much lower affinity than the standard substrate. The neutral pH reduces the β‐glucosidase activity of the specialist beetles by approximately 60% relative to its pH optimum. These low rates of aucubin hydrolysis suggest that the ability to sequester iridoid glycosides has evolved as a key to potentially preventing iridoid glycoside hydrolysis by plant‐derived β‐glucosidases. 相似文献

Stoiba Spaeth, 1909 is revised with a phylogenetic analysis of 38 adult morphological characters for nine Stoiba species and 11 outgroup species (Mesomphaliini, Ischyrosonychini, and Hemisphaerotini). Four Cuban species of Stoiba were not sampled. Parsimony analysis located the four most parsimonious trees. The strict consensus (CI=0.59, RI=0.78, Steps=83) resolved the monophyly of Stoiba. The monophyly of Stoiba is supported by pale yellow antennae, antennomere VII broader than its length, and rounded basal line of pronotum. An illustrated key to ten species of Stoiba is provided along with a distribution map of 11 species. Stoiba rufa Blake is synonymized with Stoiba swartzii (Thunberg) by a morphological comparison which includes female genitalia. 相似文献

For a study of local adaptations in the Alpine leaf beetle, Oreina elongata, we developed six microsatellite loci and screened them in 305 individuals from 13 populations. All markers were polymorphic with three to 15 alleles per locus. Average observed and expected heterozygosity values were 0.14 and 0.62, respectively. Four markers showed heterozygote deficiency and deviated significantly from Hardy–Weinberg expectations, indicating the presence of null alleles. 相似文献

The accidentally introduced beetle Ophraella communa was first found in 1996 in Japan and has rapidly expanded its distribution range to include regions with substantially different climates. During this range expansion and subsequent establishment in new habitats, the life history traits of O. communa have changed depending on new habitat environmental conditions. In this study, we investigated the photoperiodic response of O. communa controlling its reproductive diapause, an important trait that adjusts the life cycle to local climate and phenology of host‐plants. We examined temporal changes and geographical differences in this trait. In the Tsukuba population of O. communa, the diapause incidence under conditions of 12 h light : 12 h dark (LD 12:12), 13:11 and 14:10 did not consistently increase or decrease during 2005–2012, although it fluctuated yearly. The diapause incidence in this period, however, was lower than that in 1999 reported previously and the critical day‐length inducing diapause was shortened by >1 h from 1999 to 2005. These results suggest that the photoperiodic response of O. communa shifted during this period. To examine geographical differences in this trait, we compared the Hirosaki, Morioka, Tsukuba and Koshi lines. The diapause incidence at LD 13:11 was significantly different between the O. communa lines: 86.5% in the Hirosaki line (collected in 2010) , 81.4% in Morioka (collected in 2010), 45.0% in Tsukuba (collected in 2011) and 7.1% in Koshi (collected in 2009), and was positively correlated with the latitude of the origin. These results suggest that this trait shifted, responding to the environmental conditions in the colonized regions. 相似文献

The polyphagous beetle Gonioctena quinquepunctata Fabricius (Coleoptera: Chrysomelidae) is a serious leaf pest of the native European bird cherry, Prunus padus L., and the invasive alien black cherry, Prunus serotina Ehrh. (Rosaceae). In the shade, leaf damage is extensive in both species, whereas in full light, it is extensive in P. padus, but very low in P. serotina. We determined the influence of Prunus species and light conditions on differences in performance of both sexes of this folivore. In a laboratory experiment in which larvae were fed with leaves of a single species grown under particular light conditions, we measured larval, pupal, and adult mass, efficiency of conversion of ingested food (ECI), duration of development, total food eaten, and relative growth rate. In the field, we observed differences in beetle mass on shrubs of the two species growing under various light conditions. From the field observations, we hypothesised that leaves of the invasive P. serotina are not an equally good food source as leaves of P. padus for G. quinquepunctata, and the preference of these beetles for shaded shrubs is most favourable for their growth and development. Under laboratory conditions, we found that the beetle growth rate was not affected significantly by Prunus species or light conditions, despite the significant effect of light condition on the structure and chemical composition of Prunus seedlings. The lower ECI value for larvae feeding on sunlit leaves was compensated for by their higher level of consumption. In the field, adult insect mass was higher on P. padus than on P. serotina, and higher on sunlit shrubs of both species than on shaded ones. The walking and flight dispersal of marked overwintered and summer Colorado potato beetles (CPB), Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae), released in field box‐plots was monitored simultaneously in six habitats over a period of 4 days. The emigration out of plots by walking beetles was calculated from the catch in linear pitfall traps completely surrounding each box‐plot and emigration flight was estimated from the number of beetles missing from the plot or captured by the trap. Overwintered beetles dispersed sooner after release than summer beetles. Overall, the mean number of beetles retained by the habitat was significantly higher in the host habitat (potato) than in any non‐host habitat tested (soybean, pasture, bare ground, water, woodland). Unexpectedly, there was no or little difference in overall beetle retention between non‐host habitats except for higher retention in the water habitat. No difference in the ratio of flight over walking could be detected by the study between overwintered and summer CPB except in the water and woodland habitats. Twenty‐four hours after release, the highest ratios were obtained in the water and woodland habitats and the lowest in the bare‐ground habitat, but ratios were similar for all habitats, except water, after 96 h. As a population, under these experimental conditions, 96 h after release, it seems that CPB displayed a slight preference for flight over walking, with walking as a default mode. A fed and starved pre‐release treatment had no effect on dispersal rates or mode of dispersal. Essentially, our results showed that over a 96‐h period, northeastern North American CPB emigrated at similar rates from the various non‐host habitats encountered, except for water, using walking as much as flight. The host habitat retained CPB significantly longer than non‐host habitats but with a mode of dispersal ratio similar to that in non‐host habitats. The impact on dispersal of the various habitats encountered by CPB in the agro‐ecosystem was less important than expected suggesting that the interaction of environmental parameters is likely to have the most significant impact in determining dispersal rates and dispersal modes. 相似文献

The success of the current resistance management plan for transgenic maize, Zea mays L. (Poaceae), targeting the rootworm complex hinges upon high rates of mating between resistant and susceptible beetles. However, differences in the fitness of adult beetles could result in assortative mating, which could, in turn, change the rate of resistance evolution. Adult head capsule widths of naturally occurring populations of western corn rootworm, Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), were examined in a variety of refuge configurations. Beetles were classified into treatments based on the hybrid – non‐Bt refuge or Bt maize targeting larval rootworms (hereafter Bt‐RW maize) – and location – proximity to other Bt‐RW or refuge plants – of the natal host plant. Treatments included the following: a refuge plant surrounded by other refuge plants, a refuge plant located near a Bt‐RW plant, a Bt‐RW plant surrounded by Bt‐RW plants, and a Bt‐RW plant located near a refuge plant. The mean head capsule width of males emerging from Bt‐RW plants was significantly smaller than the mean head capsule width of males emerging from refuge plants. These results indicate that males emerging from Bt‐RW maize plants may be exposed to sublethal doses of the Bt toxin as larvae. No differences were detected between females emerging from refuge plants compared with Bt‐RW plants. Overall mean head capsule width decreased as the season progressed, regardless of treatment. The diminished head capsule width of western corn rootworm males emerging from Bt‐RW maize may act to enhance resistance management, particularly in a seed mix refuge system. 相似文献
