Leaves of Chinese cabbage from healthy plants or from those infected with turnip yellow mosaic virus yield protoplasts which convert methionine to protein, S-adenosylmethionine, decarboxylated S-adenosylmethionine, spermidine, spermine and 1-aminocyclopropane-1-carboxylate. The enzyme spermidine synthase is entirely cytosolic and has been purified extensively. An inhibitor of this enzyme, dicyclohexylamine, blocks spermidine synthesis in intact protoplasts, and in so doing stimulates spermine synthesis. Aminoethoxyvinylglycine blocks the conversion of S-adenosylmethionine to 1-aminocyclopropane-1-carboxylate, the precursor to ethylene, in protoplasts. This inhibitor markedly stimulates the synthesis of both spermidine and spermine. Essentially all the protoplasts obtained from new leaves of plants infected 7 days earlier are infected. On incubation, such protoplasts convert exogenous methionine to viral protein and viral spermidine whose specific radioactivity is twice that of total cell spermidine. Exogeneous spermidine is also converted to cell putrescine and viral spermidine and spermine. We report the chemically determined sequence of most of the polypeptide chain of the coat protein of tomato bushy stunt virus. Peptide locations have been determined by comparison with the high-resolution electron density map from X-ray crystallographic analysis as well as by conventional chemical overlaps. Three small gaps remain in the 387-residue sequence. Positively charged side-chains are concentrated in the N-terminal part of the polypeptide (the R domain) as well as on inward-facing surfaces of the S domain. This study investigates the behavioural and neural mechanisms involved in the oviposition behaviour of the turnip root fly,Delia floralis (Fallen). Behavioural studies showed that glucosinolates modulated the oviposition behaviour of the flies on artificial leaves
as well as the number of eggs laid in the soil at the base of these leaves. Electrophysiological responses to glucosinolates
were obtained from type A and type D sensilla on the prothoracic and mesothoracic tarsi, as well as from the long contact
sensilla on the labellum. The neural responses from these sensilla were positively correlated with the oviposition behaviour
of the flies and with the number of eggs laid. Of the eleven glucosinolates tested in the behavioural and electrophysiological
tests, the flies were most responsive to glucobrassicanapin, gluconapin and glucobrassicin. The type D tarsal sensilla were
more responsive to the glucosinolates than either the type A tarsal sensilla or the labellar sensilla. The structure-activity
investigations showed that slight modifications to the chemical composition of the glucosinolates resulted in changes in neural

Seed transmission of turnip yellow mosaic virus in winter turnip and winter oilseed rapes 总被引:1,自引:0,他引:1
This is the first record of seed transmission of turnip yellow mosaic virus (TYMV) in oilseed and turnip rapes. The seed transmission of TYMV in a naturally infected winter turnip rape (Brassica napus var. silvestris) cultivar Perko PVH was investigated. By ELISA 1.6%, 3.2% and 8.3% seed transmission of the virus was found in seed of plants from three localities. The proportion of infected seeds produced by artificially infected plants of winter oilseed rape (Brassica napus ssp. oleifera) and winter turnip rape cultivars was determined. The virus transmission rate, expressed as the proportion of virus-infected plants which germinated from the seed was for the oilseed rape cvs Jet Neuf 0.1%, Solida 0.4%, Silesia 0.8%, Darmor 1.2%, SL-507 0.2%, SL-509 0.0% and for the winter turnip rape cv. Perko 1.5%. ELISA cannot be used in direct tests on bulk seed lots to estimate proportion of infected seed, but must be used on germinated seedlings. 相似文献

To establish infection, plant viruses are evolutionarily empowered with the ability to spread intercellularly. Potyviruses represent the largest group of known plant-infecting RNA viruses, including many agriculturally important viruses. To better understand intercellular movement of potyviruses, we used turnip mosaic virus (TuMV) as a model and constructed a double-fluorescent (green and mCherry) protein-tagged TuMV infectious clone, which allows distinct observation of primary and secondary infected cells. We conducted a series of deletion and mutation analyses to characterize the role of TuMV coat protein (CP) in viral intercellular movement. TuMV CP has 288 amino acids and is composed of three domains: the N-terminus (amino acids 1–97), the core (amino acids 98–245), and the C-terminus (amino acids 246–288). We found that deletion of CP or its segments amino acids 51–199, amino acids 200–283, or amino acids 265–274 abolished the ability of TuMV to spread intercellularly but did not affect virus replication. Interestingly, deletion of amino acids 6–50 in the N-terminus domain resulted in the formation of aberrant virions but did not significantly compromise TuMV cell-to-cell and systemic movement. We identified the charged residues R178 and D222 within the core domain that are essential for virion formation and TuMV local and systemic transport in plants. Moreover, we found that trans-expression of the wild-type CP either by TuMV or through genetic transformation-based stable expression could not rescue the movement defect of CP mutants. Taken together these results suggest that TuMV CP is not essential for viral genome replication but is indispensable for viral intercellular transport where only the cis-expressed CP is functional. 相似文献

Turnip yellows virus (TuYV) is the most important virus infecting oilseed rape in the United Kingdom. The incidence and spatial distribution of TuYV in winter oilseed rape (WOSR) crops in three regions of England were determined over three growing seasons. Leaf samples were collected from three fields in each region, in autumn (November–December) and spring (April) of the three crop seasons and tested for virus presence by enzyme-linked immunosorbent assay. Infection was detected in all fields except one. Higher TuYV incidences were recorded in 2007–2008 (≤89%) and 2009–2010 (≤100%) crop seasons than in 2008–2009 (≤24%). Highest incidences were recorded in Lincolnshire (≤100%), followed by Warwickshire (≤88%), with lowest incidences in Yorkshire (1–74%). There was a significant increase in incidence detected between autumn and spring sampling in eight fields, a significant decrease in one field and no significant change in 18 fields. Rothamsted Insect Survey suction trap data for the aphid Myzus persicae in Lincolnshire, Warwickshire and Yorkshire revealed two peaks of flight activity in most years (2007–2009). The second peak (September–November) coincided with emergence of WOSR. The highest cumulative (August–November) trap catches in the three regions during the three crop seasons occurred in Lincolnshire and the lowest in Yorkshire; catches in autumn 2009 were highest and lowest in autumn 2008. Regression analysis revealed a highly significant association between the cumulative numbers of M. persicae caught in the suction traps closest to the crops between August and November each year and the incidence of TuYV detected in the WOSR crops in the autumn of each year. Relative consumption of three aphid species, Aphis gossypii Glover, Myzus persicae (Sulzer) and Lipaphis erysimi (Kaltenbach) (Homoptera: Aphididae), by larvae of the lacewing, Chrysoperla rufilabris (Burmeister) (Neuroptera: Chrysopidae), was determined in the laboratory, together with effects on lacewing development and survival. Percentages of survival of C. rufilabris from first instar to adult eclosion were significantly different among lacewing larvae fed different aphid species. When larvae were fed A. gossypii and M. persicae, all larvae developed to adulthood. All larvae died prematurely when they were fed L. erysimi. Developmental duration of C. rufilabris larvae was significantly shorter when larvae were fed A. gossypii (18.0 d) than when larvae were fed M. persicae (19.2 d). The number of fourth instar aphids consumed during development by C. rufilabris larvae differed significantly among individuals fed different aphid species. Chrysoperla rufilabris consumed an average of 168 M. persicae, followed by 141.6 A. gossypii, and only 26.6 L. erysimi. The percentage of these total number of aphids consumed by each larval stadium of C. rufilabris varied significantly among aphid species. The percentage of A. gossypii consumed by each larval stadium was similar to that for M. persicae, 12.1 and 11.4% by the first instar, 15.7 and 13.1% by the second instar, and 72.2 and 75.5% by the third instar, respectively; whereas in the case of L. erysimi, 23.3% of the total number of aphids were consumed by the first instar, 30.1% by the second instar, and 46.6% by the third instar. 相似文献

Accumulation of 60–70 % of biomass in turnip root takes place between 49–56 days after sowing. To understand the phenomenon
of rapid sink filling, the activities of sucrose metabolising enzymes and carbohydrate composition in leaf blades, petiole
and root of turnip from 42–66 days of growth were determined. An increase (2–3 folds) in glucose and fructose contents of
roots accompanied by an increase in activities of acid and alkaline invertases was observed during rapid biomass accumulating
phase of roots. The observed decrease in the activities of acid and alkaline invertases along with sucrose synthase (cleavage)
in petiole during this period could facilitate unrestricted transport of sucrose from leaves to the roots. During active root
filling period, a decrease in sucrose synthase (cleavage) and alkaline invertase activities was also observed in leaf blades.
A rapid decline in the starch content of leaf blades was observed during the phase of rapid sink filling. These metabolic
changes in the turnip plant led to increase in hexose content (35–37 %) of total dry biomass of roots at maturity. High hexose
content of the roots appears to be due to high acid invertase activity of the root. 相似文献

The translation initiation factors 4E are a small family of major susceptibility factors to potyviruses. It has been suggested that knocking out these genes could provide genetic resistance in crops when natural resistance alleles, which encode functional eIF4E proteins, are not available. Here, using the well-characterized Arabidopsis thaliana–potyvirus pathosystem, we evaluate the resistance spectrum of plants knocked out for eIF4E1, the susceptibility factor to clover yellow vein virus (ClYVV). We show that besides resistance to ClYVV, the eIF4E1 loss of function is associated with hypersusceptibility to turnip mosaic virus (TuMV), a potyvirus known to rely on the paralog host factor eIFiso4E. On TuMV infection, plants knocked out for eIF4E1 display striking developmental defects such as early senescence and primordia development stoppage. This phenotype is coupled with a strong TuMV overaccumulation throughout the plant, while remarkably the levels of the viral target eIFiso4E remain uninfluenced. Our data suggest that this hypersusceptibility cannot be explained by virus evolution leading to a gain of TuMV aggressiveness. Furthermore, we report that a functional eIF4E1 resistance allele engineered by CRISPR/Cas9 base-editing technology successfully circumvents the increase of TuMV susceptibility conditioned by eIF4E1 disruption. These findings in Arabidopsis add to several previous findings in crops suggesting that resistance based on knocking out eIF4E factors should be avoided in plant breeding, as it could also expose the plant to the severe threat of potyviruses able to recruit alternative eIF4E copies. At the same time, it provides a simple model that can help understanding of the homeostasis among eIF4E proteins in the plant cell and what makes them available to potyviruses. 相似文献
