A review of in vitro mutagenesis assessment of metal compounds in mammalian and nonmammalian test systems has been compiled.
Prokaryotic assays are ineffective or inconsistent in their detection of most metals as mutagens, with the notable exception
of hexavalent chromium. Mammalian assay systems appear to be similarly inappropriate for the screening of metal compounds
based upon the limited number of studies that have employed those compounds having known carcinogenic activity. Although of
limited value as screening tests for the detection of potentially carcinogenic metal compounds, the well-characterized in
vitro mutagenesis systems may prove to be of significant value as a means to elucidate mechanisms of metal genotoxicity. 相似文献

Interactions between the physiologically essential metals calcium, magnesium, and zinc and the carcinogenic metals nickel
and cadmium were investigated to help elucidate the mechanisms of action of the carcinogenic metals. Bioassay studies revealed
several significant findings, including: (1) the ability of magnesium and calcium to inhibit nickel-induced elevation of pulmonary
adenoma incidence in strain A mice; (2) the ability of magnesium, but not of calcium, to prevent cadmium-induced subcutaneous
sarcoma formation; and (3) the ability of magnesium, but not of calcium, to inhibit nickel-induced muscle tumor formation.
Biochemical studies indicated a direct relationship between the antitumorigenic potential of magnesium and the capacity of
this metal to: (1) inhibit nickel and cadmium uptake by the target tissues in vivo; (2) inhibit nickel-induced disturbances
in DNA synthesis in vivo; (3) inhibit nuclear and cytosolic uptake of nickel by the target tissue cells in vivo; and (4) inhibit
nickel and cadmium binding to DNA in vitro. Calcium, which in most cases did not prevent carcinogenesis, had no consistent
influence on the uptake of carcinogenic metals or their biochemical effects in the target tissues. Magnesium and zinc, but
not calcium, were also found to attenuate the acute toxic effects of nickel, indicating a possible correlation between prevention
of acute effects and reduction in tumorigenicity. Zinc, which antagonizes cadmium tumorigenicity in the rat testis, was found
to reduce markedly cadmium uptake into isolated testicular interstitial cells. Also, zinc was found to inhibit strongly cadmium

An endogenous circadian rhythm in the transpiration of Tamarix aphylla (L.) Karst. was found for plants grown in continuous light under laboratory conditions. The mean period (±SD) was 21.7±2.3 h (n = 121). No such rhythm was observed in continuous darkness, except for one small hump at the time of the first cycle. The influence of NaCl, Cd(NO3 )2 and LiCi on the rhythmic behaviour of young T. aphylla plants was investigated. NaCl concentrations of up to 150 m M reduced the overall transpiration rates of the plants, but did not change the period of the rhythm. The amplitude and the mesor of the oscillations were inversely correlated with the NaCl concentration. A similar influence was found for Cd(NO3 )2 , but with concentrations that were approximately three orders of magnitude smaller than those of the NaCl treatments. The rhythmic behaviour of the plants was not altered by 10 m M LiCl. It is suggested that the described rhythm of transpiration may have a dual effect: (a) it might cause a partial closure of the stomates during midday hours and (b) it might serve as a possible synchronizer ("master clock") for other rhythmic phenomena in the plants. 相似文献

Five plant species were cultivated on a soil from the Neckar alluvial fan near Heidelberg (FRG) polluted by the emissions
of a cement plant. Thallium, cadmium and lead concentrations in seedlings and mature plants were determined by atomic absorption
analysis. AdditionallyBrassica napus L.napus was grown on soils containing 5 different concentrations of heavy metals, achieved by mixing two similar soils, from the
same area but with different metal concentrations.
Thallium and cadmium were shown to be taken up by roots whilst lead which was also absorbed, was deposited mainly on the plant
surface. However during cultivation in the winter months, a remarkable deposit of lead via the roots was found. Thallium in
the soil from a anthrorogen source was more available to plants than thallium of geological origin.
During the lifetime of a plant concentrations of thallium and cadmium were always highest in the seedling. The decrease in
metal concentration with maturity depended on the plant species and the element, but was not a function of the metal concentration

Metallothioneins are a class of cysteine-rich and low molecular weight, metal-binding proteins that are inducible by a wide variety of agents, including metal ions, such as cadmium and zinc, glucocorticoid hormones, interferon, and tumor promoters. In an effort to delineate the regulation of the synthesis of the recently identified brain metallothionein-like protein, a study was undertaken to compare the induction of metallothionein in human neuroblastoma IMR-32 cells by zinc, cadmium, and dexamethasone using the human Chang liver cells as a control. Both cadmium (1 microM) and zinc (100 microM) significantly enhanced the incorporation of [35S]cysteine into metallothioneins isolated from both neuroblastoma and Chang liver cells. Dexamethasone in concentrations of 10 microM stimulated the synthesis of metallothionein in the Chang cells, whereas it had no effects on the synthesis of metallothionein in the neuroblastoma cells at concentrations ranging from 2.5--100 microM. The degree of stimulation of metallothionein synthesis in the Chang cells by cadmium and zinc was significantly higher than seen in neuroblastoma cells. The neuroblastoma IMR-32 exhibited less tolerance to the toxicity of both cadmium and zinc than the Chang cells, which may correlate with the inherent ability of these ions to induce metallothioneins in these dissimilar cells. The results of these studies are interpreted to indicate that the factors regulating the synthesis of metallothioneins in the Chang and neuroblastoma cells are not identical, suggesting also of the presence of dissimilar regulatory mechanisms in the liver and brain. 相似文献

Studies were conducted to determine the effect of dietary iron (Fe) levels ranging from a deficiency to an excess on the toxicity
of cadmium (Cd) in chicks. In Fe-deficient animals, cadmium was found to be more toxic than in Fe supplemented animals as
measured by growth. The liver Cd burdens were increased significantly in the presence of dietary Fe supplementation, and there
was a significant Cd−Fe interaction in the Cd concentration of the kidney, indicating that iron deficiency increased the concentration
of Cd in the kidneys of those chicks receiving this element. Cd tended to reduce the Fe concentration in both the liver and
kidney. The absorption of Cd as measured by the amount of109Cd that disappeared from an isolated duodenal segment in one h was not affected by the Fe content of the diet, but the amount
of isotope appearing in the liver compared to the amount present in the blood was increased in the Fe supplemented chicks.
Separation of the Cd binding ligands by column chromatography revealed that more of the Cd in the liver, but not the kidney,
was associated with ligands which eluted in a column volume that contained metallothionein in those chicks receiving Fe than
in the livers from Fe deficient animals. The inverse relationship between the amount of Cd bound to the metallothionein containing
fraction and toxicity may be related causally.

Evidence for the presence of the metal-binding protein metallothionein, MT, in higher plants is equivocal. Although a number of MT-like metal complexes have been isolated from plants, the chemical structures of most of these compounds have not been fully elucidated. Recently a novel class of plant peptides, poly (γ-glutamylcysteinyl) glycines, (γEC)n G, have been discovered. These peptides bind metal ions, and in the presence of such ions the amount of (γEC), G in plant cells increases. The presence of peptide bonds through the γ-carboxyl group of glutamate, rather than the α-carboxyl group, suggests that these peptides are not encoded by structural genes but are the products of biosynthetic pathways. Cells which are resistant to supra-optimal concentrations of certain metal ions over-produce (γEC)n G. (γEC)n G. may be functional analogues of MT. Whether or not some plants also produce MT is an important question which remains to be answered. 相似文献
