The PT-digest of bread wheat gliadin was very active in agglutinating undifferentiated human K562(S) cells. This activity was quantitatively, but not qualitatively, similar to that of Con A or WGA. Moreover, Con A-induced cell agglutination was inhibited by mannan and mannose, WGA-induced agglutination by NAG only, and cell agglutination induced by bread wheat gliadin peptides was inhibited by each of these three saccharides. Not only was mannan the most active saccharide in preventing cell agglutination induced by bread wheat gliadin peptides, but it was also able to dissociate agglutinated cells. As compared to the PT- digest of whole bread wheat gliadin, the digest obtained from purified A-gliadin was tenfold more active. The PT-digest of durum wheat gliadin did not show any agglutinating activity.  相似文献

Nitrate uptake and nitrite efflux patterns in Nostoc MAC showed a rapid phase followed by their saturation. Nitrite efflux was maximum in nitrate medium whereas the cells incubated in N2 and NH 4 + media exhibited a decreased nitrite efflux activity. The simultaneous presence of NH 4 + and nitrate significantly decreased nitrite efflux. L-Methionine-Dl-sulphoximine (MSX) prevented inhibition of nitrite efflux by NH 4 + . In the dark there was negligible nitrite efflux, whereas illumination increased the rate of nitrite efflux significantly. The nitrite efflux system was maximally operative at pH 8.0, 30°C and a photon fluence rate of 50 mol m-2. s-1. These results confirm that (i) the nitrite efflux system in Nostoc MAC is dependent upon nitrate uptake and assimilation and is repressible by NH 4 + ; (ii) NH 4 + itself is not the actual repressor of nitrite efflux; a product of NH 4 + assimilation via glutamine synthetase (GS) is required for repression to occur; (iii) the catalytic function of GS does not appear to be involved in nitrate assimilation-dependent nitrite efflux, and (iv) the optimum pH, temperature and illumination for maximum nitrite efflux were found to be 8.0, 30°C and 50mol m-2. s respectively.B.B. One of the principal characteristics of large scale wireless sensor networks is their distributed, multi-hop nature. Due to this characteristic, applications such as query propagation rely regularly on network-wide flooding for information dissemination. If the transmission radius is not set optimally, the flooded packet may be holding the transmission medium for longer periods than are necessary, reducing overall network throughput. We analyze the impact of the transmission radius on the average settling time—the time at which all nodes in the network finish transmitting the flooded packet. Our analytical model takes into account the behavior of the underlying contention-based MAC protocol, as well as edge effects and the size of the network. We show that for large wireless networks there exists an intermediate transmission radius which minimizes the settling time, corresponding to an optimal tradeoff between reception and contention times. We also explain how physical propagation models affect small wireless networks and why there is no intermediate optimal transmission radius observed in these cases. The mathematical analysis is supported and validated through extensive simulations.Marco Zuniga is currently a PhD student in the Department of Electrical Engineering at the University of Southern California. He received his Bachelors degree in Electrical Engineering with a four-year full-tuition scholarship from The Cooper Union for the Advancement of Science and Art in 1998. He received his Masters degree and his Ph.D. in Electrical Engineering from Cornell University in 1999 and 2002, under a four-year university graduate fellowship. Dr. Krishnamacharis previous research has included work on critical density thresholds in wireless networks, data centric routing in sensor networks, mobility management in cellular telephone systems, multicast flow control, heuristic global optimization, and constraint satisfaction. His current research is focused on the discovery of fundamental principles and the analysis and design of protocols for next generation wireless sensor networks. An allopolyploid complex with high genomic integrity has been studied. Dogroses transmit only seven chromosomes (from seven bivalents) through the pollen, whereas 21, 28 or 35 chromosomes (from seven bivalents and 14, 21 or 28 univalents) come from the egg cells. Seedlings derived from two interspecific crosses were analysed with flow cytometry and molecular markers to determine ploidy level, mode of reproduction and genomic constitution. Evidence was obtained for the formation of unreduced male and female gametes, which can take part in fertilization (producing seedlings with higher ploidy than the parental plants) or in apomictic reproduction. Random amplified polymorphic DNA (RAPD) and microsatellite analyses indicated that three seedlings (5%) were derived through apomixis, whereas the other 49 were hybrids. Bivalent formation appears to involve chromosomes that consistently share the same microsatellite alleles. Allele-sharing between the maternally transmitted and highly conserved univalent-forming chromosomes reflected the taxonomic distance between different genotypes. The frequently recombining bivalent-forming chromosomes were taxonomically less informative.  相似文献

Cytotoxic lymphocytes (CLs) are responsible for the clearance of virally infected or neoplastic cells. CLs possess specialised lysosome-related organelles called granules which contain the granzyme family of serine proteases and perforin. Granzymes may induce apoptosis in the target cell when delivered by the pore forming protein, perforin. Here we follow the perforin-granzyme pathway from synthesis and storage in the granule, to exocytosis and finally delivery into the target cell. This review focuses on the controversial subject of perforin-mediated translocation of granzymes into the target cell cytoplasm. It remains unclear whether this occurs at the cell surface with granzymes moving through a perforin pore in the plasma membrane, or if it involves internalisation of perforin and granzymes and subsequent release from an endocytic compartment. The latter mechanism would represent an example of cross talk between the endo-lysosomal pathways of individual cells. The majority of the freshwater fishes in the ornamental trade now originate from captive‐bred sources, as do a large proportion of the freshwater species exhibited in public aquariums. In contrast, commercial operators who also supply marine specimens to the ornamental trade remove directly from the wild approximately 98% of the marine fishes and invertebrates exhibited in public aquariums. The common perception prevails that captive propagation is inherently a better alternative to obtaining animals from the wild. Although captive propagation has been shown to have many benefits for terrestrial species, there are a number of features unique to marine species that challenge the idea that every species should be bred in captivity. Some of the key issues relating to the development of widespread conservation‐oriented captive propagation programs include: 1) the high taxonomic diversity in marine animals; 2) the resultant variety in their reproductive methods; 3) their ecological, behavioral, physiological, and nutritional needs; and 4) our general lack of knowledge on their husbandry and medical care. There are several characteristics of marine fish and invertebrate populations that make them suitable candidates for sustainable harvest. For instance, marine teleosts are “r‐selected,” meaning that they have an extremely high fecundity, and most marine teleosts have a wide distribution and the ability to disperse over long distances. In locations considered for fish collection, appropriate management techniques should be employed to ensure that fishes and invertebrates are collected with as little impact on the ecosystem as possible. The collection of marine fishes and invertebrates for public aquariums and the hobby trade should be managed like a fishery to ensure long‐term sustainability. The public aquarium community should support marine organism certification initiatives, such as the Marine Aquarium Council (MAC). Marine organism certification will create market incentives that encourage and support quality and sustainable practices by creating consumer demand and confidence for certified organisms, practices, and industry participants. The creation of refuges that supply propagules to harvested areas, the rotation of areas fished, species‐specific size limits and seasons, and standardization of collecting, handling, and transportation techniques should be used to manage these fisheries and harvest areas. The death of one cell can precipitate the death of nearby cells in a process referred to as the bystander effect. We investigated whether mitochondrial apoptosis generated a bystander effect and, if so, by which pathway. Microinjection with cytochrome c mimicked function of the mitochondrial apoptosis-induced channel MAC and caused apoptosis of both target and nearby osteoblasts. This effect was suppressed by inhibiting gap junction intercellular communication. A bystander effect was also observed after exogenous expression of tBid, which facilitates MAC formation and cytochrome c release. Interestingly, in connexin-43 deficient osteoblasts, microinjection of cytochrome c induced apoptosis only in the target cell. These findings indicate that a death signal was generated downstream of MAC function and was transmitted through gap junctions to amplify apoptosis in neighboring cells. This concept may have implications in development of new therapeutic approaches.  相似文献
