Stable isotopes in fish otoliths discriminate between lagoonal and oceanic residents of Taiaro Atoll (Tuamotu Archipelago, French Polynesia)

 The fully enclosed Taiaro lagoon is hypersaline (42.5 psu) and non-tidal; constant salinity and water level result from strong evaporation balanced by low percolation through the lagoon floor. Seawater can flow over the atoll rim during exceptionally high seas and may then replenish lagoonal communities with propagules of oceanic origin. The distinctive water chemistry of the lagoon suggests a possible way of identifying these immigrants. We established this potential by analysing stable isotopes of carbon and oxygen in the recent growth layers of otoliths of two adult reef fishes, Chaetodon ulietensis and Acanthurus triostegus, collected from both sides of the atoll rim. Fish from the two locations were discriminated by their isotopic signatures, suggesting that analysis of the microchemical signatures deposited during the larval development could be used in future work to determine which individuals and species complete their life-cycles in this unusual lagoon. Akt Activation Emulates Chk1 Inhibition and Bcl2 Overexpression and Abrogates G2 Cell Cycle Checkpoint by Inhibiting BRCA1 Foci

Akt is perhaps the most frequently activated oncoprotein in human cancers. Overriding cell cycle checkpoint in combination with the inhibition of apoptosis are two principal requirements for predisposition to cancer. Here we show that the activation of Akt is sufficient to promote these two principal processes, by inhibiting Chk1 activation with concomitant inhibition of apoptosis. These activities of Akt cannot be recapitulated by the knockdown of Chk1 alone or by overexpression of Bcl2. Rather the combination of Chk1 knockdown and Bcl2 overexpression is required to recapitulate Akt activities. Akt was shown to directly phosphorylate Chk1. However, we found that Chk1 mutants in the Akt phosphorylation sites behave like wild-type Chk1 in mediating G2 arrest, suggesting that the phosphorylation of Chk1 by Akt is either dispensable for Chk1 activity or insufficient by itself to exert an effect on Chk1 activity. Here we report a new mechanism by which Akt affects G2 cell cycle arrest. We show that Akt inhibits BRCA1 function that induces G2 cell cycle arrest. Akt prevents the translocation of BRCA1 to DNA damage foci and, thereby, inhibiting the activation of Chk1 following DNA damage.     

Portrait of the Patient as a Young Man: An Exploration of the Use of Photographs in Hospital

The display of personal photographs in hospital is a common practice that has yet to be rigorously examined. The photographs displayed are subject to interpretation by the viewer and may lead to misunderstandings or miscommunication if clarification of meaning is not sought. This paper explores a range of possible meanings that the display of photographs in hospital may hold, based on a case study of a 15 year old boy hospitalised with a life threatening illness. Further research is needed into the actual meanings attributed to the display of photographs in hospital by patients and family members.     

Operation of a Benchtop Bioreactor

Fermentation systems are used to provide an optimal growth environment for many different types of cell cultures. The ability afforded by fermentors to carefully control temperature, pH, and dissolved oxygen concentrations in particular makes them essential to efficient large scale growth and expression of fermentation products. This video will briefly describe the advantages of the fermentor over the shake flask. It will also identify key components of a typical benchtop fermentation system and give basic instruction on setup of the vessel and calibration of its probes. The viewer will be familiarized with the sterilization process and shown how to inoculate the growth medium in the vessel with culture. Basic concepts of operation, sampling, and harvesting will also be demonstrated. Simple data analysis and system cleanup will also be discussed.     

Phylogenetic Position of Amblyospora Hazard & Oldacre (Microspora: Amblyosporidae) Based on Small Subunit rRNA Data and Its Implication for the Evolution of the Microsporidia

ABSTRACT. Sequences of the small subunit rRNA genes of Amblyospora californica and an Amblyospora sp. from Culex salinarius were determined. These sequences were compared phylogenetically with 16 other microsporidia. The results suggest Amblyospora forms a sister taxon to the rest of the microsporidia examined. The basal position of Amblyospora is discussed with respect to the evolution of microsporidian life cycles. These sequences represent the longest microsporidian small subunit rRNA genes sequenced to date, 1,359 and 1,358 bp, respectively. Structural features and GC content (49% for both) are comparable to those of other microsporidia which have been sequenced.     

The genetic structure of a tribal population,the Yanomama Indians. XII. Biodemographic studies

The Yanomama Indians of Southern Vanezuela and Northern Brazil are one of the largest, relatively unacculturated tribes of the tropical rain forest. Over a period of eight years data have been collected from a considerable portion of their territory on estimated age, sex ratio, fertility rates (as determined by physical examination and urine tests), and infant death rates. Although it has been impossible to collect direct data on infanticide, this subject can be approached indirectly through distortions of the sex ratio and anecdotal information. Some historical data are also available as a basis for estimating tribal expansion in the past 100 years. With this material it has been possible to construct Life Tables for the Yanomama, and to explore the results of various perturbations of the input parameters. Data are also presented on patterns of mating and reproduction: number of spouses, mean and variance in number of surviving children, frequency of “extra-marital conceptions” based on the results of extensive blood group typings, and consanguinity rates as determined by observation and computer simulation. Although we do not present the Yanomama as typical, these data are seen as providing a basis for more realistic population models than have existed in the past. In addition, the data provide a basis for relatively precise estimates of such demographic measures as Fisher's Reproductive Value, Crow's Index of Total Selection, and Weiss' Index of Growth Regulation.     

厦门地区鸭鹅绦虫生活史研究

本文报告了福建厦门地区寄生于鸭鹅类的膜壳科绦虫计5属9种。实验阐明了5种绦虫的生活史,其中有三种生活史为我国首次报告。厦门地区有5种嵌水甲壳类可作膜壳绦虫的中间宿主。其中短角异剑水蚤(Apocyclops royi)是矛形剑带绦虫和片形皱缘绦虫的审间宿主,不等异介虫(Heterocypris anomala)为新发现的棘盘双睾绦虫的中间宿主,介虫(Cyprinotus sp.)为新发现的美丽膜壳绦虫的中间宿主。     

Age,growth and reproductive biology of the silky shark,Carcharhinus falciformis,and the scalloped hammerhead,Sphyrna lewini,from the northwestern Gulf of Mexico

Synopsis The silky shark, Carcharhinus falciformis, and scalloped hammerhead, Sphyrna lewini, represent >80% of the shark by-catch of the winter swordfish/tuna longline fishery of the northwestern Gulf of Mexico. This catch represents a potential supplemental fishery, yet little is known of the life histories of the two species. This report relates reproductive biology data to age and growth estimates for 135 C. falciformis and 78 S. lewini. Unlike other regional populations, C. falciformis in the Gulf of Mexico may have a seasonal 12 month gestation period. Males mature at 210–220 cm TL (6–7 yr); females at >225 cm TL (7–9 yr). Application of age at length data for combined sexes produced von Bertalanffy growth model parameter estimates of L_∞ = 291 cm TL, K = 0.153, t₀ = −2.2 yr. Adult male S. lewini outnumbered adult females in catches because of differences in the distributions of the sexually segregated population. Males mature at 180 cm TL (10 yr); females at 250 cm TL (15 yr). von Bertalanffy parameter estimates for combined sexes of this species were L_∞ = 329 cm TL, K = 0.073, t_o = −2.2 yr.
